Understanding hybridization is important for practical reasons, as the presence of hybrid trees in seed stands can influence the success of natural regeneration and reforestation. Hybridization creates new gene combinations, which can promote or enhance adaptation to new or changing environments. In the present research, we aimed, for the first time, to evaluate and compare the growth and survival of 541 putative hybrid seedlings and 455 seedlings of the pure parental trees of Pinus arizonica, P. durangensis, P. engelmannii, P. leiophylla, and P. teocote, in two reciprocal trials of duration 27 months in the Sierra Madre Occidental (SMO), Durango, Mexico. We also examined the possible correlation between needle stomatal density and seedling growth and survival. The overall analysis of the data showed that the mean height to the apical bud was significantly higher (p = 0.01) in the hybrids than in the pure trees. Considering both trials, the survival rate of P. arizonica (p = 0.002) and P. durangensis (p = 0.01) hybrids was significantly higher than that of the pure trees. The growth parameters were significantly correlated with the mean stomatal density (p < 0.01). Stomatal density and survival at the seed stand level were significantly and positively correlated in the hybrids, but not in the pure trees. In summary, Pinus hybrids generally exhibited the same ability as the pure species (or sometimes a greater ability) to withstand weather conditions, survive, and grow effectively in both growth trials. The systematic use of natural pine hybrids in Mexico could therefore be considered a possible option for sustainable management and as a component of adaptive silviculture. [ABSTRACT FROM AUTHOR]

