Birds that repeatedly visit distinct places along their migratory routes in consecutive years must be able to navigate to these places and respond appropriately to unfavourable wind conditions. This study analysed the migratory routes, repeatedly‐visited areas and responses to sidewinds of 15 GPS‐tracked adult ospreys Pandion haliaetus from northeast Germany migrating to their wintering sites in Africa and back. We determined stopovers and intermediate goal areas and performed repeatability estimations on timing and migratory paths in four regions. The orientation behaviour of the ospreys was analysed with regard to perpendicular wind components at each GPS point during autumn and spring migrations. Generalised linear mixed models were used to test the dependence of orientation behaviour on region and the distance to the next goal. The findings showed that ospreys demonstrate high fidelity to migratory paths in autumn and spring, as well as to the timing of migration in autumn; and sidewinds are predominantly compensated, especially when sidewinds are strong. Furthermore, during autumn migration, the proportion of compensation increases in most regions with decreasing distances to the next goal; however, during spring migration, drift behaviour was detected more often at smaller distances to the next goal than at higher distances in the regions Mediterranean and central Europe. In general, ospreys compensate for unfavourable sidewinds and utilise supporting tailwinds on their journeys to the wintering sites in Africa and back to central Europe. [ABSTRACT FROM AUTHOR]

Phosphorus recycling from sewage sludge will be obligatory in Germany from 2029. Phosphorus content determination in sewage sludge is crucial to assess the prescribed recycling rates. Currently, German law regards sample preparation using aqua regia digestion in a microwave or under reflux conditions as well as instrumental phosphorus determination by ICP-OES, ICP-MS, or photometric determination with ammonium molybdate as equivalent. However, it is questionable whether these methods are indeed equivalent regarding phosphorus quantification in sludges near the limit of 20 g/kg for mandatory recycling. To answer this question, 15 sewage sludges of 11 different wastewater treatment plants were investigated with all permitted method (digestion and measurement) combinations. Moreover, one sewage sludge was also examined in an interlaboratory comparison (ILC) with 28 participants. This study shows that the above-mentioned methods differ in some cases significantly but across all method combinations and sludges, phosphorus recovery was between 80 and 121% after normalization to the grand mean (average of 15 sludges between 85 and 111%). The ILC and the examination of 15 sludges produced largely similar results. There is a tendency to higher phosphorus recovery after microwave digestion compared to reflux digestion and ICP-OES measurements determine higher phosphorus contents than ICP-MS and photometric phosphorus determination. Background: To determine the repeatability of corneal topography measurements in severe dry eye disease (DED). A comparison of corneal topography parameters between severe DED and healthy subjects was a secondary goal of this study.Methods: Sixty-nine patients with severe DED and 46 healthy subjects were enrolled in the study. All participants underwent repeated corneal topography measurements with Pentacam (Oculus, Germany) within a half hour time. Both eyes of the participants were used in statistical analysis. A further subcategorization of severe DED patients was performed according to Ocular Surface Disease Index (OSDI) scores: 32-50, 51-70 and 71-100. The repeatability of corneal parameters was assessed with correlation coefficients (CC).Results: The mean age of dry eye patients and healthy subjects were 40.8 ± 13.2 (17-66) and 39.8 ± 8.2 (18-61) years (p:0.604) respectively. No significant differences were found between severe DED and control groups according to analysed corneal parameters in both eyes (p:>0.05). All CCs were greater than 0.9 in severe DED group (p:<0.001). All CCs were also greater than 0.9 in severe DED patients among different OSDI groups (p:<0.001).Conclusions: Corneal topography measurements are highly repeatable in severe DED with Pentacam. Manufacturers and diagnostic companies often recommend on-site verification of analytical performance in the clinical laboratory. The validation process used by manufacturers is rarely described in detail, and certain information on analytical performance is missing from the product sheet, especially for immunoanalytical methods. We describe an approach to the detailed validation of an ELISA method for the measurement of proprotein convertase subtilisin/kexin type 9 (PCSK9) plasma concentrations. We compared manufacturers' claims of analytical performance with data obtained in the field laboratory using several approaches. We used the Human Proprotein Convertase 9/PCSK9 Quantikine ELISA diagnostic kit (R&D systems, Bio-Techne Ltd., Abingdon Science Park, Abingdon, UK) and three levels of quality control solution Quantikine Immunoassay Control Group 235 (R&D systems, Bio-Techne Ltd., Abingdon Science Park, Abingdon, UK) to verify precision. We measured the concentration of PCSK9 using the DS2 ELISA Reader (Dynex Technologies GmbH, Denkendorf, Germany). We used analysis of variance (ANOVA) and the R statistical package (R core team, version 1.4.5). Statistical analysis and terminology were performed according to