The escalating concern of perfluoroalkyl and polyfluoroalkyl substances (PFAS), particularly at contaminated sites, has prompted extensive investigations. In this study, samples of multimedia including air, rhizosphere soil, and tissues of wheat at various growing stages were collected near a mega fluorochemical industrial park in China. Perfluorooctanoic acid (PFOA) was predominant in both air and soil with a strong correlation, highlighting air deposition as an important source in the terrestrial system. PFAS concentrations in wheat decreased in the stem and ear but increased in the leaves as wheat matured. Specifically, perfluorobutanoic acid (PFBA) dominated in the aboveground tissues in the full-life-cycle, except that PFOA surpassed and became predominant in leaves during the filling and maturing stages, hinting at an airborne source. For all PFAS, both bioaccumulation factors and translocation factors (TFs) were inversely correlated with the carbon chain length during the full-life-cycle. The obtained TF values were considerably higher than those obtained from ambient sites reported previously, further suggesting an unneglectable foliar uptake from air, which was estimated to be 25% for PFOA. Moreover, spray irrigation remarkably enhanced the absorption of PFAS in wheat via foliar uptake relative to flood irrigation. The estimated daily intake of PFBA via wheat consumption and air inhalation was 0.50 μg/kg/day for local residents, at least one magnitude higher than the corresponding threshold, suggesting an alarmingly high exposure risk.

In this study, various crops and farmland soils were collected from the Fen-Wei Plain, China, to investigate the bioavailability of perfluoroalkyl substances (PFAS), their accumulation in edible plant tissues, and the factors impacting their accumulation. PFAS were frequently detected in all of the crops, with total concentrations ranging from 0.61 to 35.8 ng/g. The results of sequential extractions with water, basic methanol, and acidic methanol indicate that water extraction enables to characterize the bioavailability of PFAS in soil to edible plant tissues more accurately, especially for the shorter-chain homologues. The bioavailability of PFAS was remarkably enhanced in the rhizosphere (RS) soil, with the strongest effect observed for leafy vegetables. The water-extracted Σ 16 PFAS in RS soil was strongly correlated with the content of dissolved organic carbon in the soil. Tannins and lignin, identified as the main components of plant root exudates by Fourier transform-ion cyclotron resonance mass spectrometry, were found to enhance the bioavailability of PFAS significantly. Redundancy analysis provided strong evidence that the lipid and protein contents in edible plant tissues play important roles in the accumulation of short- and long-chain PFAS, respectively. Additionally, the high water demand of these tissues during the growth stage greatly facilitated the translocation of PFAS, particularly for the short-chain homologues and perfluorooctanoic acid.

Per- and polyfluoroalkyl substances (PFASs) have been widely detected in various food, which has attracted worldwide concern. However, the factors influencing the transfer and bio-accumulation of PFASs from soils to wheat in normal farmland, is still ambiguous. We investigated the PFASs accumulation in agricultural soils and grains from 10 cites, China, and evaluated the health risks of PFASs via wheat consumption. Our results show that ∑PFASs in soils range from 0.34 μg/kg to 1.59 μg/kg with PFOA and PFOS dominating, whilst ∑PFASs in wheats range from 2.74 to 6.01 μg/kg with PFOA, PFBA and PFHxS dominating. The lower pH conditions and high total organic carbon (TOC) could result in the higher accumulation of PFASs in soils and subsequently in wheat grains, whilst the bioaccumulation factors of PFASs increase with increasing pH conditions but not with TOC. The estimated daily intake (EDI) values of PFBA, PFOA, and PFHxS are relatively high, but data supports that ingesting wheat grains does not result in any potential risk to the human beings. The widespread human exposure to perfluoroalkyl acids (PFAAs) has led to increasing public concern. In this study, we present a comprehensive measurement of total fluorine (TF), extractable organic fluorine (EOF), identified organic fluorine (IOF, total concentration of identified PFAAs quantified as fluorine) and 11 target PFAAs in human serum (n = 60), hair (n = 49) and nails (n = 39) collected from non-occupation exposed volunteers in 10 cities of Guangdong Province, China. The results indicated that EOF was the major form of fluorine in serum, accounting for 70-80% of TF. The levels of IOF contributed less than 10% of EOF. Perfluorooctane sulfonic acid (PFOS) was found to be the dominant PFAA with mean concentration of 23 ng·mL -1 in serum, 35 ng·g -1 in hair and 33 ng·g -1 in nail, respectively. Short-chain PFAAs (C ≤ 10) were the predominant PFAAs in three matrices. Levels of PFOS, perfluorohexane sulfonic acid (PFHxS), perfluoroundecanoic acid (PFUdA) and perfluorotridecanoic acid (PFTrDA) in males are significantly higher than those in females (p < 0.01). Significant positive correlations were observed between nail and serum for PFOS (p < 0.01), perfluorooctanoic acid (PFOA) (p < 0.05) and PFHxS (p < 0.01), suggesting that human nails, a noninvasive sample, are a promising bio-indicator for PFAA risk assessment.

