This paper aims to determine the content and bioaccumulation patterns of eight trace elements (Li, Cr, Co, As, Se, Cd, Pb and Hg) and one radioisotope (U) in kidney, liver and muscle tissues of two fish species (Squalius orpheus and Barbus strumicae) from Nestos River (northern Greece), and the potential human health risk due to the fish muscle consumption. Kidney and liver accumulate higher element concentrations, except Hg. The site closer to the river-mouth presents higher concentrations since it receives effluents from agricultural activities. The upstream station shows lower element concentrations, due to obstruction of the free transportation of sediments and suspended matter downstream the irrigation dam. The sediment inhabitant and feeder B. strumicae showed elevated concentrations compared to the water column resident S. orpheus. It was previously reported that liver glucose metabolism in rats under caloric restriction differs from that of freely-fed rats. This study hypothesized that these changes (1) were related to the expression of hypothalamic neuropeptides involved in metabolic control, and (2) were not a residual effect of litter size. To those purposes, liver glucose metabolism and hypothalamic expression of the orexigenic neuropeptides NPY (neuropeptide Y) and AgRP (agouti gene-related peptide); and of the anorexigenic neuropeptides POMC (pro-opiomelanocortin) and CART (cocaine- and amphetamine-related transcripts) were investigated. Male Wistar rats from two different litter sizes (G6 and G12, with 6 or 12 pups, respectively) were subjected to free feeding (GL, ad libitum), 50% caloric restriction (GR) or caloric restriction+ ad libitum refeeding (GRL) until the age of 90 days. Biometric values were lower in GR than in GL, while in GRL they were totally or partially recovered. Blood glucose variation during the pyruvate tolerance test (PTT) was small in GR. During in situ liver perfusion, total, basal, and adrenaline-stimulated liver glucose outputs were high in GR, but additional glucose output in the presence of alanine was negligible. Refeeding (GRL) yielded values close to those of GL. Litter size did not consistently influence any of these variables. The expression of transcripts of the hypothalamic neuropeptides was responsive to feeding regimen, litter size and/or their interaction and differed from G6 to G12, while the metabolic changes of the liver were qualitatively equal in both GR. Therefore, the changes in glucose metabolism in the liver of rats under caloric restriction were not determined by either litter size or hypothalamic neuropeptide expression and were linked only to the prevailing feeding regimen of the adult animal. Lacustrine ecosystems have been altered by accelerating pollution, excessive nutrient and organic load, water abstraction, and are susceptible to climate change. Hence, suggesting sensitive and reliable biomarkers for early assessments of their status is of urgent need. In this study, two freshwater commercial fish species, Cyprinus carpio (carp) and Carassius gibelio (prussian carp) from two lakes (i.e. Koronia and Volvi, Northern Greece) with different anthropogenic pressures were used and a battery of biochemical and molecular biomarkers related to stress response were analyzed in fish gills and liver. In parallel, water physicochemical parameters (T, DO, pH, conductivity, salinity), BOD 5 and nutrient (N-NO 3 , N-NO 2 , N-NH 4 , P-PO 4 ) concentrations were measured. Results showed that Lake Koronia had higher conductivity and salinity values and N-NO 2 concentrations. Levels of Heat Shock Response (HSR), MAPK phosphorylation, protein carbonylation, lipid peroxidation products, Bax/Bcl-2 ratio, ubiquitination and caspases were increased in gills and liver of both fish species sampled from Lake Koronia in relation to those of Lake Volvi. Likewise, liver lipid content was increased in both fish species sampled from Lake Koronia compared to those sampled from Lake Volvi. The results indicate and reflect the higher environmental degradation that prevails in Lake Koronia ecosystem in comparison to that of Lake Volvi. The fish species studied showed different susceptibility depending on the biomarkers examined. In addition, our results from both examined species provide insight into the mechanisms involved in acclimatization to stressful environments and support the role of the studied biomarkers as sensitive and reliable tools for ecological assessments of lake ecosystems in biomonitoring studies., (Copyright © 2019. Background: Collagen proportionate area (CPA) measurement is a technique that quantifies fibrous tissue in liver biopsies by measuring the amount of collagen deposition as a proportion of the total biopsy area. CPA predicts clinical outcomes in patients with HCV and can sub-classify cirrhosis., Aim: To test the ability of CPA to quantify fibrosis and predict clinical outcomes in patients with NAFLD., Methods: We assessed consecutive patients with biopsy-proven NAFLD from three European centres. Clinical and laboratory data were collected at baseline and at the time of the last clinical follow-up or death. CPA was performed at two different objective magnifications, whole biopsy macro and ×4 objective magnification, named standard (SM) and high (HM) magnification respectively. The correlation between CPA and liver stiffness was assessed in a sub-group of patients., Results: Of 437 patients, 32 (7.3%) decompensated and/or died from liver-related causes during a median follow-up of 103 months. CPA correlated with liver stiffness and liver fibrosis stage across the whole spectrum of fibrosis. HM CPA was significantly higher than SM CPA in stages F0-F3 but similar in cirrhosis, reflecting a higher ability to capture pericellular/perisinusoidal fibrosis at early stages. Age at baseline (HR: 1.04, 95% CI: 1.01-1.08), HM CPA (HR: 1.04 per 1% increase, 95% CI: 1.01-1.08) and presence of advanced fibrosis (HR: 15.4, 95% CI: 5.02-47.84) were independent predictors of liver-related clinical outcomes at standard and competing risk multivariate Cox-regression analysis., Conclusions: CPA accurately measures fibrosis and is an independent predictor of clinical outcomes in NAFLD; hence it merits further evaluation as a surrogate endpoint in clinical trials., (© 2019 John Wiley & Sons Ltd.)

