Apple (Malus Mill.) is one of the most important fruits in China, and it boasts the world's largest cultivation area and yield. It needs to be grafted onto rootstocks to maintain a variety of characteristics. China has many apple rootstock resources that exhibit high resistance and strong adaptability; for these reasons, they are highly suited to China's complex and diverse natural environment. In China, apple rootstock breeding began in the 1970s, and now, several rootstocks, such as the 'GM256' and 'SH' series, are widely used. However, domestic rootstock resources and varieties are difficult to root. This affects the selection, utilization, and promotion of apple rootstocks. Cutting is an important method of rooting for apple rootstocks. This study discusses the main factors that affect rooting in rootstock cutting propagation; it also summarizes the rooting ability of different apple rootstocks and presents analyses of the demand for rootstocks in the major areas of apple production in China. We present the apple rootstock resources that are suitable for the soil and climate conditions of this production. We also call for research on the cutting roots of these specific apple rootstock resources to be expanded and strengthened. In this study, we used genotyping to determine the genetic structure and molecular identities of apple landraces from six species of Malus Mill. in China, based on the fingerprints revealed by microsatellite markers with tailed primer M13. A total of 46 apple landraces of Malus Mill. selected from the National Apple and Pear Germplasm Repository in Xingcheng, China were genotyped with 14 SSR markers. The primers differentiated all the accessions. At least three SSR primers, CH04h02, CH01f07a and CH04g07, with a higher heterozygosity and Shannon's information index than other combinations can distinguish all the accessions. All the alleles for these three primers were arranged in descending order, and they were assigned values beginning with 01. Character strings were constituted by combining all the codes of the three primers for every accession. By such means, separate and special molecular identities were obtained for every apple landrace, which could be expressed in the form of a bar code. Using such bar codes, trees can be labeled and scanned, which aids in the identification and tracking of genebank collections. The eight-step method for establishing the molecular identities of apple landraces reported here may serve as a reference when determining the molecular identities establishment of other apple germplasms of Malus Mill. This method might also be used for the establishment of a molecular database to aid the preservation of Malus Mill., which is in imminent danger in China. Background and objective: Nationally representative reports on the characteristics and long‐term survival of pulmonary arterial hypertension (PAH) from developing countries are scarce. The applicability of the current main risk stratifications and the longitudinal changes in goal‐oriented treatments have yet to be elucidated in real‐world settings. Therefore, we aimed to provide insights into the characteristics, goal‐oriented treatments and survival of PAH in China and to explore the applicability of the main risk stratifications in our independent cohort. Methods: PAH patients were consecutively enrolled from a national prospective multicentre registry. Data on baseline, follow‐up re‐evaluation and therapeutic changes were collected. Results: A total of 2031 patients were enrolled, with congenital heart disease (CHD)‐PAH (45.2%) being the most common aetiology. The mean age was 35 ± 12 years, and 76.2% were females. At baseline, approximately 20% of the patients with intermediate or high risk received combination treatment. At follow‐up, approximately half of the re‐evaluated patients did not achieve low‐risk profiles, and even among patients who received combination therapy at baseline, 4% of them still worsened. The rate of combination therapy increased significantly from 6.7% before 2015 to 35.5% thereafter. The main risk assessment tools demonstrated good performance for predicting survival both at baseline and at follow‐up. Conclusion: Chinese PAH patients show both similar and distinct features compared to other countries. Current main risk stratifications can significantly discriminate patients at different risk levels. There were still many patients not achieving low‐risk profiles at follow‐up, indicating more aggressive treatment should be implemented to optimize the goal‐oriented treatment strategy. Rapid urbanization and economic development in coastal regions have significantly increased coastal nutrient pollution and remarkably changed the phytoplankton community and developed some species into bloom, resulting in large economic losses and serious threats to public health. Therefore, it is indispensable to reveal the shift in the phytoplankton community and