Amplicon sequencing of functional genes is a powerful technique to explore the diversity and abundance of microbes involved in biogeochemical processes. One such key process, denitrification, is of particular importance because it can transform nitrate (NO3-) to N2 gas that is released to the atmosphere. In nitrogen limited alpine wetlands, assessing bacterial denitrification under the stress of wetland desertification is fundamental to understand nutrients, especially nitrogen cycling in alpine wetlands, and thus imperative for the maintenance of healthy alpine wetland ecosystems. We applied amplicon sequencing of the nirS gene to analyze the response of denitrifying bacterial community to alpine wetland desertification in Zoige, China. Raw reads were processed for quality, translated with frameshift correction, and a total of 95,316 nirS gene sequences were used for rarefaction analysis, and 1011 OTUs were detected and used in downstream analysis. Compared to the pristine swamp soil, edaphic parameters including water content, organic carbon, total nitrogen, total phosphorous, available nitrogen, available phosphorous and potential denitrification rate were significantly decreased in the moderately degraded meadow soil and in severely degraded sandy soil. Diversity of the soil nirS-type denitrifying bacteria communities increased along the Zoige wetland desertification, and Proteobacteria and Chloroflexi were the dominant denitrifying bacterial species. Genus Cupriavidus (formerly Wautersia), Azoarcus, Azospira, Thiothrix, and Rhizobiales were significantly (P <0.05) depleted along the wetland desertification succession. Soil available phosphorous was the key determinant of the composition of the nirS gene containing denitrifying bacterial communities. The proportion of depleted taxa increased along the desertification of the Zoige wetland, suggesting that wetland desertification created specific physicochemical conditions that decreased the microhabitats for bacterial denitrifiers and the denitrification related genetic diversity. The article discusses a study which analyzed the strength anisotropy of quartz mica schist along the exploration adit of the DanBa hydroelectric power in Sichuan Province, China using the point load test. It provides information on the quartz mica schist used in the study, including its mineral composition and texture. It also mentions two types of failure modes observed during the point load test.