What is known and objective: Midazolam is a drug that is metabolized by cytochrome P450 (CYP450) enzymes, particularly CYP3A4 and CYP3A5. The presence of single‐nucleotide polymorphisms (SNPs) in the genes encoding these enzymes, such as CYP3A4*1B which is associated with low enzyme expression and activity and CYP3A5*3, has been associated with decrease in enzymatic activity and reduced drug clearance, with potential effects on drug levels and/or toxicity. The present study was conducted to determine the frequencies of the allelic variants of the CYP3A4 (rs2740574) and CYP3A5 (rs776746) genes and their effects on the plasma levels and clearance of intravenous midazolam in critically ill Mexican paediatric patients. Methods: Seventy‐two DNA samples were genotyped by real‐time PCR with TaqMan probes. Plasma midazolam levels were determined at 3 and 24 h post infusion by high‐performance liquid chromatography. Results and discussion: The allelic variant rs776746 (CYP3A5*3) was associated with high midazolam plasma levels; the median concentration in patients with the normal genotype (CC) <0.01 ng/ml (Q250.01–Q75196.09), whereas patients with the allelic variant (TT+TC) had a median midazolam concentration of 320.3 ng/ml (Q2537.51–Q75529.51), p = 0.001. The median pharmacokinetic clearance rates were 0.10 L/kg/h (Q250.01–Q750.34) in patients with the allelic variant (TT+TC) and 0.03 L/kg/h (Q250.002–Q750.13) in patients with the normal genotype (CC), p = 0.042. What is new and conclusion: This is the first study that reports the frequency of the rs776746 polymorphism in critically ill paediatric patients, which is relevant, since carriers of the *1 allele synthesizing a functional enzyme may need higher doses to achieve adequate sedation. Our results show that compared with carriers of the normal allele, patients with the CYP3A5*3 allelic variant (rs776746) had increased plasma midazolam levels at 3 h after infusion discontinuation (320.3 ng/ml) and greater clearance (0.10 L/kg/h) of the drug. Background. Spatial genetic structure (SGS) analysis is a powerful approach to quantifying gene flow between trees, thus clarifying the functional connectivity of trees at population and landscape scales. The findings of SGS analysis may be useful for conservation and management of natural populations and plantations. Pinus cembroides is a widely distributed tree species, covering an area of about 2.5 million hectares in Mexico. The aim of this study was to examine five natural seed stands of P. cembroides in the Sierra Madre Occidental to determine the SGS at population (within the seed stand) and landscape (among seed stands) levels in order to establish guidelines for the conservation and management of the species. We hypothesized that P. cembroides, in which the seeds are dispersed by birds and mammals, creates weaker SGS than species with wind-dispersed seeds. Methods DNA fingerprinting was performed using the amplified fragment length polymorphism (AFLP) technique. In order to estimate the SGS at population and landscape levels, we measured the geographical (spatial) distance as the Euclidean distance. We also estimated the genetic distances between individuals using the pairwise kinship coefficient. Results The results showed non-significant autocorrelation in four out of five seed stands studied (i.e., a mainly random distribution in the space of the genetic variants of P. cembroides at population level). Discussion SGS was detected at the landscape scale, supporting the theory of isolation by distance as a consequence of restricted pollen and seed dispersal. However, the SGS may also have been generated by our sampling strategy. We recommended establishing a close network of seed stands of P. cembroides to prevent greater loss of local genetic variants and alteration of SGS. Species diversity and genetic diversity, the most basic elements of biodiversity, have long been treated as separate topics, although populations evolve within a community context. Recent studies on community genetics and ecology have suggested that genetic diversity is not completely independent of species diversity. The Mexican Picea chihuahuana Martínez is an endemic species listed as “Endangered” on the Red List. Forty populations of Chihuahua spruce have been identified. This species is often associated with tree species of eight genera in gallery forests. This rare Picea chihuahuana tree community covers an area no more than 300 ha and has been subject of several studies involving different topics such as ecology, genetic structure and climate change. The overall aim of these studies was to obtain a dataset for developing management tools to help decision makers implement preservation and conservation strategies. However, this unique forest tree community may also represent an excellent subject for helping us to understand the interplay between ecological and evolutionary processes in determining community structure and dynamics. The AFLP technique and species composition data were used together to test the hypothesis that species diversity is related to the adaptive genetic structure of some dominant tree species (Picea chihuahuana, Pinus strobiformis, Pseudotsuga menziesii and Populus tremuloides) of the Picea chihuahuana tree community at fourteen locations. The Hill numbers were used as a diversity measure. The results revealed a significant correlation between tree species diversity and genetic structure in Populus tremuloides. Because the relationship between the two levels of diversity was found to be positive for the putative adaptive AFLP detected, genetic and species structures of the tree community were possibly simultaneously adapted to a combination of ecological or environmental factors. Genetic diversity is key to survival of species. In evolutionary ecology, the general centre–periphery theory suggests that populations of species located at the margins of their distribution areas display less genetic diversity and greater genetic differentiation than populations from central areas. The aim of this study was to evaluate the genetic diversity and differentiation in six of the main pine species of the Sierra Madre Occidental (northern Mexico). The species considered were Pinus arizonica, P. cembroides, P. durangensis, Pinus engelmannii, P. herrerae and P. leiophylla, which occur at the margins and centre of the geographic distribution. We sampled needles from 2799 individuals belonging to 80 populations of the six species. We analysed amplified fragment length polymorphisms (AFLPs) to estimate diversity and rarity indexes, applied Principal Coordinate Analysis (PCoA), and used the Kruskal–Wallis test to detect genetic differences. Finally, we calculated Spearman's correlation for association between variables. The general centre–periphery model only explained the traits in P. herrerae. The elevation gradient was an important factor that influenced genetic diversity. However, for elevation as partitioning criterion, most populations showed a central distribution. This information may be useful for establishing seed collections of priority individuals for maintenance in germplasm banks and their subsequent sustainable use. [ABSTRACT FROM AUTHOR]

The objective of this study was to determine the prevalence of gastrointestinal parasitic nematodes in sheep at necropsy at a slaughterhouse in Tabasco, Mexico. Adult parasites from gastrointestinal tract were recovered and preserved in formaldehyde for their later counting and identification. Adult nematodes were separated by species and the final count was transformed to Log + 1 to decrease the variance. GLM procedure using SAS statistic program was performed for data analysis. Provenance, sex, physiological status, and sampling month were considered as the variation sources. From a total sample of 242 slaughtered animals necropsied for monitoring gastrointestinal parasites, 57.4% including Nematoda, Trematoda or Cestoda classes. The main identified species corresponded to Haemonchus contortus in the abomasum. Cooperia curticei, Trichostongylus colubriformis, Strongyloides papillosus and Bunostomum trigonocephalum were found in the small intestine and Oesophagostomum columbianum, and Trichuris ovis in the large intestine. Fasciola hepatica and Moniezia expansa were the Trematoda and Cestoda parasites found in liver and small intestine, respectively with prevalence lower than 7%. The average of the total count of adult nematodes in the gastrointestinal tract of the infested animals was 2175 ± 445. Among the factors studied, the slaughter month and the provenance of the animals affected the prevalence of parasite infestation in sheep at slaughter. The main adult parasites found were H. contortus, C. curticei and T. colubriformis with average counts higher than 1009, 813 and 335, respectively. [ABSTRACT FROM AUTHOR]

