Background: Leukemia patients undergoing cryopreserved ovarian tissue transplantation (OTT) may carry a high risk of disease induction. Measurable residual disease (MRD) in bone marrow is linked to an elevated risk of relapse. It is controversial whether leukemia patients must be allowed to achieve measurable residual disease negative (MRD-negative) status instead of measurable residual disease positive (MRD-positive) status before ovarian tissue cryopreservation (OTC). Objective: To explore the safety and efficacy of OTT in acute leukemia patients with different MRD status by using xenotransplantation. Method: Cryopreserved ovarian tissue from 19 leukemia patients was thawed and xenotransplanted to ovariectomized BALB/C nude mice (n=36). The mice were divided into 2 groups based on the patient's MRD status before OTC: MRD-negative group (n=18) and MRD-positive group (n=18), additionally, a control group consisted of ovariectomized mice (n=9). Body weight was measured weekly and mortality, emaciation, and other abnormalities were recorded. Twenty-six weeks post-surgery, livers, spleens, uteruses, and ovarian grafts were removed for macroscopic and histological examinations to evaluate the efficacy of xenotransplantation and assess malignant cell contamination in mice. Results: Follicle growth was visible in the ovarian grafts of the MRD-negative and MRD-positive groups. Compared with the ovariectomized group, a significant decrease in body weight (p<0.01) was noted, the uterine volume was notably larger, estradiol (E2) levels were significantly higher (p<0.01), and follicle-stimulating hormone (FSH) levels were significantly lower (p<0.001) in the other two groups. Mice in the MRD-positive group showed a significantly higher incidence of death (p<0.001) and emaciation (p<0.01), compared to the MRD-negative group. Histological observation revealed the presence of malignant cells in the grafts, livers, and spleens of 3 mice in the MRD-positive group. No abnormalities were observed in the mice from the MRD-negative group in both macroscopic and histological observations except one mouse was sacrificed for ascites unrelated to leukemia relapse. Conclusion: For leukemia patients having ovarian tissue preserved in the first and only centralized human ovarian tissue cryobank in China, immunodeficient mice xenotransplantation can be a method to evaluate the safety and efficacy of OTT; the risk of malignant cell reimplantation due to OTT is higher in leukemia patients with MRD-positive status than those with MRD-negative status before OTC. The bamboo grasshopper, Ceracris kiangsu Tsai, is a serious pest of bamboo in China and Southeast Asia, and it overwinters as eggs for a period of up to 9 mo. Despite its importance, cold tolerance has not been thoroughly examined in this species. Thus, the present study investigated the low-temperature tolerance of overwintering eggs collected in the field, particularly the effects of rapid cold-hardening (RCH) and cold acclimation (ACC) on egg survival, and changes in the cryoprotectant contents. The supercooling point of overwintering eggs collected from the field was −23.9 °C, and their survival rate after exposure to −22 °C for 6 h was 26.6%, thereby suggesting that the cold tolerance strategy employed by C. kiangsu is freeze avoidance. After exposure to 0 and 4 °C for 12 and 24 h, or 0, 4, and 8 °C for 5 days, the survival rate of eggs subjected to −22 °C increased to approximately 50%, with significant effects of RCH and ACC. The glycerol levels in RCH and ACC treated eggs were also significantly higher than those in the control group, but the protein and trehalose contents were either reduced or not significantly altered. These results indicate that glycerol functions as a low molecular weight cryoprotectant in C. kiangsu eggs, and that RCH and ACC treatment facilitate its accumulation, thereby enhancing cold tolerance. Survival rate and emergence timing of the overwintering generation of many temperate agricultural pests is expected to affect their population dynamics and damage potential. However the impact of fluctuating winter and spring conditions on the successful emergence of insects post-diapause is generally poorly known. Here we characterize diapause responses in the peach fruit moth (PFM) pest, Carposina sasakii Matsumura, which overwinters at the larval stage in soil. Temperatures at a depth of 5 cm fluctuated markedly in early spring during the critical PFM post-diapause period (late December to mid-April). By removing outdoor larval samples over this period, we show that the completion of diapause for PFM in northern China starts from late January and continues until March. This extended developmental period is accompanied by an ongoing loss of cold resistance. Temperature conditions experienced in the field were associated with cold tolerance and emergence times, and reduced cold tolerance was associated with shorter emergence time. Cryoprotectants declined from late December, and levels were associated with changes in the supercooling point (SCP) of the larvae, but both correlated weakly to survival under cold stress during the post-diapause period. These findings suggest that diapause stage and soil temperatures should be taken into account when predicting field dynamics of soil-dwelling overwintering insects based on degree day accumulation models and other approaches. Hu sheep is a valuable sheep breed in China, and semen cryopreservation of Hu sheep is important for sustainable development of the agri-food industry. This study aimed to evaluate the effect of thawing rate and antioxidants (procyanidins [PC] and mitoquinone [MitoQ]) on the quality and antioxidant enzyme activity of post-thaw sperm in Hu sheep. Our results showed that the highest sperm quality was obtained from the group thawed at 70°C for 5 seconds. Furthermore, addition of 150 nM MitoQ in the extender could enhance motility, integrity of the membrane and acrosome, and mitochondrial activity, whereas only sperm motility and membrane integrity were increased with 10 μg/mL of PC supplementation, compared with the control group. Meanwhile, both PC (10 μg/mL) and MitoQ (150 nM) supplementation increased the levels of superoxide dismutase and glutathione peroxidase and decreased the levels of reactive oxygen species and malondialdehyde. In conclusion, the optimal thawing protocol of semen cryopreservation in Hu sheep was 70°C for 5 seconds. MitoQ supplementation (150 mM) in the extender could improve sperm quality and reduce the level of oxidative stress in Hu sheep semen after cryopreservation. Further studies are needed to evaluate fertility of the post-thaw semen using MitoQ.