A total of 54 rhizobial strains were isolated from faba bean root nodules in 21 counties of Sichuan hilly areas in China, and their symbiotic effectiveness, genetic diversity and phylogeny were assessed. Only six strains increased the shoot dry mass of the host plant significantly ( P ≤ 0.05). Based on the cluster analysis of combined 16S rDNA and intergenic spacer region (IGS) PCR-RFLP, the strains were divided into 31 genotypes in 11 groups, indicating a high degree of genetic diversity among the strains. The sequence analysis of three housekeeping genes ( atpD , glnII and recA ) and 16S rDNA indicated that the strains represented two R. leguminosarum , two Rhizobium spp., R. mesosinicum , Agrobacterium sp. and A. tumefaciens . The strains representing four Rhizobium species were divided into two distinct nodC and nifH genotypes. However, the phylogeny of housekeeping genes and symbiotic genes was not congruent, implying that the strains had been shaped by vertical evolution of the housekeeping genes and lateral evolution of the symbiotic genes. A novel bacterial strain, TRM F109, was isolated from hypersaline habitat in Sichuan Province, China. It was a Gram-positive, aerobic, non-motile, halotolerant, filamentous bacterium. Phylogenetic analysis based on 16S rRNA gene sequences revealed levels of similarity of 97.0-98.4 % to the type strains of recognized species of the genus Isoptericola. Chemotaxonomic data also supported the placement of strain TRM F109 within the genus Isoptericola. The low levels of DNA-DNA relatedness between the novel strain and the type strains of recognized species of the genus Isoptericola, in combination with differential phenotypic data, demonstrate that strain TRM F109 represents a novel species of the genus Isoptericola, for which the name Isoptericola salitolerans sp. nov. is proposed. Pteroceltis tatarinowii Maxim. (Ulmaceae), one of the most widespread temperate canopy trees in mainland China, is the sole representative of the genus Pteroceltis. To illuminate the biogeographic and demographic history of this temperate tree species, we carried out a survey of chloroplast DNA sequence variation ( trnS-trnG and psbA-trnH) within and among 28 populations (284 individuals in total) representing most of the distributional range of the species. Based on a total of 13 haplotypes identified, P. tatarinowii was found to harbor surprisingly high levels of haplotype and nucleotide diversity ( hT= 0.71; πT= 2.83 × 10−3), possibly associated with its long evolutionary history and wide-scale geographical distribution. Significant chloroplast DNA population subdivision was detected ( GST= 0.898; NST= 0.938), suggesting low levels of recurrent gene flow through seeds among populations and significant phylogeographical structure ( NST > GST, P < 0.05). The ancestral haplotypes show clear-cut geographical distribution, and most regions possess a unique set of haplotypes, suggesting multiple potential refugia of the species occurring in montane areas of South China. The haplotype mismatch distributions analysis indicates that populations from North China underwent a spatial northward expansion, which might reflect one of the repeated Pleistocene south-to-north shifts of temperate deciduous forest in North China following cold periods. The taxonomic status of Primula ranunculoides and other members of section Ranunuculoides is reappraised here based on data from morphological, reproductive and molecular characters. Multivariate analysis of morphological characters indicates that P. ranunculoides is a coherent species that can be distinguished from its sectional congeners P. cicutariifolia and P. merrilliana by the characters of simple kidney-shaped outer leaves and the unique clonal reproductive ability by which apices of the scape differentiate into bulblets at the late phases of flowering. Recognition of P. ranunculoides at the specific level is also supported by palynological characters, breeding system, cross pollination results and molecular phylogenetic analysis of nrDNA internal transcribed spacer sequences. Pteroceltis tatarinowii (Cannabaceae), a relic tree endemic to China, is mainly distributed in limestone mountains and has a wide geographical range. In this study, 12 microsatellite primer pairs were assayed to analyse the genetic pattern and gene flow among 461 individuals sampled from 23 wild populations of P. tatarinowii. A high level of genetic diversity was detected based on high values of total alleles (159), the number of alleles (NA = 6.373), expected heterozygosity (HE = 0.696) and observed heterozygosity (HO = 0.679). The high genetic diversity in this species may be attributed to its long‐life history, wide geographical distribution and wind dispersal. Only low genetic differentiation (GST = 0.137, FST = 0.138) was found among populations. Gene flow (migrants per generation, Nm) was estimated to be 1.56. This moderate level of gene flow possibly decrease interpopulation differentiation by buffering against genetic drift and improving gene exchange. However, spatial genetic structure was detected throughou the sampled range of the species (r = 0.311, p < 0.05) as well as in southern China (r = 0.453, p < 0.05), and may be related to terrain heterogeneity and the demographic history of P. tatarinowii. The east‐west high mountains of southern China might serve as physical barriers to seed and pollen flow. The isolation and local adaptation of different refugia may further limit gene flow. In addition, geographically remote populations might fail to effectively disperse pollen and seeds. Abstract: The perennial herb Primula cicutariifolia Pax is an endangered and endemic species with narrow distribution in eastern Anhui and Zhejiang provinces of China. In this study, the levels of genetic variation and the pattern of genetic structure in five natural populations of P. cicutariifolia were assessed by inter-simple sequence repeats (ISSR) and simple sequence repeat (SSR) markers. Both markers revealed that there was remarkably low genetic variation within populations (e.g., H e = 0.19 and 0.18, for ISSR and SSR respectively) and high differentiation among populations (G ST = 0.714 and 0.611; Φ ST = 0.698 and 0.599, for ISSR and SSR respectively). The level of population genetic diversity was correlated to population size only detected by ISSR markers. The genetic structure of P. cicutariifolia may be explained by limited gene flow that was caused by habitat fragmentation and limited seeds and pollen dispersal ability, self breeding system and biennial life form. To protect and avoid disappearance of P. cicutariifolia, much more attentions should be paid to protect all the remnant populations and their habitats, and three management units, i.e. Tianmushan, Damodao, and Panan units, were proposed. Abstract: Primula merrilliana Schltr. is an endangered and narrowly-distributed endemic species of southern Anhui Province in China. In this study, the level of genetic variation and the pattern of genetic structure in six natural populations of P. merrilliana were assessed by using ISSR (inter-simple sequence repeats) markers. Based on ten primers, 137 clear and reproducible DNA fragments were generated, of which 109 were polymorphic. The statistical results indicated that there was a relatively low genetic diversity within populations, and a high genetic differentiation among populations (G ST = 0.53, Φ ST = 0.49). The level of population genetic diversity was correlated to habitat type and the gene flow (N m) was low with only 0.45. The unexpected genetic structure of P. merrilliana may be explained by limited gene flow that was caused by habitat fragmentation and limited seeds and pollen dispersal ability, self-compatible breeding system and biennial life form. Due to complexity of operational control and difficulty of operational scheduling brought by the hybrid cascaded MTDC system and the lack of prior related research works, this paper establishes the mathematical model of the hybrid cascaded MTDC system and then proposes a system operational dispatching and scheduling strategy, which includes: 1) feasible coordinated operational control strategies investigation; 2) security operating range of the whole hybrid cascaded MTDC system analysis; 3) operating points of the hybrid cascaded MTDC system calculation. The aim of this study was to analyze the carbon sink effect under natural vegetation restoration and the influence of changes in vegetation community characteristics on ecosystem carbon density in ecologically fragile areas of the Loess Plateau. In this study, the changes in carbon sequestration of a vegetation-soil system under eight successional stages[slope cropland, abandoned cropland for 10 years, abandoned cropland for 20 years, Sophora davidii (Franch.) Skeels., Betula platyphylla Suk., Pinus tabulaeformis Carr., Quercus wutaishanic Mary+ P. tabulaeformis Carr mixed forests, and Q. wutaishanic Mary] in Ziwuling area over 150 restoration periods were investigated using the common method of spatial and temporal substitution. This study also discussed the relationship between changes in vegetation community characteristics and vegetation-soil system carbon density. The results showed that the community coverage of the investigated vegetation fluctuated from 85% in the slope cropland stage to 100% in the arbor stage. The number of species, Margalef index, Shannon-Wiener index, Pielou index, and Simpson index initially increased rapidly, then declined slowly until becoming stable, and reached a peak in the middle of the succession ( B. platyphylla Suk.). The biomass and carbon density of vegetation components (above-ground biomass, below-ground roots, and litter) increased exponentially during the succession, i.e., increased slowly before B. platyphylla Suk. but increased significantly in B. platyphylla Suk. and P. tabulaeformis Carr.( P <0.05). The biomass and carbon density reached the maximum values of 27858.08 g·m -2 and 13232.51 g·m -2 , respectively, in Q. wutaishanic Mary+ P. tabulaeformis Carr mixed forests and tended to be stable in the late succession stage. Soil organic carbon density showed a power function relationship with vegetation restoration, with the greatest increase in the stages of abandoned cropland for 10 years and B. platyphylla , but no significant changes in the subsequent stages ( P >0.05). In the early succession stage, the carbon density of the farmland ecosystem was the lowest (4395.70 g·m -2 ), whereas the other seven stages increased by 55.54%, 40.37%, 69.96%, 202.48%, 326.35%, 357.43%, and 351.07%, respectively, compared with the farmland ecosystem. Community coverage, Margalef index, Shannon-Wiener index, above-ground biomass, root biomass, and litter biomass were significantly positively correlated with vegetation-soil system carbon density ( P <0.05). The carbon sink effect of long-term natural restoration in Ziwuling Region was significant, and the carbon density of the vegetation-soil system under interspecific competition tended to be stable in the late succession stage. Dynamic changes in the vegetation community structure and plant diversity during the succession process increased vegetation carbon density and soil carbon density. This study helps to clarify the carbon sink effect of natural vegetation restoration in ecologically fragile areas of the Loess Plateau and provides a theoretical basis for promoting natural forest conservation and achieving carbon neutrality.

