Plastic is a ubiquitous environmental contaminant, resulting in widespread exposure across terrestrial and marine spaces. In the environment, plastics can degrade into microparticles where exposure has been documented in a variety of fauna at all trophic levels. Human epidemiological studies have found relationships between inhaled microplastics and oxidative stress and inflammation. Previous studies of bottlenose dolphins (Tursiops truncatus) have reported prevalent exposure to plasticizing chemicals (e.g., phthalates) as well as particle loads in gastrointestinal tracts, but exposure from inhalation has not yet been studied. The objective of this study was to determine if inhalation is a viable route of microplastic exposure for free-ranging dolphins. Exhalation samples were opportunistically collected from dolphins residing in Sarasota Bay, Florida (n = 5) and Barataria Bay, Louisiana (n = 6) during catch-and-release health assessments to screen for microplastic particles. All dolphin samples contained at least one suspected microplastic particle, and polymer composition was determined for 100% of a subset (n = 17) of samples. Since late 2021, highly pathogenic avian influenza (HPAI) viruses of A/goose/Guangdong/1/1996 (H5N1) lineage have caused widespread mortality in wild birds and poultry in the United States. Concomitant with the spread of HPAI viruses in birds are increasing numbers of mammalian infections, including wild and captive mesocarnivores and carnivores with central nervous system involvement. Here we report HPAI, A(H5N1) of clade 2.3.4.4b, in a common bottlenose dolphin (Tursiops truncatus) from Florida, United States. Pathological findings include neuronal necrosis and inflammation of the brain and meninges, and quantitative real time RT-PCR reveal the brain carried the highest viral load. Virus isolated from the brain contains a S246N neuraminidase substitution which leads to reduced inhibition by neuraminidase inhibitor oseltamivir. The increased prevalence of A(H5N1) viruses in atypical avian hosts and its cross-species transmission into mammalian species highlights the public health importance of continued disease surveillance and biosecurity protocols., (© 2024. Mortality patterns in cetaceans are critical to understanding population health. Common bottlenose dolphins (Tursiops truncatus truncatus) inhabiting the Indian River Lagoon (IRL), Florida have been subjected to four unusual mortality events (UMEs), highlighting the need to evaluate morbidity and mortality patterns. Complete gross examinations were conducted on 392 stranded dolphins and histopathological analyses were conducted for 178 animals (2002-2020). The probable causes of mortality were grouped by etiologic category: degenerative, metabolic, nutritional, inflammatory (infectious and noninfectious disease), and trauma. Probable cause of mortality was determined in 57% (223/392) of cases. Inflammatory disease (infectious/noninfectious) and trauma were the most common. Inflammatory disease accounted for 41% of cases (91/223), with the lungs (pneumonia) most commonly affected. Trauma accounted for 36% of strandings (80/223). The majority of trauma cases were due to anthropogenic activities (entanglement, fishing gear or other debris ingestion, and propeller strikes), accounting for 58% of trauma cases (46/80). Natural trauma (prey-associated esophageal obstruction or asphyxiation, shark bites, and stingray interactions) accounted for 12% of all cases (26/223), and trauma of undetermined origin was identified in 4% of cases (8/223). Starvation or inanition (nutritional) were the probable cause of mortality in 17% of cases and peaked during the 2013 UME (61% of cases). Degenerative and metabolic etiologies accounted for 5% of cases. This study represents the most comprehensive evaluation of morbidity and mortality patterns in IRL dolphins. Intoxications of captive bottlenose dolphins (Tursiops truncatus) in the Florida Keys have been linked to observed interactions with marine macrophytic algal and cyanobacterial communities within enclosures. Taxonomic characterization of these communities coupled, in turn, to available observational data collected during intoxication events point to a contribution of filamentous cyanobacterial assemblages comprised of members of the polyphyletic genus, Lyngbya sensu lato. To identify toxic metabolites possibly relevant to these intoxications, chemical screening for known neurotoxins from cyanobacteria, as well as other regionally relevant harmful algal bloom (HAB) taxa, was combined with toxicity testing, and subsequent