Background Many novel tick-borne viruses have been discovered by deep-sequencing technology in recent years; however, their medical significance is unknown. Methods We obtained clinical data of a patient from Xinjiang, China. Possible pathogens were detected by metagenomic analysis; the causative pathogen Tacheng tick virus 1 (TcTV-1) was found and further confirmed by reverse transcriptase–polymerase chain reaction, viral culture, and sequence analyses. Epidemiological investigation was conducted in the local human population, domestic animals, and ticks by serological/molecular methods. Results A 62-year-old woman with a history of tick bite in Qinghe, Xinjiang, presented with fever and rashes. These symptoms were relieved after clinical treatment. TcTV-1 (strain QH1) was isolated from the patient's cerebrospinal fluid, throat swabs, and urine on day 47 after illness onset. Although the blood and urine showed viral RNA positive on day 73 after illness onset, the virus was only isolated from urine. Serological detection revealed a virus neutralizing antibody titer of 1:40 and 1:80 on day 47 and 73 after illness onset, respectively. No coinfection with other pathogens was detected, suggesting TcTV-1 may be the potential causative pathogen. We detected anti–TcTV-1 antibodies (immunoglobulin G: 10.1%; immunoglobulin M: 4.8%) in the local human population. The viral RNA was also found in cattle (4.9%), sheep (9.2%), and ticks, including Dermacentor marginatus (14.3%), Dermacentor silvarum (11.8%), Dermacentor nuttalli (6.7%), and Hyalomma asiaticum (4.8%). Conclusions TcTV-1 may be associated with a febrile illness syndrome, and epidemiological data of the virus in humans and animals necessitate disease surveillance of TcTV-1 infection in China. Bean pyralid (BP; Lamprosema indicata Fabricius) is one of the major leaf-feeding insects that affect soybean crops in central and southern China. Four recombinant inbred line populations (KY, WT, XG and SX) were tested during 2004-2006 in Nanjing, China, to identify quantitative trait loci (QTL) for resistance to BP on the basis of data for rolled leaflet percentage under field infestation conditions. The mapping was performed using QTL Network V2.0 and checked with Windows QTL Cartographer V2.5 and IciMapping V2.2. The results showed that 81-92 % of the phenotypic variation was accounted for by additive QTL (27-43 %), epistatic QTL pairs (5-13 %), and collective unmapped minor QTL (38-58 %). In total, 17 QTL were detected on 11 linkage groups, of which two had additive effects, six had both additive and epistatic effects, and nine had only epistatic effects. Eight epistatic QTL pairs were observed, of which three pairs involved two QTL with additive effects, one involved one QTL with additive effect, and four involved no QTL with additive effects. Different genetic structures for BP resistance were found among the populations. Eight QTL (five additive and three epistatic pairs) were detected in KY, ten QTL (four additive and five epistatic pairs) were detected in WT, and only one additive QTL was detected in both the XG and the SX populations. BP12- 1 and BP1- 1 are major QTL, with the former accounting for 15, 31, and 50 % of the total genetic variation (including epistasis) in KY, WT, and XG, respectively, and the latter accounting for 13 and 32 % of the total genetic variation in KY and SX, respectively. The additive × year and epistasis × year interaction effects were negligible, indicating that the QTL were stable over the years. Because 41-68 % of the total genetic variation could not be accounted for by these QTL, the use of both identified QTL and unmapped minor QTL in breeding for BP resistance should be considered. Background: can carry and transmit various zoonotic pathogens. Some studies were conducted to investigate a few zoonotic pathogens in Guangzhou, China, but no coinfections were investigated or specifically mentioned. Studies on the infections and the influencing factors of various zoonotic pathogens in Rattus norvegicus can carry and transmit various zoonotic pathogens. Studies on the infections and the influencing factors of various zoonotic pathogens in R. norvegicus along the Zengjiang River in Guangzhou have not been carried out. Materials and Methods: In this study, R. norvegicus was captured in November 2020 and September 2021 along the Zengjiang River, and was tested for Bartonella spp., Leptospira spp., Orientia tsutsugamushi , Borrelia burgdorferi , Hantavirus (HV), Ehrlichia spp., and severe fever with thrombocytopenia syndrome virus (SFTSV) by the RT-PCR. Logistic regression analysis was used to determine the impact of habitat and demographic factors on the infections and coinfections of the surveyed pathogens. Results: In 119 R. norvegicus , the detection rates of Bartonella spp., Leptospira spp., O. tsutsugamushi , B. burgdorferi , and HV were 46.2%, 31.9%, 5%, 0.8%, and 18.5%, respectively. Ehrlichia spp. and SFTSV were negative. The triple coinfection rate of Bartonella spp., Leptospira spp., and HV was 11.8%. In addition, the coinfection of Bartonella spp., Leptospira spp., and B. burgdorferi was 0.8%. Dual coinfection of Bartonella spp. and Leptospira spp., Leptospira spp. and HV, Bartonella spp. and O. tsutsugamushi , Leptospira spp. and O. tsutsugamushi , and HV and O. tsutsugamushi was 9.2%, 3.4%, 1.7%, 1.7%, and 0.8%, respectively. Infections of these pathogens in R. norvegicus were found in habitats of banana plantation, grassland, and bush. Weight affected the infection of Bartonella spp., Leptospira spp., or HV in R. norvegicus. Conclusions: R. norvegicus along the Zengjiang River not only carried various potentially zoonotic pathogens but also had a variety of coinfections. Surveillance of the density and pathogens in R. norvegicus should be strengthened to reduce the incidence of relevant zoonotic diseases.

