Background: Escherichia coli cause a wide range of infections in humans and animals in hospitals and community associated with multidrug-resistance, and serves as a bioindicator of fecal contamination as well as antimicrobial resistance (AMR). Aim: Early detection of this bioindicator from humans, animals, and environment facilitates characterization with respect to AMR and bacterial forensics for effective public health and infection control measurements. Method: We investigated a simple and rapid species-specific end-point and real-time PCR assays targeting malate dehydrogenase (mdh) gene to detect E. coli isolated from livestock, human, and environmental water in Midnapore, West Bengal, India, in a timely and cost-effective approach. Results: Of 126 E. coli isolates, 53, 41, and 32 were cultured from livestock, human, and environmental water. The mdh gene was present in 120 (95.2%) isolates, including all environmental isolates. Two (3.8%) animal and four human (9.8%) isolates were discordant between culture and PCR results. The PCR assays produced results in about 90 minutes with a running cost of US$3-5 per sample. Classification of water quality is a valuable method for reporting quality status of a water body that can help to control pollution in monitored sites. In this study water quality of rivulet Holy Kali Bein in the state of Punjab, India was analysed by using water quality index (WQI) and multivariate statistical analysis on the bases of measured water quality parameters from five sampling sites (K1-K5) during the period 2017–2018. Discriminant analysis (DA) and WQI were applied to examine spatiotemporal changes and to spot possible pollution sources. These changes in water quality parameters were analysed by using stepwise DA method. The first spatial discriminate function (DF1) accounted for 96.9%, second discriminate function (DF2) accounted 2.1%, DF3 accounted for 0.8% and DF4 accounted only 0.2% of the total spatial variance. Similarly, the first temporal DF1 accounted for 80.1% and DF2 accounted for 19.9% of total temporal variance. The water quality index (WQI) score showed good water quality at sites K1 (30.67), K2 (43.98) and K3 (51.25). The sites K4 (64.50) and K5 (62.89) showed poor water quality status. The present investigation represents the first of its type undertaken on the rivulet Holy Kali Bein and shows that the water quality of the rivulet is good at origin and when we move towards its confluence point at Harike wetland (Ramsar site) water quality becomes poor due to input of municipal wastes and agricultural activities discharge at the bank of the rivulet. From decades it has been observed that male infertility has boomed over time as the industrialisation is augmented. Explanation of this downfall has been associated to environmental factors and escalation of the AQI in industrialised cities. Currently, it has been hypothesised that the defect in foetal development can also be from the origin of fragmented DNA of sperms in semen sample. To determine the effect of AQI on DNA fragmentation, this study aims to evaluate the differences in sperm DNA fragmentation in places with diverse levels of AQI. A retrospective analysis of 3222 patients was performed of the patients aged between 21-40 years during January 2023 to January 2024 at 120 centres of Indira IVF Hospital Private Limited, India. The patients were divided based on the AQI levels of the places – less polluted (50-100) (n=1384), moderately polluted (101-150) (n=1838), highly polluted (>151) (n=1334). The semen analysis and DNA fragmentation testing at all the centres were done based on the WHO recommendations. The patients were divided based on their DNA fragmentation score into – normal (<25%) and abnormal (>25%). Subsequently, the impact of AQI on sperm chromosome integrity was analysed. Our examinations disclosed that areas with less pollution presented 69.3% normal sperms and 30.7% abnormal sperms. However, as the pollution escalates, the area with moderate pollution showed a significant uprise in abnormal sperms and significant downfall of normal sperms by 8.8% compared with sperms of the population found in less polluted region. In the highly polluted zone, it was discovered that the abnormal sperm has a significant increase and normal sperms has a significant decrease by 11% compared with results found in less polluted area. Our findings underscore a pollution bias determining that as the pollution escalates the abnormal sperms exponentially augments. However, the normal sperms inversely corelates to the enhancement of pollution. Triterpenoids and fatty alcohols were identified from the sediments of five mangrove ecosystems (Kunjimangalam, Pazhayangadi, Pappinissery, Thalassery, and Kadalundi) in the northern Kerala coast, India, to illustrate the organic matter sources. Straight chain fatty alcohols in the range C12–32 were detected, and among these the long chain fatty alcohols (C20–32) were exclusively even chained compounds. Branched fatty alcohols (6,10,14-trimethyl-pentadecan-2-ol and 3,7,11,15-tetramethyl-2-hexadecanol) of bacterial origin were detected in a few observations. Twenty triterpenoids were