Aflatoxin contamination of corn reduces grain quality and can negatively impact the economic well-being of Mississippi's corn farmers. Mitigation of aflatoxin contamination in corn through pre-harvest field application of non-aflatoxigenic strains of Aspergillus flavus holds promise as an effective management strategy. In this study, field trials were conducted at fourteen sites over three years, in experimental plots and within commercial fields, to evaluate the efficacy of two biocontrol products at 11 or 22 kg ha −1 application rates and at various timings, including two vegetative-stages (V5 and V10), tasseling (VT), a reproductive application made at blister (R2) and two split-applications. The greatest reduction in aflatoxin generally occurred with applications at the earliest developmental stages. Furthermore, six treatments proved more cost-effective across multiple fields and years. These observations support the use of atoxigenic strains of A. flavus to promote the safety and marketability of corn grown in areas susceptible to aflatoxin contamination. [ABSTRACT FROM AUTHOR]

The effect of corn-soybean rotation on mycotoxin contamination in corn (Zea mays L.) and soybean (Glycine max L. Merrill.) grains has not been fully evaluated. Therefore, this research investigated the effect of corn-soybean rotation on aflatoxin and fumonisin contamination in respective grains. The results showed that aflatoxin levels in soybean averaged 2.3, < 0.5, 0.6, and 6.8 ng/g in 2005, 2006, 2007, and 2008, while corn aflatoxin levels were 16.7, 37.1, 2.4, and 54.8 ng/g, respectively. Aspergillus flavus colonization was significantly greater (P ⩽ 0.05) in corn (log 1.9, 2.9, and 4.0 cfu/g) compared to soybean (< 1.3, 2.6, and 2.7 cfu/g) in 2005, 2007, and 2008, respectively. Aflatoxigenic A. flavus isolates were more frequent in corn than in soybean in all four years. Higher fumonisin levels were found in corn (0.2 to 3.6 μg/g) than in soybean (< 0.2 μg/g). Rotating soybean with corn reduces the potential for aflatoxin contamination in corn by reducing A. flavus propagules in soil and grain and reducing aflatoxigenic A. flavus colonization. These results demonstrated that soybean grain is less susceptible to aflatoxin contamination compared to corn due to a lower level of colonization by A. flavus with a greater occurrence of non-aflatoxigenic isolates. This study compared cultural and analytical methods for detecting aflatoxin production by Aspergillus species. Aspergillus isolates were obtained from various Mississippi Delta crops (corn, peanut, rice, cotton) and soils. Most of the isolates (99%) were A. flavus and the remainder comprised A. parasiticus and A. nomius. The following three cultural methods were evaluated on potato dextrose agar: fluorescence (FL) on β-cyclodextrin-containing media (CD), yellow pigment (YP) formation in mycelium and medium, and color change after ammonium hydroxide vapor exposure (AV). Aflatoxins in culture extracts were confirmed by thin-layer chromatography (TLC) and quantified by enzyme-linked immunosorbent assay (ELISA). Of the 517 isolates, 314 produced greater than 20 ng/g of total aflatoxin based on ELISA, and 180 produced greater than 10 000 ng/g of aflatoxin in the medium. Almost all the toxigenic isolates (97%) were confirmed by TLC as producers. Of the toxigenic isolates, as determined by ELISA, 93%, 73%, and 70% gave positive FL, YP, and AV responses, respectively. Of the 203 isolates producing less than 20 ng/g of aflatoxin, 20%, 6%, and 0% of respective FL, YP, and AV methods gave false-positive responses. The 9% false-positive results from TLC fall within this range. This study showed good agreement among all tested cultural methods. However, these cultural techniques did not detect aflatoxin in all cultures that were found to produce aflatoxins by ELISA, LC/MS, and TLC. The best results were obtained when the AV color change and CD fluorescence methods were used together, yielding an overall success rate comparable to TLC but without the need for chemical extraction and the time and expense of TLC. Microsatellite loci were identified from the charcoal rot fungus ( Macrophomina phaseolina). Primer pairs for 46 loci were developed, and of these, 13 were optimized and screened using genomic DNA from 55 fungal isolates collected predominantly from two soybean fields in Mississippi. Twelve of the optimized loci were polymorphic and the number of alleles per locus ranged from 6 to 22. These microsatellites will be useful in population and pathogenicity studies to correspond with development of potential disease-resistant soybean and other susceptible crops. [ABSTRACT FROM AUTHOR]

