The ornamental Tagetes minuta is a herbaceous plant of the Asteraceae family. T. minuta, a species native to southern South America, is used as a condiment, as a refreshing beverage, and for medicinal purposes. In 2011, disease symptoms of yellowing, root and foot rot, drying of leaves, and plant death were observed in an ornamental marigold (T. minuta) greenhouse in Fars province. The infected plants were collected and transferred to a laboratory. Samples were washed, cut into small pieces, surface disinfested with a 0.5% NaClO solution, and cultured on Potato Dextrose Agar (PDA) acidified to pH 4.5 with 0.5% lactic acid. Based on morphological characters, the causal agent was identified as Cylindrocarpon destructans. To confirm morphological identification, DNA was extracted from isolates using a genomic DNA purification Kit. The region of internal transcribed spacers 1, 2, and 5.8S genes of rDNA were amplified using the ITS4 and ITS1 universal primer set. Fragments of 600 bp were recovered from PCR, purified, sequenced, edited, and deposited in GenBank. The isolates had a 100% identity with all the compared C. destructans sequences. The pathogenicity tests were done with a suspension of 1 x 106 conidia per ml homogenised in sterile water. The symptoms on inoculated plants were similar to those previously observed and the fungus was reisolated from the inoculated plants. Eight Cylindrocarpon isolates recovered from the trunk bases of 10-year-old grapevines showing decline symptoms from two vineyards in Bavanat (Fars province, south-western Iran) were studied. Based on phenotypical characteristics, mating experiments and molecular data, they were identified as Cylindrocarpon liriodendri. Pathogenicity was confirmed with selected isolates inoculated into 8-month-old dormant rooted cuttings of grapevine rootstock cv. 110 Richter. Root and crown rot of corn and rape seed is one of the most important disease attack to these crops at various stages of their growth and cause considerable damage to these crops. For study and identification of the Cylindrocarpon species associated with corn and rape seed plants in Khuzestan province, this study was conducted during growing season of 2006-2007. Samples were collected from different field in different region of Khuzestan In that study 207 isolates of Cylindrocarpon were recovered, isolates belonged to the 5 species which based on there frequency are as follow: C. destructans (96), C. didymum(69), C. hederae(25),), C. obtusisporum(14), C. macrodidymum (3). This is the first report of the association of these species of Cylindrocarpon with corn and rape plants in the World. Cylindrocarpon isolates specific primers ITS4 & ITS5 were used. specific primers amplified a 500-560 bp band which is a confirmation of Cylindrocarpon species. Comparsion of the generated sequences from the mentioned species with the avalaible sequences in the gene Bank, using Blast Browers indicated that all of the generated sequences beloneged to the Cylindrocarpon species. Cylindrocarpon grouping using ITS sequences was corresponding to the classification of this geneus based on morfhological characteristics.