PFAAs have emerged as persistent organic pollutants (POPs) in a variety of environmental matrixes and biota, posing potential hazards for wildlife and humans. Diet has been considered as an important source for human exposure to PFAAs and seafood accounts for a relatively large proportion in human diets, especially for coastal residents. In an effort to clarify the impact of PFAAs in seafood on human health, 17 PFAAs were determined in 42 pooled seafood samples (14 species) from the Yellow Sea areas, China. The concentrations of total PFAAs (sum of 17 PFAAs, ∑PFAAs) were in the range of 1.10-1067 ng/g dry weight (dw), with the highest concentration found in swimming crab (Portunus trituberculatus) and the lowest in silvery pomfret (Pampus argenteus). Concentrations and composition profiles of PFAAs varied significantly among different species, suggesting that bioaccumulation potential of PFAAs differed from species to species. The distinct spatial distribution of PFAAs in four categories of seafood could be mainly attributed to the contamination patterns of PFAAs in three nearby cities. PFBA was presented as the most abundant PFAA in this study, which was different from the findings in many other studies where PFOS was the predominant compound. Perfluoroalkyl substances (PFASs) are a family of synthetic, fluorinated organic compounds. They have been widely used in industrial applications and consumer products and widespread in the environment, wildlife and human. Experimental and epidemiologic evidence suggested that PFASs are capable of interfering with endocrine processes and have potential reproductive and developmental toxicities. Polycystic ovarian syndrome (PCOS), one of the main reasons of female infertility, is a common endocrine disorder in reproductive age women. We performed a case-control study to evaluate associations between PCOS-related infertility and PFASs concentrations in plasma. A total of 180 infertile PCOS-cases and 187 healthy controls were recruited from the Center for Reproductive Medicine of Shandong University. Blood specimens were collected at enrollment and analyzed for ten PFASs using liquid chromatography-tandem mass spectrometry. Multivariable logistic regression procedure was used to estimate odds ratios (ORs) and 95% confidence intervals (CIs) for each PFAS. Perfluorooctanoic acid (PFOA) and perfluorooctane sulfonate (PFOS) were the dominant PFASs in the plasma of participants, with the median concentration of 5.07 ng/mL and 4.05 ng/mL, respectively. The median levels of individual PFAS were not significantly different between PCOS-cases and controls. While adjusted for the potential confounders (age, BMI, household income, education level, employment status, age at menarche, menstrual volume), the plasma concentration of perfluorododecanoic acid (PFDoA), a 12 carbons lengths of perfluorocarboxylic acids, was associated with a significantly increased risk of PCOS-related infertility (medium vs low tertile: OR = 2.36, 95% CI: 1.12, 4.99, P = 0.02; high vs low tertile: OR = 3.04, 95% CI: 1.19, 7.67, P = 0.02), with the P trend 0.01. No significant relationship was observed between PCOS-related infertility and other PFAS analytes in the adjusted model, despite perfluoroundecanoic acid showed a negative association (P trend 0.03). Sixteen per- and polyfluoroalkyl substances (PFASs) were measured in liver (n = 52) and kidney (n = 18) tissues of Indo-Pacific humpback dolphins (Sousa chinensis) stranded in the Pearl River Estuary (PRE) of China between 2004 and 2016. The average concentrations of perfluorooctane sulfonate (PFOS), perfluorooctanoic acid (PFOA) and most of other PFASs in the liver samples were respectively greater than any records previously reported in cetaceans globally. PFOS levels in 46% of dolphin liver samples exceeded the hepatic toxicity threshold in cetaceans. For the first time, we found a U-shaped trend for the distribution pattern of perfluorinated carboxylic acids (PFCAs) between liver and kidney with increasing carbon chain lengths (C5-C16), whereas a descending trend (C4-C10) was found for perfluoroalkane sulfonic acids (PFASs), which may be explained by binding efficiencies of PFAS analogues to proteins. Dolphins with the highest levels of ∑PFASs (age-corrected) were clustered near the river outlets in