Mucor species are a group of common soil-borne fungi, known to cause infections on humans and animals, interfere in food production, and act as useful agents in biotechnological applications. This study reports one new Mucor species, M. yunnanensis, which was found to be fungicolous on an Armillaria sp. from southwest China. Further, M. circinelloides on Phlebopus sp., M. hiemalis on Ramaria sp. and Boletus sp., M. irregularis on Pleurotus sp., M. nederlandicus on Russula sp., and M. yunnanensis on Boletus sp. are reported as new host records. Mucor yunnanensis and M. hiemalis have been collected from Yunnan Province in China, whereas M. circinelloides, M. irregularis, and M. nederlandicus have been collected from Chiang Mai and Chiang Rai Provinces in Thailand. All the Mucor taxa reported herein were identified based on both morphology and phylogenetic analyses of a combined nuc rDNA internal transcribed spacer region ITS1-5.8S-ITS2 (ITS) and partial nuc 28S rDNA (28S) sequence matrix. Comprehensive descriptions, illustrations, and a phylogenetic tree are provided for all the taxa reported in the study to show the placements of taxa, and the new taxon is compared with its sister taxa. Corn is widely planted in China, but corn stalks have not been adequately utilized for a long time. Here, the potential of corn stalks to serve as an ingredient in the composition of substrates to cultivate king oyster mushroom (Pleurotus eryngii) was studied. Corn stalks were added to the matrix at a ratio of 10.5%, 21%, or 42% to replace sawdust or sugarcane bagasse in the typical matrix. Analysis of the mycelia growth rate, production days, agronomic traits, and nutrient content confirmed the feasibility of using corn stalk instead of sawdust and bagasse for the cultivation of P. eryngii. The samples grown on Y5 (without sawdust and bagasse) has more crude fiber (8.3%) and polysaccharide (5.05%) content compared to those of the control substrate (7.1% and 4.16%, respectively), moreover, it also conteined more zinc and calcium than others. Therefore, these findings indicate that corn stalks could be used as an alternative to sawdust and sugarcane bagasse for P. eryngii production, which would not only reduce costs and improve food quality, but also improve waste utilisation. Most of the sequenced wood-rotting edible mushroom produce fruiting body at relatively low temperatures. Little information has been known about the high-temperature wood-rotting mushroom. Here, we performed de novo sequencing and assembly of the genome of a high-temperature edible mushroom Pleurotus giganteus from a monokaryotic strain zhudugu2 using the Illumina and Pac-Bio CLR sequencing technologies. P. giganteus, also known as Zhudugu in China, is a well-known culinary edible mushroom that has been widely distributed and cultivated in China, Southeast Asia, and South Asia. The genome consists of 40.00 Mb in 27 contigs with a contig N50 of 4.384 Mb. Phylogenetic analysis reveals that P. giganteus and other strains in Pleurotus clustered in one clade. Phylogenetic analysis and average nucleotide identity analysis indicated that the P. giganteus genome showed a closer relationship with other Pleurotus species. Chromosome collinearity analysis revealed a high level of collinearity between P. ostreatus and P. giganteus. There are 12,628 protein-coding genes annotated in this monoploid genome. A total of 481 enzymes accounting for 514 carbohydrateactive enzymes (CAZymes) terms were identified in the P. giganteus genome, including 15 laccases and 10 class II peroxidases predicted in the genome, which revealed the robustness of lignocellulose degradation capacity of P. giganteus. The mating-A type locus of P. giganteus consisted of a pair of homeodomain mating-type genes HD1 and HD2. The mating-B type locus of P. giganteus consisted of at least four pheromone receptor genes and three pheromone genes. Oyster mushroom (Pleurotus geesteranus Singer) is grown widely in China withut problems, but occasionally severe bacterial contamination occurred in cultivation bags of Pleurotus geesteranus during high summer temperatures in some companies in the Chunan county of Zhejiang province in 2018, belonging to the so-called Bacillus pumilus group by morphological characteristics, 16S rRNA, GyrB and aroE genes sequencing, fatty acid analysis and MALDI analysis. The inoculation tests confirmed that Bacillus pumilus strains were contaminants causing mycelial disappearance of P. geesteranus. The dual culture tests demonstrated that several strains of B. pumilus were able to inhibit mycelial growth of P. geesteranus. In order to find the sources of contamination the, process of mushroom cultivation was investigated. A 41.0% contamination rate occurred in cultivation bags with sponge plugs, as opposed to a very low contamination rate (<4%) in cultivation bags with cotton plugs. It was also shown that a 13.0% bacterial isolation rate was obtained from autoclaved sponge pieces, as opposed to 2.0% or 0.0% from cotton pieces, respectively. Fifteen strains isolated all were identified by MALDI-Biotyper as Bacillus spp., belonging