Mesopelagic water from the deep Eastern Mediterranean Sea (EMS) was collected under disrupted (REPRESS) or undisturbed (HP) pressure conditions and was acclimated to oil (OIL) or dispersed-oil (DISPOIL) under in situ pressure and temperature (10 MPa, 14 °C). Decompression resulted in oil-acclimatised microbial communities of lower diversity despite the restoration of in situ pressure conditions during the 1-week incubation. Further biodiversity loss was observed when oil-acclimatised communities were transferred to ONR7 medium to facilitate the isolation of oil-degrading bacteria. Microbial diversity loss impacted the degradation of recalcitrant oil compounds, especially PAHs, as low-abundance taxa, linked with PAH degradation, were outcompeted in the enrichment process. Thalassomonas, Pseudoalteromonas, Halomonas and Alcanivorax were enriched in ONR7 under all experimental conditions. No effect of dispersant application on the microbial community structure was identified. The response of Posidonia oceanica meadows to global warming of the Eastern Mediterranean Sea, where the increase in sea surface temperature (SST) is particularly severe, is poorly investigated. Here, we reconstructed the long-term P. oceanica production in 60 meadows along the Greek Seas over two decades (1997-2018), using lepidochronology. We determined the effect of warming on production by reconstructing the annual and maximum (i.e. August) SST, considering the role of other production drivers related to water quality (i.e. Chla, suspended particulate matter, Secchi depth). Grand mean (±SE) production across all sites and the study period was 48 ± 1.1 mg DW per shoot yr -1 . Production over the last two decades followed a trajectory of decrease, which was related to the concurrent increase in annual SST and SST aug . Annual SST > 20°C and SST aug  > 26.5°C was related to production decline (GAMM, P < 0.05), while the rest of the tested factors did not help explain the production pattern. Chronic discharge of surplus organic matter is a typical side effect of fish aquaculture, occasionally leading to coastal eutrophication and excessive phytoplankton growth. Owing to their innate filter-feeding capacity, marine sponges could mitigate environmental impact under integrated multitrophic aquaculture (IMTA) scenarios. Herein, we investigated the clearance capacity of four ubiquitous Mediterranean sponges ( Agelas oroides , Axinella cannabina , Chondrosia reniformis and Sarcotragus foetidus ) against three microalgal substrates with different size/motility characteristics: the nanophytoplankton Nannochloropsis sp. (~3.2 μm, nonmotile) and Isochrysis sp. (~3.8 μm, motile), as well as the diatom Phaeodactylum tricornutum (~21.7 μm, nonmotile). In vitro cleaning experiments were conducted using sponge explants in 1 L of natural seawater and applying different microalgal cell concentrations under light/dark conditions. The investigated sponges exhibited a wide range of retention efficiencies for the different phytoplankton cells, with the lowest average values found for A. cannabina (37%) and the highest for A. oroides (70%). The latter could filter up to 14.1 mL seawater per hour and gram of sponge wet weight, by retaining 100% of Isochrysis at a density of 10 5 cells mL -1 , under darkness. Our results highlight differences in filtering capacity among sponge species and preferences for microalgal substrates with distinct size and motility traits.

The invasion of the tetrodotoxin (TTX)-bearing silver-cheeked toadfish and potential poisoning due to its consumption (tetrodotoxication) threatens public safety in the Mediterranean Sea. In this study, TTX and TTX analogues of Lagocephalus sceleratus (Gmelin, 1789) were measured using liquid chromatography tandem mass spectrometry (LC-MS/MS) in fish collected off the island of Crete (Southern Mediterranean). We tested the synergistic effect of a suite of factors potentially affecting toxins' levels and tetrodotoxication risk using general and generalized linear models, respectively. The type of tissue, geographic origin (Cretan Sea, Libyan Sea), sex, and fish maturity stage were significant predictors of toxin concentrations. Mean TTX was higher in gonads and lower in muscles, higher in the Libyan Sea and in female fish, and lower in juvenile (virgin) fish. The concentration of TTX was also significantly and positively correlated with the concentration of several TTX analogues (4-epiTTX, 4,9-anhydroTTX, 11-deoxyTTX, 5,11/6,11-dideoxyTTX, 5,6,11-trideoxyTTX, 11-norTTX-6-ol). The analysis showed that fish originating from the Libyan Sea had significantly higher probability to cause tetrodotoxication in case of consumption. The variability explained by the models developed in this study was relatively low, indicating that toxin levels are hard to predict and the consumption of L. sceleratus should therefore be avoided.