Lingdingyang area, which agrees with the spatial distribution of PFASs in the environment. Significant temporal trends were observed for many PFASs. Concentrations of PFOA, perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA) and perfluoroheptanoic acid (PFHpA) all peaked in year 2011, followed by a decreasing trend, while a consistently descending trend was shown for perfluoroundecanoic acid (PFUdA) and perfluorodecane sulfonate (PFDS). Our findings contribute to the knowledge of tissue distribution and spatiotemporal trends of PFASs in the PRE dolphins, which are valuable for us to understand the PFASs exposure risk and their industrial emission in Southern China., (Copyright © 2018. Perfluoroalkyl and polyfluoroalkyl substances (PFASs) can be transferred from a mother to her fetus during pregnancy and adversely affect fetal development. However, the efficiency and influencing factors of PFASs maternal-fetal transfer remain unclear. We measured the levels of six perfluoroalkylcarboxylates, three perfluoroalkylsulfonates, and one sulfonamide in 369 pairs of maternal and umbilical cord serum and examined the transplacental transfer efficiency (TTE) of PFASs by the functional group and carbon chain length in a prospective birth cohort in Shandong, China. All ten PFASs were detected in both maternal and umbilical cord serum in nearly all samples. Maternal and cord levels were closely correlated (the correlation coefficient [r] ranging from 0.485 to 0.908) in most PFASs except perfluorobutane sulfonic acid (PFBS) (r = 0.159). TTE was significantly affected by the functional group and carbon chain length. Compared to perfluoroalkylcarboxylates, perfluoroalkylsulfonates had a lower ratio of maternal to fetal transfer. A U-shaped relationship between carbon chain length and TTE was observed for perfluoroalkylcarboxylates while a monotonic descending trend was identified between TTE and the increasing carbon chain length for perfluoroalkylsulfonates. PFASs can readily pass through the placenta. The functional group and carbon chain length are important determinants for the TTE of PFASs.

Non-invasive samples present ethical and practical benefits for investigating human exposure to hazardous contaminants, but analytical challenges and difficulties to interpret the results limit their application in biomonitoring. Here we investigated the potential for using hair, nail and urine samples as a measure of internal exposure to an array of legacy and emerging per- and polyfluoroalkyl substances (PFASs) in two populations with different exposure conditions. Paired urine-serum measurements of PFASs from a group of highly exposed fishery employees displayed strong correlations for PFASs with three to eight perfluorinated carbons (ρ > 0.653; p < 0.01). Consistent statistical correlations and transfer ratios in nails and hair from both populations demonstrated that these non-invasive samples can be used as a measure of internal exposure to perfluorooctane sulfonic acid and C8 chlorinated polyfluoralkyl ether sulfonic acid (C8 Cl-PFESA). Contrastingly, the infrequent detections and/or lack of consistent transfer ratios for perfluorooctanoic acid, perfluorononanoic acid and short-chain PFASs in hair and nail samples indicate passive uptake from the external environment rather than uptake and internal distribution. Collectively, the study supports the use of urine samples as a valid measure of internal exposure for a range of short- and medium-chain PFASs, while the validity of nail and hair samples as a measure of internal exposure may vary for different PFASs and populations. Fluorotelomer alcohol (6:2 FTOH) is a polyfluoalkyl substance that has been widely used in industry and consumer products in recent years, causing potential harm to the environment. However, currently the impact of 6:2 FTOH and its degradation products on microbial communities in sediment is unclear. The purpose of this study is to explore the impact of the biodegradation of 6:2 FTOH on bacterial community structures in surface sediment based on gene analysis. Surface sediment and river water were collected from Hai river, Tianjin, and a microcosm experiment was performed in the laboratory. The concentration of 6:2 FTOH and its degradation products were analyzed by liquid chromatograph-mass spectrometry/mass