bioassay-guided fractionation, employing early life stages (i.e., embryos and larvae) of zebrafish (Danio rerio) as a well-established aquatic vertebrate toxicological model. Chemical analyses did not detect (within analytical limits) any of the known algal or cyanobacterial neurotoxins. Toxicity testing, alongside bioassay-guided fractionation, however, identified several chemical fractions with a range of potentially relevant bioactivities in both zebrafish embryos and post-hatch larvae including, in particular, behavioral (e.g., aberrant swimming) and physiological (e.g., altered heart rate) endpoints indicative of possible neurotoxicity, and subsequent chemical characterization of fractions suggested a contribution of the previously identified bioactive metabolite, eudesmacarbonate, in the observed toxicity. Comparative toxicological assessment with PbTx-2, as a positive control for neurotoxicity in the zebrafish model, further supported neurotoxic activity of cyanobacterial metabolites potentially relevant, in turn, to a contribution of these metabolites to dolphin intoxications. These findings suggest, in general, that marine zoological facilities may be affected by regional HABs, and assessments of potentially toxigenic algae and cyanobacteria should be included in management strategies in these facilities., (Copyright © 2021 Elsevier Ltd. Esophageal and pharyngeal obstruction are commonly reported in marine mammals, but asphyxiation from blowhole and nasal cavity obstruction has been reported only rarely: in two long-finned pilot whales (Globicephala melas), several harbor porpoises (Phocoena phocoena), and one common bottlenose dolphin (Tursiops truncatus). We describe two cases of blowhole obstruction and subsequent asphyxiation in bottlenose dolphins caused by eels. A whip eel (Bascanichthys scuticaris) was found obstructing the blowhole of a deceased dolphin from the Indian River Lagoon, Florida, US (2011) and a shrimp eel (Ophichthus gomesii) was found obstructing the blowhole of a deceased dolphin from Tampa Bay, Florida (2020). Normally, the respiratory and digestive tracts of cetaceans do not communicate. Consuming large or oddly shaped prey can result in laryngeal displacement and subsequent interaction between the two systems. It is likely the eels entered the oral cavity while the dolphins were consuming or playing with prey, and laryngeal displacement enabled the eels to slither into and become stuck in the nasal passage, causing asphyxiation. These novel findings underscore the importance of continued investigation into causes of mortality in stranded marine mammals and can contribute to the knowledge of feeding ecology in bottlenose dolphins. Health assessments were conducted on bottlenose dolphins in Barataria Bay, Louisiana, USA, during 2011 to 2018, to assess potential health effects following the Deepwater Horizon oil spill, compared to the unoiled Sarasota Bay, Florida, USA, reference dolphin population. We previously reported significant increases in T-lymphocyte proliferation, as well as lower T helper 1 (Th1) cytokines, higher Th2 cytokine IL-4, and lower T regulatory (Treg) cytokine IL-10 in Barataria Bay in 2011 compared to Sarasota Bay, consistent with Deepwater Horizon oil exposure. Although values between 2013 and 2016 were more similar to those observed in Sarasota Bay, T-cell proliferation was again elevated and cytokine balance tilted toward Th2 in Barataria Bay during 2017-2018. In 2018, Barataria Bay dolphins had significantly more circulating Treg cells than Sarasota Bay dolphins. Mice experimentally exposed to oil also had significantly increased T-lymphocyte proliferation and circulating Treg cell number, including effects in their unexposed progeny. In vitro stimulation resulted in greater Th2 responsiveness in Barataria Bay compared to Sarasota Bay dolphins, and in vitro oil exposure of Sarasota Bay dolphin cells also resulted in enhanced Th2 responsiveness. Evidence points to Treg cells as a potential target for the immunomodulatory effects of oil exposure. The immunological trends observed in Barataria Bay appeared exaggerated in dolphins born after the spill, suggesting the possibility of continued oil exposure or multigenerational health consequences of exposure to oil, as observed in mice. Harmful algal blooms produced by the phytoplankton species Karenia brevis and its associated neurotoxin, brevetoxin (PbTx), occur throughout the Gulf of Mexico and have had devastating impacts on co-occurring populations of bottlenose dolphins (Tursiops truncatus), an important marine sentinel species. The majority