Goats play an important role in the livestock sector in Greece. The national herd consists mainly of two indigenous breeds, the Eghoria and Skopelos. Here, we report the population structure and genomic profiles of these two native goat breeds using Illumina's Goat SNP50 BeadChip. Moreover, we present a panel of candidate markers acquired using different genetic models for breed discrimination. Quality control on the initial dataset resulted in 48,841 SNPs kept for downstream analysis. Principal component and admixture analyses were applied to assess population structure. The rate of inbreeding within breed was evaluated based on the distribution of runs of homozygosity in the genome and respective coefficients, the genomic relationship matrix, the patterns of linkage disequilibrium, and the historic effective population size. Results showed that both breeds exhibit high levels of genetic diversity. Level of inbreeding between the two breeds estimated by the Wright's fixation index FST was low (Fst = 0.04362), indicating the existence of a weak genetic differentiation between them. In addition, grouping of farms according to their geographical locations was observed. This study presents for the first time a genome-based analysis on the genetic structure of the two indigenous Greek goat breeds and identifies markers that can be potentially exploited in future selective breeding programs for traceability purposes, targeted genetic improvement schemes and conservation strategies. The main objective of this study was to assess the genetic background of colostrum yield and quality traits after calving in Holstein dairy cows. The secondary objective was to investigate genetic and phenotypic correlations among laboratory-based and on-farm–measured colostrum traits. The study was conducted in 10 commercial dairy herds located in northern Greece. A total of 1,074 healthy Holstein cows with detailed pedigree information were examined from February 2015 to September 2016. All cows were clinically examined on the day of calving and scored for body condition. All 4 quarters were machine-milked, and a representative and composite colostrum sample was collected and examined. Colostrum total solids (TS) content was determined on-farm using a digital Brix refractometer. Colostrum fat, protein, and lactose contents were determined using an infrared milk analyzer, and energy content was calculated using National Research Council (2001) equations. Dry period length (for cows of parity ≥2), milk yield of previous 305-d lactation (for cows of parity ≥2), age at calving, parity number, season of calving, time interval between calving and first colostrum milking, and milk yield were recorded. Each trait (colostrum yield and quality traits) was analyzed with a univariate mixed model, including fixed effects of previously mentioned factors and the random animal additive genetic effect. All available pedigrees were included in the analysis, bringing the total animal number to 5,662. Estimates of (co)variance components were used to calculate heritability for each trait. Correlations among colostrum traits were estimated with bivariate analysis using the same model. Mean percentage (±SD) colostrum TS, fat, protein, and lactose contents were 25.8 ± 4.7, 6.4 ± 3.3, 17.8 ± 4.0, and 2.2 ± 0.7%, respectively; mean energy content was 1.35 ± 0.3 Mcal/kg and mean colostrum yield was 6.18 ± 3.77 kg. Heritability estimates for the above colostrum traits were 0.27, 0.21, 0.19, 0.15, 0.22, and 0.04, respectively. Several significant genetic and phenotypic correlations were derived. The genetic correlation of TS content measured on-farm with colostrum protein was practically unity, whereas the correlation with energy content was moderate (0.61). Fat content had no genetic correlation with TS content; their phenotypic correlation was positive and low. Colostrum yield was not correlated genetically with any of the other traits. This simulation study investigated the farm-level economic benefits of a genetic improvement scheme using artificial insemination (AI) with fresh ram semen in dairy sheep of the Chios breed in Greece. Data were collected from 67 farms associated with the Chios Sheep Breeders' Cooperative 'Macedonia', describing the percentage of ewes that would be artificially inseminated in the flock, pregnancy rate, annual ram costs that could be saved using AI rather than natural mating, expected improvement in milk production, annual costs of semen and feed, milk price and number of years of AI usage. The study considered 77 760 possible scenarios in a 3 x 4 x 4 x 3 x 3 x 3 x 4 x 15 factorial arrangement. Analysis of variance was used to investigate the effect of each factor on farm profitability. All factors considered