Background: Malaria was once widespread in Guangzhou, China. However, a series of control measures have succeeded in eliminating local malaria infections. Based on the analysis of the characteristics of malaria epidemics in Guangzhou, China, from 1950 to 2022, the changes and effectiveness of malaria control strategies and surveillance management in Guangzhou from 1950 to 2022 are described., Methods: Data on malaria prevention and treatment in Guangzhou from 1950 to 2022 were collected, and descriptive epidemiological methods were used to analyse the prevalence of malaria, preventive and control measures taken, and the effectiveness of prevention and treatment in different periods. Data on malaria cases were obtained from the Guangzhou Centre for Disease Control and Prevention (CDC) and the China Communicable Disease Reporting System., Results: The development of the malaria control system in Guangzhou has gone through four periods: 1. High malaria prevalence (1950-1979), 2. Intensive prevention and control stage (1980-2000), 3. Consolidating gains in malaria control (2001-2008), and 4. Preventing reestablishment of transmission (2009-2022). During Period 1, only medical institutions at all levels and the local CDCs, the Guangzhou CDC participated in the malaria prevention and control system, establishing a three-tier health system on malaria prevention and control. During Period 2, other types of organizations, including the agricultural sector, schools and village committees, the construction department and street committee, are involved in the malaria control system. During Period 3, more and more organizations are joining forces to prevent and control malaria. A well-established multisectoral malaria control mechanism and an improved post-elimination surveillance management system are in place. Between 1950 and 2022, a total of 420,670 cases of malaria were reported. During Period 1, there was an epidemic of malaria in the early 1950s, with an annual incidence rate of more than 10,000/100,000, including a high rate of 2887.98/100,000 in 1954. In Period 2 malaria was gradually brought under control, with the average annual malaria incidence rate dropping to 3.14/100,000. During Period 3, the incidence rate was kept below 1/100,000, and by 2009 local malaria infections were eliminated., Conclusion: For decades, Guangzhou has adopted different malaria control strategies and measures at different epidemic stages. Increased collaboration among civil organizations in Guangzhou in malaria control has led to a significant decline in the number of malaria cases and the elimination of indigenous malaria infections by 2009.The experience of Guangzhou can guide the development of malaria control strategies in other cities experiencing similar malaria epidemics., (© 2023. Thyroid volume was proposed as a factor for malignancy in evaluating thyroid nodules. Previous studies have demonstrated the endocrine disrupting effect of polycyclic aromatic hydrocarbons (PAHs), but studies on the association between internal exposure of PAHs and thyroid volume are still scarce. In this work, we evaluated the association of polycyclic aromatic hydrocarbon internal exposure and urinary iodine concentration with thyroid volume in 590 school-age children without thyroid disease in Guangzhou, China. Urinary hydroxylated polycyclic aromatic hydrocarbons (OH-PAHs), urinary iodine concentrations, and thyroid volumes were measured. The mean concentrations of urinary iodine and ΣOH-PAHs were 271.1 μg/L and 3.27 μg/L, respectively, and the mean thyroid volume was 2.4 mL. The associations of urinary iodine and OH-PAH concentrations with thyroid volume were investigated by multivariable linear regression and the Bayesian kernel machine regression models. Urinary ΣOH-PAHs were observed to be significantly positively associated with thyroid volume in multivariable linear regression models. The increase in each unit in the log-transformed concentration of ΣOH-PAHs caused 3.88% change in thyroid volume. The Bayesian kernel machine regression model demonstrated a positive joint effect of increased urinary ΣOH-PAHs on thyroid volume. Objectives: Scrub typhus is an increasingly serious public health problem, which is becoming the most common vector-borne disease in Guangzhou. This study aimed to analyse the correlation between scrub typhus incidence and potential factors and rank the importance of influential factors., Methods: We collected monthly scrub typhus cases, meteorological variables, rodent density (RD), Normalised Difference Vegetation Index (NDVI), and land use type in Guangzhou from 2006 to 2019. Correlation analysis and a random forest model were used to identify the risk factors for scrub typhus and predict the importance rank of influencing factors related to scrub typhus incidence., Results: The epidemiological results of the scrub typhus cases in Guangzhou between 2006 and 2019 showed that the incidence rate was on the rise. The results of correlation analysis revealed that a positive relationship between scrub typhus incidence and meteorological factors of mean temperature (T mean ), accumulative rainfall (RF), relative humidity (RH), sunshine hours (SH), and NDVI, RD, population density, and green land coverage area (all p < 0.001). Additionally, we tested the relationship between the