Toxins have been proposed to facilitate fungal root infection by creating regions of readily-penetrated necrotic tissue when applied externally to intact roots. Isolates of the charcoal rot disease fungus, Macrophomina phaseolina, from soybean plants in Mississippi produced a phytotoxic toxin, (−)-botryodiplodin, but no detectable phaseolinone, a toxin previously proposed to play a role in the root infection mechanism. This study was undertaken to determine if (−)-botryodiplodin induces toxic responses of the types that could facilitate root infection. (±)-Botryodiplodin prepared by chemical synthesis caused phytotoxic effects identical to those observed with (−)-botryodiplodin preparations from M. phaseolina culture filtrates, consistent with fungus-induced phytotoxicity being due to (−)-botryodiplodin, not phaseolinone or other unknown impurities. Soybean leaf disc cultures of Saline cultivar were more susceptible to (±)-botryodiplodin phytotoxicity than were cultures of two charcoal rot-resistant genotypes, DS97-84-1 and DT97-4290. (±)-Botryodiplodin caused similar phytotoxicity in actively growing duckweed (Lemna pausicostata) plantlet cultures, but at much lower concentrations. In soybean seedlings growing in hydroponic culture, (±)-botryodiplodin added to culture medium inhibited lateral and tap root growth, and caused loss of root caps and normal root tip cellular structure. Thus, botryodiplodin applied externally to undisturbed soybean roots induced phytotoxic responses of types expected to facilitate fungal root infection. [ABSTRACT FROM AUTHOR]

Pseudomonas chlororaphis strain JP1015 and P. fluorescens strain JP2175 were previously isolated from Mississippi cornfield soil samples and selected for their growth inhibition of Aspergillus flavus in laboratory culture. In this study, the antifungal activity of these bacterial strains against A. flavus in soil coculture was determined. Growth of A. flavus was inhibited up to 100-fold by P. chlororaphis strain JP1015 and up to 58-fold by P. fluorescens strain JP2175 within 3 days following soil coinoculation. A. flavus propagule densities after 16 days remained 7- to 20-fold lower in soil treated with either bacterial strain. Using a bench-scale wind chamber, we demonstrated that treatments of soil with P. chlororaphis strain JP1015 and P. fluorescens strain JP2175 reduced airborne spores dispersed across a 1 m distance by 75- to 1,000-fold and 10- to 50-fold, respectively, depending on soil type and inoculum level. These results suggest that application of these bacterial strains may be effective in reducing soil populations of mycotoxigenic fungi, thereby reducing fungal spore formation, and ultimately reducing the potential for crop plant infection via airborne transmission. Aflatoxin contamination of corn can occur when developing kernels are infected by the plant pathogen Aspergillus flavus. One route of infection is from airborne conidia. We executed a series of experiments within the corn canopy during two growing seasons and in two states to document the abundance and dynamics of the airborne A. flavus population. We did not observe any significant diurnal changes in the conidial density (p = 0.171) or any effect of sampler height (p = 0.882) within the canopy. Significant changes (p < 0.001) were noted during the season, with a trend towards increased airborne populations with later stages of corn development and more than a 20-fold increase from July to August. The median aflatoxigenicity of airborne isolates from a corn canopy in Texas was about 50 times higher than the corresponding population in Mississippi. It was also noteworthy that highly aflatoxigenic, weakly sporulating S-morphotypes accounted for 14-30% of the airborne isolates in Mississippi at a site with historically rare abundance of S-morphotypes. The genetic diversity was high among the 140 analyzed airborne isolates, with 76 unique haplotypes identified and 55 haplotypes occurring only in 1 isolate. Even in the context of this highly diverse population, a haplotype matching that of a commercial biocontrol strain was found in 13 of the 70 isolates from Mississippi and 1 of the 70 isolates from Texas. The airborne A. flavus population is genetically diverse (Shannon's index = 1.4 to 1.6), similar to grain samples in other surveys, and much less aflatoxigenic in Mississippi than in Texas., Competing Interests: Declarations. Two isolates of Beauveria bassiana (Balsamo) Vuillemin, including the commercial strain GHA and the Mississippi Delta native NI8 strain, and two emulsifiers, Tween-80 and a starch-based sprayable bioplastic, were evaluated in the laboratory and field for pathogenicity and infectivity against the tarnished plant bug, Lygus lineolaris (Palisot de Beauvois) (Heteroptera: Miridae). The effect on fruit