The proprietary Chinese medicine (PCM) has become a significant supplement of modern medicine. Nevertheless, the absence of quality control standard of compatible materials in PCM has led to serious adulteration, which has an extremely bad effect on safety of drug use and clinical efficacy. Here, a quality tracing evaluation strategy of compatible materials in 32 Aconitum proprietary Chinese medicines (APCMs) was established, including data normalization, model development, model verification, and unknown prescription cracking. The model was delimited based on the weighted content of total 9 key alkaloids in 24 APCMs, which were 5.65-57.10 μg/g for extract medicines and 42.62-380.61 μg/g for powder medicines. Three newly published commercial APCMs, including Wangbi Tablet, Wangbi Granule, and Fengshigutong Capsule, were used to verify its reliability and the results proved to be positive. Moreover, a novel prescription cracking approach was proposed to decode the content of each material in five unknown prescriptions including Yaoxitong Capsule, Tongrendahuoluo Pill, Xinbao Pill, Dahuoluo Capsule, and Mugua Pill. Ultimately, the single or two compatible Aconitum materials in APCMs was successfully decoded and the processed level of the materials were effectively judged. Coronavirus disease 2019 (COVID-19) is a novel and lethal infectious disease, posing a threat to global health security. The number of cases has increased rapidly, but no data concerning kidney transplant (KTx) recipients infected with COVID-19 are available. To present the epidemiological, clinical, and therapeutic characteristics of KTx recipients infected with COVID-19, we report on a case series of five patients who were confirmed as having COVID-19 through nucleic acid testing (NAT) from January 1, 2020 to February 28, 2020. The most common symptoms on admission to hospital were fever (five patients, 100%), cough (five patients, 100%), myalgia or fatigue (three patients, 60%), and sputum production (three patients, 60%); serum creatinine or urea nitrogen levels were slightly higher than those before symptom onset. Four patients received a reduced dose of maintenance immunosuppressive therapy during hospitalization. As of March 4, 2020 NAT was negative for COVID-19 in three patients twice in succession, and their computed tomography scans showed improved images. Although greater patient numbers and long-term follow-up data are needed, our series demonstrates that mild COVID-19 infection in KTx recipients can be managed using symptomatic support therapy combined with adjusted maintenance immunosuppressive therapy., (Copyright © 2020 European Association of Urology. To examine the effects of management measures on carbon and nitrogen contents, as well as their distribution and structural characteristics of different soil fractions in Moso bamboo plantations, we compared three types of the bamboo forests (undisturbed, extensively managed, and intensively managed) and the control secondary broadleaved evergreen forest using the methods of physical fractionation, chemical and biological analysis and Fourier-transform infrared spectroscopy (FTIR). The results showed that soil total organic carbon (TOC) and total nitrogen (TN) content, as well as free particulate organic carbon and nitrogen, soluble organic carbon and nitrogen (DOC, DON), and mineral-associated organic carbon and nitrogen in the undisturbed and extensively managed Moso bamboo plantations were significantly increased compared with that in the control. The distribution ratio of free particulate organic carbon and nitrogen in the undisturbed Moso bamboo plantation significantly increased, with mineral-associated organic carbon being the largest reservoir of soil organic carbon (67.6%). Intensive management resulted in the decrease of soil organic carbon, total nitrogen storage, and the contents of each component, but significantly increased DOC/TOC, the ratio of microbial biomass nitrogen to TN as well as the ratio of microbial biomass carbon to TOC (microbial quotient). Management measures significantly affected the chemical structure of SOC. Compared with the control, the relative intensities of phenolic and alcoholic-OH, aliphatic methyl and methylene, aromatic C=C, and carbonyl C=O absorption were higher in the SOC of undisturbed and extensively managed Moso bamboo plantations, and soil hydrophobicity was significantly increased. Results from correlation analysis showed that soil hydrophobicity and the content of aliphatic and aromatic groups were negatively correlated with microbial quotient and positively correlated with TOC and TN content. In conclusion, the increased inputs of organic matter residues (such as litter and roots) could contribute to the relative accumulation of chemical resistance compounds with reduced human disturbance, which significantly enhanced chemical stability of soil organic carbon. Soil clay minerals played a key role in protecting soil organic carbon through the formation of mineral-organic compounds, which facilitate the stability of soil carbon storage and the long-term preservation of soil carbon.