detected from the study region in which 13 of them were sterols. Triterpenoids originated from higher plants such as stigmasterol, campesterol, β-sitosterol, taraxerol, lupeol, germanicol, β-amyrin, α-amyrin, and betulin were identified in the study. Brassicasterol, a diatom marker and dinosterol, a dinoflagellates marker were also detected in the study region. An elevated % composition of dinosterol and dissolved oxygen super-saturation were noticed in Kunjimangalam during pre-monsoon, which may suggest elevated input of algal organic matter in the mangrove system. Cholesterol transformation products such as coprostanol and epi-coprostanol were detected, which indicated the presence of fecal contamination in the sedimentary organic matter. Introduction: Outbreaks of acute watery diarrhoea are common in developing countries having poor access to safe drinking water and sanitation. An outbreak of acute watery diarrhoea in a village in North India was investigated with the aim to initiate and recommend necessary actions to control it. Methodology: A house to house survey using a specially designed case record form was conducted covering all the households. Stool samples of some of the affected individuals and twenty water samples during and after the outbreak from various sites were collected for laboratory analysis. Results: Out of 624 inhabitants surveyed, 118 were found to be suffering from acute watery diarrhoea (Overall attack rate of 18.9%) with two suspected deaths following diarrhoea. Males and females were affected equally and the age group of 15-44 years was affected predominantly. A peculiar epidemic curve with single peak was noted. One of the stool samples collected during the outbreak grew Vibrio Cholera O1(Ogawa) and twelve out of the twenty water samples including the samples from both the tube wells had high coliform counts indicating fecal contamination. Chlorine levels in all the water samples were found to be inadequate. Conclusions: Local cultural practices such as indiscriminate defecation in public places, using tullu pumps to extract water from the public supply line, poor engineering design and maintenance of the water supply system having leakages at many sites along with inadequate chlorination of the supply water from the tube wells were the risk factors that could have contributed to this outbreak. Contamination of drinking water due to fluoride (F−) is a major concern worldwide. Although fluoride is an essential trace element required for humans, it has severe human health implications if levels exceed 1.5 mg. L−1 in groundwater. Several treatment technologies have been adopted to remove fluoride and reduce the exposure risk. The present article highlights the source, geochemistry, spatial distribution, and health implications of high fluoride in groundwater. Also, it discusses the underlying mechanisms and controlling factors of fluoride contamination. The problem of fluoride-contaminated water is more severe in India's arid and semiarid regions than in other Asian countries. Treatment technologies like adsorption, ion exchange, precipitation, electrolysis, electrocoagulation, nanofiltration, coagulation-precipitation, and bioremediation have been summarized along with case studies to look for suitable technology for fluoride exposure reduction. In this study presence of virulence genes in multidrug resistant Escherichia coli isolated from Mula-Mutha river, Pune, India was undertaken. The objective was to understand whether the isolates were of diarrhoeagenic or of environmental origin. This was essential since the river flows through urban and rural parts of Pune and its water is used not only for industrial and agricultural purposes but also for domestic usage. One hundred and two multidrug E. coli isolates were selected from our previous study which detected genes coding for antibiotic resistance as well as identified integrons associated with multidrug resistance. Isolates were subjected to multiplex PCR to detect presence of virulence genes, set1A, set1B, sen astA, aggA, aafA, pet, stx1 and stx. Sequencing was performed to confirm the amplified PCR product. Seven of the 102 E. coli isolates showed gene set1A alone identifying them as Enteroaggregative E. coli. Thus, the findings revealed that majority of drug resistant E. coli were environmental in origin. The presence of antibiotic resistant genes, integrons in the environment as well as diarrhoeagenic E. coli isolates is a warning and calls for efficient public health measures to ensure that untreated sewage and industrial waste does not enter the Mula-Mutha river. Haryana is one of the leading states in India in the agricultural and industrial production. With the expansion of these sectors, a continuous increase in water demand is leading to water crises arising from overexploitation and quality deterioration of the available water. Contamination of aquifer resources is a significant concern, because majority of population depends on the groundwater for various agricultural, industrial, and domestic needs. This review article provides an overview of groundwater contamination, associated health risks with different