incidence of scrub typhus and the lagging meteorological factors through cross-correlation function, and found that incidence was positively correlated with 1-month lag T mean , 2-month lag RF, 2-month lag RH, and 6-month lag SH (all p < 0.001). Based on the random forest model, we found that the T mean was the most important predictor among the influential factors, followed by NDVI., Conclusions: Meteorological factors, NDVI, RD, and land use type jointly affect the incidence of scrub typhus in Guangzhou. Background: Wenzhou virus (WENV), a newly discovered mammarenavirus in rodents, is associated with fever and respiratory symptoms in humans. This study was aimed to detect and characterize the emerging virus in rodents in Guangzhou, China., Results: A total of 100 small mammals, including 70 Rattus norvegicus, 22 Suncus murinus, 4 Bandicota indica, 3 Rattus flavipectus, and 1 Rattus losea, were captured in Guangzhou, and their brain tissues were collected and pooled for metagenomic analysis, which generated several contigs targeting the genome of WENV. Two R. norvegicus (2.9%) were further confirmed to be infected with WENV by RT-PCR. The complete genome (RnGZ37-2018 and RnGZ40-2018) shared 85.1-88.9% nt and 83.2-96.3% aa sequence identities to the Cambodian strains that have been shown to be associated with human disease. Phylogenetic analysis showed that all identified WENV could be grouped into four different lineages, and the two Guangzhou strains formed an independent clade. We also analyzed the potential recombinant events occurring in WENV strains., Conclusions: Our study showed a high genetic diversity of WENV strains in China, emphasizing the relevance of surveillance of this emerging mammarenavirus in both natural reservoirs and humans., (© 2021. This study quantified the association of rodent fruit damage and the microbiological quality of irrigation water on the risk of microbiological contamination of strawberries collected from 18 U-pick farms across five different districts in the Guangzhou metropolitan region of southern China. Fifty-four composite strawberries samples, with or without evidence of rodent or avian foraging damage (i.e., bitten), along with 16 irrigation water samples, were collected during the spring of 2014 and winter of 2015 from our cohort of 18 farms. Composite strawberry samples and irrigation water were analyzed for total coliforms, E. coli , Salmonella , E. coli O157, Giardia , and Cryptosporidium . Total coliforms and E. coli were detected in 100% and ~90% of irrigation water samples, respectively. In contrast, Cryptosporidium was detected in only two water samples, while Salmonella , E. coli O157, and Giardia were not detected in any water samples. Strawberries with signs of being bitten by wildlife had significantly higher concentrations of total coliforms and E. coli, compared to strawberries with no physical evidence of rodent damage ( p < 0.001). Similarly, Cryptosporidium was detected in 7/18 (39%) of bitten, 4/18 (22%) of edge, and 5/18 (28%) of central strawberry samples, respectively. Concentration of E. coli on strawberries ( p < 0.001), air temperature ( p = 0.025), and presence of Cryptosporidium in irrigation water ( p < 0.001) were all associated with the risk of Cryptosporidium contamination on strawberries. Salmonella and Giardia were detected in <4% strawberry samples and E. coli O157 was not detected in any samples. These results indicate the potential food safety and public health risks of consuming unwashed strawberries from U-pick farms, and the need for improved rodent biosecurity of U-pick strawberry fields and enhanced microbiological quality of irrigation water used at these facilities.

Objective: To describe changes in rabies surveillance and management in Guangzhou, China between 1951 and 2015, and to analyse human rabies cases over that period., Methods: Rabies control policies and strategies implemented by the Guangzhou government were reviewed for three periods: 1951 to 1978, 1979 to 2000 and 2001 to 2015. Data on human rabies deaths and exposure were obtained from Guangzhou and national health and disease records. The demographic characteristics of human cases are reported using descriptive statistics., Findings: Between 1951 and 2015, the number of organizations cooperating on rabies control increased: there were two between 1951 and 1978, six between 1979 and 2000, and nine between 2001 and 2015. The number of human rabies cases reported in these periods was 331, 422 and 60, respectively. Organizations involved included city and district centres for disease control and prevention, rabies outpatient clinics, medical institutions and police. Overall, 88% (713/813) of cases occurred in rural districts, though, between 1951 and 2015, the distribution shifted from being predominantly rural to being both urban and rural. The number of people exposed to rabies increased annually. The biggest increases were among those injured by a pet dog or other animal: 3.26 and 4.75 times, respectively, between 2005 and 2015., Conclusion: Increased cooperation on rabies control between civil organizations in Guangzhou over decades was associated with a marked decrease in the number of human rabies cases. The Guangzhou experience could thereby provide guidance for other cities experiencing similar rabies epidemics.