Objective: Type 2 diabetes mellitus is increasing in young adults, and greater adiposity is considered a major risk factor. However, whether there is an association between obesity and diabetes and how this might be impacted by age is not clear. Therefore, we investigated the association between body mass index (BMI) and diabetes across a wide range of age groups (20-30, 30-40, 40-50, 50-60, 60-70 and ≥70 years old)., Design: We performed a retrospective cohort study using healthy screening programme data., Setting: A total of 211 833 adult Chinese persons >20 years old across 32 sites and 11 cities in China (Shanghai, Beijing, Nanjing, Suzhou, Shenzhen, Changzhou, Chengdu, Guangzhou, Hefei, Wuhan, Nantong) were selected for the study; these persons were free of diabetes at baseline., Primary and Secondary Outcome Measures: Fasting plasma glucose levels were measured and information regarding the history of diabetes was collected at each visit. Diabetes was diagnosed as fasting plasma glucose ≥7.00 mmol/L and/or self-reported diabetes. Patients were censored at the date of diagnosis or the final visit, whichever came first., Results: With a median follow-up of 3.1 years, 4174 of the 211 833 participants developed diabetes, with an age-adjusted incidence rate of 7.35 per 1000 persons. The risk of incident diabetes increased proportionally with increasing baseline BMI values, with a 23% increased risk of incident diabetes with each kg/m 2 increase in BMI (95% CI 1.22 to 1.24). Strong associations between HLA alleles and infectious and autoimmune diseases are well established. Although obesity is also associated with these diseases, the relationship between HLA and obesity has not been systematically investigated in a large cohort. In the current study, we analyzed the association of HLA alleles with BMI using data from 1.3 million healthy adult donors from the Chinese Marrow Donor Program (CMDP). We found 23 HLA alleles, including 12 low-resolution and 11 high-resolution alleles, were significantly associated with BMI after correction for multiple testing. Alleles associated with high BMI were enriched in haplotypes that were common in both Chinese and European populations, whereas the alleles associated with low BMI were enriched in haplotypes common only in Asians. Alleles B*07, DRB1*07, DRB1*12, and C*03:02 provided the strongest associations with BMI ( P = 6.89 × 10 -10 , 1.32 × 10 -9 , 1.52 × 10 -9 , and 4.45 × 10 -8 , respectively), where B*07 and DRB1*07 also had evidence for sex-specific effects ( P heterogeneity = 0.0067 and 0.00058, respectively). The pit mud (PM) in fermentation cellar is a complex ecosystem that hosts diverse microbial communities that contribute to the production of Chinese strong-flavor Baijiu (CSFB). However, the microbial ecology of PM, particularly the extent of their phylogenetic novelty remains poorly understood. Here we conducted Illumina MiSeq sequencing to explore the diversity and novelty patterns of PM bacterial communities from Luzhou Laojiao cellars in use for 40 and 400years. High diversity indices were found in the PM with 16 phyla and 105 genera. Interestingly, the compositions of dominant genera of the PM were significantly different than that reported previously for PM sampled from other geographic sites, suggesting greater microbial diversity of PM. The dominant genus of Caproiciproducens, a caproic acid-producing bacterium, is the first reported for Chinese Baijiu production. Our results demonstrate that the PM hosts a large number of novel taxa, with 26% of the total OTUs (operational taxonomic units) distant to cultured counterparts. The class Clostridia within Firmicutes presented the highest proportion of novel OTUs. Most novel OTUs were initially isolated from diverse environments, the most abundant of which came from Chinese Baijiu brewing ecosystems, highlighting the huge culturing gap within the PM, but at the same time suggesting the importance of these OTUs in CSFB production. The data presented in this study significantly increases the number of bacteria known to be associated with CSFB production and should help guide the future exploration of microbial resources for biotechnological applications., (Copyright © 2017. Objective: To understand the status of Entamoeba histolytica infection in diarrhea patients in general hospitals, so as to provide the evidences for the prevention and control of the disease., Methods: The diarrhea patients in intestinal disease clinics of 3 general hospitals in Shanghai City were chosen as the investigation objectives, and their fecal and blood samples were collected, and then were detected by the normal saline direct smear method and iodine solution staining, immunochromatographic method and ELISA respectively to understand the infection status of E. histolytica, and the characteristics of the infected persons were analyzed. RESULTS Totally 1 015 fecal samples were detected, and among which 36 positive ones were detected by parasitological examinations, with a general positive rate of 3.55%. There were no statistically significant differences among the positive rates of patients from the three hospitals (P > 0.05), nor between or among those of the patients with different sexes, ages, occupations and education levels (all P > 0.05). The positive rate of E. histolytica in bloody purulent stools was higher than those in loose stools and watery stools (both P < 0.01). The peak period of infection was from July to September. Among the 36 infected people detected by parasitological examination, 88.90% of them complained about abdominal pain, and the red blood cells and leucocyte cells were found in the stool samples of 75.00% and 22.23% of the cases, respectively. The positive rates of E. histolytica were 8.18% (83/1 015) and 7.12% (48/675) respectively when detected by the immunochromatographic method and ELISA., Conclusions: Summer and autumn are the high risk seasons for E. histolytica infection, and the surveillance should be strengthened in this period. The positive rate of E. histolytica in samples of bloody purulent stools is high, and the combined application of several detection methods can increase the detection rate.

A novel halophilic, filamentous actinomycete, designated TRM 4064(T), was isolated from a hypersaline habitat in Sichuan Province, China. Phylogenetic analysis based on an almost-complete 16S rRNA gene sequence of strain TRM 4064(T) showed that it was most closely related to Actinopolyspora mortivallis (99.1 % sequence similarity). The sequence similarities between strain TRM 4064(T) and other Actinopolyspora species with validly-published names were <97.0 %. However, it had relatively low mean values for DNA-DNA relatedness with the A. mortivallis DSM 44261(T) (23.2 %). Optimal growth occurred at 37 °C, pH 7.0 and in the presence of 13 % (w/v) NaCl. The whole-cell sugar pattern consists of xylose, glucose, ribose and arabinose. The predominant menaquinones are MK-10(H4) (38.2 %), MK-9(H4) (25.1 %), MK-9(H2) (28.6 %) and MK-8(H4) (7.3 %). The major fatty acids are anteiso-C17:0 (36.9 %) and iso-C17:0 (19.3 %). The diagnostic phospholipids detected were diphosphatidylglycerol (DPG), phosphatidylglycerol (PG), phosphatidylcholine (PC), phosphatidylinositol (PI) and two unknown phospholipids. The G+C content of the genomic DNA of the type strain is 66.3 mol%. Strain TRM 4064(T) therefore represents a novel species of the genus Actinopolyspora, for which the name Actinopolyspora dayingensis sp. nov. is proposed. The type strain is TRM 4064(T) (= KCTC 19979(T) = CCTCC AA 2010010(T)).