contaminants with regions severely affected by poor water quality, and delves in identifying the sources, by observing and recognising the types of industries dominant in the state with types of effluents discharge. It further suggests the possible mitigation measures such as advanced remedial technologies and proper management practices from the consequent contamination sources. It has been observed during the perusal of various studies and data that the degree of contamination was considerably higher in districts with heavy agro-industrial activities. The groundwater resources in three highly industrialized districts were found to be gravely contaminated with toxic heavy metals. Alongwith heavy metals, the salinity, hardness, nitrate, and fluoride are also posing significant problems in the aquifer resources of Haryana state. The present study was designed to analyse the microbiological quality of groundwater in open-defaecation-free (ODF) villages of the Kurukshetra district in Haryana. Twenty-seven groundwater samples were collected from borewells of nine ODF villages located in three different blocks of the Kurukshetra district in three seasons and analysed for total and faecal coliform count. The presence of gram-negative, rod-shaped bacteria was observed in all the groundwater samples and the dominating genera found were Klebsiella sp. All strains were found to be resistant to clindamycin (100%) followed by gentamicin (66.6%), azithromycin and streptomycin (50%) and chloramphenicol (33.3 %) antibiotics. Minimum resistance of isolates was observed for antibiotic norfloxacin, ciprofloxacin (16.66%) and zero resistance were observed against tetracycline. The Multiple Antibiotic Resistance (MAR) index values indicated that in a majority of the samples (66.6%), the source of contamination was non-human, but nearly 33.3% were found to be contaminated by some human sources. Efforts to eradicate open defecation and improve sanitation access are unlikely to achieve health benefits unless interventions reduce microbial exposures. This study assessed human fecal contamination and pathogen exposures in rural India, and the effect of increased sanitation coverage on contamination and exposure rates. In a cross-sectional study of 60 villages of a cluster-randomized controlled sanitation trial in Odisha, India, human and domestic animal fecal contamination was measured in community tubewells and ponds (n = 301) and via exposure pathways in homes (n = 354), using Bacteroidales microbial source tracking fecal markers validated in India. Community water sources were further tested for diarrheal pathogens (rotavirus, adenovirus and Vibrio cholerae by quantitative PCR; pathogenic Escherichia coli by multiplex PCR; Cryptosporidium and Giardia by immunomagnetic separation and direct fluorescent antibody microscopy). Exposure pathways in intervention and control villages were compared and relationships with child diarrhea examined. Human fecal markers were rarely detected in tubewells (2.4%, 95%CI: 0.3–4.5%) and ponds (5.6%, 95%CI: 0.8–10.3%), compared to homes (35.4%, 95%CI: 30.4–40.4%). In tubewells, V. cholerae was the most frequently detected pathogen (19.8%, 95%CI: 14.4–25.2%), followed by Giardia (14.8%, 95%CI: 10.0–19.7%). In ponds, Giardia was most often detected (74.5%, 95%CI: 65.7–83.3%), followed by pathogenic E. coli (48.1%, 95%CI: 34.8–61.5%) and rotavirus (44.4%, 95%CI: 34.2–54.7%). At village-level, prevalence of fecal pathogen detection in community drinking water sources was associated with elevated prevalence of child diarrhea within 6 weeks of testing (RR 2.13, 95%CI: 1.25–3.63) while within homes, higher levels of human and animal fecal marker detection were associated with increased risks of subsequent child diarrhea ( P = 0.044 and 0.013, respectively). There was no evidence that the intervention, which increased functional latrine coverage and use by 27 percentage points, reduced human fecal contamination in any tested pathway, nor the prevalence of pathogens in water sources. In conclusion, the study demonstrates that (1) improved sanitation alone may be insufficient and further interventions needed in the domestic domain to reduce widespread human and animal fecal contamination observed in homes, (2) pathogens detected in tubewells indicate these sources are microbiologically unsafe for drinking and were associated with child diarrhea, (3) domestic use of ponds heavily contaminated with multiple pathogens presents an under-recognized health risk, and (4) a 27 percentage point increase in improved sanitation access at village-level did not reduce detectable human fecal and pathogen contamination in this setting. We compared host-associated Bacteroidales qPCR assays developed in the continental United States and Europe for the purpose of measuring the effect of improved sanitation on human fecal exposure in rural Indian communities where both human and animal fecal loading are high. Ten candidate Bacteroidales qPCR assays were tested against fecal samples (human, sewage, cow, buffalo, goat, sheep, dog and chicken) from a test set of 30 individual human, 5 sewage, and 60 pooled animal samples collected