protocol CLSI EP15-A3, and the reference interval was checked according to CLSI/IFCC C28-A3c. We found a significant difference between the manufacturer's claims of analytical performance and real data measured in the routine clinical laboratory. The calculated CV (%) for repeatability (calculated by simple estimation of the mean and SD, as used by the manufacturer) was between 5.5% and 7.4%, but the manufacturer's claim was between 4.1% and 6.5%. Using ANOVA, the true repeatability was between 5.0% and 6.9%. Similarly, ANOVA revealed values of CV (%) for within-laboratory imprecision between 6.5% and 9.1%, while the manufacturer's claims were between 4.1% and 5.9%. The recovery ranged from 105.5% to 121.8%. The manufacturer's recommended reference interval was checked and we didn't find any significant difference between men and women. We describe a comprehensive approach to verify the analytical performance of an ELISA method using the measurement of PCSK9 plasma concentration as an example. We found differences between the results of this approach based on the CLSI EP15-A3 protocol and data provided by the manufacturer. Purpose: One of the most frequently cited radiomics investigations showed that features automatically extracted from routine clinical images could be used in prognostic modeling. These images have been made publicly accessible via The Cancer Imaging Archive (TCIA). There have been numerous requests for additional explanatory metadata on the following datasets — RIDER, Interobserver, Lung1, and Head–Neck1. To support repeatability, reproducibility, generalizability, and transparency in radiomics research, we publish the subjects' clinical data, extracted radiomics features, and digital imaging and communications in medicine (DICOM) headers of these four datasets with descriptive metadata, in order to be more compliant with findable, accessible, interoperable, and reusable (FAIR) data management principles. Acquisition and validation methods: Overall survival time intervals were updated using a national citizens registry after internal ethics board approval. Spatial offsets of the primary gross tumor volume (GTV) regions of interest (ROIs) associated with the Lung1 CT series were improved on the TCIA. GTV radiomics features were extracted using the open‐source Ontology‐Guided Radiomics Analysis Workflow (O‐RAW). We reshaped the output of O‐RAW to map features and extraction settings to the latest version of Radiomics Ontology, so as to be consistent with the Image Biomarker Standardization Initiative (IBSI). Digital imaging and communications in medicine metadata was extracted using a research version of Semantic DICOM (SOHARD, GmbH, Fuerth; Germany). Subjects' clinical data were described with metadata using the Radiation Oncology Ontology. All of the above were published in Resource Descriptor Format (RDF), that is, triples. Example SPARQL queries are shared with the reader to use on the online triples archive, which are intended to illustrate how to exploit this data submission. Data format: The accumulated RDF data are publicly accessible through a SPARQL endpoint where the triples are archived. The endpoint is remotely queried through a graph database web application at http://sparql.cancerdata.org. SPARQL queries are intrinsically federated, such that we can efficiently cross‐reference clinical, DICOM, and radiomics data within a single query, while being agnostic to the original data format and coding system. The federated queries work in the same way even if the RDF data were partitioned across multiple servers and dispersed physical locations. Potential applications: The public availability of these data resources is intended to support radiomics features replication, repeatability, and reproducibility studies by the academic community. The example SPARQL queries may be freely used and modified by readers depending on their research question. Data interoperability and reusability are supported by referencing existing public ontologies. The RDF data are readily findable and accessible through the aforementioned link. Compared to force-plates, pressure-detecting insoles have the advantage that vertical ground reaction force (vGRF) can be estimated under field rather than laboratory conditions. However, the question arises whether insoles also provide valid and reliable results compared to a force-plate (i.e., the gold standard). The study aimed to investigate the concurrent validity and test-retest reliability of pressure-detecting insoles during static and dynamic movements. Twenty-two healthy young adults (12 females) performed standing, walking, running, and jumping movements while simultaneously collecting pressure (GP MobilData WiFi, GeBioM mbH, Münster, Germany) and force (Kistler ® ) data twice, 10 days apart. Concerning validity, ICC values showed excellent agreement (ICC > 0.75), irrespective of the test condition. Further, the insoles underestimated (mean bias: -4.41 to -37.15%) most of the vGRF variables. Concerning reliability, ICC values for nearly all test conditions also showed excellent agreement, and the SEM was rather low. Lastly, most of the MDC 95% values were low (≤5%). The predominantly excellent ICC values for between-devices (i.e., concurrent validity) and between-visits (i.e., test-retest reliability) comparisons suggest that the tested pressure-detecting insoles can be used under field-based conditions for a valid and reliable estimation of relevant vGRF variables during standing, walking, running, and jumping.