Twenty-four endophytic actinomycetes strains were isolated from the Salvia przewalskii in Tibetan Plateau of China by tablet coating method. Fusarium moniliforme, Helminthosporium turcicum and Bipolaris maydis were selected as indicator fungi to test the antimicrobial activities of these endophytic actinomycetes by tablet confrontation method. The results showed that 21 strains can produce antimicrobial substances which accounts for 85.7% of the total separates number. Four strains of endogenous actinomyces have more obvious antifungi activity. According to results of morphology and culture properties and 16S rDNA sequences of endophytic actinomyces, it is concluded that all of the isolates were streptomycetes trains.

Objective: To understand the survival of imported Oncomelania snails in new environments with different densities in waterway net region, Qingpu District, Shanghai., Methods: The snails collected from Guichi, Anhui Province were put into the ponds of 4 square meters and each had 25, 50, 100, 200, 400 and 800 pairs of snails, respectively. During the next 2 years, the temperature, humidity, the activities of snails and their second birth snails were investigated each day. All the data were analyzed statistically., Results: The highest number of snails appeared from May to June on the surface of soils in different densities. The average numbers of snails were 2.0%-12.7% of whole put in snails, and the numbers of offspring were less than 2% of the whole put in snails., Conclusion: The survival and reproduction of artificially imported Oncomelania snails from Guichi, Anhui Province is not good in Qingpu District, Shanghai.

Aiming at the delayed sowing of winter wheat induced by the drought and water logging often occurred in Huanghuai Plains of China, six sowing dates (15 October, normal sowing; 30 October, moderate delay; 15 November, delay; 30 November, seriously delay; 15 February, early spring sowing; and 1 March, spring sowing) were designed to investigate the effects of different sowing dates on the shoot type morphology and growth characteristics of winter wheat. With the delay of sowing date, the winter wheat grew and developed faster, and the growing period of the wheat sown in early spring and spring was 115-130 days shorter than that with normal sowing. As compared with those of the wheat with normal sowing, the shoot height, spike number per unit area, and productive spikelets per unit ear of the wheat sown delayed had a decrease, leaf position and canopy moved down, and leaf area reduced. When the sowing was delayed from the date 15 October (normal sowing) to 1 March (spring sowing), the harvest index increased from 0.46 to 0.53. Delaying sowing date also resulted in the significant reduction of grain yield, with the maximum decrement as high as 43. 6%. The spring-sown winter wheat not going through vernalization could still form yield.

Taxus chinensis var. mairei is an endemic and endangered plant species in China. The resources of T. chinensis var. mairei have been excessively exploited due to its anti-cancer potential, accordingly, the extant T. chinensis var. mairei population is decreasing. In this paper, ultrasonic extraction and HPLC were adopted to determine the contents of active components paclitaxel, 7-xylosyltaxol and cephalomannine in cultivated and wild T. chinensis var. mairei plants, with the content distribution of these components in different parts of the plants having grown for different years and at different slope aspects investigated. There existed obvious differences in the contents of these active components between cultivated and wild T. chinensis var. mairei plants. The paclitaxel content in the wild plants was about 0.78 times more than that in the cultivated plants, whereas the 7-xylosyltaxol and cephalomannine contents were slishtly higher in the cultivated plants. The differences in the three active components contents between different parts and tree canopies of the plants were notable, being higher in barks and upper tree canopies. Four-year old plants had comparatively higher contents of paclitaxel, 7-xylosyltaxol and cephalomannine (0.08, 0.91 and 0.32 mg x g(-1), respectively), and the plants growing at sunny slope had higher contents of the three active components, with significant differences in the paclitaxel and 7-xylosyltaxol contents and unapparent difference in the cephalomannine content of the plants at shady slope. It was suggested that the accumulation of the three active components in T. chinensis var. mairei plants were closely related to the sunshine conditions. To appropriately increase the sunshine during the artificial cultivation of T. chinensis var. mairei would be beneficial to the accumulation of the three active components in T. chinensis var. mairei plants.

Objective: To understand the contamination status of food with parasites in Shanghai market, so as to provide the evidence for formulating the surveillance strategy in parasitic diseases and provide the technical support in the food safety., Methods: The freshwater fish products, marine products, vegetables, snails and frogs were sampled with the cluster random method in the farmer-trades and supermarkets of the 18 districts in Shanghai City during 2005-2010 period, and all the specimen were screened by the digestion method, or crushing method or dissection method or saline floatation method to check the parasite metacercaria or larvae or eggs., Results: A total of 5 185 specimen in 23 species of fishes were screened in fresh-water products, with parasite infection rate of 1.93%. About 4 033 specimens of 20 species of fishes and shrimps were screened and 1.76% of samples were infected with Clonorchis sinensis. Among all kind of fishes, the highest infection rate was 7.83% (48/613) in Pseudorasbora parve. No any infected specimen was found among 1 152 specimen of fresh water crustacean screened. Anisakis spp. were found in 12.7% of 433 specimens of 23 species of seawater products, among them, the higher infection rate of Anisakis spp. was found in Pneumatophorus japonicas and Trichiurus haumela with their infection rates of 50.00% (13/26) and 23.46% (42/179), respectively, which much higher than those found in other seawater products (P < 0.01). In 37 kinds of vegetables, the parasite eggs were found in one of 428 specimens with its infection rate of 0.47%, while no any parasite egg was found in 103 specimens of 10 kinds of fruits. No any Angiostrongylus cantonensis larvae were found in 330 snails, 31.37% of 102 frogs were found infected with Spirometra mansoni spargana. No any contamination with parasites was found in 116 meat specimens of pigs and cattle. In the same time, the intestinal parasite infection rate of residents was 0.42% (131/31 239)., Conclusions: It is found that some of foods in Shanghai markets are contaminated with parasites. Therefore, it is necessary to enforce the activities in health education as well as to take integrated prevention measures in order to ensure the food safety.