phytoplankton abundance, and phytoplankton's environmental drivers. However, previous studies could not present the details of the environmental drivers of phytoplankton due to samples being collected with low temporal resolution. Here, high-temporal-resolution (daily) samples were collected to investigate the influence of environmental factors on phytoplankton in Qinhuangdao for 44 days. Phytoplankton communities showed a rapid succession, with predominant genera changing in the order Skeletonema–Chaetoceros–Skeletonema–Thalassiosira. Similarly, Thalassiosira pacifica, Skeletonema costatum, Chaetoceros tortissimus, and Chattonella marina were identified as the dominant species and were abundant in 0–1.27 × 107 cells·L−1, 0–9.34 × 106 cells·L−1, 0–6.49 × 106 cells·L−1, and 0–3.64 × 106 cells·L−1, respectively. Moreover, inflows facilitate the rapid succession of the phytoplankton community. Dissolved inorganic phosphorus (DIP) was found to remarkably influence the succession of phytoplankton communities and the bloom of the top three dominant species, i.e., Thalassiosira pacifica, Skeletonema costatum, and Chaetoceros tortissimus. Overall, our results provide high-temporal-resolution observations of phytoplankton community succession and reveal its environmental drivers. Lonicera ruprechtiana Regel is widely used as a greening tree in China and also displays excellent pharmacological activities. The phylogenetic relationship between L. ruprechtiana and other members of Caprifoliaceae remains unclear. In this study, the complete cp genome of L. ruprechtiana was identified using high-throughput Illumina pair-end sequencing data. The circular cp genome was 154,611 bp long and has a large single-copy region of 88,182 bp and a small single-copy region of 18,713 bp, with the two parts separated by two inverted repeat (IR) regions (23,858 bp each). A total of 131 genes were annotated, including 8 ribosomal RNAs, 39 transfer RNAs, and 84 protein-coding genes (PCGs). In addition, 49 repeat sequences and 55 simple sequence repeat loci of 18 types were also detected. Codon usage analysis demonstrated that the Leu codon is preferential for the A/U ending. Maximum-likelihood phylogenetic analysis using 22 Caprifoliaceae species revealed that L. ruprechtiana was closely related to Lonicera insularis. Comparison of IR regions revealed that the cp genome of L. ruprechtiana was largely conserved with that of congeneric species. Moreover, synonymous (Ks) and non-synonymous (Ka) substitution rate analysis showed that most genes were under purifying selection pressure; ycf3, and some genes associated with subunits of NADH dehydrogenase, subunits of the cytochrome b/f complex, and subunits of the photosystem had been subjected to strong purifying selection pressure (Ka/Ks < 0.1). Background: At present, there is no generally accepted comprehensive prognostic risk prediction model for medically treated chronic thromboembolic pulmonary hypertension (CTEPH) patients.Methods: Consecutive medically treated CTEPH patients were enrolled in a national multicenter prospective registry study from August 2009 to July 2018. A multivariable Cox proportional hazards model was utilized to derive the prognostic model, and a simplified risk score was created thereafter. Model performance was evaluated in terms of discrimination and calibration, and compared to the Swedish/COMPERA risk stratification method. Internal and external validation were conducted to validate the model performance.Results: A total of 432 patients were enrolled. During a median follow-up time of 38.73 months (IQR: 20.79, 66.10), 94 patients (21.8%) died. The 1-, 3-, and 5-year survival estimates were 95.5%, 83.7%, and 70.9%, respectively. The final model included the following variables: the Swedish/COMPERA risk stratum (low-, intermediate- or high-risk stratum), pulmonary vascular resistance (PVR, ≤ or > 1600 dyn·s/cm5), total bilirubin (TBIL, ≤ or > 38 µmol/L) and chronic kidney disease (CKD, no or yes). Compared with the Swedish/COMPERA risk stratification method alone, both the derived model [C-index: 0.715; net reclassification improvement (NRI): 0.300; integrated discriminatory index (IDI): 0.095] and the risk score (C-index: 0.713; NRI: 0.300; IDI: 0.093) showed improved discriminatory power. This is the first multicentre prospective registry reporting baseline characteristics and estimated survival of patients with chronic thromboembolic pulmonary hypertension (CTEPH) in China. A total of 593 patients with CTEPH were included. The long‐term survival rates are similar to those of patients in the international and Spanish registries. Methods: Patients diagnosed with CTEPH were included in the registry that was set up in 18 centres from August 2009 to July 2018. The characteristics and survival of patients with CTEPH receiving the different treatments were reported. Prognostic factors were evaluated by Cox regression model. Results: A total of 593 