The aim of vaccination in an eradication campaign is not only to induce clinical protection, but primarily to stop transmission of infections within and between herds by inducing herd immunity. Consequently, vaccines should be evaluated for their capacity to reduce virus transmission in the population. Glycoprotein E (gE) negative marker vaccines against Pseudorabies virus (PRV) infections in pigs have been evaluated this way in experiments and field studies. PRV infection in groups of (vaccinated) pigs was determined by measuring antibodies against gE of PRV from regularly taken serum samples. For the statistical analysis of the experiments a stochastic susceptible-infectious-removed (SIR) model was used. A measure for the transmission of virus is the reproduction ratio R, which is defined as the average number of secondary cases caused by one typical infectious individual. This implies that an infection will always fade out in a population when R < 1, but the infection can spread massively when R > 1. From several experiments it was shown that R < 1. Field studies showed that the R within herds was still > 1, but by reducing further contacts the R could be reduced to a value below one. This would imply that PRV could be eradicated by means of vaccination. In The Netherlands, an eradication campaign was launched in 1993, and in 2002 the virus was eradicated, as shown by a negligible number of gE-positive pigs. Farmers' organizations have to decide whether or not to stop vaccination.

Pseudorabies virus will be eradicated from the Netherlands if a typical infectious pig (Rind) infects, on average, less than one other pig. In this review, we used a stochastic SIR model to estimate Rind using data from the field and from experiments. Rind in sow herds was estimated to be significantly less than 1 and in rearing and finishing pigs Rind was higher than 1. However, if Rind is higher than 1, PRV can still be eradicated if one infectious herd infects less than one other herd during the period that the herd is infectious(Rherd <1). Some future developments in Dutch pig husbandry (e.g. group-housing of sows) and possible risks after halting vaccination are also quantitatively evaluated.

The Aujeszky's disease virus (ADV) eradication campaign in The Netherlands is based on compulsory vaccination and certification of ADV-free herds and consists of three stages: reduction of transmission of ADV, tracing and elimination of remaining sources of ADV, and, in the last stage prohibition of vaccination. This paper summarizes the progress made in the first two stages of the eradication. Between 1994-1999, ADV seroprevalence decreased sharply in all four regions in The Netherlands, both in the sow and the finishing pig population. In the sow population, the mean ADV prevalence in the northern, eastern and western regions decreased to less than 0.5% in the first quarter of 1998, and to approximately 1% in the southern region. For the finishing pig population, the mean ADV prevalence decreased to less than 0.6% in all four regions in the first quarter of 1998. The proportion of sow herds that were certified ADV-free increased steadily from approximately 40% in the last quarter of 1996 to 96% in the first quarter of 1999. The proportion of finishing pig herds that were certified ADV-free showed the same spectacular increase as in the sow population: from approximately 14% in the last quarter of 1996 to 90% in the first quarter of 1999. It is anticipated that the last stage of the ADV eradication campaign, stopping vaccination, will be achieved very soon. Monitoring wild boars for ADV between 1994 and 1999 indicated that ADV is uncommon within the wild boar population. Therefore, it seems that ADV infection in the wild boar population is not an important reservoir in The Netherlands.

A surveillance programme for swine vesicular disease (SVD) and Aujeszky disease was set up in 1993 in the Netherlands. Blood samples are taken from pigs by local veterinarians to enable testing for the presence of antibodies against these viruses. A programme to guarantee the identity of pigs tested for these diseases has been in operation since late 1995. In this programme, pigs are identified on the basis of the DNA profiles of blood and hair samples. The hair samples are collected by the department of Animal Health Control. Analysis proves whether blood and hair samples are from the same animal. A total of 655 animals have been tested from 94 farms. In 38.6% of these farms differences were found between blood and hair samples.

Simple mathematical models based on experimental and observational data were applied to evaluate the feasibility of eradicating pseudorabies virus (PRV) regionally by vaccination and to determine which factors can jeopardise eradication. As much as possible, the models were uncomplicated and our conclusions were based on mathematical analysis. For complicated situations, Monte-Carlo simulation was used to support the conclusions. For eradication, it is sufficient that the reproduction ratio R (the number of units infected by one infectious unit) is < 1. However, R can be determined at different scales: at one end the region with the herds as units and at the other end compartments with the pigs as units. Results from modelling within herds showed that contacts between groups within a herd is important whenever R between individuals (R(ind)) is > 1 in one or more groups. This is the case within finishing herds. In addition, if the R(ind) is more than 1 within a herd, the size of the herd determines whether PRV can persist in the herd and determines the duration of persistence. Moreover, when reactivation of PRV in well-vaccinated sows is taken into account, R(ind) in sow herds is still less than 1. In sow herds with group-housing systems, it is possible that in those groups R(ind) is > 1. Results from modelling between herds showed that whether or not Rherd is < 1 in a particular region is determined by two factors: (1) the transmission of infection between nucleus herds and rearing herds through transfer of animals and (2) contacts among finishing herds and among rearing herds. The transmission between herds can be reduced by reduction of the contact rate between herds. reduction of the herd size, and reduction of the transmission within herds.