Background: To compare the measurement error of Cirrus HD-OCT and Heidelberg Retina Tomograph 3 (HRT3) in patients with early glaucomatous visual field defect. Methods: Thirty-nine consecutive patients (39 eyes) with early glaucomatous visual field defect were included. One eye of a patient was examined with Cirrus HD-OCT and HRT3 in one session. Each instrument was used by two operators, each taking two measurements in turn. We performed measurements of average retinal nerve fiber layer (RNFL) thickness and RNFL thickness in four quadrants with Cirrus HD-OCT and 13 stereometric parameters of the optic nerve head and RNFL with HRT3. Results: Cirrus HD-OCT parameters performed much better than HRT3 stereometric parameters. Mean (for both operators) intraoperator within-subject coefficient of variation of the best Cirrus HD-OCT parameter (average RNFL thickness) was 2.6-2.7 times lower than the best HRT3 parameters [mean cup depth and rim area ( P < 0.001)]. Mean intraoperator variability of RNFL thickness in quadrants (except nasal quadrant) was also significantly lower with OCT than with HRT. The interoperator within-subject coefficients of variation for both average RNFL thickness and RNFL thickness in all quadrants were significantly lower than the interoperator variability of best HRT3 parameter [mean cup depth ( P < 0.001)]. The within-subject coefficient of variation of the average/mean RNFL thickness assessed by both instruments was 5.4-7.3 times lower for Cirrus HD-OCT. Among HRT3 parameters, mean cup depth, rim area and linear cup/disk ratio were the least variable, while cup volume, cup area and cup/disc area ratio were the most variable parameters. Conclusions: Cirrus HD-OCT provides excellent intrasession intra- and interoperator repeatability of the RNFL measurements, especially of the average RNFL thickness in primary open-angle glaucoma (POAG) patients and POAG suspects with early glaucomatous visual field defect. The measurement error (expressed as within-subject coefficient of variation) of RNFL measurements by Cirrus HD-OCT is much lower than the error of HRT3 measurements of stereometric parameters of the optic nerve head and RNFL. Use of a digital preparation assistant system may improve considerably the quality of preclinical dental education, provided the system works reliably. Thus, the purpose of this pilot study was to quantitatively assess the reliability of a new preclinical digital preparation assistant system (PREPassist, KaVo, Germany). The system was used to repeatedly scan four different unprepared and four different prepared teeth both with and without repositioning. Corresponding measurements were made to quantify accuracy, repeatability, and reproducibility. This was done by estimating the measurement error. Based on this estimation, respective limits of agreement were calculated. We used these ranges, along with assessments of the measurement's accuracy, to judge whether the results satisfy our expectations for clinically acceptable measurements. For preclinical laboratory instruction, the results indicate an acceptable accuracy (mean accuracy of 89 μm) of the measurements. This assessment applies as well to repeatability, given by the range of the respective limits of agreement (range <200 μm). However, in the case of reproducibility, the limits revealed discrepancies of practical importance (range >200 μm). Reproducibility of tooth repositioning in the available mounting device is unacceptable for preclinical laboratory instruction because of the observed range >200 μm. Thus, there is a need for the manufacture of new and more reliable mounting devices because reproducibility procedures are mostly encountered in preclinical instruction in restorative techniques. In contrast to reproducibility, accuracy and repeatability are acceptable for practical purposes. Simple Summary: Infestation with the parasitic mite Varroa destructor is a serious cause of bee colony (Apis mellifera) losses on a global level. However, the presence of untreated survivor populations in many different regions indicates that selection for resistance might lead to a long-term solution. The success of selection depends on suitable testing criteria. To be effective, results must show repeatable effects of the individual genotype and correlate with the breeding goal. As colony survival is difficult to measure, selective breeding for Varroa resistance can be based on differences in mite infestation and specific behavioral traits. In this paper we look into different definitions of mite infestation and link these with brood hygiene (pin test), brood recapping and suppressed mite reproduction (SMR). Due to the large dataset (489 colonies) from Austria, Croatia and Germany and four seasons (2016–2019), our study arrogates high representability. Repeatability analysis depicts different infestation parameters, brood hygiene and recapping data as characteristic colony traits while SMR results are very strongly influenced by environmental effects. Brood hygiene and recapping data correlate weakly but significantly with mite infestation. We therefore recommend combining them with estimates of mite population increase and brood infestation for an effective selection on resistance. As colony survival is difficult or impossible to measure, differences in mite infestation levels and tests for specific behavioral traits are used for selective breeding for Varroa resistance. In this paper we looked into different definitions of mite infestation and linked these with brood hygiene (pin test), brood recapping and suppressed mite reproduction. We based our analyses on datasets of Apis mellifera carnica from three countries: Austria (147 records), Croatia (135) and Germany (207). We concluded that bee infestation in summer, adjusted for the level of natural mite fall in spring, is a suitable trait in the breeding objective, and also suggested including brood infestation rate and the increase rate of bee infestation in summer. Repeatability for bee infestation rate was about 0.55, for cells opened in pin test about 0.33, for recapping 0.35 and for suppressed mite reproduction (SMR) virtually zero. Although in most cases we observed correlations with the expected sign between infestation parameters and behavioral traits, the values were generally low (<0.2) and often not significantly different from zero.