El objetivo de esta investigación fue conocer la prevalencia de parásitos en ovinos sacrificados en un rastro en el estado de Tabasco. Se realizó la recuperación y conservación en formol de parásitos adultos presentes en el tracto gastrointestinal, para su posterior conteo e identificación. Los conteos de nematodos adultos por especie se transformaron a Log + 1 para disminuir la varianza, y se realizó el análisis con el procedimiento GLM del SAS en el cual se incluyeron como fuentes de variación: origen, sexo, estado fisiológico y mes de muestreo. De una muestra total de 242 animales sacrificados, se observó que 57.4% se encontraban parasitados con alguna especie de las clases Nematoda, Trematoda o Cestoda. Las principales especies identificadas correspondieron a Haemonchus contortus en abomaso. Cooperia curticei, Trichostongylus colubriformis, Strongyloides papillosus y Bunostomum trigonocephallum en intestino delgado. Oesophagostomum columbianum, y Trichuris ovis en intestino grueso. De los trematodos se encontró Fasciola hepatica en hígado y de los cestodos Moniezia expansa se localizó en intestino delgado con prevalencia menor a 7%. El conteo total de los nematodos adultos en el tracto gastrointestinal de los animales parasitados fue, en promedio, 2175 ± 445. De los factores estudiados, el mes de sacrificio y el origen de los animales afectaron la prevalencia de parasitosis en los ovinos al sacrificio. Background Physicians have frequently encountered difficulties when prescribing drugs not available in doses suitable for pediatric age groups. Furthermore, children have difficulty swallowing tablets. This study aimed to determine the stability of an oral propafenone suspension made from commercial tablets with a syrup vehicle and to establish its reliable use with children. Methods An extemporaneous suspension of propafenone 1.5 mg/ml was prepared with commercial tablets. Its physicochemical and microbiologic stability was established by constant monitoring during 90 days at room temperature (15 ± 5°C) and at refrigeration (3–5°C). Plasma levels of propafenona were measured in two children with supraventricular tachycardia at steady state. Results The suspension was stable, maintaining its original physicochemical and microbiologic properties. Moreover, no apparent changes in color or odor were observed. Plasma levels of propafenone in patients demonstrated therapeutic concentrations after they had taken the suspension, with no unwanted outcome. Conclusions The conservation of both physicochemical and microbiologic stability of the suspension represents an option for the administration of propafenone to children. [ABSTRACT FROM AUTHOR]

Suicide is a complex behavior with biological, psychological and social causes. It has predisposing family factors such as domestic violence, psychiatric disorders and parents' alcohol abuse. Suicides have increased in children and adolescents in the last decades and it is now the third leading cause of death. Mexico has the second place in mortality in adolescents between 10-14 years old, of which 21.9 % are suicides. This study aimed to determine the frequency of suicide attempts using drugs in children and adolescents in a tertiary care hospital and to identify the causes. A retrospective and descriptive study was performed considering socioeconomic, cultural and biological issues related to suicide attempts using drugs in children and adolescents who were attended at the National Institute of Pediatrics from January 1995 to March 2005. A total of 141 drug intoxication cases were detected including 47 suicide attempts; 25 girls and 22 boys, with a median of 13 years of age. The most frequently drug classes used were analgesics 21 of 47 cases and antidepressants 11 of 47 cases. Risk factors were parents' divorce and family dysfunction in 16 of 47 cases of the population. Suicidal behavior in children and adolescents can be a way to attract attention, as well as an alarm signal asking for help. Analgesics and antidepressants are drugs commonly used in suicide attempts.

In Mexico, more than 70 % of acute pediatric poisoning is caused by medicines. The age groups at greatest risk of drug poisoning are those between 2 to 5 years and 14 to 18 years; although in this last group, drug ingestion is usually intentional. The purpose of our study was to determine the frequency of unintentional drug poisoning in the pediatric population attended in a tertiary care hospital in Mexico, and to review the rescue procedures applied in specific cases. A retrospective and descriptive study was performed through revision of clinical records, obtained from patients attended at the National Pediatrics Institute from January 1995 to June 2005. One hundred and thirty nine (139) records, 62 females and 77 males, median age 2 years with clinical diagnosis of drug poisoning were reviewed. Poisoning was confirmed in 23.7% of the cases by determination of drug plasma concentration. The most frequent causes of drug poisoning were analgesics (42.3 %), from which 60 % corresponded to acetylsalicylic acid and 40 % to acetaminophen; antiepileptics (22.9 %), anxiolytics (17.9 %) and other drugs (16.3 %). From our results, we concluded that self-medication was unlikely due to the early age of patients, unless ingestion of the drug was accidental. No case needed more than 24 h of hospitalization, and no patient died due to poisoning. Specific cause of poisoning was that, at early ages, doses must be administered according to the infant's weight, which poses a risk of poisoning.