Concentrations of 27 metals and elements were investigated in the tissues of 28 demersal and pelagic marine fish species from the Eastern Mediterranean Sea. The analysis indicated highest concentrations of accumulation for the majority of elements in the gills, liver, and scales; whereas, lowest assimilation of elements was reported in the muscle. Pelagic fish accumulated higher concentrations of elements than demersal fish. An inverse correlation due to the growth dilution effect was revealed among element concentrations in the gills, liver, muscle, and skin tissues, and fish size. Patterns of biomagnification for Cs solely were recorded in the muscle and patterns of biodilution for most elements were evident in the gills, liver, muscle, and skin tissues. A positive correlation was determined between trophic level and element concentrations in the scales which could not only be associated with fish diet, but also with metal concentrations in the ambient environment and scales size., (Copyright © 2021 Elsevier Ltd. Smaller sized plastics (microplastics or MPs <5 mm) are ubiquitous in nature and have been found to interact in diverse ways with most biotic and abiotic systems globally. Most MPs in the seas have a land-based source, however, little is known about how the transfer occurs. In our study, we used three sandy beaches to describe the process of how MPs travel from accumulation points at the backshore of the beach to the sea, and vice versa. MPs differed significantly in all three beaches (both in quantitative and qualitative terms) between the summer and the winter samplings. During the summer, heavy MPs are the majority, while during the winter, lightweight microplastics are predominant, and the ratio of heavy per lightweight MPs is affected by the sediment median diameter after the summer sampling. Chemical residues released from aquaculture farms may persist in the environment and may pose adverse ecological effects. The aim of this study was to assess the pollution status of marine sediments underneath or close to aquaculture farms in the Eastern Mediterranean Sea, the factors that affect the elemental distribution, and the exceedance of environmental quality standards and factors. To this end, surface sediment samples were collected (underneath fish cages, at 25 m distance and from a reference station) from 48 fish farms in Greece with variable environmental and geochemical characteristics. The content of 29 metals and major and trace elements, and three antibiotics (oxytetracycline, florfenicol, and flumequine), was determined in the sediment samples. Most of the elements found in the sediments originated from geological sources and their concentrations were close to Earth's Crust content. Below and close to fish farm cages, the sediment was enriched with P, Cu, Zn, Mo, and Cd mainly due to the aquaculture biosolid deposition, and changes in environmental conditions (increased organic matter, low redox potential). Cr and As were found to exceed twice the upper threshold limit of the available sediment quality guidelines in 13.5% and 7.3% of sampling stations, respectively. The potential ecological risk of V, Cr, Mn, Co, Ni, Cu, Zn, As and Pb was found to be low in the sediment close to fish cages except for Cd, which may pose a moderate to considerable risk in 12.5% of sampled fish farms. However, the estimated risks for this metal may be influenced by the high background levels found in the investigated areas. The planktonic food web in the oligotrophic Mediterranean Sea is dominated by small-sized (<20 μm) microbes, with nanoflagellates being the major bacterial grazers and the main participants in nutrient cycling. Phosphate is a key nutrient in the P-limited Cretan Sea (NE Mediterranean) and P-availability can affect its trophic dynamics. Here, we examined the grazing potential of heterotrophic (HF) and pigmented (PF) nanoflagellates as a response mechanism to phosphate amendment. Flagellate grazing effect on bacteria was quantified in P-amended nutrient-depleted water from the Cretan Sea over the course of 4 days using microcosm experiments. P-addition positively affected HF abundance, while PF abundance remained unchanged. At the community level, P-addition had a negative effect on PF bacterial removal rates. In the control, PF-grazing rate was significantly higher than that of HF throughout the experiment. Pigment analysis showed no changes in phytoplankton community composition as a result of P-addition, indicating that PF grazing rate declined as a physiological response of the cells. The present study emphasizes the dominant grazing role of PF under P-depleted conditions and reveals that during the late stratified season PF respond to P-addition by lowering their grazing rates, enhancing the relative importance of bacterial removal by HF., (© FEMS 2020.)

