Plantations actively participate in the global carbon cycle and play a significant role in mitigating global climate change. However, the influence of forest management strategies, especially planting density management, on the biomass carbon storage and production value of plantations for ensuring carbon sink benefits is still unclear. In this study, we estimated the carbon sequestration and economic value of Pinus massoniana plantations with various stand densities and rotation ages using a growth model method. The results revealed that with increasing stand age, low-density plantations at 2000 trees·ha−1 (358.80 m3·ha−1), as well as high-density plantations at 4500 trees·ha−1 (359.10 m3·ha−1), exhibited nearly identical standing volumes, which indicated that reduced inter-tree competition intensity favors the growth of larger trees during later stages of development. Furthermore, an increase in planting density led to a decrease in the average carbon sequestration rate, carbon sink, and number of trees during the rapid growth period, indicating that broader spacing between trees is favorable for biomass carbon accumulation. Further, extending the rotation period from 15 to 20 years or 25 years and reducing the optimal planting density from 3000 to 2000 trees·ha−1 increased the overall benefits of combined timber and carbon sink income by 2.14 and 3.13 times, respectively. The results highlighted that optimizing the planting density positively impacts the timber productivity and carbon sink storage of Pinus massoniana plantations and boosts the expected profits of forest managers. Drought stress poses a serious threat to Pinus massoniana seedling growth in southern China. Trichoderma species, as beneficial microorganisms, have been widely used in agriculture to enhance plant growth and drought tolerance, but the interaction mechanisms remain unclear. To investigate the effect of drought-resistant Trichoderma longibrachiatum inoculation on P. massoniana growth under drought stress, the plant physiological indicators and rhizosphere microbiome diversity were measured to identify Trichoderma-activated mechanisms. Trichoderma longibrachiatum inoculation significantly promoted P. massoniana growth under drought treatment, and enhanced nitrogen, phosphorus, and potassium absorption compared with those of non-inoculated seedlings. Trichoderma longibrachiatum treatment alleviated the damage to cell membranes and needle tissue structure, and significantly increased antioxidant enzyme activities, osmotic substance contents, and photosynthesis in P. massoniana in response to drought stress. Soil nutrient contents, activities of sucrase, phosphatase, and urease as well as the relative abundances of the dominant genera Burkholderia, Rhodanobacter, and Trichoderma were elevated in the rhizosphere soil of P. massoniana inoculated with T. longibrachiatum under drought stress. A network analysis showed that certain crucial dominant taxa driven by T. longibrachiatum inoculation, including Penicillium, Trichoderma, Simplicillium, Saitozyma, Burkholderia, Bradyrhizobium, Sinomonas, and Mycobacterium, had more correlations with other microorganisms in the soil. Aims: Pinus massoniana Lamb. is a widely distributed native species and afforestation pioneer tree in the subtropical forests of China; however, the species' quality and productivity are significantly affected by phosphorus (P) deficiency. Although ectomycorrhizal (ECM) fungi can enhance plant adaptation to nutrient deficiency, the interactive effects of ECM fungi colonization and P availability on physiological responses and nutrient uptake in P. massoniana seedlings remain unknown. Method: We compared the physiological response and nutrient absorption of uninoculated P. massoniana seedlings and seedlings inoculated with Suillus luteus or S. bovinus under three P supplies (P deficiency, 0.01 mM/L; medium P, 0.1 mM/L; and P sufficiency, 1 mM/L) in sand culture. Results: P deficiency limited the seedlings growth and nutrient content but increased the antioxidant enzyme activity. S. luteus and S. bovinus significantly increased the seedlings growth under P deficiency. The activities of superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) as well as the contents of malondialdehyde (MDA) and proline of inoculated and uninoculated seedlings were significantly higher under P deficiency than P sufficiency. Under P deficiency, POD and SOD activities increased with time and were higher in inoculated seedlings; however, proline contents were lower in inoculated seedlings. P deficiency decreased the P and potassium (K) contents in the roots, stems, and leaves of seedlings. Moreover, under P deficiency, S. luteus