were statistically significant (P < 0.001), but their effect varied. The number of years using AI had the greatest effect on profitability and farmers should become aware that using AI is a long-term investment. Semen price, pregnancy rate and improvement in milk production also had substantial effects. The price of milk and feed had a considerably lower effect on profitability, as did the annual cost of maintaining rams that would be replaced by AI. A positive annual and cumulative return was achieved in the model within the first 6 years. The cost of semen was estimated at 8h to 10€ per dose for the first 5 years. Where the annual improvement in milk production was 1% of annual phenotypic mean (e.g. 3.0 kg) profitability of the scheme was improved greatly. [ABSTRACT FROM AUTHOR]

A retrospective study was conducted to evaluate temporal patterns of environmental heat stress during the in-uterus period of development (IUP) and the 3-month post-natal (PN) period of dairy heifers, and to estimate their association with the age at first calving (AFC). Data from 30 dairy herds in Northern Greece including 9098 heifers were extracted from National Cattle Database. Data (2005–2019) regarding 230,100 farm-specific ambient daily temperature and relative humidity records, were obtained from ERA5-Land. Average monthly Temperature-Humidity-Index values (THI; low≤68, and high>68) were calculated and matched for each heifer to their IUP and PN. Subsequently, Cluster Analysis was used with monthly THIs as predictors to allocate heifers to THI clusters. The association of clusters with AFC was assessed with Generalized Linear Mixed Model analysis, an extended form of multiple linear regression. Finally, 8 Heat Stress Clusters (HSC; namely HSC-1 to HSC-8) were identified. Compared to HSC-8 (8th-9th IUP months and 1st PN month) heifers of HSC-5 (4th-7th IUP months) and HSC-6 (6th-8th IUP months) calved 13.8 and 17.8 days later, respectively (P < 0.01–0.001). Moreover, when AFC was treated as a binary variable, heifers of HSC-5 and HSC-6 had 1.15 and 1.34 (P < 0.01–0.001) higher risk of calving for the first time later than 787 days compared to HSC-8, respectively. • Temporal heat stress patterns identified in heifers during their fetal life. • Temporal heat stress patterns identified in heifers during post-natal period. • Specific temporal heat stress patterns can increase age at first calving. • Heat stress mitigation practices should be adopted by farmers for all dairy cows. Simple Summary: Age at first calving is an important reproductive trait of dairy cows, with long-term implications on milk yield, health, reproduction, and profitability of dairy herds. Heat stress can adversely affect the welfare and productivity of cattle. Temperature-Humidity-Index values that quantifies the degree of heat stress in dairy cattle can be easily calculated based on climatic data. However, the effect of heat stress imposed during fetal and early life on age at first calving has not been examined, yet. In the present study the temporal patterns of environmental heat stress imposed on future Holstein dairy heifers during their in-uterus development and 3-month post-natal period and the association of those patterns with age at first calving were investigated. Results offer a deeper understanding in the future impacts of heat stress in subsequent reproductive life of young heifers. Exposure of heifers to heat stress during their very early stages of life can later increase age at first calving from one up to three weeks. This important finding warrants caution in evaluating the long-term implications of heat stress in reproduction. Heat stress mitigation practices should be adopted by dairy farmers. The aim was to assess the intensified dairy sheep farming systems of the Chios breed in Greece, establishing a typology that may properly describe and characterize them. The study included the total of the 66 farms of the Chios sheep breeders' cooperative Macedonia. Data were collected using a structured direct questionnaire for in-depth interviews, including questions properly selected to obtain a general description of farm characteristics and overall management practices. A multivariate statistical analysis was used on the data to obtain the most appropriate typology. Initially, principal component analysis was used to produce uncorrelated variables (principal components), which would be used for the consecutive cluster analysis. The number of clusters was decided using hierarchical cluster analysis, whereas, the farms were allocated in 4 clusters using k-means cluster analysis. The identified clusters were described and afterward compared using one-way ANOVA or a chi-squared test. The main differences were evident on