patients with CTEPH were included. Eighty‐one patients were treated with PEA, 61 with BPA and 451 with drugs. The estimated survival rates at 1, 3, 5 and 8 years were, respectively, 95.2%, 84.6%, 73.4% and 66.6% in all patients; 92.6%, 89.6%, 87.5% and 80.2% in surgical patients; and 95.4%, 88.3%, 71.0% and 64.1% in medically treated patients. The estimated survival rates at 1, 3, 5 and 7 years in patients treated with BPA were 96.7%, 88.1%, 70.0% and 70.0%, respectively. For all patients, PEA was an independent predictor of survival. Other independent risk factors were CHD, cardiac index, PVR, big endothelin‐1, APE and 6MWD. Gramineae and Leguminosae forages are widely grown in the semiarid region of northwest China. To understand the changes in root morphology in forage seedlings under drought stress, a nursery experiment was conducted with and without water stress using four Gramineae species (Bromus inermis, Agropyron mongolicum, Lolium perenne, and Bothriochloa ischaemum) and four Leguminosae species (Lespedeza bicolor, Medicago sativa, Astragalus adsurgens and Melilotus suaveolens). The forages were grown in rhizoboxes under drought stress (15% polyethylene glycol stress) and well‐watered (85% of field capacity) conditions. The results showed that the root length, root area, and specific root length of the Gramineae species were higher than those of the Leguminosae species at the 30–60 cm soil depth. The fine roots of eight species in the 0~0.5 mm diameter class had the largest total root length and total root area, and the total root volume and root tips in the 0.5~1.0 mm diameter class were the highest. Based on the subordinate function method results, B. inermis had the highest comprehensive score, while M. suaveolens had the lowest. Our results implied that Gramineae seedlings distributed more roots than Leguminosae seedlings in the 30–60 cm soil layer under drought stress. Under drought stress, fine roots in the 0~0.5 mm and 0.5~1.0 mm diameter classes were the main organs sustaining the growth of the eight species when suffered drought stress. In this study, we investigated the environmental impacts of scallop culture on two coastal estuaries adjacent the Bohai Sea including developing a quantitative PCR assay to assess the abundance of the bacterial pathogens Escherichia coli and Vibrio parahaemolyticus . Scallop culture resulted in a significant reduction of nitrogen, Chlorophyll a , and phosphorous levels in seawater during summer. The abundance of bacteria including V. parahaemolyticus varied significantly across estuaries and breeding seasons and was influenced by nitrate as well as nutrient ratios (Si/DIN, N/P). Bacterioplankton diversity varied across the two estuaries and seasons, and was dominated by Proteobacteria , Cyanobacteria , Actinobacteria , Bacteroidetes . Overall, this study suggests a significant influence of scallop culture on the ecology of adjacent estuaries and offers a sensitive tool for monitoring scallop contamination. [ABSTRACT FROM AUTHOR]

A total of ten marine yeast strains isolated from Bohai Sea, Northern China were identified to be members of three genera Rhodosporidium, Rhodotorula, and Cryptococcus. Two representative strains Rhodosporidium TJUWZ4 and Cryptococcus TJUWZA11 with high lipid content based on Nile red staining method were further characterized. A wide range of culture conditions (C and N sources, pH, temperature, salinity and C/N ratio) were tested to characterize the biomass and lipid production (yield and productivity) of these strains. Results indicated that Rhodosporidium TJUWZ4 was capable of achieving lipid yield up to 44% and 0.09 g/l-h productivity on glucose and peptone medium at pH 4, 20 °C, 30% salinity, and C/N 80. Three fatty acids, namely oleic acid (18:1), palmitic acid (C16:0) and linoleic acid (18:2) were the major intracellular fatty acids, which accounted for 90% of total lipids. With promising features for intracellular lipid accumulation, Rhodosporidium TJUWZ4 is a robust strain with great potentials for application in biodiesel production from renewable feedstocks. Thraustochytrids are ubiquitous marine osmo-heterotrophic fungi-like microorganisms with only about 40 identified species till now. In this study, a total of 60 thraustochytrid strains were isolated from marine coastal habitats. Analysis of 18S rRNA gene sequences revealed that they belonged to three genera, i.e., Schizochytrium, Aurantiochytrium, and Thraustochytrium. All of the isolates were found to show considerable cellulolytic and lipolytic activities. Strains of Aurantiochytrium sp. and Thraustochytrium sp. were found to produce the highest levels of extracellular polysaccharides (EPS), which reached 345 μg ml in the growth media. Fourier transform infrared (FTIR) spectra of the EPS samples derived from two thraustochytrids (PKU#Sed1 and #SW1) displayed peaks for carbohydrates, proteins, lipids, uronic acids, and nucleic acids. Fatty