Background: The main subtypes of idiopathic inflammatory myopathies (IIMs)--polymyositis (PM) and dermatomyositis (DM)--are often presented as interstitial lung disease (ILD) in clinical practice; therefore, many researchers have combined the three studies into PM/DM with ILD. Methods: Using bibliometrics, the research status, progress, and hotspots of PM/DM with ILD between 2000 and 2022 were studied. Literature data on PM/DM with ILD were retrieved from the Web of Science (WoS) database for the research period. Visualization software, including VOSviewer, Pajek, CiteSpace, and Scimago Graphica were used for bibliometric analysis. Results: A total of 1555 relevant articles were obtained, and the overall research in this field showed an increasing trend. Regarding contributing countries and venues, Japan published the most articles while Rheumatology was the most prolific journal. Regarding authors, the most published article was by Wang Guochun from Changchun University of Technology in China. Keyword analysis and cocited literature cluster analysis showed that diagnosis, classification, autoantibodies, antibodies, prognosis, complications, and treatment of PM/DM with ILD have been hot topics in this field recently. Moreover, our study shows that anti-mda5 antibody, mortality, gene 5 antibody, IIMs, double-blind, and prognostic factors, among others, may be new hot topics. Conclusion: This study found that research on PM/DM with ILD has increased over time, and scholars are paying more attention to this field. The development of new drugs for the management, treatment, and prevention of PM/DM with ILD is the primary task of researchers and a direction for future research in this field. Abstract: Association between polymorphism of the major histocompatibility complex (MHC) genes and disease resistance has been documented for few teleosts. In this study, we first investigate the genetic variation at the MHC IIα gene in Japanese flounder (Paralichthys olivaceus) for survival after challenge with bacterial infection. To explore the specific allele associated with disease resistance, about 6000 individuals from 60 families challenged with Vibrio anguillarum, which causes significantly different mortality in flounder families. 15–20 individuals from each of six high-resistance (HR) and six low-resistance (LR) families were screened for their MHC class IIα genotypes using sequence analysis. High polymorphism of MHC IIα gene and at least two loci were discovered in Japanese flounder and the rate of d N occurred at a significantly higher frequency than that of d S in PBR and non-PBR, the balancing selection hypothesis could explain the high polymorphism of MHC IIα gene in flounder. From the 212 individuals, a total of 55 MHC class IIα alleles were identified, and 9 alleles were used to study association between alleles and resistance to disease. Three alleles, Paol-DAA*1301, Paol-DAA*1401 and Paol-DAA*2201 were significantly associated with resistance against V. anguillarum, and Paol-DAA*1001 and Paol-DAA*1501 alleles were significantly associated with increased susceptibility to V. anguillarum. This study confirmed the association between alleles of MHC class IIα gene and disease resistance or susceptibility to bacterial infection in flounder, and the disease resistance-related MHC markers could be used for molecular marker-assisted selective breeding in the flounder. The construction of genetic linkage map and evaluation of population genetic diversity both require large numbers of polymorphic molecular markers. In the study, 60 microsatellite loci were isolated from a dinucleotide- enriched genomic library of Japanese halfbeak (Hyporhamphus sajori). And 30 polymorphic microsatellite loci were found to be polymorphic between 2 and 11 alleles. The number of observed, expected heterozygosity and polymorphism information content per locus in 24 individuals ranged from 0.1667 to 1.000, 0.1828 to 0.9220, 0.1828 to 0.8945, respectively. Three loci significantly deviated from Hardy-Weinberg equilibrium after Bonferroni correction analysis and there was no significant linkage disequilibrium found between pairs of loci. As a result, 30 microsatellite loci probably should provide sufficient level of genetic diversity to investigate the fine-scale population structure, stock management and enhancement, genetic linkage map construction and molecular marker-assisted breeding in H. sajori. Abstract: Inter-simple sequence repeat (ISSR) analysis was performed in order to evaluate one common population and two selected hatchery populations of Japanese flounder Paralichthys olivaceus. A total of 105 individuals belonging to three populations were screened using 12 different ISSR primers. A total of 112 loci were produced in the three studied populations. 33.92%, 28.15% and 27.45% of these loci were polymorphic over all the genotypes tested in common, susceptible and resistant populations, respectively. The total number of loci and polymorphic loci detected by single primer ranged from six to twelve and two to five. The average heterozygosities of common, susceptible and resistant populations were 0.1086, 0.0930 and 0.0921, respectively. Compared with the common population, the two selected hatchery populations, susceptible and resistant, showed significant genetic differences including a smaller number of total loci (P <0.05), a smaller number of total polymorphic loci (P <0.05), a smaller number of genotypes (P <0.05) and a smaller Shannon index (P <0.05). The ISSR fingerprinting technique used was confirmed to be a reproducible and sensitive tool for the study of population genetics of Japanese flounder. The present study suggests that the genetic variability of domestic hatchery populations has implications to the conservation of natural Japanese flounder resources. [Copyright &y& Elsevier]

A member of the NF-κB signaling pathway, PoAkirin1, was cloned from a full-length cDNA library of Japanese flounder (Paralichthys olivaceus). The full-length cDNA comprises a 5'UTR of 202 bp, an open reading frame of 564 bp encoding a 187-amino-acid polypeptide and a 521-bp 3'UTR with a poly (A) tail. The putative protein has a predicted molecular mass of 21 kDa and an isoelectric point (pI) of 9.22. Amino acid sequence alignments showed that PoAkirin1 was 99% identical to the Scophthalmus maximus Akirin protein (ADK27484). Yeast two-hybrid assays identified two proteins that interact with PoAkirin1: PoHEPN and PoC1q. The cDNA sequences of PoHEPN and PoC1q are 672 bp and 528 bp, respectively. Real-time quantitative reverse-transcriptase polymerase chain reaction analysis showed that bacteria could induce the expressions of PoAkirin1, PoHEPN and PoC1q. However, the responses of PoHEPN and PoC1q to the bacterial challenge were slower than that of PoAkirin1. To further study the function of PoAkirin1, recombinant PoAkirin1 and PoHEPN were expressed in Escherichia coli and would be used to verify the PoAkirin1-PoHEPN binding activity. These results identified two proteins that potentially interact with PoAkirin1 and that bacteria could induce their expression.

A continuous cell line, the flounder embryonic cell line (FEC), was established from gastrula-stage embryos of a marine cultured fish, the Japanese flounder Paralichthys olivaceus and cultured for more than 200 d with more than 60 passages. FEC cells were cultured in Dulbecco's Modified Eagle's Medium (DMEM) supplemented with antibiotics, fetal bovine serum (FBS), sea perch serum (SPS), and basic fibroblast growth factor (bFGF). The cells were small and round, and grew actively and stably in culture. The effect of temperature, FBS concentration and bFGF on FEC cell growth was examined. Cells grew well between 24 and 30 degrees C, but had a reduced growth rate below 18 degrees C. The growth rate of FEC cells in medium containing 15% FBS was higher than that in medium containing 7.5% FBS. Addition of bFGF to the medium also significantly increased the growth rate. Chromosome analysis revealed that FEC cells have a normal diploid karyotype with 2n = 48. High survival rate was obtained after cryopreservation of cell cultures. The susceptibility of the cell line to piscine viruses was examined. Two viruses tested were shown to induce CPE (cytopathic effect) on FEC cells. FEC cell culture infected with fish iridovirus was further elucidated by electron microscopy. Many virus particles were found in the cytoplasm of the virus-infected FEC cells. These results indicated that the FEC cell line could be potentially used to isolate and study fish viruses.
