Bovine spongiform encephalopathy (BSE) surveillance programs have been employed in numerous countries to monitor BSE prevalence and to protect animal and human health. Since 1999, the European Commission (EC) authorized the evaluation and approval of 20 molecular based tests for the rapid detection of the pathological prion protein (PrPsc) in BSE infection. The diagnostic sensitivity, convenience, and speed of these tests have made molecular diagnostics the preferred method for BSE surveillance. The aim of this study was to determine the analytical sensitivity of 4 commercially available BSE rapid-test kits, including the Prionics®-Check WESTERN, the Prionics® Check-PrioSTRIP™, the BioRad® TeSeE™ ELISA, and the IDEXX® HerdChek™ EIA. Performances of these tests were then compared to 2 confirmatory tests, including the BioRad® TeSeE™ Western Blot and the modified Scrapie Associated Fibrils (SAF)/OIE Immunoblot. One 50% w/v homogenate was made from experimentally generated C-type BSE brain tissues in ddH2O. Homogenates were diluted through a background of BSE-negative brainstem homogenate. Masses of both positive and negative tissues in each dilution were calculated to maintain the appropriate tissue amounts for each test platform. Specific concentrated homogenization buffer was added accordingly to maintain the correct buffer condition for each test. ELISA-based tests were evaluated using their respective software/detection platforms. Blot-protocols were evaluated by manual measurements of blot signal density. Detection limitations were determined by fitted curves intersecting the manufacturers' positive/negative criteria. The confirmatory SAF Immunoblot displayed the highest analytical sensitivity, followed by the IDEXX® HerdChek™ EIA, Bio-Rad® TeSeE™ Western Blot, the Bio-Rad® TeSeE™ ELISA, Prionics®-Check PrioSTRIP™, and Prionics®-Check WESTERN™, respectively. Although the tests performed at different levels of sensitivity, the most sensitive and least sensitive of the rapid tests were separated by 2 logs in analytical sensitivity, meeting European performance requirements. The epidemiology and possibly the etiology of bovine spongiform encephalopathy (BSE) have recently been recognized to be heterogeneous. In particular, three types [classical (C) and two atypical (H, L)] have been identified, largely on the basis of characteristics of the proteinase K (PK)-resistant core of the misfolded prion protein associated with the disease (PrPres). The present study was conducted to characterize the 17 Canadian BSE cases which occurred prior to November 2009 based on the molecular and biochemical properties of their PrPres, including immunoreactivity, molecular weight, glycoform profile and relative PK sensitivity. Two cases exhibited molecular weight and glycoform profiles similar to those of previously reported atypical cases, one corresponding to H-type BSE (case 6) and the other to L-type BSE (case 11). All other cases were classified as C-type. PK digestion under mild and stringent conditions revealed a reduced protease resistance in both of these cases compared to the C-type cases. With Western immunoblotting, N-terminal-specific antibodies bound to PrPres from case 6 but not to that from case 11 or C-type cases. C-terminal-specific antibodies revealed a shift in the glycoform profile and detected a fourth protein fragment in case 6, indicative of two PrPres subpopulations in H-type BSE. No mutations suggesting a genetic etiology were found in any of the 17 animals by sequencing the full PrP-coding sequence in exon 3 of the PRNP gene. Thus, each of the three known BSE types have been confirmed in Canadian cattle and show molecular characteristics highly similar to those of classical and atypical BSE cases described from Europe, Japan and the USA. The occurrence of atypical cases of BSE in countries such as Canada with low BSE prevalence and transmission risk argues for the occurrence of sporadic forms of BSE worldwide. [ABSTRACT FROM AUTHOR]