damage based on within-season cotton plant mapping was also examined. The highest mortality 10 d after treatment was found with insects caged on cotton terminals sprayed with NI8 + Tween-80, followed by those exposed to NI8 + bioplastic. Similarly, sporulation was shown to be higher in NI8 + Tween-80 than in other treatments. Plots sprayed with B. bassiana showed at least a twofold decrease in tarnished plant bug adults 3 d after treatment compared with control plots. Little to no variation was observed in tarnished plant bug nymph populations between treated and untreated plots. Within-season plant mapping provided clear evidence of damage to cotton caused by tarnished plant bug. The highest percentage retention of all first position fruiting structures was observed in plots treated with NI8 + Tween-80 (93.41 ± 1.51) followed by NI8 + bioplastic (90.25 ± 1.52). Both treatments were significantly different when compared with GHA + Tween-80 (82.89 ± 2.26) and GHA + bioplastic (70.48 ± 3.19), and both GHA formulations did not differ from the control (63.61 ± 2.96). Overall, these results indicated that B. bassiana application resulted in >50% mortality of tarnished plant bug regardless of the isolates by direct spray or by contact. Background: Applying non-aflatoxin-producing Aspergillus flavus isolates to the soil has been shown to be effective in reducing aflatoxin levels in harvested crops, including peanuts, cotton and corn. The aim of this study was to evaluate the possibility of controlling aflatoxin contamination using a novel sprayable formulation consisting of a partially gelatinized starch-based bioplastic dispersion embedded with spores of biocontrol A. flavus strains, which is applied to the leaf surfaces of corn plants., Results: The formulation was shown to be adherent, resulting in colonization of leaf surfaces with the biocontrol strain of A. flavus, and to reduce aflatoxin contamination of harvested kernels by up to 80% in Northern Italy and by up to 89% in the Mississippi Delta. The percentage of aflatoxin-producing isolates in the soil reservoir under leaf-treated corn was not significantly changed, even when the soil was amended with additional A. flavus as a model of changes to the soil reservoir that occur in no-till agriculture., Conclusions: This study indicated that it is not necessary to treat the soil reservoir in order to achieve effective biocontrol of aflatoxin contamination in kernel corn. A nontoxigenic Aspergillus flavus strain, K49, is currently being tested as a biological control agent in corn fields in the Mississippi Delta. However, little is known about the overall genetic diversity of A. flavus from year to year in corn fields and specifically in Mississippi. Our objective was to assess the genetic variability of A. flavus isolates from different seasons, inoculum sources, and years, from a no-till corn field. Of the 175 A. flavus isolates examined, 74 and 97 had the typical norB-cypA type I (1.5 kb) and type II (1.0 kb) deletion patterns, respectively. Variability in the sequence of the omtA gene of the majority of the field isolates (n = 118) was compared to strain K49. High levels of haplotypic diversity (24 omtA haplotypes; Hd = 0.61 ± 0.04) were found. Among the 24 haplotypes, two were predominant, H1 (n = 71), which consists of mostly toxigenic isolates, and H49 (n = 18), which consists of mostly atoxigenic isolates including K49. Toxigenic isolates were prevalent (60%) in this natural population. Nonetheless, about 15% of the population likely shared the same ancestral origin with K49. This study provides valuable information on the diversity of A. flavus. This knowledge can be further used to develop additional biological control strains.

Mycotoxin and Aspergillus flavus levels in soil-surface corn debris left by no-till agriculture methods (stover, cobs, and cobs with grain) were determined during the December-March fallow period for near-isogenic Bt and non-Bt hybrid corn. By December, average mycotoxin levels in non-Bt corn were many times higher in cobs with grain than in grain harvested in September (total aflatoxins, 774 vs 211 ng/g; total fumonisins, 216 vs 3.5 microg/g; cyclopiazonic acid, 4102 vs 72.2 microg/g; zearalenone, 0.2 vs < 0.1 microg/g). No trichothecenes were detected. Levels of mycotoxins and A. flavus propagules were approximately 10- to 50-fold lower in cobs without grain and stover, respectively, for all mycotoxins except zearalenone. Mycotoxin levels in corn debris fractions decreased during winter but began to rise in March. Levels of all mycotoxins and A. flavus propagules were lower in harvested grain and debris from Bt than non-Bt corn, but differences were significant (p < 0.05) only for aflatoxins.