A novel actinomycete strain, designated TRM 40137(T), was isolated from a hypersaline habitat in Xinjiang Province, north-west China, and subjected to a polyphasic taxonomic study. The strain was aerobic, Gram-positive and the optimum NaCl concentration for growth was 4-5% (w/v). Phylogenetic analysis showed that strain TRM 40137(T) has a 16S rRNA gene sequence similarity of 95.02% with the described species Glycomyces sambucus E71(T) and can be distinguished from all previously described representatives of the genus Glycomyces. The whole-cell sugar pattern consisted of xylose and galactose. The predominant menaquinone was MK-10(H(2)) and the major fatty acids were anteiso-C15:0 and anteiso-C17:0. The phospholipid pattern consists of phosphatidylglycerol, diphosphatidylglycerol, three unknown aminophospholipids and two unknown phospholipids. The G+C content of the genomic DNA was 68.8 mol%. A novel species Glycomyces halotolerans sp. nov. is proposed, with strain TRM 40137(T) (=CCTCC AA 2010013(T) = KCTC 19988(T)) as the type strain of G. halotolerans.

A novel actinomycete strain, designated TRM 40133(T), was isolated from a hypersaline habitat of Tarim basin in Xinjiang Province, north-west China. Its taxonomic status was determined using a polyphasic approach. Phylogenetic analysis based on an almost-complete 16S rRNA gene sequence of the strain showed that it formed a well-seperated sub-branch within the radiation of the genus Saccharopolyspora. The highest levels of 16S rRNA gene sequence similarity was found between the strain TRM 40133(T) and Saccharopolyspora qijiaojingensis YIM 91168(T) (96.5%). The chemotaxonomic characteristics of the isolate are typical for the genus Saccharopolyspora. It contained meso-DAP as the diagnostic diamino acid. Whole cell hydrolysate contained arabinose, xylose, ribose and glucose. The diagnostic phospholipids were phosphatidylcholine, phosphatidylglycerol, phosphatidylinositol and two unknown phospholipids. The main menaquinone was MK-9(H(6)) and MK-9(H(4)). No mycolic acid was detected. The predominant cellular fatty acids were iso-C(16:0) and anteiso-C(17:0). The G+C content of the genomic DNA was 68.2 mol%. In addition, the strain TRM 40133(T) had a phenotypic profile that readily distinguished it from the recognized representatives of the genus Saccharopolyspora. The strain TRM 40133(T) therefore represents a novel species of the genus Saccharopolyspora, for which the name Saccharopolyspora lacisalsi sp. nov. is proposed. The type strain is TRM 40133(T) (=KCTC 19987(T) =CCTCC AA 2010012(T)).

Based on the analysis of the coercing and dynamic evolution relationships between agro-economic system and agro-ecological system, and by using modified entropy method, the weight of each evaluation index for the agro-ecolomic system in Changwu County of Shaanxi Province was determined, and, by employing coupling degree model, the coupling processes of agro-ecolomic system in the gullyarea of Loess Plateau were investigated. From 1949 to 2008, the agro-ecolomic system in the County experienced three stages of "traditional extensive agriculture", "agricultural mechanization", and "agricultural pre-modernization". In corresponding to the implement of national macro policies, the coupling degree of the agro-ecolomic system reached two peaks standing for utmost increasing phase and three valleys standing for coordinate development phase. Periods 1955-1961 and 1984-1992 were the utmost increasing phases of the coupling degree; periods 1962-1965 and 1981-1983 were the rapid transition periods responding to the national-wide policies controlling; while periods 1949-1954 and 1966-1980 were the low-level coordinated development phases. Since 1993, the system had been in the harmonious phase by a spiral development. At present, the system had a potential risk of changing from the harmonious phase to the utmost development phase. Appropriate agricultural policies and better external environment could promote the stable development of the agro-ecolomic system on a harmonious level; otherwise, a retrogressive system might be expected.

Objective: To establish a FTA-polymeras chain reaction (FTA-PCR) method in detection of Cryptospordium spp. in different sources of water., Methods: The semi automated immunomagnetic separation (IMS) of Cryptospordium oocysts in environmental water samples was performed firstly, and then genomic DNA of Cryptospordium oocysts was extracted by FTA filters disk. Oligonucleotide primers were designed based on the DNA fragment of the 18 S rRNA gene from C. parvum. Plate DNA was amplified with primers in PCR. The control DNA samples from Toxoplasma gondii,Sarcocystis suihominis, Echinococcus granulosus, and Clonorchis sinensis were amplified simultaneously. All PCR products were detected by agar electrophoresis dyed with ethidium bromide., Results: The 446 bp fragment of DNA was detected in all samples of C. parvum, C. andersoni, and C. baileyi, while it was not detected in control groups in laboratory. No positive samples were found from 10 samples collected from tape water in 5 districts of Shanghai City by FTA-PCR. Nine positive samples were detected totally from 70 different environmental water samples, there were 0 out of 15 samples from the source of tape water, 2 out of 25 from the Huangpu River, 5 out of 15 from rivers around the animal farmers, 1 out of 9 from output water of contaminating water treatment factory, 1 out of 6 from the out gate of living contaminating water. The 446 bp fragment was detected from all the amplified positive water samples., Conclusions: FTA-PCR is an efficient method for gene detection of Cryptospordium oocysts, which could be used in detection of environmental water samples. The contamination degree of Cryptospordium oocysts in the river water around animal farms is high.