land availability and use, facility and equipment availability and type, expansion rates, and application of preventive flock health programs. In general, cluster 1 included newly established, intensive, well-equipped, specialized farms and cluster 2 included well-established farms with balanced sheep and feed/crop production. In cluster 3 were assigned small flock farms focusing more on arable crops than on sheep farming with a tendency to evolve toward cluster 2, whereas cluster 4 included farms representing a rather conservative form of Chios sheep breeding with low/intermediate inputs and choosing not to focus on feed/crop production. In the studied set of farms, 4 different farmer attitudes were evident: 1) farming disrupts sheep breeding; feed should be purchased and economies of scale will decrease costs (mainly cluster 1), 2) only exercise/pasture land is necessary; at least part of the feed (pasture) must be home-grown to decrease costs (clusters 1 and 4), 3) providing pasture to sheep is essential; on-farm feed production decreases costs (mainly cluster 3), and 4) large-scale farming (feed production and cash crops) does not disrupt sheep breeding; all feed must be produced on-farm to decrease costs (mainly cluster 3). Conducting a profitability analysis among different clusters, exploring and discovering the most beneficial levels of intensified management and capital investment should now be considered. Body condition score (BCS), energy content (EC), cumulative effective energy balance (CEEB), and blood serum concentrations of glucose, β-hydroxybutyrate (BHBA), and nonesterified fatty acids (NEFA) were measured throughout first lactation in 497 Holstein cows raised on a large commercial farm in northern Greece. All these traits are considered to be indicators of a cow's energy balance. An additional measure of BCS, EC, and blood serum glucose, BHBA, and NEFA concentrations were taken approximately 2 mo (61 ± 23 d) before first calving. During first lactation, first service conception rate, conception rate in the first 305 d of lactation, interval from calving to conception, number of inseminations per conception, incidence of metritis, and incidence of reproductive problems of these cows were recorded; interval between first and second calving, and second lactation first service conception rate were also recorded. Random regression models were used to calculate weekly animal breeding values for first lactation BCS, EC, CEEB, glucose, BHBA, and NEFA. Single trait animal models were used to calculate breeding values for these traits measured on pregnant heifers before calving. Reproductive records were then regressed on animal breeding values for these energy balance-related traits to derive estimates of their genetic correlations. Several significant estimates were obtained. In general, traits that are known to be positively correlated with energy balance (BCS, EC, CEEB, and glucose) were found to have a favorable genetic relationship with reproduction, meaning that increased levels of the former will lead to an enhancement of the latter. On the other hand, traits known to be negatively correlated with energy balance (BHBA and NEFA) were found to have an unfavorable genetic association with reproductive traits. Body condition score, BHBA, and NEFA recorded early in lactation, and glucose concentrations measured in pregnant heifers had the highest genetic correlation with future reproductive performance. Results suggest that genetic selection for body energy and blood metabolites could facilitate the genetic improvement of fertility and overall reproductive efficiency of dairy cows. The objectives of this study were to characterize the changes of body condition score (BCS), energy content (EC), cumulative effective energy balance (CEEB), and blood serum concentrations of glucose, β-hydroxybutyrate (BHBA), and nonesterified fatty acids (NEFA) across the first lactation of Holstein cows, and to estimate variance components for these traits. Four hundred ninety-seven cows kept on a commercial farm in Greece that had calved during 2005 and 2006 were used. Body condition score, estimated live weight, and blood metabolic traits were recorded weekly for the first 3 mo of lactation and monthly thereafter until the end of lactation. Body condition score and estimated live weight records were used to calculate EC and CEEB throughout the first lactation. Estimates of fixed curves and genetic parameters for each trait, by week of lactation, were obtained with the use of random regression models. The estimated fixed curves were indicative of changes in the metabolic process and energy balance of the cows. Significant genetic variance existed in all studied traits, and was particularly high