Aim: The purpose of the present study is to determine the relationship between the anticonvulsant drug use during pregnancy and the presence of malformations in the newborns., Methods: The frequency of malformations in the neonates of epileptic mothers under anticonvulsant treatment was analyzed in two periods, one from 1988 to 1992, which included 76 epileptic mothers, and another from 1996 to 2003 with 170 patients., Results: In the first period, 51 (67.1%) of mothers received monotherapy and 25 (32.9%) received polytherapy of phenytoin with carbamazepine, valproic acid or phenobarbital. In this period, 4 newborns (16%) with congenital malformations were registered. In the second period, 159 (93.5%) of the epileptic mothers received monotherapy and 11 (6.5%) received polytherapy of valproic acid with carbamazepine or phenytoin. During this period only 3 newborns 27.3% with malformations were registered., Discussion: Clinical treatment should consider the risk of using polytherapy, mainly if phenytoin or valproic acid are combined with other anticonvulsants.

Although the drug-metabolizing enzyme CYP2D6 has been studied extensively in subjects of differing ethnicities, limited CYP2D6 pharmacogenetic data are available for the Amerindian population and Mestizos of Mexico. Dextromethorphan hydroxylation phenotype was studied in Tepehuano Amerindian (n = 58) and Mestizo (n = 88) subjects, and 195 individuals (85 Tepehuano Amerindians and 110 Mestizos) were genotyped by polymerase chain reaction-restriction fragment length polymorphism methods to identify the frequencies of the CYP2D6*3, *4, *6, and *10 alleles. Tepehuano Amerindian subjects lacked the poor metabolizer (PM) phenotype, whereas in Mestizos the PM phenotype frequency was 6.8%. The CYP2D6*3, *6, and *10 alleles were not found in Tepehuano Amerindians. The CYP2D6*4 allele had a low frequency (0.006) in this Amerindian group. In the Mestizo group, the CYP2D6*3, *4, and *10 alleles had frequencies of 0.009, 0.131, and 0.023, respectively. The CYP2D6*6 allele was not found in Mestizos. The genotype-phenotype association was strongly statistically significant (r(2) = .45; P = .005) in Mestizos. The Tepehuano population was found to have a low phenotypic and genotypic CYP2D6 diversity and differed from other Amerindian groups. On the other hand, the frequencies of the CYP2D6 variant alleles in Mestizos were similar to those reported for whites.

Cytochrome P4502D6 (CYP2D6) shows genetic polymorphism, which is clinically important in the metabolism of drugs and other xenobiotics. Dextrometorphan (DM) has been used as a test compound to evaluate the in vivo activity of CYP2D6. Phenotypical frequencies of CYP2D6 have been determined in some populations, but little is known about them in native populations. The object of this study was to characterize the phenotypical expression of CYP2D6 in Amerindian subjects of Tepehuano origin from the State of Durango, using DM as metabolic marker, as well as the effect of age, sex and nutritional status on this activity. Three hr after oral administration of a single 30 mg dose of DM, the plasma concentration of DM and its metabolite dextrophan (DX) were determined with HPLC in 55 Tepehuano subjects. All subjects were extensive metabolizers (metabolic ratio MR < 0.3). No correlation of age, sex or nutritional status was found with the DM/DX metabolic ratio. However, we found a monoexponential relationship between the metabolic ratio of DM and DX, and their concentrations respectively, which can have clinical applications, since metabolic ratio can be predicted from a known DM or DX concentration. Three hr after ingestion of DM, 18 individuals showed DM plasma concentrations of 5 to 10 ng/mL, 15 subjects of 11 to 20 ng/mL, 8 subjects of 21 to 50 ng/mL and 14 subjects >51 ng/mL pointing out that DM concentrations and MR must be determined to establish toxicity risk levels.