of documented impacts, however, are from the eastern Gulf of Mexico, with a critical lack of information on the degree and frequency of PbTx exposure in bottlenose dolphins from Texas coastal waters. This study documents PbTx exposure in Texas bottlenose dolphins between 2007 and 2017 and their association with co-occurring K. brevis blooms. PbTx was detected in 60% (n = 112) of the animals tested. Liver tissue samples had the highest frequency of detection (62%), followed by feces (41.4%) and gastric contents (30.4%). PbTx was not detected in urine or intestinal tissue. The concentration ranges of PbTx detected in feces (1.2-216, mean 38.4 ng/g), gastric contents (3.3-1016, mean 158 ng/g) and liver (0.6-52.4, mean 8.5 ng/g) samples were an order of magnitude less than values reported for Florida dolphins for the same sample types. The proportion of dolphins recovered within 4 weeks of a bloom that tested positive for PbTx ('Bloom' group; 75%) was significantly higher compared to those that were recovered 5-8 weeks after termination of a bloom ('Post-Bloom' group; 36%; p = 0.004). The proportion of PbTx-positive animals with no observed bloom association ('Baseline' group; 60%) was also significantly greater than the Post-Bloom group (p = 0.012). No significant difference in proportion of PbTx-positive animals was detected between Bloom and Baseline groups (p = 0.242). No significant differences in liver PbTx concentrations were observed between any pairwise combinations of the 3 exposure groups (p = 0.261). Due to their long life-span and top trophic position, odontocetes can accumulate high concentrations of mercury (Hg) in their tissues. This study measured the concentration of total Hg (THg) in the blubber and skin of bottlenose dolphins (Tursiops truncatus) that stranded along the Florida (FL) panhandle and Louisiana (LA) coasts and investigated the relationship between total Hg (THg) concentration and sex, body length, age, stranding location, diet/trophic position (δ 13 C and δ 15 N, respectively), and foraging habitat (δ 34 S). Additionally, we compared models using body length and age as explanatory variables to determine which was a better predictor of THg concentration. In both tissues, sex was not an influential predictor of THg concentration and there was a positive relationship between body length/age and THg concentration (p < 0.001). Florida dolphins had greater mean blubber and skin THg concentrations compared to LA dolphins (p < 0.001). There was a modest improvement in model fit when age was used in place of body length. δ 13 C, δ 15 N, and δ 34 S differed between stranding locations and together with age were significant predictors of THg concentrations (R 2  = 0.52, P < 0.001). Florida dolphins were δ 13 C enriched compared to LA dolphins (p < 0.001) and THg concentrations were positively correlated with δ 13 C (R 2  = 0.22, p < 0.001). Our results demonstrate spatial variability in THg concentrations from stranded bottlenose dolphins from the northern Gulf of Mexico; however, future research is required to understand how fine-scale population structuring of dolphins within FL and LA impacts THg concentrations, particularly among inshore (bay, sound, and estuary) stocks and between inshore and offshore stocks, as variations in biotic and abiotic conditions can influence both stable isotope ratios and THg concentrations., (Copyright © 2019 Elsevier Inc. Temporal trends in plasma concentrations of per- and polyfluoroalkyl substances (PFAS) in free-ranging bottlenose dolphins ( Tursiops truncatus ) inhabiting two geographic areas: Indian River Lagoon, Florida over the years 2003-2015 and the waters surrounding Charleston, South Carolina over 2003-2013, were examined. Nine PFAS met the inclusion criteria for analysis based on percent of values below level of detection and sampling years. Proportionate percentiles parametric quantile regression assuming lognormal distributions was used to estimate the average ratio of PFAS concentrations per year for each chemical. Plasma concentrations decreased over time for perfluorodecanoate (PFDA), perfluorohexane sulfonate (PFHxS), perfluorooctanoate (PFOA), perfluorooctane sulfonate (PFOS), and perfluoroundecanoate (PFUnDA) in both locations. Perfluorononanoate (PFNA) decreased with time in Indian River Lagoon dolphins. Perfluorododecanoate (PFDoDA) concentrations significantly increased over time among female Indian River Lagoon dolphins. Regulation and phaseout of specific PFAS groups may have led to the decreasing levels of those PFAS and increasing levels of other replacement PFAS.