Background: Henneguya Thélohan, 1892 (Myxobolidae) is one of the most species-rich genera of myxosporean parasites infecting fish. Although common in nature, there are few reports of these parasites causing important disease in aquaculture. In this paper, we describe a new species of Henneguya infecting Pagrus major (Temminck & Schlegel), a fish host introduced to the Mediterranean Sea from Japan in the late 1980s., Results: Large plasmodia of the parasite were found in the bulbus arteriosus and in the ventricle of the infected fish. Spores were found mainly in the kidney and heart and were accompanied by melanized macrophages or vascular intimal proliferation mixed with a mild non-suppurative response, respectively. Comparisons of morphometric data for spore and polar capsule length and width, suggest a unique combination of features in the newly described species. Molecular analysis, based on 18S rDNA sequence of the parasite, followed by phylogenetic analysis, indicated that the parasite described here is a novel species of Henneguya, clustered with the marine congeneric species., Conclusions: Henneguya aegea n. sp. infects in aquaculture P. major, a host introduced as eggs to the Mediterranean from Japan. Despite the high host specificity of the myxobolid parasites, H. aegea n. sp. seems to be able to use P. major as a host and propagate successfully, causing morbidity and mortality. This could result in spillback of the new species from high density cultured non-native P. major to native fish hosts.

The potential effects of climate change on the distribution of benthic species commonly used in marine ecological quality assessment were investigated using a spatial modelling approach. In this work, the relevance of the ecological groups that macrofaunal molluscs are assigned according to their sensitivity or tolerance to environmental disturbance was examined under the scope of the RCP 8.5 severe emissions scenario. The effects of climate change were more profound on species that are indicative of a specific suite of climatic conditions regarding temperature and salinity. Significant loss of habitat suitability was observed for the tolerant species Corbula gibba and Abra prismatica whereas the sensitive species Moerella donacina was least affected. In contrast, an overall expansion of the distributional potential was observed for the sensitive species Flexopecten hyalinus as newly suitable habitats are formed. As hypothesised, the current ecological grouping that depicts the sensitivity of a benthic species to an environmental stressor is irrelevant when assessing the effects of climate change. Marine habitat assessment using indicator species through Species Distribution Modeling (SDM) was investigated. The bivalves: Corbula gibba and Flexopecten hyalinus were the indicator species characterizing disturbed and undisturbed areas respectively in terms of chlorophyll a concentration in Greece. The habitat suitability maps of these species reflected the overall ecological status of the area. The C. gibba model successfully predicted the occurrence of this species in areas with increased physical disturbance driven by chlorophyll a concentration, whereas the habitat map for F. hyalinus showed an increased probability of occurrence in chlorophyll-poor areas, affected mainly by salinity. We advocate the use of C. gibba as a proxy for eutrophication and the incorporation of this species in monitoring studies through SDM methods. For the Mediterranean Sea we suggest the use of F. hyalinus in SDM as an indicator of environmental stability and a possible forecasting tool for salinity fluctuations., (Copyright © 2018 Elsevier Ltd. Uric acid is a promising hydrophobic nitrogen source for biostimulation of microbial activities in oil-impacted marine environments. This study investigated metabolic processes and microbial community changes in a series of microcosms using sediment from the Mediterranean and the Red Sea amended with ammonium and uric acid. Respiration, emulsification, ammonium and protein concentration measurements suggested a rapid production of ammonium from uric acid accompanied by the development of microbial communities containing hydrocarbonoclastic bacteria after 3 weeks of incubation. About 80 % of uric acid was converted to ammonium within the first few days of the experiment. Microbial population dynamics were investigated by Ribosomal Intergenic Spacer Analysis and Illumina sequencing as well as by culture-based techniques. Resulting data indicated that strains related to Halomonas spp. converted uric acid into ammonium, which stimulated growth of microbial consortia dominated by Alcanivorax spp. and Pseudomonas spp. Several strains of Halomonas spp. were isolated on uric acid as the sole carbon source showed location specificity. These results point towards a possible role of halomonads in the conversion of uric acid to ammonium utilized by hydrocarbonoclastic bacteria.