This article proposes a reflection on the creation of the Bureau des affaires francophones et francophiles from Simon Fraser University. The institution's mission is to foster the influence of French in a predominantly English-Speaking environment. The Bureau attributes its success to the joint efforts of Francophone and Francophile communities from British Columbia and the Provincial and Federal governments. The research aims to identify the contribution of each group to the success of the project. Our article is based on Kingdon's theory of the "window of opportunity" and our findings were as a result of 17 interviews of stakeholders. Marine invaders have become a significant threat to native biodiversity and ecosystem function. In this study, the invasion of the varnish clam (Nuttallia obscurata) in British Columbia, Canada, is investigated using a matrix modeling approach to identify the life history characteristics most crucial for population growth and to investigate population differences. Mark-recapture analyses and field collections from 2003 to 2004 were used to determine individual growth, survival rates, and fecundity for two sites. A multi-state matrix model was used to determine population growth rates and to conduct sensitivity and elasticity analyses. A life table response experiment was also used to determine what life history stage contributed most to observed differences in population growth rates. Population survey data were used in conjunction with the matrix model to determine plausible recruitment levels and to investigate recruitment scenarios. Both populations are currently declining but are likely sustainable because of the pulsed nature of large recruitment events. Survival of larger clams (>40 mm) is the most important for population growth based on elasticity and sensitivity analyses. Adult survival also had the largest influence on observed differences between site- specific population growth rates. The two populations studied differed in recruitment dynamics; one experiencing annual recruitment with higher post-settlement mortality and the other, episodic recruitment and lower post-settlement mortality. The most influential factor for the successful invasion of the varnish clam appears to be survival of the larger size classes. Therefore, any process that decreases adult survival (e.g., predation, commercial harvest) will have the greatest impact on population growth. [ABSTRACT FROM AUTHOR]

Approximately 20% of Canada's oil supply now comes from the extraction of bitumen from the oil sands deposits in northeastern Alberta. The oil sands are strip-mined, and the bitumen is typically separated from sand and clays by an alkaline hot water extraction process. The rapidly expanding oil sands industry has millions of cubic metres of tailings for disposal and large areas of land to reclaim. There are estimates that the consolidation of the mature fine tails (MFT) in the settling ponds will take about 150 years. Some of the settling ponds are now evolving microbially produced methane, a greenhouse gas. To hasten consolidation, gypsum (CaSO[sub 4] ·2H[sub 2] O) is added to MFT, yielding materials called consolidated or composite tailings (CT). Sulfate from the gypsum has the potential to stimulate sulfate-reducing bacteria (SRB) to out-compete methanogens, thereby stopping methanogenesis. This investigation examined three MFT and four CT samples from three oil sands extractions companies. Each was found to contain methanogens and SRB. Serum bottle microcosm studies showed sulfate in the CT samples stopped methane production. However, if the microcosms were amended with readily utilizable electron donors, the sulfate was consumed, and when it reached approximately 20 mg/L, methane production began. Some unamended microcosms were incubated for 372 days, with no methane production detected. This work showed that each MFT and CT sample has the potential to become methanogenic, but in the absence of exogenous electron donors, the added sulfate can inhibit methanogenesis for a long time.Key words: consolidated tailings, composite tailings, methanogens, oil sands, sulfate-reducing bacteria.Environ 20 % de l'approvisionnement en pétrole au Canada provient actuellement de l'extraction de bitume issu de dépôts de sables bitumineux dans le nord-est de l'Alberta. Les sables bitumineux sont minés à ciel ouvert et le bitume est normalement séparé du sable et des argiles par une procédé d'extraction à l'eau chaude alcaline. L'industrie des sables bitumineux est en expansion rapide; elle dispose de millions de mètres cube de résidus à rejeter, et de vastes régions à réclamer. On estime qu'il faudra environ 150 ans pour consolider les résidus fins terminaux (RFT) dans les bassins de décantation. Certains bassins de décantation développent désormais du méthane d'origine microbienne, un gaz à effet de serre. Pour accélérer la consolidation, du gypse (CaSO[sub 4] ·2H[sub 2] O) est ajouté au RFT, produisant des matériaux nommés résidus consolidés ou composites (RC). Le sulfate du gypse a le potentiel de stimuler des bactéries réductrices de sulfate (BRS) qui supplanteraient les méthanogènes, mettant fin à la méthanogénèse. Cette étude a examiné trois échantillons de RFT et quatre de RC provenant de trois compagnies d'extraction de sables bitumineux. Chacun contenait des méthanogènes et des BRS.