acid profiles of four thraustochytrids comprised of palmitic acid (C16:0) and docosahexaenoic acid (DHA) as their major constituents. Schizochytrium sp. demonstrated the highest DHA production at 44 % of total fatty acids (TFA) with biomass and DHA yield of 7.1 and 1.6 g l, respectively, on the fourth day of growth. All the four isolates exhibited considerable production of palmitic acid (16:0) in their fatty acid profiles ranging from 35 to 50 % TFA. Viburnum sargentii Koehne is widely used for garden greening and also displays excellent medicinal value in China. However, the phylogenetic relationship between V. sargentii and other Adoxaceae members remains unknown. In this research, the complete chloroplast genome of V. sargentii was obtained by the high-throughput Illumina pair-end sequencing data. The chloroplast genome shows a typically quadripartite structure with 158,524 bp in size, including a large single-copy (LSC) region of 87,087 bp and a small single-copy (SSC) region of 18,489 bp, which were separated by two inverted repeat (IR) regions (26,474 bp each). A total of 128 genes were predicted, including 8 ribosomal RNAs (rRNAs), 37 transfer RNAs (tRNAs), and 83 protein-coding genes (PCGs). Maximum-likelihood phylogenetic analysis revealed that V. sargentii was clustered in the Viburnum genus and in a sister position to Viburnum japonicum, Viburnum erosum, Viburnum fordiae, and Viburnum betulifolium. Viburnum burejaeticum Regel et Herd is widely cultivated in botanical gardens. However, as a member of Adoxaceae, few studies have been carried out on its phylogenetic relationship with other family members. Here we report the first complete chloroplast (cp) genome sequence of V. burejaeticum collected from China. The circular cp genome is 158,381 bp in size, including a large single-copy (LSC) region of 87,067 bp and a small single-copy (SSC) region of 18,212 bp, which were separated by two inverted repeat (IR) regions (26,551 bp each). A total of 126 genes were annotated, including 8 ribosomal RNAs (rRNAs), 36 transfer RNAs (tRNAs), and 82 protein-coding genes (PCGs). The sequence comparison of two V. burejaeticum collected from Korea and China revealed 101 single nucleotide polymorphisms (SNPs) and 16 insertions/deletions (InDels). In addition, maximum-likelihood (ML) phylogenetic analysis indicated V. burejaeticum species collected in Korea and China are clustered together. • Highter genetic diversity was observed in Malus landraces and genetic differentiation of them in China mainly occurred within their respective populations. • Population structure and PCoA results showed that Malus domestica subsp. chinensis, Malus asiatica and Malus prunifolia may be derived from similar genetic sources. • The 216 germplasms were divided into five groups based on Nei's genetic distance, and the cluster analysis results were correlated with the geographical distribution of the accessions to some degree. • BIO4 (Temperature seasonality) was the greatest factor influencing the distribution of Malus landraces, and the suitable habitat area of Malus landraces under the current climate situation accounted for 33.40 % of the total area of China. • With climate change, the distribution of Malus landraces showed a tendency to move to higher latitudes and the total potential distribution area was projected to grow. China is the largest center of genetic diversity of apple (Malus Mill.), with rich germplasm resources. In the process of evolution and domestication, a large number of apple landraces appeared, and they played a very important role before the introduction of modern cultivated apple varieties to China. However, the habitat of Malus landraces is under serious threat. In this study, 19 pairs of polymorphic simple sequence repeat primers were used to analyze the genetic structure and genetic relationships of Malus landraces and the potential distribution was predicted according to eight environmental variables using MaxEnt modeling. The results showed that the genetic variation of Malus landraces was high (Ho = 0.728 and He = 0.884), and the genetic differentiation was mainly distributed within respective populations and with almost no differentiation among populations. The five population groups were ranked, from highest genetic diversity level to lowest, as follows: Malus domestica subsp. c hinensis > Malus prunifolia > Malus robusta > Malus asiatica > Malus micromalus. The 216 germplasms were divided into five groups based on Nei's genetic distance, whereas the Bayesian structure analysis categorized them into two groups. Population structure and principal coordinate analyses showed that Malus domestica subsp. chinensis, Malus asiatica and Malus prunifolia may be derived from similar genetic sources. In the modeling analysis, the cumulative contribution of temperature to the distribution of Malus landraces in suitable growth areas reached 66.1 %, and BIO4 (temperature seasonality) was