to four species (Bacillus circulans, B. pumilus, B.cereus and B. halosaccharovorans). Dual culture tests showed that B. circulans, B. cereus and B. halosaccharovorans strains had not inhibitory effect on mycelial growth of P. geesteranus. Pantoea beijingensis, a gram-negative pathogenic bacterium, causes soft rot disease in the fungus Pleurotus eryngii in China. However, the taxonomic classification of this pathogen is controversial due to close relationships between bacteria of the genera Pantoea and Erwinia. This study aimed to resolve the identity of P. beijingensis using phylogenomic and systematic analyses of Pantoea and Erwinia by whole-genome sequencing. Single-copy orthologs identified from the Erwinia/Pantoea core genomes were used to delineate Erwinia/Pantoea phylogeny. P. beijingensis LMG27579T clustered within a single Erwinia clade. A whole-genome-based phylogenetic tree and average nucleotide and amino-acid identity values indicate that P. beijingensis LMG27579T should be renamed Erwinia beijingensis. The hrp/hrc genes encoding type III secretion system (T3SS) proteins in Erwinia and Pantoea were divided into five groups according to gene contents and organization. Neighbor-joining-inferred phylogenetic trees based on concatenated HrcU, HrcN, and HrcR in the main hrp/hrc cluster showed that E. beijingensis T3SS proteins are closely related to those in Ewingella americana, implying that E. beijingensis and E. americana have a recent common hrp/hrc gene ancestor. Furthermore, T3SS proteins of Erwinia and Pantoea were clustered in different clades separated by other bacterial T3SS proteins. Thus, T3SS genes in Pantoea and Erwinia strains might have been acquired by horizontal gene transfer. Overall, our findings clarify the taxonomy of the bacterium causing soft rot in P. eryngii, as well as the genetic structure and classification of the hrp/hrc T3SS virulence factor. Pleurotus eryngii (King Oyster) is one of the most highly prized edible mushrooms. Among the diverse varieties within P. eryngii , P. eryngii var. eryngii is the commonest one, with a worldwide distribution, while P. eryngii var. ferulae is only distributed in Europe and China, and is especially adapted to the Gobi Desert in Xinjiang Autonomous Region of China. However, little is known about the genome-wide pattern of evolution and adaptation to the divergent environments of P. eryngii. Here, we present the high-quality genome sequences of P. eryngii var. eryngii strain PEE81 originating from Europe and P. eryngii var. ferulae strain PEF12 originating from the Gobi Desert of China. The assembled genome sizes of PEE81 and PEF12 were 53.6 and 48.0 Mbp, respectively, which are larger than other reported genomes in the genus Pleurotus. We propose that the selective amplification of long terminal repeat (LTR) retrotransposons increases the genome size of the genus Pleurotus , and may play a key role in driving their rapid species diversification. Molecular clock analyses of five Pleurotus species, namely PEE81, PEF12, P. tuoliensis, P. ostreatus and P. cf. floridanus suggest that the divergence estimates of the genus Pleurotus over time scales ranged from ∼4 to ∼38 million years ago (Mya), and PEE81 and PEF12 diverged at ∼13 Mya. The whole genome resequencing of 33 geographically diverse strains of P. eryngii var. eryngii and var. ferulae was then performed and the genome variation among and within these two populations were investigated. Comparative analyses of these two populations detected several candidate genes related to stress responses and DNA repair that are putatively involved in adaptation to the Gobi Desert environment. Pleurotus giganteus (Berk.) Karunarathna & Hyde is one of the largest edible mushrooms which can be found either in groups or solitary and in soil and buried woods, but seldom in grassland. It has been recorded in Southeast Asia, Australasia, and China by either its common names or local names. Previously known as Lentinus/Panus giganteus, its unique physical characteristics and the utilization of molecular tools have since anchored its taxonomical position in the Pleurotus genus. A sawdust-based substrate coupled with soil casing method is described as a standard cultivation procedure for P. giganteus. The basidiocarp of P. giganteus is found to be rich in carbohydrates, proteins, polyunsaturated fatty acids, and polysaccharides. Both ethanol and aqueous extracts of P. giganteus have been studied in vitro for its antioxidative, antifungal, anticancer, hepatoprotective, and neurite outgrowth capabilities. The review covers the optimum cultivation methods of P. giganteus, as well as its various biological activities and medicinal properties that have been studied by different approaches. Available data in regards to the chemical compounds present in P. giganteus are also compiled for future references. Pleurotus tuoliensis, a kind of valuable and favorable edible mushroom in China, is always subjected to high environmental temperature during cultivation. In our previous study with P. tuoliensis, trehalose proved to be effective for tolerating heat