Trace element concentrations in sediment were investigated at four fish farms in the Eastern Mediterranean Sea. Fish farms effects were negligible beyond 25-50 m from the edge of the cages. Based on elemental distribution, sediments from the farms were separated into coarse oxidized and silty reduced ones. Fish feed is richer in P, Zn and Cd than reference and impacted stations. Comparison among impacted stations and the respective reference stations shows that, in anoxic sediments, all elements had higher concentrations at the impacted stations than at reference stations while in oxic sediments, many elemental concentrations were lower at impacted stations than at reference stations. The behavior of elements and therefore their distribution is affected by changes in sediment grain size, organic content and redox regime. Elements in sediments around fish farms can be clustered into five groups according to these environmental variables. In silty and anoxic sediments, element concentrations were higher than in coarse and oxic ones. Several approaches were used to assess potential sediment toxicity (enrichment factors, geoaccumulation indices, contamination factors) as well as to assess the potential danger to aquatic life (Sediment Quality Guidelines, SQG). Cu, Zn and Fe can cause from threshold to extreme effects on aquatic life in anoxic, fine-grained sediments and As can cause threshold effects in all types of sediment around fish farms. The aim of the present study was to examine the species-specific stress response for seven Mediterranean fishes in culture. Also, to evaluate the method of measuring free cortisol concentration in the rearing water as a non-invasive and reliable indicator of stress in marine species, of aquaculture importance. Gilthead sea bream, Sparus aurata (Sparidae); common dentex, Dentex dentex (Sparidae); common Pandora, Pagellus erythrinus (Sparidae); sharpsnout sea bream, Diplodus puntazzo (Sparidae); dusky grouper, Epinephelus marginatus (Serranidae); meagre, Argyrosomus regius (Sciaenidae) and European sea bass, Dicentrarchus labrax (Moronidae) were subjected to identical acute stress (5-6 min chasing and 1-1.5 min air exposure) under the same environmental conditions and samples were analyzed by the same procedures. Results indicated that there was a clear species-specificity in the magnitude, timing and duration of the stress response in terms of cortisol, glucose and lactate. European sea bass showed a very high response and dusky grouper and meagre a very low response, except plasma glucose concentrations of dusky grouper which was constantly high, while sharpsnout sea bream presented a protracted stress response, up to 8h. The aim of this paper was to test how benthic fauna and biogeochemical properties of sediment will vary in response to similar levels of organic enrichment (induced by fish farming) as a function of bottom-habitat type (i.e., mud versus seagrass/coarse sediments), distance from the enrichment source and depth. Our results showed that samples from silty sediments in the vicinity of fish farms have higher TOC and TON values, higher oxygen consumption, higher PO(4) release and lower benthic diversity. In this context muddy sites are more likely to be identified as impacted/critical, than coarse sediment ones.

We investigated 16S rRNA gene diversity at a high sediment depth resolution (every 5 cm, top 30 cm) in an active site of the Kazan mud volcano, East Mediterranean Sea. A total of 242 archaeal and 374 bacterial clones were analysed, which were attributed to 38 and 205 unique phylotypes, respectively (> or = 98% similarity). Most of the archaeal phylotypes were related to ANME-1, -2 and -3 members originating from habitats where anaerobic oxidation of methane (AOM) occurs, although they occurred in sediment layers with no apparent AOM (below the sulphate depletion depth). Proteobacteria were the most abundant and diverse bacterial group, with the Gammaproteobacteria dominating in most sediment layers and these were related to phylotypes involved in methane cycling. The Deltaproteobacteria included several of the sulphate-reducers related to AOM. The rest of the bacterial phylotypes belonged to 15 known phyla and three unaffiliated groups, with representatives from similar habitats. Diversity index H was in the range 0.56-1.73 and 1.47-3.82 for Archaea and Bacteria, respectively, revealing different depth patterns for the two groups. At 15 and 20 cm below the sea floor, the prokaryotic communities were highly similar, hosting AOM-specific Archaea and Bacteria. Our study revealed different dominant phyla in proximate sediment layers.