Mucocutaneous lesions were biopsied from free-ranging Atlantic bottlenose dolphins Tursiops truncatus inhabiting the Indian River Lagoon (IRL), Florida, and estuarine waters of Charleston (CHS), South Carolina, USA, between 2003 and 2013. A total of 78 incisional biopsies from 58 dolphins (n=43 IRL, n=15 CHS) were examined. Thirteen dolphins had 2 lesions biopsied at the same examination, and 6 dolphins were re-examined and re-biopsied at time intervals varying from 1 to 8 yr. Biopsy sites included the skin (n=47), tongue (n=2), and genital mucosa (n=29). Pathologic diagnoses were: orogenital sessile papilloma (39.7%), cutaneous lobomycosis (16.7%), tattoo skin disease (TSD; 15.4%), nonspecific chronic to chronic-active dermatitis (15.4%), and epidermal hyperplasia (12.8%). Pathologic diagnoses from dolphins with 2 lesions were predominately orogenital sessile papillomas (n=9) with nonspecific chronic to chronic-active dermatitis (n=4), TSD (n=3), lobomycosis (n=1), and epidermal hyperplasia (n=1). Persistent pathologic diagnoses from the same dolphins re-examined and re-biopsied at different times included genital sessile papillomas (n=3), lobomycosis (n=2), and nonspecific dermatitis (n=2). This is the first study documenting the various types, combined prevalence, and progression of mucocutaneous lesions in dolphins from the southeastern USA. The data support other published findings describing the health patterns in dolphins from these geographic regions. Potential health impacts related to the observed suite of lesions are important for the IRL and CHS dolphin populations, since previous studies have indicated that both populations are affected by complex infectious diseases often associated with immunologic disturbances and anthropogenic contaminants.

Differences in priority organic pollutants (POPs), analyzed by gas chromatography-mass spectrometry, and stable isotope ratios (δ(13)C, δ(34)S, and δ(15)N; analyzed by isotope ratio-mass spectrometry), divide 77 bottlenose dolphins (Tursiops truncatus) from the Florida Gulf Coast into three distinct groups. POP levels reflect human population and historical contamination along the coast. In the least disturbed site, concentrations of ΣPOP in male dolphins were 18,000 ng g(-1)±6000 (95% confidence interval here and throughout); in the intermediate bay, males had ΣPOP concentrations of 19,000 ng g(-1)±10,000. St Andrews Bay was home to dolphins with the highest ΣPOP concentrations: 44,000 ng g(-1)±10,300. δ(34)S and δ(15)N, differed significantly between St. George Sound dolphins and those frequenting each of the other two bays, but not between St. Andrews and St. Joseph Bays. ΣPOP concentrations were statistically higher in dolphins frequenting St. Andrews Bay, but were not significantly different between dolphins occupying St. Joseph Bay and St. George Sound. Thus, using either POP or isotope values alone, we would only be able to identify two dolphin groups, but when POP and isotope data are viewed cumulatively, the results clearly define three distinct communities occupying this region., (Copyright © 2012 Elsevier B.V. Lacaziosis (lobomycosis; Lacazia loboi) is a fungal skin disease that naturally occurs only in humans and dolphins. The first reported case of lacaziosis in a bottlenose dolphin Tursiops truncatus occurred in 1970 in Sarasota Bay, Florida, USA, and subsequent photo-ID monitoring of the Sarasota Bay dolphin population has revealed persistence of the disease. The objectives of this study were to estimate lacaziosis prevalence (P) in 2 bottlenose dolphin populations on the west coast of Florida (Sarasota Bay and Charlotte Harbor) and compare disease occurrence to other published estimates of lacaziosis in dolphin populations across the globe. Historic photographic records of dolphins captured and released for health assessment purposes (Sarasota Bay) and photo-ID studies (Charlotte Harbor) were screened for evidence of lesions consistent with lacaziosis. Health assessment data revealed a prevalence of lacaziosis in the Sarasota Bay bottlenose dolphin population between 2 and 3%, and analyses of photo-ID data provided a lacaziosis-like prevalence estimate of 2% for Charlotte Harbor dolphins. With the exception of lacaziosis prevalence estimates for dolphins inhabiting the Indian River Lagoon (P = 0.068; P = 0.12), no statistically significant differences were seen among Sarasota Bay, Charlotte Harbor, and other published estimates. Although lacaziosis is a rare disease among these dolphin populations, studies that assess disease burden among different populations can assist with the surveillance of this zoonotic pathogen.