Objective: To understand the contamination status of Cryptosporidium sp. and Giardia lamblia in drinking water, source water and environmental water in Shanghai., Methods: All water samples collected from drinking water, source water and environmental water were detected by a procedure of micromembrane filtration, immune magnetic separation (IMS), and immunofluorescent assay (IFA)., Results: Cryptosporidium oocysts and Giardia cysts were not found in 156 samples of the drinking water including finished water, tap water, or pipe water for directly drinking in communities. Among 70 samples either source water of water plants (15 samples), environmental water from Huangpu River(25), canal water around animal sheds(15), exit water from waste-water treatment plants(9), or waste water due to daily life(6), Cryptosporidium oocysts were detected in 1(6.7%), 2(8.0%), 7(46.7%), 1(11.1%), and 1(16.7%) samples, respectively; and Giardia cysts were detected in 1(6.7%), 3(12.0%), 6 (40.0%), 2(22.2%), and 2(33.3%), respectively. The positive rate of Cryptosporidium oocysts and Giardia cysts was 17.1% (12/70) and 20.0% (14/70), respectively., Conclusion: No Cryptosporidium oocysts and Giardia cysts have been detected in drinking water, but found in source water and environmental water samples in Shanghai.

Background: There has been little literature about leukemia epidemiology in Nanjing in recent years. We aimed to explore the incidence rate, gender and age distribution of leukemia in Nanjing using the leukemia database of the city., Results: The average yearly incidence rate of leukemia was 3.68/105 during 2003 - 2007 in Nanjing. There were no significant difference in gender (x2 = 3.266, p > 0.05) or seasons (x2 = 11.36, p > 0.05). The incidence rate was the highest in group aged 80~ years (18.64/105). AML accounted for approximately 36.8% of all leukemias., Conclusions: The incidence rate of leukemia, especially in the aged population, was relatively high in Nanjing. Leukemia is the major malignant tumor in children. Therefore, more attention should be paid to leukemia in children and the aged people.

By using O-ring statistics in point pattern analysis and Programita software, this paper studied the spatial distribution patterns and associations of different age-class individuals of Taxus chinensis var. mairei populations at Xianyu and Xiaxiang in the mountainous area of southern Anhui Province. The Xianyu population had a random spatial distribution at the scales from 2 m to 25 m but a clumped distribution at the scales < 2 m, while the Xiaxiang population distributed randomly at all the scales. As for Xianyu population, its individuals of age-class I had a clumped distribution at the scales < 5 m, while the individuals of elder age-classes tended to be non-randomly distributed at more scales than < 5 m, which might be related to the features of population regeneration. With increasingly enlarged age-class differences, the spatial association between younger and elder age-classes individuals turned to be negative, but less spatial association was observed between adjacent age-classes individuals. Such a spatial relationship between different age-classes individuals of T. chinensis was beneficial to the effective use of resources. With the growth of the individuals, T. chinensis population had a transition from clumped to random distribution.

A novel bacterium, TRM 0175(T), belonging to the genus Halomonas, was isolated from a soil sample taken from a salt lake in Xinjiang Province, north-west China. The isolate was Gram-negative, aerobic, rod-shaped and motile by means of peritrichous flagella. It was catalase-positive and oxidase-negative. Growth occurred at NaCl concentrations of 0-20 % (optimum at 10-13 %), at 15-50 degrees C (optimum at 37 degrees C) and at pH 6.0-9.0 (optimum at pH 7.0). Metabolism was respiratory with oxygen as terminal electron acceptor. Acid was produced from D-ribose, D- and L-arabinose, D-xylose, D-galactose, D-mannose, L-rhamnose, cellobiose, maltose, trehalose and D- and L-fucose and was produced weakly from aesculin. The predominant ubiquinone was Q-9. The major fatty acids were C(18 : 1)omega7c and C(19 : 0) cyclo omega8c. The G+C content of the genomic DNA was 60.0 mol%. The affiliation of strain TRM 0175(T) with the genus Halomonas was confirmed by 16S rRNA gene sequence comparisons. The most closely related species was Halomonas anticariensis; 16S rRNA gene sequence similarity between H. anticariensis FP35(T) and strain TRM 0175(T) was 95.3 %. Phenotypically, some characteristics of TRM 0175(T) differed from those of H. anticariensis. On the basis of data from this polyphasic study, strain TRM 0175(T) represents a novel species of the genus Halomonas, for which the name Halomonas xinjiangensis sp. nov. is proposed; the type strain is TRM 0175(T) (=CCTCC AB 208329(T) =KCTC 22608(T)).