This research is aimed at elucidating the removal mechanisms of nutrients due to natural attenuation in drainage canals in Evrotas River delta in Greece. We investigated nutrients fluxes in groundwater, sediments, and reeds (Phragmites Australis and Arundo Donax) of the drainage canal. Groundwater fluxes indicated that the rate of mineralization was 37.6 mg N/m(2) day. The accumulation of toxic ammonia was prevented through the nitrification process (26.6 mg N m(-2) day(-1)). The decrease of NO(3)-N flux in groundwater in the riparian zone was calculated to be 56.1 mg N m(-2) day(-1) (20.48 g N m(-2) year(-1)). Phosphate was adsorbed to sediments and its load to the drainage canal was minimized. Harvesting of above ground reed biomass in mid June, when maximum standing stock of nutrients was attained for both plants, would remove 2.73 g P m(-2) and 11.2 g N m(-2). All the phosphorous (1.39 g P/m(2) year(-1)) and 76.5% of the nitrate nitrogen (14.64 g N m(-2) year(-1)) entering the drainage canal was taken up by plants. Drainage canal management is suggested as an efficient low cost-high gain agri-environmental measure, which is easy to be adapted by farmers, to reduce diffuse nutrient pollution.

By using the Grob closed loop stripping analysis technique a wide variety of disinfection by-products (DBPs) was determined, in finished drinking water of 15 important cities of Greece distributed in the southern, northern, west continental land and in the islands of Aegean and Ionian Seas. Trihalomethanes, haloacetonitriles, haloacetic acids, chloropicrin, halogenated ketons and chloral hydrate were the main DBPs determined. In chlorinated drinking water of coastal cities brominated DBPs were most abundant than their chlorinated homologues due to the higher bromide concentration in raw waters of these areas. Factor analysis with principal component extraction of the results demonstrated a clear differentiation between the examined drinking waters on the basis of their DBPs content and vicinity to the sea.

We have examined mitochondrial DNA (mtDNA) variation in samples of the mussel Mytilus galloprovincialis from the Black Sea, the Mediterranean and the Spanish Atlantic coast by scoring for presence or absence of cleavage at 20 restriction sites of a fragment of the COIII gene and at four restriction sites of the 16S RNA gene. This species contains two types of mtDNA genomes, one that is transmitted maternally (the F type) and one that is transmitted paternally (the M type). The M genome evolves at a higher rate than the F genome. Normally, females are homoplasmic for an F type and males are heteroplasmic for an F and an M type. Occasionally molecules from the F lineage invade the paternal transmission route, resulting in males that carry two F-type mtDNA genomes. These features of the mussel mtDNA system give rise to a new set of questions when using mtDNA variation in population studies and phylogeny. We show here that the two mtDNA types provide different information with regard to amounts of variation and genetic distances among populations. The F genome exhibits higher degrees of diversity within populations, while the M genome produces higher degrees of differentiation among populations. There is a strong differentiation between the Atlantic and the Black Sea. The Mediterranean samples have intermediate haplotype frequencies, yet are much closer to the Black Sea than to the Atlantic. We conclude that in this species gene flow among the three Seas is restricted and not enough to erase the combined effect of mutation and random drift. In one sample, that from the Black Sea, the majority of males did not contain an M mtDNA type. This suggests that a molecule of the maternal lineage has recently invaded the paternal route and has increased its frequency in the population to the point that the present pool of paternally transmitted mtDNA molecules is highly heterogeneous and cannot be used to read the population's history. This liability of the paternal route means that in species with doubly uniparental inheritance, the maternal lineage provides more reliable information for population and phylogenetic studies.