This study assessed microbiological safety of water from public swimming pools in Guangzhou, China. Water samples from 39 outdoor municipal swimming pools were collected from late June to early September, 2013 and subjected to detection of protozoa ( Giardia and Cryptosporidium ) and bacteria ( Pseudomonas aeruginos , total coliforms, E. coli , E. coli O157, Shigella , and Salmonella ). Cryptosporidium and Giardia were both detected in 5 (12.8%) swimming pools. Total coliforms were detected in 4 (10.3%) samples with concentrations ranging from 1.3 to 154.0 MPN/100 mL while E. coli was detected in 4 (10.3%) samples with concentrations ranging from 0.5 to 5.3 MPN/100 mL. P. aeruginosa was detected in 27 (69.2%) samples but E. coli O157, Shigella and Salmonella were not detected. Among these swimming pools, 9 (23%) met the Chinese National Standard of residual chlorine levels and 24 (62%) were tested free of residual chlorine at least once. The multi-locus sequence typing (MLST) analysis showed that all P. aeruginosa isolates belonged to new sequence types (STs) with dominant ST-1764 and ST-D distributed in different locations within the area. Some P. aeruginosa strains were resistant to medically important antibiotics. Results indicate potential public health risks due to the presence of microbiological pathogens in public swimming pools in this area.

Strains of Francisella spp. were isolated from cooling water from an air conditioning system in Guangzhou, China. These strains are Gram negative, coccobacilli, non-motile, oxidase negative, catalase negative, esterase and lipid esterase positive. In addition, these bacteria grow on cysteine-supplemented media at 20 °C to 40 °C with an optimal growth temperature of 30 °C. Analysis of 16S rRNA gene sequences revealed that these strains belong to the genus Francisella. Biochemical tests and phylogenetic and BLAST analyses of 16S rRNA, rpoB and sdhA genes indicated that one strain was very similar to Francisella philomiragia and that the other strains were identical or highly similar to the Francisella guangzhouensis sp. nov. strain 08HL01032 we previously described. Biochemical and molecular characteristics of these strains demonstrated that multiple Francisella species exist in air conditioning systems.

Cases of brucellosis were diagnosed in 3-month-old twins and their mother. An epidemiologic survey suggested that raw sheep or goat meat might be the source of Brucella melitensis infection. This finding implies that the increasing threat of brucellosis might affect low-risk persons in urban settings in China.

Objective: To identify and characterize the strain 08H101032 was isolated from air condition systems in the routine investigations of Legionella in Guangzhou, China, in 2008., Methods: We adopted several phenotypic and genotypical methods, such as the growth status on various media, morphological, physical and biochemical characteristics, animal test, antibiotic susceptivities, PCR identification, sequence analysis of 16S RNA and RNA polymerase beta-subunit (ropB) gene etc, to determinate the phylogenetic position and outline the basic biological characteristics., Results: Strain 08H101032 was Gram-negative with polymorphic short rods or coccobacillus; with no flagella; devoid of spores; well growth on buffered charcoal yeast extraction (BCYE) agar and BCYE supplemented with glycine (3 g/L), polymyxin B sulfate (80000 iu/L), vancomycin (1 mg/L) and cycloheximide (80 mg/L) (GVPC medium) within 2 days, but delayed growth on ordinary sheep blood agar untill 5 - 7 days; catalase positive; oxidase negative; no reduction of nitrate; no hydrolysis of urea; delayed fermention of glucose to produce acid; which was primarily considered as Legionella. It was lastly identified to the genus Fransicella, characterized by a variety of biochemical and molecular phylogenetic tests, which shared the highest similarities to F. Philomiragia with 95.3% to 16S rRNA gene of 1377 oligo nucleotides and 87.3% to ropB gene of 367 oligo nucleotides (GenBank accession number: FJ591095, FJ939309). Growth were observed after a treatment for 10 minutes with the KCl-HCl buffer of pH 2.2, 20 degrees C, and at 25 degrees C, 37 degrees C (optimum 25 degrees C - 28 degrees C), but not at 42 degrees C. The cells had capsule-like construction by transmission electron microscopy, however no virulence found to mice., Conclusions: Strain 08H101032 was a potential new species of the genus Fransicella with a typical characteristic of L-cysteine growth stimulating activity, distinguishingly to Legionella with L-cysteine growth dependent activity.
