COTE1 was recently described as an oncogene in hepatocellular carcinoma and gastric cancer. However, the roles of COTE1 in intrahepatic cholangiocarcinoma (ICC) are little known. Our study is aimed at clarifying novel functions of COTE1 in ICC progression, including proliferation, invasion, and autophagy. By using quantitative real-time PCR, immunohistochemistry staining, and western blotting, we found that COTE1 expression was frequently upregulated in ICC tissues, compared to paracarcinoma tissues. High COTE1 expression was significantly correlated with aggressive clinical features and predicted poor prognosis of ICC patients. Functional experiments revealed that ectopic COTE1 expression promoted ICC cell proliferation, colony formation, cellular invasion, migration, and in vivo tumorigenicity; in contrast, COTE1 knockdown resulted in the opposite effects. At molecular mechanism in vitro and vivo, our study revealed that COTE1 overexpression suppressed autophagy via Beclin1 transcription inhibition; conversely, COTE1 silencing facilitated autophagy through promoting Beclin1 expression. Furthermore, the suppression of COTE1 knockdown on cellular growth and invasion was rescued/aggravated by Beclin1 inhibition/accumulation. Our data, for the first time, illustrate that COTE1 is an oncogene in ICC pathogenesis, and the ectopic COTE1 expression promotes ICC proliferation and invasion via Beclin1-dependent autophagy inhibition. Boron carbide (B4C) has excellent high-temperature oxidation resistance, high hardness, low density, high melting point and excellent abrasive resistance, which is widely used in fields such as refractories, wear-resistant materials and lightweight protective materials. The research progress and the application of B4C materials in China and at abroad in recent years were summarized. The influences of sintering processes (pressureless sintering, hot pressing sintering, hot isostatic pressing sintering, spark plasma sintering and microwave sintering) and sintering additives (simple substances, oxides and carbides) on the B4C densification were analyzed. Background: The prognostic benefits and safety of extended lymphadenectomy for hilar cholangiocarcinoma remain uncertain. The available evidence is still insufficient concerning its retrospective aspect. The aim of this study is to explore the clinical effect and safety of extended lymphadenectomy compared to regional lymphadenectomy in patients with hilar cholangiocarcinoma.Methods: The Relay-HC trial is a prospective, multicenter, and randomized controlled trial. Seven hundred and thirty-four eligible patients with resectable perihilar cholangiocarcinoma across 15 tertiary hospitals in China will be randomly assigned (1:1) to receive either regional lymphadenectomy or extended lymphadenectomy. The primary objective is to determine the overall survival after the two approaches. Secondary objectives of the study include the evaluation of perioperative mortality, postoperative complication, and disease-free survival. This study has been approved by the ethics committee of each participating hospital.Discussion: The Relay-HC trial is designed to investigate the prognostic benefits and safety of expanded lymphadenectomy for hilar cholangiocarcinoma. Clarifying the relationship between urban expansion and ecosystem services (ESs) is critical for sustainable management of land resources and ecosystems. However, little is known about the relationship between the two at the cross-scale (particularly at the national-provincial scale). Therefore, we conducted a systematic assessment of the spatiotemporal dynamics and the relationship between urban expansion and ESs including food production (FP), soil conservation (SC), carbon sequestration (CS), and water yield (WY) in China from 1992 to 2020 on the national-provincial scale. The results show that China's urban expansion took up a large amount of cropland, accounting for 79.35% of the newly-added built-up land. Shandong had the largest expansion scale and the highest speed, Shanghai had the most pronounced expansion intensity, and more than 50% of the provinces were dominated by outlying expansion pattern. In terms of total change, the three ESs of FP, SC, and WY increased by 286.5 × 106 t, 1893.61 × 106 t, and 8337.20 × 106 mm, respectively, and CS decreased by 683.90 × 106 Mg C. However, in the urban expansion area, FP and CS net decreased by 1757.6 × 104 t and 19,640.19 × 104 Mg C, respectively, while SC and WY net increased by 347.52 × 104 t and 20,264.11 × 104 mm, respectively. Shandong contributed the most to changes in ESs in urban expansion areas. Urban expansion was significantly negatively correlated with FP and CS with the correlation coefficients > -0.8; it was significantly positively correlated with SC and WY, with coefficients of 0.714 and 0.413, respectively, and urban expansion had a lagged effect on ESs. The impact of urban expansion on ESs had a spatial spillover effect and showed prominent spatial clustering in Anhui, Henan, and Shandong. Based on these results, we proposed urban planning countermeasures grounded in the perspective of ES improvement, which would provide policy references for the sustainable management of the ecological environment and land resources. • Urban expansion and ESs at the national-provincial scale were quantified. • The relationship between urban expansion and ESs varied by type of ESs. • Shandong was the largest contributor to ESs changes in urban expansion areas. • There were time lags and spatial spillovers between urban expansion and ESs. Aim: To determine whether genetic variations in DNMT1 and DNMT3A could be associated with gastric cancer risk., Materials & Methods: A total of 466 patients and 452 healthy controls were enrolled in this study. Genotypes were examined by using the Sequenom MassARRAY platform., Results: No significant differences in the distribution frequencies were observed between the groups. However, subgroup analysis revealed that, in the group aged ≤60 years, DNMT3A rs13420827 was associated with reduced gastric cancer risk, and that carrier of GC/GG genotype was associated with reduced risk of gastric cancer with low differentiation or at the T3-T4 stage., Conclusion: This study suggests that five genetic variations of interest in DNMT1 and DNMT3A are not associated with the presence of gastric cancer, but that rs13420827 may contribute to the gastric cancer risk for those younger individuals, the risk of which may be influenced by the characteristics of tumor.