identified as the major factor influencing the distribution of Malus landraces. Under four future emission scenarios, the potential distribution range of Malus landraces basically unchanged, but the total potential distribution area is projected to grow greatly over time and will tend to shift to higher latitudes. Objective: Information about epidemiology on burns is rare in China. The aim of this article is to describe the pattern of burns in East China during a 10-year time period.Method: A retrospective data analysis was performed on all hospitalized patients to the burn center at the Changhai hospital, one of major burn centers in East China, from 2005 to 2014.Results: We included 3376 patients in this study. Among them, 48.1% were from 27 provinces out of Shanghai and nearly 90% were from East China. August saw the most admissions and November saw the fewest. Spring and summer separately dominated in number of female and male patients. Children aged 2-5 and working-age adult were the most commonly treated. Home was the commonest place of injury, followed by industrial-related places, outdoors, public buildings, and vehicles or roads. Scalds remained the primary reason, followed by fire, contact burns, electricity, and chemicals. The average %TBSA of male patients was 14.2±21.3, significantly different from that of female patients (10.4±16.9). Extremities were the most vulnerable body region burned, followed by the trunk, face and hands. The average hospital length of stay in male patients was 25.4±72.4 days, significantly different from that of females' 19.9±27.6 days. The total mortality was 1.8% and the lethal area burned resulting in 50% mortality was 96.5% TBSA.Conclusion: Compared with published data, these result are encouraging, which demonstrate that burn care and treatment has made significant progress. Abstract: This study describes identification, cultivation, monitoring of carbohydrate accumulation and bioethanol production from microalgal strains from the coastal waters of Pearl River Delta. Eighteen identified strains belong to the families Chlorellaceae, Scotiellocystoidaceae, Neochloridaceae, Selenastraceae and Scenedesmaceae. Of isolated strains Mychonastes afer PKUAC 9 and Scenedesmus abundans PKUAC 12 were selected for further biomass and ethanol production analysis. Comparison of three cultivation modes (stationary, shaken and aerated) resulted in the highest biomass productivity obtained for aerated cultures that yielded 0.09g and 0.11g dry weight per day per litre of medium for M. afer PKUAC 9 and S. abundans PKUAC 12, respectively. Carbohydrate accumulation monitored by FTIR showed that early stationary phase is optimal for biomass harvest. Microalgal biomass was successfully used as a carbohydrate feedstock for fermentative bioethanol production. S. abundans PKUAC 12 was superior feedstock for bioethanol production when pre-treated with the combination of dilute acid treatment and cellulase. [Copyright &y& Elsevier]

Abstract: This study is aimed to systematically estimate marginal land resources with different grades (total area; land with certain eco-environmental-economic feasibility; centralized reserve land) in China, and evaluate potential energy production from microalgae on marginal lands in the long-, mid- and near-term, based on a model. The annual potential energy production from algae in total marginal land of China (APEMC) was estimated to 4.19 billion standard coal equivalent (tce), far more than total annual energy consumption equivalent in China (TECCE) in 2007. For microalgae with 35% lipid content, the APEMC in the mid-term would be 37.6–65.8% of the TECCE in 2007. The corresponding annual CO2 emission mitigation by replacement of fossil fuels by algal bioenergy would be 4.27–7.44billiont. Although Southwest China provides the highest potential algae production in the long-term, Northwest China provides the highest value in the near-term. [Copyright &y& Elsevier]

• High squalene-producing thraustochytrid strains from coastal waters were identified. • Newly isolated strain Aurantiochytrium sp. Newly-isolated thraustochytrid strains from coastal waters of China were characterized as bioresource of squalene and the culture condition for the top producer was systematically optimized. Phylogenetic analysis revealed that eight squalene-producing isolates were closely related to genus Aurantiochytrium and one to genus Labyrinthula. The top producer, Aurantiochytrium sp. TWZ-97, produced squalene up to 188.6 mg/L at 28 °C in a 5-L bioreactor containing optimal medium (glucose: 40 g/L, monosodium glutamate: 3 g/L, yeast extract: 25 g/L, and NaCl: 6 g/L), which was 6-fold higher than that under unoptimized condition. Transcriptome analysis revealed for the first time the presence of seven key genes of mevalonate pathway for squalene biosynthesis in strain TWZ-97. Medium optimization yielded a 2.23-fold higher expression of the squalene synthase