during the first weeks of lactation (except for the genetic variance of CEEB, which was not significant at the beginning of lactation). Significant heritability estimates for BCS ranged from 0.34 to 0.79, for EC from 0.19 to 0.87, for CEEB from 0.58 to 0.93, for serum glucose from 0.12 to 0.39, for BHBA from 0.08 to 0.40, and for NEFA from 0.08 to 0.35. Genetic correlations between different weeks of lactation were near unity for adjacent weeks and decreased for weeks further apart, becoming practically zero for measurements taken more than 3 to 4 mo apart, especially with regard to blood metabolic traits. Significant heritability estimates were also obtained for BCS recorded before first calving. Abstract: The aim of this study was to develop an efficient strategy for ration formulation for milking ewes of the Chios breed in Greece. The strategy involved two and three groupings according to production level and challenge feeding using lead factors. Lead factors, that adjust upwards the average production of a flock or a group of ewes, were calculated for the 83rd and the 90th percentile, using 49,237 milk test-day records from 549 flock-test-days, referring to 64 flocks and 97 complete lactations. Lead factors were 1.25 for the single-group, 1.14 and 1.17 for the two-group, and 1.11, 1.07 and 1.15 for the three-group strategy for the 83rd percentile. Regarding the 90th percentile, these were: 1.33 for the single-group, 1.18 and 1.19 for the two-group, and 1.15, 1.09 and 1.16 for the three-group strategy. Analysis of variance was used to assess the influence of several effects on lead factors. Flock-year and mean and standard deviation milk yield were significant (P <0.05) in nearly all cases, leading to calculation of different lead factors for high (>250 kg of milk/ewe/year) and low (<250 kg of milk/ewe/year) producing flocks. Higher producing flocks were associated with somewhat lower than average lead factors, while the opposite was true for lower producing flocks. In order to allow the sufficient expression of the genetic potential of the best ewes and accurately estimate their genetic value under Greek conditions, the 90th percentile strategy can be adopted for the higher producing groups and the 83rd for the lower ones. Abstract: The effect of meat appearance on consumers’ preferences for pork chops was assessed using images manipulated for appearance characteristics. Data were collected from 412 consumers in Greece and Cyprus. Consumers were asked for their preference for pork chops from a book of computer-modified images and then completed a questionnaire of socio-demographic information, including eating and purchasing behaviour. Consumers under the age of 35 years showed preferences for dark red, lean pork, while consumers aged 35 years and older preferred either dark or light red pork. Gender appeared to be an important selection factor as men showed an increased preference for dark red pork while women preferred the light red. Consumers who stated that they like pork for its taste (91%) preferred either dark or light red pork chops while those who like pork for reasons other than taste preferred dark red, lean pork. Urban consumers preferred light red, fatty pork chops while the rural consumers preferred the dark red pork chops. [Copyright &y& Elsevier]

As future climate challenges become increasingly evident, enhancing performance resilience of farm animals may contribute to mitigation against adverse weather and seasonal variation, and underpin livestock farming sustainability. In the present study, we develop novel seasonal resilience phenotypes reflecting milk production changes to fluctuating weather. We evaluate the impact of calendar season (autumn, winter and spring) on animal performance resilience by analysing 420,534 milk records of 36,908 milking ewes of the Chios breed together with relevant meteorological data from eastern Mediterranean. We reveal substantial seasonal effects on resilience and significant heritable trait variation (h 2  = 0.03-0.17). Resilience to cold weather (10 °C) of animals that start producing milk in spring was under different genetic control compared to autumn and winter as exemplified by negative genetic correlations (- 0.09 to - 0.27). Animal resilience to hot weather (25 °C) was partially under the same genetic control with genetic correlations between seasons ranging from 0.43 to 0.86. We report both favourable and antagonistic associations between animal resilience and lifetime milk production, depending on calendar season and the desirable direction of genetic selection. Concluding, we emphasise on seasonal adaptation of animals to climate and the need to incorporate the novel seasonal traits in future selective breeding programmes.