National registries constitute an invaluable source of information and contribute to the improvement of hemoglobinopathy management. Herein, we present the second updated report of the National Registry for Haemoglobinopathies in Greece (NRHG) and critically discuss the time trends in demographics, affected births, and causes of mortality. Thirty-eight Greek hemoglobinopathy units reported data from diagnosis to the last follow-up or death by retrospectively completing an electronic form. Four thousand thirty-two patients were eligible for inclusion; more than half of them had thalassaemia major. Compared to the previous report, a reduction in the total number of all hemoglobinopathies except for hemoglobinopathy "Η" was evident. The total number of affected births was also reduced; most of them were attributable to diagnostic errors and lack of awareness. Importantly, data on iron overload are reported for the first time; although most patients had low or moderate liver iron concentration (LIC) values, a non-negligible proportion of patients had high LIC. The burden due to heart iron overload was less prominent. Cardiac- and liver-related complications are the major causes of morbidity and mortality. From 2000 to 2015, a decrease in heart-related deaths along with an increase in liver-associated fatalities was observed. The Hellenic Prevention Program along with advances in chelation regimens and iron status monitoring have resulted in improved patient outcomes. The NRHG gives insight into the effectiveness of prevention programs, the therapeutic management of hemoglobinopathies and associated outcomes. NRHG may contribute to the formulation of a roadmap for hemoglobinopathies in Europe and promote the implementation of effective public health policies.