This study represents the first systematic study of lacaziosis (lobomycosis) in bottlenose dolphins Tursiops truncatus in the Atlantic Ocean along the east-central coast of Florida, USA. Lacaziosis is a chronic infection of the skin caused by the fungus Lacazia loboi, which affects only dolphins and humans. Previous studies have shown a high prevalence (6.8 to 12.0%) of lacaziosis in resident dolphins from the adjacent Indian River Lagoon Estuary (IRL), where the disease is endemic. We examined the prevalence of lacaziosis in this coastal area using photo-identification data collected between 2002 and 2008 to determine the prevalence of lacaziosis in coastal dolphins using photographic methodology shown to have high sensitivity and specificity in prior research. The prevalence of skin lesions compatible with lacaziosis estimated from photographic data was 2.1% (6/284), approximately 3 times lower than that described for the estuarine population using similar methods. To exclude potential bias introduced by differences in study duration and survey effort among areas, an 18 mo period when effort was most equal (January 2006 to June 2007) was chosen for statistical comparison. The prevalence of lacaziosis estimated from photographic data was significantly lower (3.8%: n = 6/160) in the Atlantic Ocean compared to the IRL (12.0%: n = 20/167) (risk ratio = 3.19, 95% CI 1.32 to 7.75, p < 0.01 by chi-square analysis). The lower prevalence of lacaziosis in dolphins found in the Atlantic Ocean and the overall lack of movement of dolphins between these habitats suggests that environmental conditions within the estuary may favor viability of L. loboi, and/or that immune compromise in resident estuarine dolphins is a precursor to the disease.

Trained odontocetes appear to have good control over the timing (pulse rate) of their echolocation clicks; however, there is comparatively little information about how free-ranging odontocetes modify their echolocation in relation to their environment. This study investigates echolocation pulse rate in 14 groups of free-ranging bottlenose dolphins (Tursiops truncatus) at a variety of depths (2.4-30.1 m) in the Gulf of Mexico. Linear regression models indicated a significant decrease in mean pulse rate with mean water depth. Pulse rates for most groups were multi-modal. Distance to target estimates were as high as 91.8 m, assuming that echolocation was produced at a maximal rate for the target distance. A 5.29-ms processing lag time was necessary to explain the pulse rate modes observed. Although echolocation is likely reverberation limited, these results support the hypotheses that free-ranging bottlenose dolphins in this area are adapting their echolocation signals for a variety of target detection and ranging purposes, and that the target distance is a function of water depth.

Photo-identification surveys conducted between 2002 and 2005 were used to determine dolphin home ranges and site fidelity within the Indian River Lagoon (IRL), Florida. The IRL was divided into six segments based on hydrodynamics and geographic features for purposes of characterization. Among the 615 dolphins with identifiable dorsal fins, 339 had > or =6 sightings and were used in segment and linear range analyses. The majority (98%) of dolphins were seen in < or =3 consecutive segments (331/339); of these, 44% (144/331) occurred in two segments, and 33% (109/331) in one segment. No dolphins were observed in all six segments. The largest number of dolphins was sighted in segment 1C (North Indian River). However, the highest density of dolphins was found in segment 2 (North-Central Indian River). Re-sighting rates for dolphins with > or =6 sightings ranged from 2.8 to 8.7 times observed. The mean linear home range varied from 22 to 54 km. Distributional analyses indicated that at least three different dolphin communities exist within the IRL: Mosquito Lagoon, and the North and South Indian River. No statistically significant correlations were found between the total number or density per km(2 )of dolphins and surface water area, salinity, or contaminant loads within segments of the lagoon. These results suggest that dolphins do not selectively avoid areas with relatively unfavorable water quality. IRL dolphins should be studied on smaller spatial scales than currently practiced, and potential anthropogenic impacts should be evaluated based on geographic partitioning.