Kirengeshoma comprises two species inhabiting warm temperate-deciduous forests in East China/South Japan (Kirengeshoma palmata) and South Korea (Kirengeshoma koreana). A survey of chloroplast (cp) DNA and inter-simple sequence repeats (ISSRs) variation in Kirengeshoma was carried out to determine the population history of a plant taxon around the East China Sea (ECS). CpDNA and ISSRs revealed lower genetic divergence between China and Japan relative to the other contrasts, in line with intrageneric classification. Molecular dating suggests that K. koreana diverged at the Plio-Pleistocene boundary from the Japanese populations, whereas the latter migrated into China during the early-to-mid Pleistocene via the ECS basin. Vicariant segregation of Chinese and Japanese populations likely occurred during the mid-Pleistocene. Mismatch distributions and neutrality tests indicated that Chinese populations expanded their range during the Late Pleistocene, probably during a cold period, whereas those from Japan showed no significant population growth. We conclude that the current distribution and differentiation of components of presently isolated warm temperate-deciduous forests in China, Japan and Korea likely resulted from a combination of relatively ancient vicariant and immigration events, and those from Japan were particularly sensitive to range fragmentation and long-term refugial isolation throughout the Late Pleistocene.

Based on the field investigation data obtained from the typical plots of four community types, i. e. , secondary shrub, Phyllostachys edulis forest, Cunninghamia lanceolata forest, and Pinus massoniana forest, in the Zongli Village of Qimen County in Anhui Province, this paper studied the mean basal diameter and structure of Monimopetalum chinense population, and the effects of environmental factors on the population characteristics. The results showed that the mean basal diameter of M. chinense in the communities was in the order of P. edulis forest > P. massoniana forest > C. lanceolata forest > secondary shrub, and significantly larger in the two former forests than in the others (P < 0.05). The population structure of M. chinense also differed with habits. In secondary shrub and P. massoniana forest, the structure was a aptypical pyramid-like form, suggesting that the population was stable; in P. edulis forest, it was a spindle type, indicating that the population was at the early stage of declining; whereas in C. lanceolata forest, it was a typical pyramid-like form, with most young individuals in the population. The survival curve of the whole population belonged to Deevey II, suggesting that the population was in developing tendency with no declination. M. chinense preferred the sites with low altitude, high soil moisture and organic matter contents, gentle slope, and high coverage of tree layer; while frequent human disturbance decreased its natural regeneration and stability. Based on the results obtained, some preliminary protection suggestions were proposed.

The prevalence and geographical distribution of the intestinal protozoa Blastocystis in humans across China is unknown, and the relative importance of different subtypes has yet to be investigated. We assessed the community prevalence and relative frequencies of different Blastocystis subtypes in four epidemiological settings in China, i.e., Shanghai municipality, Yongjia county (Zhejiang province), Eryuan county, and Menghai county (both Yunnan province). Blastocystis infection was detected with the culture method, and the subtype was identified with polymerase chain reaction using a set of subtype-specific primers. The prevalence at the four study settings was 1.9, 5.9, 18.4, and 32.6%, respectively. People aged greater than or equal to 60 years had a higher prevalence in the former two settings, Shanghai and Yongjia, whereas the highest infection rate was found among individuals aged 10-17 years in the latter two settings, Eryuan and Menghai. A higher prevalence was found in men in the former two settings but in women in the latter two settings. Five different Blastocystis subtypes were identified from the 192 isolates. Subtype 3 was the predominant type, followed by subtype 1. In conclusion, the epidemiology of Blastocystis varies across China.

Objective: To understand the potential risk for schistosomiasis transmission caused by introduction of infection source from mobile population in Shanghai., Methods: Field investigation was conducted in the suburb of Shanghai City by screening the mobile population living in Shanghai for more than 1 month and over 1 years old in a procedure of interviewing, serum indirect hemagglutination (IHA) test, and then fecal examination to detect the eggs with nylon sedimentation approach for those IHA positives., Results: Among 2,931 mobile people investigated, 1,575 were male (53.74%) and 1356 were female(46.26%); 138 out of 2931 were positive in IHA test (4.71%). 1938 (66.12%) out of 2 931 came from Schistosoma japonicum-endemic provinces and its positive rate in mobile population (5.99%) was significantly higher than those from the transmission-interrupted provinces (2.6%) (chi2=10.28, P<0.01), and those from non-endemic provinces (1.68%) (chi2=12.86, P<0.01). The 138 IHA positives all showed negative in fecal examination. In accordance with the serum positive rate and egg-infection rate in the national reporting system in 2004, it was estimated that there would be about 13 356 and 1 699 potential serum positive cases respectively from endemic area and transmission controlled area, and about 2,168 and 255 egg-positive cases from the two kind areas respectively, majority of the cases were from Anhui Province., Conclusion: Schistosomiasis transmission risks potentially exist in Shanghai suburb due to the introduction of infected mobile people from other endemic provinces, and a surveillance system and quick response are needed for the possible re-emergence of the disease.

Objective: To further study concentration-response relationship between daily mortality and particulate air pollution, to provide scientific basis for making policy., Methods: We applied spline model to daily time-series data for Taiyuan city, China from 2004-2005, using concentration of particulate matter less than 10 microm in aerodynamic diameter (PM10) as the exposure measure, and controlling confounding factors, such as season, meteorological factors, day-of-the-week, other air pollutions, etc., Results: The spline model showed a nonlinear relationship for the relative risks of death for all causes (total deaths) and for specific causes in relation to PM10 concentration, and obtained threshold levels. For total mortality, malignant tumor mortality and cardiovascular-cerebrovascular-respiratory mortality, the threshold levels were 85.00 microg/m3, 89.59 microg/m3, 122.54 microg/m3, respectively using the value of differential deduction., Conclusion: Our nonlinear models are appropriate for assessing the effect of particulate air pollution on daily mortality at current ambient levels in Taiyuan, China,in contrast to those of Europe and America.