gene under optimal condition compared to unoptimized. This study provides a potential thraustochytrid strain TWZ-97 as bioresource of squalene and uncovers novel information about its squalene biosynthesis pathway for future strain improvement. Pathogens in coastal waters cause infectious diseases and endanger public sanitation safety in humans and animals worldwide. To avoid these risks, timely detection of human-associated pathogens in waters is crucial. In this study, the decay kinetics of the molecular markers for human-associated pathogens, including enteric bacteria (Escherichia coli, Enterococcus, and Bacteroides), non-enteric bacteria (Staphylococcus aureus), crAssphage, and polyomavirus, were monitored over time at different temperatures and background microbes in seawater microcosms. The results indicated that temperature and native marine microbes were the main influential factors in attenuating bacterial pathogens. Remarkably, the effect of native microorganisms was more evidentially striking. Furthermore, Enterococcus was a more reliable and suitable fecal indicator bacterium than E. coli for the marine environment. The decay of crAssphage was like that of polyomavirus, indicating that it may be a good indicator of enterovirus in seawater. More importantly, the 16S amplicon sequencing data highlighted the decay kinetics of multiple bacterial pathogens in parallel with the dynamic changes of the whole bacterial communities. Marine suspended particles are unique micro-habitats for diverse microbes and also hotspots of microbially metabolic activities. However, the association of bacterial pathogens, especially those carrying antibiotic resistance genes (ARGs), with these particles remain largely unknown in coastal habitats. This study investigated the distribution of pathogen-related bacteria and ARGs in particle-associated (PA) and free-living (FL) fractions of samples collected at three coastal beaches using NextGen sequencing and qPCR. Suspended particles were found to harbor significantly higher abundances of bacteria of pathogen-related genera and ARGs than their counterparts. Functional analysis of microbial community suggested that antibiotic biosynthetic pathways were also more abundant among PA microbiome comparing to FL microbial community, which further facilitated the spread of ARGs. Additionally, 13 pathogen-related genera co-occurred with ARG in PA fraction while only 2 pathogen-related genera co-occurred with ARGs in FL fraction. Overall, our research revealed suspended particles harbored more abundant pathogen-related genera and ARGs comparing with surrounding waters. Two Gram-stain-negative, short rod-shaped and non-flagellated strains, designated 17-4A T and L52-1-41, were isolated from the surface seawater of the Indian Ocean and South China Sea, respectively. The 16S rRNA genes of the two strains shared sequence similarity of 99.45 %. Strain 17-4A T shared the highest 16S rRNA gene similarity of 98.02 % with Pusillimonas caeni EBR-8-1 T , followed by Pusillimonas noertemannii BN9 T (97.47 %), Pusillimonas soli MJ07 T (96.93 %), Parapusillimonas granuli Ch07 T (96.68 %), Pusillimonas ginsengisoli DCY25 T (96.65 %), Eoetvoesia caeni PB3-7B T (96.63 %), Paracandidimonas caeni 24 T (96.34 %), Castellaniella defragrans 54Pin T (96.28 %) and Pusillimonas harenae B201 T (96.05 %). L52-1-41 shared the highest 16S rRNA gene similarity of 97.74 % with Pusillimonas caeni EBR-8-1 T , followed by Pusillimonas noertemannii BN9 T (97.47 %), Pusillimonas soli MJ07 T (96.65 %), Parapusillimonas granuli Ch07 T (96.41 %), Pusillimonas ginsengisoli DCY25 T (96.37 %), Eoetvoesia caeni PB3-7B T (96.35 %), Pusillimonas harenae B201 T (96.28 %), and Paracandidimonas caeni 24 T (96.06 %). The results of phylogenetic analyses indicated that 17-4A T and L52-1-41 formed a stable, distinct and highly supported lineage affiliated to the genus Pusillimonas . The results of the digital DNA-DNA hybridization (dDDH) and average nucleotide identity (ANI) analyses indicated that they represented a single species. They featured similar genomic DNA G+C contents of 53.2-53.4 mol%. Activities of catalase and oxidase were negative for both strains. The fatty acids patterns of 17-4A T and L52-1-41 were most similar, mostly comprised of C 16 : 0 , C 17 : 0 cyclo, C 18 : 0 , C 18 : 1 ω9 c and summed feature 8 (C 18 : 1 ω7 c and/or C 18 : 1   ω6 c ). The major polar lipids of the two strains were phosphatidylethanolamine, phosphatidylglycerol, diphosphatidylglycerol and unidentified aminolipids. The respiratory quinone of the two strains was Q-8. Hence, on the basis of the phenotypic, chemotaxonomic and genotypic data presented in this study, we proposed the classification of both strains as representatives of a novel species named Pusillimonas maritima sp. nov., with the type strain 17-4A T (=MCCC 1A12670 T =KCTC 62121 T =NBRC 113794 T ), and another strain L52-1-41 (=MCCC 1A05046=KCTC 52313).