stress. Trehalose-6-phosphate synthase (TPS; EC2.4.1.15) plays a key role in the biosynthesis of trehalose in fungi. In this study, a full-length of cDNA with 1,665 nucleotides encoding TPS (PtTPS) in P. tuoliensis was cloned. The PtTPS amino acid was aligned with other homologues and several highly conserved regions were analyzed. Thus, the TPS protein was expressed in Escherichia coli and purified by affinity chromatography to test its biochemical properties. The molecular mass of the enzyme is about 60 kDa and the optimum reaction temperature and pH is 30° C and 7, respectively. The UDP-glucose and glucose-6-phosphate were the optimum substrates among all the tested glucosyl donors and acceptors. Metal cations likeMg2+, Co2+, Mn2+, Ni2+, K+, Ag+ stimulated PtTPS activity significantly. Metal chelators such as sodium citrate, citric acid, EDTA, EGTA and CDTA inhibited enzyme activity. Recently, Pantoea beijingensis has become an emerging pathogen against Pleurotus eryngii, causing soft rot disease in China and Korea. A qPCR assay targeting hrpK, which is predicted to be one of the key genes for the pathogenicity and virulence of P. beijingensis, was developed and evaluated. Approximately 0.1 fg of purified plasmid or 3.23x10² copies µl-1 in pure cultures could be detected. Moreover, the use of an enrichment procedure could induce 100-fold enhancement of the sensitivity. The qPCR assay developed here is a rapid, reliable and sensitive tool for the detection of P. beijingensis at an early stage of infection and it will be useful in the development of management practices. “Spawn-burning” syndrome has become one of the most serious problem for edible mushroom cultivation in mushroom bags at high growth temperatures in China. The symptoms of spawn-burning are that the mushroom mycelia ceases growth, dies, and becomes contaminated with mould and bacteria. This article used Pleurotus eryngii as material to study the cause of spawn-burning syndrome. Spawn-burning of P. eryngii occurred at 34 °C, which was far lower than maximum growth temperature of 38 °C or the lethal temperature of 41 °C. Aerobic respiration increased at high growth temperature. At 34 °C, there was increased oxygen consumption by aerobic respiration and decreased dissolved oxygen content in the mushroom bags resulting in anoxia. Anaerobic respiration increased with higher anaerobic respiratory enzyme activities for alcohol dehydrogenase (ADH), pyruvate decarboxylase (PDC), and lactate dehydrogenase (LDH), increased. The anaerobic respiration metabolites of ethanol, acetaldehyde and lactate accumulated, which have an inhibitory effect on mushroom mycelia growth. In conclusion, inadequate oxygen supply in the bags to the mycelia was correlated to spawn-burning at high growth temperature. Pleurotus eryngii, the second largest industrial cultivation mushroom in China, is usually cultivated on substrates mainly consisting of sawdust and corncob. In this study, experiments were performed to determine the effects of different carbon sources and C/N values on nonvolatile taste components of P. eryngii. The effects of different carbon sources on nonvolatile taste components levels revealed that sawdust was beneficial to high levels of crude protein, amino acids, 5′-nucleotides and equivalent umami concentration, while corncob was beneficial to high contents of carbohydrate, polysaccharides and trehalose. At the similar C/N values, relatively higher sawdust content was beneficial to umami amino acid production, while relatively higher corncob content was beneficial to high contents of carbohydrate, polysaccharides and mannitol. Higher C/N value was beneficial to high levels of crude protein, amino acids, 5′-nucleotides and equivalent umami concentration, while lower C/N value was beneficial to high contents of carbohydrate, polysaccharides and trehalose. Pleurotus eryngii (DC. Ex. Fr.) Quél is a rare precious edible fungus which belongs to the family Pleurotaceae. This mushroom has highly nutritional, pharmaceutical, economic and ecological values. In the present study, combined randomly amplified polymorphic DNA (RAPD)/inter-simple sequence repeat (ISSR) was used to assess the genetic diversity of P. eryngii strains cultivated in China. For the RAPD and ISSR analyses, 404 and 392 polymorphic bands were obtained from 32 P. eryngii strains using 28 and 24 selected primers, respectively. A combined RAPD/ISSR dendrogram grouped the 32 strains into five clades with coefficient of 0.770. The comparison of RAPD and ISSR was also elucidated in the present study. The results of our study obtained by combined RAPD/ISSR analysis contributed to a better understanding of the genetic relationships among the P. eryngii strains and provide orientation for the strain improvement of P. eryngii species. Abstract: Experiments were carried out to test the viability of growing Pleurotus geesteranus on media containing varying amounts of crushed water hyacinth slices, which were soaked in pig farm biogas fluid and dried. The water hyacinth material was utilized to substitute sawdust in the media for mushroom cultivation. Mushroom