Definition and establishment of a fixed reference lactation length could provide useful tools for on-farm comparison of ewes and flock management as well as genetic evaluations for the breeding programme. The objectives of this study were to (i) evaluate different reference lactation lengths for the Chios dairy sheep and (ii) define the most suitable reference length for the breed. A total of 260 042 test-day milk records from 24 474 ewes in 130 flocks collected between 2003 and 2014 were used; 15 different lactation lengths were evaluated ranging from 120 to 260 days, defined at 10-day intervals as reference for the Chios sheep. The evaluation criteria included: (a) heritability and repeatability of milk yield in each reference lactation, (b) genetic correlation of reference lactation milk yield with actual lactation milk yield and yield at first test-day record and (c) correlated response in reference lactation milk yield from selection based on first test-day milk yield. The latter emulates genetic gains achieved for milk yield based on early lactation selection. Heritability and repeatability estimates of reference lactation milk yield and genetic correlation with actual lactation yield favoured long reference lactations (180 to 230 days). On the contrary, correlation with first test-day record milk yield was higher for short lactations (120 to 170 days). Moreover, selection on first test-day record milk yield would lead to a correlated response in reference yield in 220 days equal to 85% of the highest estimate achieved in the maximum reference length of 260 days (190 days when only considering first lactation milk yield). Based on the results of the present study, an overall reference lactation length for the Chios breed of 220 days post-lambing and a first lactation reference length of 190 days post-lambing are recommended.

Although experimental data regarding cross-protection of horse West Nile virus (WNV) vaccines against lineage 2 infections exist, the cross-protective efficacy of these vaccines under field conditions has not been demonstrated. This study was conducted to evaluate the capability of an inactivated lineage 1 vaccine (Equip WNV) to protect against natural infections from the Nea Santa-Greece-2010 lineage 2 strain. In total, 185 WNV-seronegative horses in Thessaloniki, Greece, were selected during 2 consecutive years (2011 and 2012); 140 were immunized, and 45 were used as controls. Horses were examined for signs compatible with WNV infection. Neutralizing antibody titers against the Greek strain and the PaAn001/France lineage 1 strain were determined in immunized horses. WNV circulation was detected during both years in the study area. It was estimated that 37% and 27% of the horses were infected during 2011 and 2012, respectively. Three control animals developed clinical signs, and the WNV diagnosis was confirmed. Signs related to WNV infection were not observed in the vaccinated animals. The nonvaccinated animals had a 7.58% ± 1.82% higher chance of exhibiting signs than immunized animals (P < 0.05). Neutralizing antibodies raised against both strains in all immunized horses were detectable 1 month after the initial vaccination course. The cross-protective capacity of the lowest titer (1:40) was evident in 19 animals which were subsequently infected and did not exhibit signs. Neutralizing antibodies were detectable until the annual booster, when strong anamnestic responses were observed (geometrical mean titer ratio [GMTR] for lineage 1 of 30.2; GMTR for lineage 2 of 27.5). The objective of this study was to examine the prion protein gene locus (PRNP) in Chios sheep. PRNP is linked with scrapie resistance in small ruminants. Here, its impact on milk production (test-day and total lactation yield) and reproduction (age at first lambing, conception rate at first service, and prolificacy) was assessed. Genotyping at codons 136, 154 and 171 (classical scrapie) and 141 (atypical scrapie) was performed using DNA from milk somatic cells and PCR-RFLP analysis. A total of 1013 Chios ewes raised in 23 flocks were used. This constituted a random sample of the national breeding population. A total of 15 genotypes and 6 alleles linked to codons 136, 154 and 171 were detected. All animals were homozygous for the leucine allele at codon 141. Linear mixed models were used to assess the impact of PRNP genotypes and alleles on milk production and reproduction traits. The TRQ allele, whose association with such traits was assessed for the first time, had an adverse effect on age at first lambing. All other PRNP alleles, including ARR, which is associated with increased resistance