significantly improved P content in stems and leaves, whereas S. bovinus significantly improved K content in leaves. Aims: Soil water deficits have presented challenges to vegetation restoration in rocky desertification areas. In the field, small volumes of soil resources are present only in near-surface crevices, fissures, and other similar features. Water stored in the soil in near-surface fissures can help plants grow in such areas. The goals of this study are to discuss the soil water retention functions of near-surface fissures in terms of soil structure, water infiltration and water storage capacity and to define the role of fissure water in the growth of plants in rocky desertification areas. Methods: Several near-surface fissures with four types of vegetation (i.e., crops, grass, shrubs and trees) within a rocky karst desertification area on the Karst Plateau in Guizhou Province, China, were examined. Soil physicochemical property analysis and stable isotope techniques were applied. Results: Fissures with shrubs and trees present high levels of soil porosity, while fissures with crops and grasses present low levels of soil porosity. The water infiltration rates of the soil in all of the examined fissures are higher than the rainfall intensity of the maximum daily rainfall for this province. Consequently, most rainwater infiltrates through the fissure soils. Compared to the other fissures, fissures with crops present higher levels of usable soil storage capacity in the surface soils (0–20 cm), which are affected by tillage (ploughing), but exhibit lower capacities in the bottom soil layer. Additionally, tree and shrub fissures present higher usable soil storage capacities in bottom soil layer than other types of fissures. Conclusions: The main source of water for Ligustrum and Pyracantha in the dry season is fissure water, which accounts for 44.7% and 58.2% of all the water utilized by these species, respectively. With increasing age, plants will cause changes in soil physicochemical properties. The objective of this study was to investigate differences in the soil physicochemical properties in different-aged Masson pine forest plantations (i.e., 10, 20, 40, and 60 years old). Soil samples were collected in a pure Masson pine forest plantation in Southwest China. The soil determination indexes included organic carbon, nitrogen, phosphorus and potassium contents, water content, bulk density, and pH. The soil pH of a 20-year-old forest was significantly (p < 0.05) higher than that of a 10, 40, and 60-year-old forest. In addition, soil-available phosphorus in a 60-year-old forest was significantly (p < 0.05) higher than that in the other three age forest groups. With increasing forest age, available phosphorus increased, while available nitrogen decreased at 20 years old and then increased at 40 years old. There was a significant positive correlation (p < 0.05) between total nitrogen and available potassium; no significant correlation (p> 0.05) between total phosphorus and total potassium, organic carbon, bulk density, and pH; and a significant negative correlation (p < 0.05) between available phosphorus and the water content. The availability and utilization efficiency of soil nutrients in young forests were higher than those in old forests and the intermediate forest age was an important time point that affected the soil properties. To improve the availability of soil nutrients and ensure the sustainable utilization of soil resources, it is necessary to increase the input of nitrogen and especially phosphorus. Masson pine is an important afforestation species in southern China, where seasonal drought is common. The present study focused on the effects of Suillus placidus, an ectomycorrhizal fungus, inoculation on the growth and physiological and biochemical performance of masson pine seedlings under four different watering treatments (well-watered, mild drought, moderate drought, and severe drought) to evaluate the symbiotic relationship between S. placidus and masson pine seedlings. Ectomycorrhizal-inoculated (ECM) and non-inoculated (NM) seedlings were grown in pots and maintained for 60 days using the weighing method. Results showed that seedlings' growth, dry weight, RWC, chlorophyll content, PSII efficiency, and photosynthesis decreased as drought stress intensified in both ECM and NM plants. This suggests that drought stress significantly limits the growth and photosynthetic performance of masson pine seedlings. Nevertheless, increased An/gs and proline contents in both NM and ECM prevented oxidative damage caused by drought stress. In addition, increased peroxidase (POD) activity is an essential defense mechanism of ECM seedling under