fruiting bodies harvested were evaluated for yield, amino acid and heavy metal contents. Among the eight treatment groups, the greatest yield and highest amino acid content in the mushrooms were obtained when the proportions of water hyacinth and sawdust in the medium were equal. The concentrations of heavy metals, Hg, Pb and Cd, in most of the present mushroom samples did not exceed the maximum allowed levels according to the limits set forth by the food hygienic and safety regulations for edible mushrooms in China. The proposed waste utilization of water hyacinth could conceivably benefit the environment in various aspects including conservation of forest by reducing the demand on natural woods for mushroom production. Pleurotus eryngii is a valuable new edible mushroom variety cultivated on a large scale in China. The polysaccharides found in this mushroom are strong bioactive. This study used simulated digestion and fermentation model to study the digestion and fermentation characteristics of Pleurotus eryngii polysaccharide (PEP) and its effect on gut microbiota. The results showed that the molecular weight of PEP remained unchanged after simulated digestion, and the overall structure of PEP was not destroyed, indicating that PEP was not decomposed during digestion. However, during fermentation, PEP was degraded and utilized by intestinal flora to produce a variety of short-chain fatty acids (SCFAs), which reduced the pH value in fecal cultures. Meanwhile, PEP regulated the composition of intestinal flora, and the relative abundance of Firmicutes increased significantly. Pleurotus eryngii, a highly valued edible fungus, is one of the major commercially cultivated mushrooms in China. The development of P. eryngii, especially during the stage of primordium differentiation, is easily affected by light. However, the molecular mechanism underlying the response of primordium differentiation to light remains unknown. In the present study, primordium expression profiles under blue-light stimulation, red-light stimulation, and exposure to darkness were compared using high-throughput sequencing. A total of 16,321 differentially expressed genes (DEGs) were identified from three comparisons. GO enrichment analysis showed that a large number of DEGs were related to light stimulation and amino acid biosynthesis. KEGG analyses demonstrated that the MAPK signaling pathway, oxidative phosphorylation pathway, and RNA transport were most active during primordium differentiation. Furthermore, it was predicted that the blue-light photoreceptor WC-1 and Deoxyribodipyrimidine photolyase PHR play important roles in the primordium differentiation of P. eryngii. Taken together, the results of this study provide a speculative mechanism that light induces primordium differentiation and a foundation for further research on fruiting body development in P. eryngii. For decades, the edible mushroom Pleurotus eryngii (P. eryngii) has been cultivated as important raw materials for food and pharmaceutical industries in most of Asian countries, especially in China. Unfortunately, the generation and improvement of new cultivars are very difficult since there are many barriers which have not been solved thoroughly by gene editing tools, even though the CRISPR-Cas9 technique has been widely applied in other species. In this study, we identified the point-mutated variant of the endogenous sdhB gene (cbxr) as a more stable selection marker than hygromycin B resistance gene (hph) in P. eryngii. Furthermore, using a codon-optimized Cas9, a predicted native U6 promoter-guided sgRNA, as well as an optimized protoplast transformation system, a highly efficient pyrG gene editing system was established in P. eryngii , that incorporated varied insertions and deletions (indels) by non-homologous end joining (NHEJ) and homology-directed repair (HDR). Mushrooms accumulate arsenic (As), yet As concentrations, speciation, and localization in cultivated mushrooms across a large geographic distribution are unknown. We characterized 141 samples of nine species from markets in nine capital cities in China, with samples of Lentinula edodes, Pleurotus ostreatus, and Agaricus bisporus being analyzed for As speciation and localization. Total As concentrations ranged from 0.01 to 8.31 mg kg -1 dw, with A. bisporus (0.27-2.79 mg kg -1 ) containing the most As followed by P. ostreatus and L. edodes (0.04-8.31 and 0.12-2.58 mg kg -1 ). However, As in A. bisporus was mostly organic including nontoxic arsenobetaine, while P. ostreatus and L. edodes contained mainly inorganic As (iAs). On the basis of in situ imaging using LA-ICP-MS, As in L. edodes was localized to the surface coat of the cap, while As in P. ostreatus was localized to the junction of the pileus and stipe. When As speciation and daily mushroom consumption (1.37 g d -1 dw) are considered, daily mushroom consumption may result in elevated iAs exposure, with increased bladder and lung cancer rates up to 387 cases per 100000. Our study showed that market mushrooms could be a health risk to the general public so its production should be monitored.