Fungal communities in the coastal waters have long been known to be dynamic with a significant role in organic matter cycling. However, the effects of environmental gradients on their community structures are poorly described. Here we studied three coastal sites off the South China Sea, namely Pearl River Estuary (PE), Shenzhen Bay (SB), and Daya Bay (DB) with contrasting trophic status and heterogenous local influences. Environmental analysis of these sites suggested higher nutrient and low salinity levels at PE and SB with wide variability compared to DB. Average molecular abundances (18S rRNA gene copy numbers) at sites PE (1.05 ± 0.27 × 10 7 copies L -1 ) and SB (1.2 ± 0.69 × 10 7 copies L -1 ) were similar and significantly higher (P < 0.05) than that at site DB (5.5 ± 9.5 × 10 5 copies L -1 ). Although planktonic fungi were molecularly abundant at the three sites, live fungal biomass based on ergosterol assay was detected only at some stations of PE and SB. Both molecular abundance and live biomass were significantly correlated with chemical oxygen demand, nutrients, and phytoplankton biomass, supporting their role in detritus turnover. The fungal communities were unprecedently diverse with the ubiquitous dominance of Dikarya and the occasional predominance of Glomeromycota, Mucoromycota, Mortierellomycota, and Chytridiomycota. A total of 24 classes, 46 orders, 71 families, 59 genera, and eight species were classified within the eight detected phyla, including the new finding of ascomycetous class Geoglossomycetes in coastal waters. Salinity and nitrate were the significant (r 2  = 0.70, P < 0.05) factors that determined the β-diversity of fungal communities. Overall, this study suggests that although planktonic fungi are ubiquitous in coastal habitats, their molecular abundances and diversities (both α and β) are significantly determined by environmental gradients, particularly the salinity, COD and nitrate levels of coastal waters., (Copyright © 2019 Elsevier B.V. Labyrinthulomycete protists have gained significant attention in the recent past for their biotechnological importance. Yet, their lipid profiles are poorly described because only a few large-scale isolation attempts have been made so far. Here, we isolated more than 200 strains from mangrove habitats of China and characterized the molecular phylogeny and lipid accumulation potential of 71 strains. These strains were the closest relatives of six genera namely Aurantiochytrium , Botryochytrium , Parietichytrium , Schizochytrium , Thraustochytrium , and Labyrinthula . Docosahexaenoic acid (DHA) production of the top 15 strains ranged from 0.23 g/L to 1.14 g/L. Two labyrinthulid strains, GXBH-107 and GXBH-215, exhibited unprecedented high DHA production potential with content >10% of biomass. Among all strains, ZJWZ-7, identified as an Aurantiochytrium strain, exhibited the highest DHA production. Further optimization of culture conditions for strain ZJWZ-7 showed improved lipid production (1.66 g/L DHA and 1.68 g/L saturated fatty acids (SFAs)) with glycerol-malic-acid, peptone-yeast-extract, initial pH 7, 28 °C, and rotation rate 150 rpm. Besides, nitrogen source, initial pH, temperature, and rotation rate had significant effects on the cell biomass, DHA, and SFAs production. This study provides the identification and characterization of nearly six dozen thraustochytrids and labyrinthulids with high potential for lipid accumulation.

With the escalating coastal development and loss of vegetated landscape, the volume of storm runoff increases significantly in Chinese coastal cities. To protect human health and valuable recreational resources, it is necessary to develop a quantitative understanding of coastal pollution. Here we studied the influence of storm runoff on the nutrients and microbial pathogens at two popular bathing beaches in northern China. Dongshan Beach, located near the mouth of an urban river, is influenced by non-point source pollution while Tiger-Rock Beach, a coastal beach, is primarily influenced by a point source from a storm drain outfall. Storm runoff significantly (P < 0.001) decreased the salinity and Chl a post-storm at both the beaches, but only reduced the concentration of dissolved inorganic N at Tiger-Rock Beach. Escherichia coli decreased by 68.7% at Dongshan Beach, possibly due to the dilution effect of the stormflow, contradicting the notion of elevated fecal contamination in coastal beaches from storm runoff. Vibrio parahaemolyticus increased at both beaches post-storm, by 155.7% at Dongshan Beach and 136.7% at Tiger-Rock Beach. Regardless of storm impact, both E. coli and V. parahaemolyticus were much higher at Dongshan Beach than that at Tiger-Rock, suggesting the influence of different surrounding topographies. Lastly, the statistical models developed based on the environmental and microbial parameters regression showed predictive power (adjusted R 2  > 0.5) to estimate the concentration of E. coli at Dongshan Beach and V. parahaemolyticus at Tiger-Rock Beach. Overall, the results suggest the unique role of the individual beaches in attenuating the effect of rainfall on the concentration of microbial pathogens in bathing water quality and provide unique predictive models for recreational water management and public health protection., (Copyright © 2019 Elsevier B.V. Thraustochytrids are unicellular fungi-like (heterotrophic) marine protists that have long been considered to play an important role in the biogeochemical cycles of the coastal oceans. However, the significance of their ecological functions and their diversity in marine ecosystems remain largely unknown. In this report, we examined the spatial and temporal variations of planktonic thraustochytrids, their relationship with other environmental factors, and their diversity in the subtropical coastal waters of China. The abundance of planktonic thraustochytrids ranged from 2.56 × 105 to 17.57 × 105 cells L-1 with highest abundance detected in polluted coastal water in the Spring (March) season. The thraustochytrid biomass was greater than the bacterial biomass in most seawater samples, ranging from 32.29 to 359.51% that of bacterioplankton. The abundance of thraustochytrids appeared to be largely related to that of bacterioplankton and to chemical oxygen demand in water columns. High-throughput sequencing analyses revealed a total of 105 OTUs (97% similarity), which were members of genera Thraustochytrium, Aplanochytrium, Oblongichytrium, Ulkenia, Labyrinthula and undescribed novel phylotypes. Results of this study indicated unprecedented high diversity of labyrinthulomycetes as well as the presence of novel labyrinthulomycete and thraustochytrid lineages, and also provided new information on the significant role of thraustochytrids in microbial food webs in a coastal marine ecosystem.