drought stress. Compared with NM, ECM seedlings showed faster growth, higher RWC, and photosynthetic performance, and lower lipid peroxidation in cell membranes under drought stress, as indicated by higher POD activity and lower proline and malondialdehyde (MDA). Our experiment found that S. placidus inoculation can enhance the drought resistance of masson pine seedlings by increasing antioxidant enzyme activity, water use efficiency, and proline content, thereby enhancing growth under water-deficiency conditions. The effects of biochar (BC) on soil erosion and nutrient output have attracted widespread attention; however, the role of BC in soil and water conservation remains debated. In particular, the effect of BC on underground erosion and nutrient output in soil-mantled karst areas has not been clearly determined. The objective of this study was to investigate the effects of BC on soil and water conservation and nutrient output in surface-underground dual erosion structures in soil-mantled karst areas. Eighteen runoff plots (2 m × 1 m) were established at the Guizhou University research station. Two BC treatments (T1 = 30 t/ha; T2 = 60 t/ha) and a control treatment (CK = 0 t/ha) were used. The BC material was produced from corn straw. The experiment ran from January to December 2021 and a total of 1132.64 mm of rainfall was measured. Runoff and soil and nutrient loss at the surface and underground were collected during natural rainfall. The results showed that 1) compared to CK, the BC application significantly increased surface runoff (SR, P < 0.05) and reduced subsurface runoff (SF, P < 0.05), and underground fissure runoff (UFR) decreased in general but did not reach a significant level (P > 0.05). The total amount of SR collected in each treatment during the test period accounted for 51 %-63 % of the total amount of all collected outlet runoff (SR, SF and UFR); 2) total organic carbon (TOC), total nitrogen (TN) and total phosphorus (TP) were mainly exported through the UFR, and total potassium (TK) was mainly exported through the SR; and 3) compared to CK, the BC reduced TOC, TN and TP output through runoff but had no significant effect on TK output regardless of surface runoff or underground runoff. Thus, BC application reduces nonpoint source (NPS) pollution, and more importantly, it can inhibit TN and TP flow into groundwater through bedrock fissures. Our results provide further evidence for evaluating the soil and water conservation benefits of BC. Therefore, BC in soil-mantled karst agricultural areas can prevent groundwater pollution in karst regions. In general, BC enhances surface erosion and inhibits underground runoff and nutrients loss on soil-mantled karst slopes. Background: Pinus massoniana Lamb. is an important afforestation tree species with high economic, ecological and medicinal values. Aluminum (Al) toxicity driven by soil acidification causes dieback of P. massoniana plantations. Previous studies showed that ectomycorrhizal fungi alleviate Al stress damages in Pinus, but the underlying molecular mechanisms and key genes induced by ectomycorrhizal fungi inoculation under Al stress in Pinus have not been explored. Herein, we applied Al stress for 60 days to P. massoniana seedlings inoculated with Suillus luteus (SL) and those non-inoculated. Then, we compared their growth parameters and transcriptome in order to detect candidate genes induced by SL conferring Al tolerance in P. massoniana., Result: Our results showed that SL inoculation confers Al stress tolerance in P. massoniana through improved growth performance, strong antioxidant enzyme activities and reduced malondialdehyde accumulation as compared to non-inoculated seedlings. Transcriptome sequencing further supported these findings as very few genes (51 genes) were transcriptionally altered by Al in SL inoculated plants as compared to non-inoculated plants (2140 genes). We identified three core genes (cox1, cox3 and Nd1) that were strongly up-regulated by Al in the SL inoculated plants but were down-regulated in the non-inoculated plants. We also identified 42 genes specifically regulated by SL inoculated plants under Al stress, which are involved in a wide range of biological processes such as antioxidative response, transporters, hormone signaling and plant pathogen infection responses., Conclusions: Altogether, our data suggest that SL inoculation induces priming of key stress response pathways and triggers specific genes that efficiently alleviate Al stress effects in P. massoniana. The candidate genes resources generated in this study are of utmost importance for functional characterization and molecular studies aiming at improving Al tolerance in plants.