Pesticide residue in mushrooms is less known. In this study, the risks of beta-cypermethrin, pyriproxyfen, avermectin, and diflubenzuron in oyster and shiitake mushrooms were evaluated using two different treatments: substrate mixture and surface spraying. Almost all the concentrations of these pesticides at day 90 were higher than 80% of the initial concentrations, while it was less than 45% for all cases within 35 days by spraying. For surface spraying, the residues of beta-cypermethrin were 0.0843-1.22 mg kg -1 in shiitake mushrooms and below 0.005 mg kg -1 in oyster mushrooms; the residues of pyriproxyfen, avermectin, and diflubenzuron were 0.122-4.84, 0.00501-0.111, and 0.0681-1.91 mg kg -1 , respectively. The residues of beta-cypermethrin, pyriproxyfen, and diflubenzuron in oyster mushrooms (in shiitake mushrooms) at interval of 0, 3, 5 days (1, 5, 7 days) were below their MRLs in China or Japan. The residue of avermectin in both mushrooms was lower than its limit of detection. These results provide information to safe and proper use of the pesticides in oyster and shiitake mushrooms.

Four Gram-negative-staining, facultatively anaerobic bacterial isolates were obtained from the fruiting bodies of the edible mushroom Pleurotus eryngii showing symptoms of bacterial blight disease in Beijing, China. Nearly complete 16S rRNA gene sequencing placed these isolates in the genus Pantoea. Multilocus sequence analysis based on the partial sequences of atpD, gyrB, infB and rpoB revealed Pantoea agglomerans as their closest phylogenetic relatives. DNA-DNA hybridization and phenotypic tests confirmed the classification of the new isolates as a novel species. The name Pantoea pleuroti sp. nov. [type strain KCTC 42084(T) = CGMCC 1.12894(T) = JZB 2120015(T)] is proposed.

This is the first study on the co-pyrolysis of spent substrate of Pleurotus ostreatus and coal tar pitch, and the activated carbon prepared from the pyrolytic char. Thermogravimetry (TG) analysis was carried out taking spent substrate, coal tar pitch and spent substrate-coal tar pitch mixture. The activation energies of pyrolysis reactions were obtained via the Flynn-Wall-Ozawa and Kissinger-Akahira-Sunose methods. The kinetic models were determined by the master-plots method. The activated carbons were characterised by N2-adsorption, Fourier transform infrared spectroscopy and X-ray diffraction. Experimental results demonstrated a synergistic effect happened during co-pyrolysis, which was characterised by a decreased maximum decomposition rate and an enhanced char yield. The average activation energies of the pyrolysis reactions of spent substrate, coal tar pitch and the mixture were 115.94, 72.92 and 94.38 kJ mol(-1) for the Flynn-Wall-Ozawa method, and 112.17, 65.62 and 89.91 kJ mol(-1) for the Kissinger-Akahira-Sunose method. The reaction model functions were f(α) = (1-α)(3.42), (1-α)(1.72) and (1-α)(3.07) for spent substrate, coal tar pitch and the mixture, respectively. The mixture char-derived activated carbon had a Brunauer-Emmett-Teller surface area up to 1337 m(2) g(-1) and a total pore volume of 0.680 cm(3) g(-1). Mixing spent substrate with coal tar pitch led to the creation of more micropores and a higher surface area compared with the single spent substrate and coal tar pitch char. Also, the mixture char-derived activated carbon had a higher proportion of aromatic stacking. Four Gram-negative-staining, facultatively anaerobic bacterial isolates were obtained from a fruiting body of the edible mushroom Pleurotus eryngii showing symptoms of soft rot disease in Beijing, China. Partial 16S rRNA gene sequencing, together with partial rpoB sequencing, placed these isolates in the genus Pantoea. Multilocus sequence analysis based on the partial sequences of gyrB, rpoB, infB and atpD revealed Pantoea dispersa and Pantoea gaviniae as their closest phylogenetic relatives and indicated that these isolates constituted a possible novel species. DNA-DNA hybridization studies confirmed the classification of the new isolates as a novel species and phenotypic tests allowed for differentiation from the closest phylogenetic neighbours. The name Pantoea beijingensis sp. nov. [type strain LMG 27579(T) = KCTC 32406(T) = JZB2120001(T) (deposited at Institute of Plant and Environment Protection, Beijing Academy of Agriculture and Forestry Sciences)] is proposed.