spectrometry (LC-MS/MS). The bacterial community structure was analyzed by denaturing gradient gel electrophoresis and high-throughput sequencing. The results showed that 6:2 FTOH could be degraded by microorganisms (half-life was less than 3 d), producing transient products such as 6:2 fluorotelomer carboxylic acid (FTCA) and 6:2 fluorotelomer unsaturated carboxylic acid (FTUCA) and stable products such as 5:2 fluorotelomer (FT) ketone, 5:2 fluorotelomer alcohol (sFTOH), perfluorohexanoic acid(PFHxA), perfluoro-n-pentanoic acid (PFPeA), perfluorobutanoic acid (PFBA) and 5:3 polyfluorinated acid. At different stages of 6:2 FTOH degradation, a change of bacteria and the predominant population became somewhat different. Based on the experimental results for 100 d, at the Phylum level, the biodegradation of 6:2 FTOH greatly increases the abundance of Chloroflexi (+24.8%) and decreases the abundance of Proteobacteria (-17.8%) and Firmicutes (-15.9%). At the Class level, due to the biodegradation of 6:2 FTOH, bacteria with notable increases included Anaerolineae (+19.6%) and δ -Proteobacteria (+4.3%), while bacteria with notable decreases included ε -Proteobacteria (-20.0%), Clostridia (-10.1%), Bacilli (-5.8%) and γ -Proteobacteria (- 4.2%). At the Genus level, due to the biodegradation of 6:2 FTOH, bacteria with notable increases included Anaerolineaceae&#95;(uncultured) (+19.1%) and Thioalkalispira (+13.3%), while bacteria with notable decreases included Vibrio (-14.1%), Sulfurimonas (-13.2%), Bacillus (-5.1%), Sulfurovum (-4.2%) and Fusibacter (-4.1%). These results are helpful for predicting the response of bacteria to the contamination of polyfluoalkyl substances and isolating the bacteria capable of the biodegradation of polyfluoalkyl substances.

In this study, 90 human breast milk samples collected from Japan, Korea, and China were analyzed for perfluorooctanoic acid (PFOA) (C8), perfluorononanoic acid (PFNA) (C9), perfluorodecanoic acid (PFDA) (C10), perfluoroundecanoic acid (PFUnDA) (C11), perfluorododecanoic acid (PFDoDA) (C12), and perfluorotridecanoic acid (PFTrDA) (C13). In addition, infant formulas (n = 9) obtained from retail stores in China and Japan were analyzed. PFOA was the predominant compound and was detected in more than 60% of samples in all three countries. The PFOA, PFNA, PFDA, and PFUnDA levels in Japan were significantly higher than those in Korea and China (p<0.05). The PFTrDA level was highest in Korea (p<0.05). The median PFOA concentrations were 89 pg mL(-1) (48% of total perfluorinated carboxylic acids (PFCAs) (C8-C13)) in Japan, 62 pg mL(-1) (54%) in Korea, and 51 pg mL(-1) (61%) in China. The remaining ∑PFCAs (C9-C13) were 95 pg mL(-1) in Japan, 52 pg mL(-1) in Korea, and 33 pg mL(-1) in China. Among the long-chain PFCAs, odd-numbered PFCAs were more frequently detected than even-numbered PFCAs, except for PFDA in Japan. There were no evident correlations between the mother's demographic factors and the PFCA concentrations. PFOA, PFNA, and PFDA were frequently detected in both Japan and China, but there were no significant differences between the two countries. The total PFCA concentrations in the infant formulas were lower than those in the breast milk samples in Japan (p<0.05), but not in China (p>0.05). The accumulation of perfluorinated compounds (PFCs) in the sera of captive wildlife species Bengal tigers (Panthera tigris tigris) and African lions (Panthera leo Linnaeus) from Harbin Wildlife Park, Heilongjiang Province, in China were analyzed by high-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Perfluorooctanesulfonate (PFOS) was the predominant contaminant with a mean serum concentration of 1.18 ng mL(-1) in tigers and 2.69 ng mL(-1) in lions. Perfluorononanoic acid (PFNA) was the second most prevalent contaminant in both species. The composition profiles of the tested PFCs differed between tigers and lions, and the percentages of perfluorooctanoic acid (PFOA) were greater in lions than in tigers, indicating different exposures and/or metabolic capabilities between the two species. Assessments of the risk of PFC contamination to the two species were obtained by comparing measured concentrations to points of departure or toxicity reference values (TRVs). Results suggest no risk of PFOS exposure or toxicity for the two species.