The South China Sea is one of the largest marginal seas, with relatively frequent passage of eddies and featuring distinct spatial variation in the western tropical Pacific Ocean. Here, we report a phylogenetic study of bacterial community structures in surface seawater of the northern South China Sea (nSCS). Samples collected from 31 sites across large environmental gradients were used to construct clone libraries and yielded 2,443 sequences grouped into 170 OTUs. Phylogenetic analysis revealed 23 bacterial classes with major components α-, β- and γ-Proteobacteria, as well as Cyanobacteria. At class and genus taxon levels, community structure of coastal waters was distinctively different from that of deep-sea waters and displayed a higher diversity index. Redundancy analyses revealed that bacterial community structures displayed a significant correlation with the water depth of individual sampling sites. Members of α-Proteobacteria were the principal component contributing to the differences of the clone libraries. Furthermore, the bacterial communities exhibited heterogeneity within zones of upwelling and anticyclonic eddies. Our results suggested that surface bacterial communities in nSCS had two-level patterns of spatial distribution structured by ecological types (coastal VS. oceanic zones) and mesoscale physical processes, and also provided evidence for bacterial phylogenetic phyla shaped by ecological preferences.

Mariculture is one of the major seafood supplies worldwide and has caused serious environmental concerns on the coastal zone. Its rapid development has been shown to disrupt the sediment ecosystems and thus influence the benthic bacterial communities. Bacterial diversity and community structure within both adjacent farms and non-cultured zones intertidal sediments along the coasts of Qinhuangdao and Dalian, China, were investigated using full-length 16S rRNA gene-based T-RFLP analyses and clone library construction. Richness and Shannon-Wiener index were significantly increased at sites adjacent the mariculture farm with mean values of 29 and 2.97 from peak profiles of T-RFLP result. Clustering analyses suggested that impacts of mariculture on bacterial diversity of sediment were significantly larger than those resulted from temporal and spatial scales. Upon comparisons of RFLP patterns from 602 clones from libraries of the selected five samples, 137 OTUs were retrieved. Members of γ- and δ-Proteobacteria, Bacilli, Flavobacteria, and Actinobacteria were recorded in all libraries. In addition, γ-Proteobacteria were dominant in all samples (21.7~45.0 %). Redundancy analysis revealed that the distribution of bacterial composition seemed to be determined by the variables of salinity, PO4 (3-)-P, NH4 (+)-N, and Chlorophyll a content. The phyla of γ-Proteobacteria, Clostridia, Flavobacteria, Bacilli, and Planctomycetes were principal components to contribute to the bacterial differences of clone libraries. Our finding demonstrated that these phyla could display variations of bacterial composition linked to environmental disturbance resulted from mariculture.

Objective: Cyclooxygenase-2 (COX-2), one of the rate-limiting enzymes in the metabolism of arachidonic acid which is reported to be involved in the pathogenesis of many human tumors. As well, Vascular endothelial growth factor (VEGF) is well known to be involved in the infiltration and metastasis of many kinds of cancers. The aim of this study was to further elucidate the clinicopathologic significance of the immunohistochemical expressions of COX-2 and VEGF in thyroid carcinoma., Methods: Eighty-five patients with thyroid neoplasms were enrolled in our study from December 2003 to January 2010 from the authors' institution retrospectively. Their tumors were examined in the Department of Pathology, the First Bethune Hospital of Jilin University. Immunohistochemistry was performed on paraffin-embedded tissues sections using monoclonal anti-human COX-2 and VEGF antibodies. The tissues were classified into four types: papillary, follicular, medullary and undifferentiated. The patients ranged in age from 23 to 71 years. Breast cancer slides acted as control slides. The immunohistochemical stains were quantified by staining intensity and by the proportion of positively stained cells which were stained brown or yellow., Results: The results were analysed by χ(2) test. COX-2 and VEGF expressions were stronger in thyroid carcinoma than in thyroid adenomas and normal tissues (P<0.01). COX-2 and VEGF expressions in thyroid carcinoma correlated with the tumor type and TNM stage., Conclusion: Our results suggest that expression of COX-2 and VEGF may promote angiogenesis of thyroid carcinoma, its infiltration, and metastasis.