Non-alcoholic fatty liver disease (NAFLD) in children has been recognized as a major health burden. The high prevalence of NAFLD is probably due to the contemporary epidemics of obesity, unhealthy dietary pattern, and sedentary lifestyle. The purpose of this study was to investigate anthropometric, biochemical and dietary intake parameters of obese Greek children with and without NAFLD. Eighty two obese children aged 8-15 (45 boys/37 girls) participated in the study. Ultrasonography (US) was used to diagnose NAFLD in all participated subjects. Liver indexes were measured in all children. A 3-day dietary was recorded for all subjects. Data for continuous variables is expressed as mean values±standard deviation. Thirty-five out of eighty two subjects (42.6%) had NAFLD. Body Mass Index levels (P<0.001) and Waist Circumference (P<0.046) levels were statistically higher in the children with severe NAFLD (37.2kg/m(2)±6.2 and 102.9cm±14) compared to children with mild NAFLD (26.6kg/m(2)±3.3 and 86.1cm±9.9) and absent of fatty liver (25.3kg/m(2)±3.6 and 85.2cm±11.2), respectively. Total carbohydrates intakes were significantly higher in subjects with NAFLD (288.8g±70.6) compared to children without NAFLD (244.5g±67.5), (P<0.001). Saturated fatty acids (SFAs) intake was proportionally increased to the degree of hepatic steatosis, while n-3 fatty acids (n-3 FA) consumption was inversely related with NAFLD. In multiple regression analysis of factors associated with NAFLD, HOMA-IR (OR: 1.260, 95%CI: 1.110-1.470, P<0.001) and n-3FA (OR:1.921, 95%CI:1.132-2.187, P<0.001) were the most significant ones. Background: Primary biliary cirrhosis (PBC) is an organ specific autoimmune disease of still unidentified genetic etiology. We have shown that endothelins (ETs), produced by the liver endothelial cells are increased in PBC and may play a major pathogenetic role., Aims: To study gene polymorphisms related to the endothelial cells (eNOS, EDN-1 genes) and, to investigate whether the previously reported association of CTLA4 gene polymorphisms is replicated in a genetically homogeneous Greek population., Patients and Methods: Genomic DNA was extracted from 100 PBC patients (83 females, 93% AMA+, 74/100 Ludwig stage I-II) and 158 healthy controls. eNOS, CTLA4 and ET1 polymorphisms were determined by PCR-RFLPs analysis., Results: Both eNOS intron4 VNTR and eNOS exon7 G894T SNP were significantly associated with increased risk in PBC. EDN-11 rs2071942 "A" and rs5370 "T" alleles appeared a tendency for association with disease progression. No association was found between PBC and the CTLA4 SNPs analyzed., Conclusions: We demonstrated that eNOS, a gene related to the liver endothelium function is associated with PBC. Contrarily, the important in adaptive immunity gene CTLA4 was not associated with the disease in the homogeneous population analyzed. These results are compatible partially with our previous hypothesis that defects of the liver endothelial system, leading to endothelin overproduction, may be a fundamental early pathogenetic mechanism in PBC., (Copyright © 2012 American Society for Histocompatibility and Immunogenetics. The concentrations of persistent organic pollutants, such as DDT and its metabolites (DDTs), hexachlorocyclohexane isomers (HCHs), cyclodienes (Cycls), and polychlorinated biphenyls (PCBs), were measured in livers and subcutaneous fat tissues of six Accipitridae and four Falconidae bird species from different areas in Greece. This is the first report of persistent organochlorine (OC) pollutants in birds of prey tissues presented for Greece and the Eastern Mediterranean region. Accumulation patterns of OCs found in birds suggested that the predominant contaminants were p,p'-dichlorodiphenyldichloro-1,1-ethylene (DDE) and PCBs, whereas Cycls and HCHs occurred at low concentrations only. Concentration values of p,p'-DDE ranged from nondetected to 19,518.72 ng/g wet wt in livers and from nondetected to 2679.19 ng/g wet wt in fat. Total PCB levels ranged from 1.01 to 7419.43 ng/g and from 3.25 to 490.10 ng/g wet weight for liver and fat samples, respectively. Higher-chlorinated PCBs such as 118, 138, 153, and 180 predominated in both the liver and subcutaneous fat samples, a pattern comparable to that observed in birds from other European countries. No significant differences in mean concentrations of OCs are detected between species. Hepatic concentrations were in general higher than the fat concentrations showing depleted fat stores in most birds. Concentration ranges were also found in lower or similar levels to those reported for birds in other regions. Variation of OCs levels in bird tissues could be due to different causes of death, with a subsequent effect on body lipid levels, and different feeding and migration habits. The liver PCB levels reported in this study are below the concentrations currently believed to exert mortality or ecotoxicological effects. On the contrary, in some cases p,p'-DDE concentrations were higher than the reported effect values for birds of the same families and could be associated with sublethal effects.

Copper liver and kidney concentrations were evaluated from sheep grazing at 2 areas: A (sampling points A1, A2, A3) and B. One hundred forty specimens were measured by flame atomic absorption spectroscopy after wet digestion for the February through April 1995 sampling period. Mean liver concentrations were: Area A (A1 = 99.48, A2 = 62.28, A3 = 23.02 mg/kg ww) and Area B 74.07 mg/kg ww. Copper concentrations in kidneys from all areas were in the normal range.

Ten cases of acute paraquat poisonings with lethal (n = 5) and non-lethal (n = 5) outcomes are presented. Paraquat plasma and urine levels of the patients on admission (from 2 to 41 h after ingestion) at the peripheral hospitals varied from 0.4 to 6.0 and 0.5 to 12.8 micrograms/ml, respectively. Besides standard supportive treatment, hemoperfusion/hemodialysis and/or plasmapheresis were performed for therapy, which in 1 case lasted for 28 d. The pharmacokinetic data from patient paraquat elimination confirmed considerable paraquat rebound from tissues to blood circulation and marked the need for continued therapy even with undetectable paraquat plasma levels. Postmortem analysis showed that while paraquat was mainly distributed in kidney (807 micrograms/g), lung (479 micrograms/g) and liver (206 micrograms/g), amounts were also detected in thyroid (64 micrograms/g), testis (21 micrograms/g), vitreous humor (45 micrograms/ml) and cerebrospinal fluid (7.4 micrograms/ml). Paraquat crossed the placenta and concentrated there to higher levels than in the mother's blood. The fetus seemed to tolerate the mother's paraquat intoxication while dependent on placental circulation.

During prehibernating season (September-November) a part of lipid is mobilized from fat bodies to other body sites or is transformed to other metabolites. 4. During wintering this frog consumes mainly glycogen. 5. In February the lipid is accumulated in the fat bodies and the liver mass shows a second peak, probably as a result of glycogen accumulation. 6. The greatest decrease of metabolites was observed during the breeding season and this is the result of the intensive activities related to the reproduction and maintenance.