to classical scrapie, had no significant effect on the traits studied. No significant associations of the PRNP genotypes with production and reproduction traits were observed. The association between PRNP variation and scrapie incidence was investigated in a highly affected Greek goat herd. Four mutations were identified at codons 171Q/R, 211R/Q, 222Q/K and 240P/S. Lysine at codon 222 was found to be associated with the protection from natural scrapie (P=0.0111). Glutamine at codon 211 was observed in eight animals, all of them being scrapie-negative, indicating a possible protective role of this polymorphism although statistical analysis failed to support it (P=0.1074). A positive association (P=0.0457) between scrapie-affected goats and the wild-type Q(171)R(211)Q(222)S(240) allele is presented for the first time. In addition, a novel R(171)RQS allele, which is identical to the A(136)R(154)R(171) allele that has been associated with resistance to classical scrapie in sheep, was observed in low frequency. Resistant alleles that include K(222) and Q(211) are absent or rare in sheep and can provide the basis for the development of a feasible breeding programme for scrapie eradication in goats.

The objective of this study was to examine the impact of polymorphisms in the acyl-CoA:diacylglycerol acyltransferase (DGAT1), leptin and growth hormone receptor genes on body energy (body condition score, total body energy content and cumulative effective energy balance) and blood metabolic traits (levels of beta-hydroxybutyrate, glucose and non-esterified fatty acids), measured once before the first calving and then repeatedly throughout first lactation in 497 Holstein cows. The influence of the same polymorphisms on cow reproductive performance and health during the first and second lactations was also assessed. Several reproductive traits were considered including interval, conception and insemination traits, as well as incidence of metritis and reproductive problems. Genotyping was performed using PCR-RFLP (DGAT1, leptin) or allele-specific PCR (growth hormone receptor). For each locus, the effect of allele substitution on body energy and blood metabolic traits was estimated using random regression models. The same effect on reproductive traits was assessed with single-trait mixed linear models. Significant (P<0.05) effects on specific reproductive traits were observed. DGAT1 and growth hormone receptor alleles responsible for significant increases in milk production were found to have an adverse effect on reproduction, while the leptin allele responsible for significant increase in milk production was linked to marginally increased metritis frequency. Furthermore, the three studied loci were also found to significantly (P<0.05) affect certain body energy and blood metabolic traits. Several associations are published for the first time, but these should be confirmed by other investigators before the polymorphisms are used in gene-assisted selection.

Organising the breeding plan of a seasonally breeding species, such as sheep, presents a challenge to farmers and the industry as a whole, since both economical and biological considerations need to be carefully balanced. Understanding the breeding activity of individual animals becomes a prerequisite for a successful breeding program. This study set out to investigate the sources of variation and the genetic profile of the spontaneous, out-of-season ovulatory activity of ewes of the Chios dairy sheep breed in Greece. The definition of the trait was based on blood progesterone levels, measured before exposing the ewes to rams, which marks the onset of the usual breeding season. Data were 707 records, taken over two consecutive years, of 435 ewes kept at the Agricultural Research Station of Chalkidiki in northern Greece. When all available pedigree was included, the total number of animals involved was 1068. On average, 29% of all ewes exhibited spontaneous, out-of-season ovulatory activity, with no substantial variation between the years. Significant sources of systematic variation were the ewe age and live weight, and the month of previous lambing. Older, heavier ewes, that had lambed early the previous autumn, exhibited more frequent activity. Heritability estimates were 0.216 (+/-0.084) with a linear and 0.291 with a threshold model. The latter better accounts for the categorical nature of the trait. The linear model repeatability was 0.230 (+/-0.095). The results obtained in this study support the notion that spontaneous out-of-season ovulatory activity can be considered in the development of a breeding plan for the Chios sheep breed.