in coastal Odisha, India. The two universal/general Bacteroidales assays tested (BacUni, GenBac3) performed equally well, achieving 100% sensitivity on the test set. Across the five human-associated assays tested (HF183 Taqman, BacHum, HumM2, BacH, HF183 SYBR), we found low sensitivity (17 to 49%) except for HF183 SYBR (89%), and moderate to high cross-reactivity with dog (20 to 80%) and chicken fecal samples (60 to 100%). BacHum had the highest accuracy (67%), amplified all sewage samples within the range of quantification (ROQ), and did not cross-react with any fecal samples from cows, the most populous livestock animal in India. Of the ruminant- and cattle-associated assays tested (BacCow, CowM2), BacCow was more sensitive in detecting the full range of common Indian livestock animal fecal sources, while CowM2 only detected cow sources with 50% sensitivity. Neither assay cross-reacted with human sources. BacCan, the dog-associated assay tested, showed no cross-reactivity with human sources, and high sensitivity (90%) for dog fecal samples. Overall, our results indicate BacUni, BacHum, HumM2, BacCan and BacCow would be the most suitable MST assays to distinguish and quantify relative amounts of human-associated and livestock/domestic animal-associated contributions to fecal contamination in Odisha, India. In this article the authors document evolving attitudes, policies and roles of stakeholders in wastewater and faecal-sludge management in India, China and Ghana. In each country there is momentum for expanding not just access to sanitation at the household/community levels, but also for greater treatment and safe end-of-life management of human excreta. Governments are increasingly looking to engage the private sector, but models of engagement that make a compelling business case and instil confidence in cost recovery will have to emerge before the private sector takes an active role in wastewater and faecal sludge treatment in low-income countries. Water-borne illness, primarily caused by fecal contamination of drinking water, is a major health burden in the state of Andhra Pradesh, India. Currently drinking water is treated at the reservoir level and supplied on alternate days, necessitating storage in households for up to 48 hrs. We hypothesized that fecal contamination occurs principally during storage due to poor water handling. In this study we tested for coliform bacteria in water samples collected at distribution points as household storage containers were filled, and then tested containers in the same households 24-36 hours after collection. We also conducted an observational survey to make an assessment of water handling and hygiene. Ninety-two percent (47/51) of samples tested at supply points were adequately chlorinated and bacterial contamination was found in two samples with no residual chlorine. Samples collected from household storage containers showed an increase in contamination in 18/50 houses (36%). Households with contaminated stored samples did not show significant differences in demographics, water handling, hygiene practices, or sanitation. Nevertheless, the dramatic increase in contamination after collection indicates that until an uninterrupted water supply is possible, the point at which the biggest health impact can be made is at the household level. Twenty-two water samples from the Thirumanimuthar River course in southern India were collected before COVID-19 lockdown and during COVID-19 lockdown periods and were analyzed for microbiological parameters (fecal coliform bacteria, total coliform bacteria, Escherichia coli, and fecal streptococci) and heavy metals (Fe, Mn, Zn, Cu, Cd, Ni, Pb and Cr). The lockdown has decreased microbial populations and heavy metals. Fe, Cu, Cd, Ni, Pb and Cr exceeded the drinking water limits, respectively, in 77%, 45%, 27%, 18%, 9% and 91% of the pre-lockdown samples. During the lockdown period, Fe, Cu and Cd concentrations in 23% and Cr in 50% of the samples exceeded the limits. Heavy Metal Pollution Index (PI) expressed that 27%, 64% and 9% of the pre-lockdown samples represented 'low', 'medium' and 'high' pollution categories, respectively, but 68% and 32% of the lockdown period samples represented 'low' and 'medium' categories, respectively. The Metal Index (MI) exposed that all samples of pre-lockdown were under the seriously affected category, whereas 54% and 46% of lockdown samples were under strongly and seriously affected categories, respectively. Health risk evaluation predicted that 95%, 91% and 86% of pre-lockdown samples and 45%, 36% and 33% of lockdown period samples were at risk among children, teenagers and adults, respectively. As there is no integrated study on river water quality of COVID-19 lockdown this work is uniquely carried out by combining heavy metal pollution, microbial contamination and human health risk evaluation. The present investigation was conducted to find the possible chromium contamination in groundwater and the related health risks in a leather industrial region of south India using Sobol sensitivity modeling. Thirty-five groundwater samples were sampled from the field sites and were analyzed for pH, TDS (Total Dissolved Solids), EC (Electrical Conductivity), F− (Fluoride), NO 3 − (Nitrate) and Cr (Chromium). The concentration of nitrate varied from 3 to 81 mg/L with a mean of 48.6 mg/L. About 57% (n = 20) of the wells surpassed the drinkable limit (45 mg/L) for NO 3 − as per World Health Organization (WHO). The fluoride ion ranged from 0.1 to 2.7 mg/L with a mean of 1.5 mg/L. Around 51% (n = 18) of the samples crossed the recommended limit of WHO for F− (1.5 mg/L). The chromium varied from 0.01 to 0.19 mg/L in groundwater with a mean of 0.1 mg/L. About 66% (n = 23) of the samples overshoot the permissible limit of WHO standards (0.05 mg/L) for Cr. The spatial distribution map of chromium in the groundwater showed that 271.76 km2 area is under risk. Based on total hazard index (THI), 66%, 46%, and 43% of the groundwater samples surpassed the allowable limit (THI > 1) for children, women and men, correspondingly. Children pose severe health risks than women and men in this region. Using Sobol sensitivity indices, three different categories of risk effects were assessed: first order effect (FOE), total effect (TE) and second order effect (SOE). In the oral sensitivity model, concentration of Cr (C w) in water and ingestion rate (IR) had the dominant role, whereas in the dermal model, skin surface area (SA) and contact fraction by skin (F) had vital role in addition to the concentration (C w). We evaluate the imprints of urbanization, landuse and lifestyle on the prevalence and provenance of antibiotic resistance in the tropical rivers of Sri Lanka (Kelani and Gin) and India (Sabarmati, and Brahmaputra River). The prevalence of E. coli in the Kelani, Sabarmati, and Brahmaputra Rivers was in the range of 10–27, 267–76,600, and <50 CFU ml−1 respectively. Isolated E. coli colonies were subjected to six antibiotics to assess their resistance. We found higher resistance to old generation antibiotics like tetracycline (TC), and sulfamethoxazole (ST) transcends the resistance for fluoroquinolones like norfloxacin (NFX), ciprofloxacin (CIP), and levofloxacin (LVX). Interestingly, both Indian rivers had exhibited relatively higher resistance to TC and ST than the Kelani river or Gin River, implying that the Sri Lankan situation is relatively less critical. At genetic level the resistance for β-lactams, fluoroquinolones and sulphonamides, were detected in many samples, as reported globally. While the resistance genes for aac-(6')-1b-cr, qnrS and sul1 were detected in both Sri Lankan and Indian Rivers, bla TEM and ampC were specific to the Indian Rivers only. Decoupling of the prevalence of metal contamination and antibiotic resistance has been noticed in India and Sri Lanka. Background: In 2016, India reported 709 acute diarrheal disease (ADD) outbreaks (> 25% of all outbreaks). Tribal populations are at higher risk with 27% not having accessibility to safe drinking water and 75% households not having toilets. On June 26, 2017 Pedda-Gujjul-Thanda, a tribal village reported an acute diarrheal disease (ADD) outbreak. We investigated to describe the epidemiology, identify risk factors, and provide evidence-based recommendations.Methods: We defined a case as ≥3 loose stools within 24 h in Pedda-Gujjul-Thanda residents from June 24-30, 2017. We identified cases by reviewing hospital records and house-to-house survey. We conducted a retrospective cohort study and collected stool samples for culture. We assessed drinking water supply and sanitation practices and tested water samples for faecal-contamination.Results: We identified 191 cases (65% females) with median age 36 years (range 4-80 years) and no deaths. The attack-rate (AR) was 37% (191/512). Downhill colonies (located on slope of hilly terrains of the village) reported higher ARs (56%[136/243], p < 0.001) than others (20%[55/269]). Symptoms included diarrhea (100%), fever (17%), vomiting (16%) and abdominal pain (13%). Drinking water from five shallow bore-wells located in downhill colonies was significantly associated with illness (RR = 4.6, 95%CI = 3.4-6.1 and population attributable fraction 61%). In multi-variate analysis, drinking water from the shallow bore-wells located in downhill colonies (aOR = 7.9, [95% CI =4.7-13.2]), illiteracy (aOR =6, [95% CI = 3.6-10.1]), good hand-washing practice (aOR = 0.4, [95%CI = 0.2-0.7]) and household water treatment (aOR = 0.3, [95%CI = 0.2-0.5]) were significantly associated with illness. Two stool cultures were negative for Vibrio cholerae. Heavy rainfall was reported from June 22-24. Five of six water samples collected from shallow bore-wells located in downhill colonies were positive for faecal contamination.Conclusion: An ADD outbreak with high attack rate in a remote tribal village was associated with drinking water from shallow downhill bore-wells, likely contaminated via runoff from open defecation areas after heavy rains. Based on our recommendations, immediate public health actions including repair of leakages at contaminated water sources and alternative supply of purified canned drinking water to families, and as long-term public health measures construction of house-hold latrines and piped-water supply initiated.
