Freshwater red algae of the order Batrachospermales are poorly studied in India and Nepal, especially on a molecular basis. During a survey in northeast India and east Nepal, six populations of the genus Sheathia were found and analyzed using molecular and morphological evidence. Phylogenetic analyses based on the rbcL gene sequences grouped all populations in a large clade including our S. arcuata specimens and others from several regions. Sheathia arcuata represents a species complex with a high sequence divergence and several smaller clades. Samples from India and Nepal were grouped in three distinct clades with high support and representing new cryptic species: a clade formed by two samples from India, which was named Sheathia assamica sp. nov.; one sample from India and one from Nepal formed another clade, named Sheathia indonepalensis sp. nov.; two samples from Nepal grouped with sequences from Hawaii and Indonesia (only 'Chantransia' stages) and gametophytes from Taiwan, named Sheathia dispersa sp. nov. Morphological characters of the specimens from these three species overlap one another and with the general circumscription of S. arcuata, which lacks the heterocortication (presence of bulbous cells in the cortical filaments) present in other species of the genus Sheathia. Although the region sampled is relatively restricted, the genetic diversity among specimens of these three groups was high and not closely related in the phylogenetic relationship with the other clades of S. arcuata. These data corroborate information from other groups of organisms (e.g., land and aquatic plants) that indicates this region (Eastern Himalaya) as a hotspot of biodiversity. [ABSTRACT FROM AUTHOR]

A diethylcarbamazine (DEC)-fortified salt intervention programme was implemented between 1982 and 1986 in Karaikal district, Union territory of Pondicherry, south India, to control Culex transmitted bancroftian filariasis. The intervention reduced the microfilaria (Mf) rate from 4.49% to 0.08%. To eliminate the residual microfilaraemia, the health department detected and treated Mf carriers from 1987 to 2005 and mass-administered drugs in 2004 and 2005. Surveillance from 1987 to 2005 revealed persistent microfilaraemia in 0.03–0.42% of the population. In 2006, we conducted a more detailed Mf survey and a child antigenaemia (Ag) survey in 15 urban wards and 17 rural villages. These surveys showed an overall Mf rate of 0.46% in the high-risk urban areas and 0.18% in the rural areas; none of the sampled children was positive for Ag. All detected Mf carriers were >20 years old. The age of the youngest Mf carrier was 30 years in urban and 21 years in rural areas, which suggests that transmission was interrupted and there was no incidence of new Mf case after cessation of DEC salt programme. Eleven of 15 urban and 15 of 17 villages were totally free from microfilaraemia. Nevertheless, three of 15 surveyed urban localities and two of 17 villages showed >1% Mf rate. Thus, it seems that (i) post-intervention very low levels of microfilaraemia can continue as long as 20 years; (ii) 0.60–0.70% Mf rate is a safe level and at this level recrudescence of infection may not occur; (iii) there can be isolated localities with >1% Mf rate and their detection for further intervention measures could be challenging in larger control/elimination programmes and (iv) the residual infection mostly gets concentrated in the adult population, in underdeveloped urban areas and in historically highly endemic or large endemic rural areas. Objective: To quantify circulating antigens in individuals with lymphatic filariasis by means of an ELISA using blood on filter strips.Method: Circulating antigens in filarial patients and normal individuals living in an area endemic for W. bancrofti infection in Madras, India were estimated using a monoclonal antibody-based ELISA.Results: All microfilaraemics showed positivity to circulating antigens whereas people with chronic pathology and 80% of the endemic normals tested negative. The antigen levels in the blood collected in the night and during day time showed positivity and there was no difference in the antigen concentration. The results of the antigen levels collected onto filter strips correlated with their corresponding plasma antigen levels (r = 0.83). In microfilaraemics, DEC treatment did not alter the levels of circulating antigens for up to one month.Conclusions: We conclude that this monoclonal antibody-based ELISA using filter strips may be used in day time and replace the existing routine night blood surveys in our endemic area in India.