A further integration of international markets makes a coordinated policy against contagious animal infections increasingly important. In the future, stricter demands are to be expected concerning the control and eradication of such infections. To anticipate these demands, a computer simulation model is created in which scenarios can be evaluated with respect to epidemiological and economic effects of the infections and control strategies. In this paper, the simulation model is described for Pseudorabies in swine. In the model, the population of herds is subdivided into two main herd types: breeding and finishing. Each herd is in one of 24 states per herd type. The states are based on (1) the reproduction ratio R which is the number of secondary cases caused by one infectious herd, (2) the prevalence for each value of R and (3) the expected number of infectious animals in an infectious herd within each prevalence range and for each R. The different values of R are based on experiments and field data in which different vaccination strategies were used. The transition matrix with the probabilities of every transition from one state to another is calculated on a weekly base. With this matrix the distribution of herds over states from week to week is derived. To include a dynamic element in the transition probabilities, the number of newly infectious herds per week is a function of animal and other contacts, including aerial, material and personal contacts. Calculations show that the infection in the Dutch swine population will not disappear without vaccination. With a vaccination scheme in which sows are vaccinated 3 times per year and fattening pigs 1 time per cycle the infection will ultimately be eradicated, but 2 vaccinations per cycle for fattening pigs are needed to eradicate the infection within an acceptable timespan (i.e. 2 to 3 years). The latter strategy will become compulsory in the Netherlands from October 1st 1995.

The Dutch Aujeszky's disease virus (ADV) eradication campaign is based on vaccination with glycoprotein E deleted vaccines. In the first stage of the programme, that was started in September 1993, the transmission of ADV must be reduced sharply. Subsequently, the remaining sources of virus need to be traced and eliminated. During the final stage, vaccination should be forbidden. This paper summarizes the observations made during a field study on the eradication of ADV by vaccination and reports the design and preliminary results of the first stage of the Dutch eradication campaign.

From February to June 1995, 5-12 blood samples were collected in each Dutch pig herd and tested for antibodies against pseudorabies virus (PRV). The percentage of PRV-seropositive pig-finishing herds in three regions with more than 1,000 pigs/km2 (regions 2, 4 and 7) was 6% (region 2), 12% (region 7), and 25% (region 4). The percentage of PRV-seropositive pig-finishing herds in five regions with fewer than 1,000 pigs/km2 (regions 1, 3, 5, 6, and 8) was 3% (region 1), 9% (region 3), 6% (region 5), 0% (region 6), and 4% (region 8). The small sample size allows only the detection of major outbreaks and the percentages of PRV-seropositive herds therefore under-estimate the actual virus circulation in the regions. The fraction of PRV introductions that will result in a major outbreak depends on the herd immunity and thus on the vaccination programme of the herds. By combining for each herd the occurrence or absence of a major outbreak with the herd immunity induced by the vaccination programme, we estimated the average rate at which PRV was introduced into finishing herds in the eight regions. The average number of PRV introductions per finishing herd per finishing period (16 weeks) in the pig-dense regions was estimated at 0.20 in region 2, 0.83 in region 4, and 0.48 in region 7. In the less densely populated regions this rate was estimated at 0.08 in region 1, 0.34 in region 3, 0.17 in region 5, 0.00 in region 6, and 0.09 in region 8. The eight regions could be classified into four areas with a statistically different (P < 0.05 in Mann Whithey U test) rate of PRV introduction: i) regions 1, 6, and 8; ii) regions 2 and 5; iii) regions 3 and 7; and iv) region 4.

The breeding and finishing pig populations in four Animal Health Service regions in the Netherlands were monitored with respect to pseudorabies virus (PRV) seroprevalence. Analysis of data of the seroprevalence survey of 1994 indicated that two samples per herd was sufficient to estimate the seroprevalence in both the sow and finishing pig populations. In the northern, eastern, southern, and western regions, 115, 645, 940, and 218 sow herds and 114, 1036, 954, and 323 pig finishing herds were sampled, respectively. In the breeding pig population, the PRV-seroprevalence decreased from 27% to 17% in the eastern region, from 32% to 17% in the southern region, from 18% to 12% in the western region, and from 10% to 6% in the northern region. In the finishing pig population, the PRV-seroprevalence decreased from 15% to 6% in the eastern region, from 19% to 6% in the southern region, and from 12% to 5% in the western region. There was no significant difference in PRV seroprevalence in finishing pigs in the northern region between the survey of 1994 and 1995.

In January 1990, a 6-year program was initiated to eliminate endemic Aujeszky's Disease virus (ADV) infection from the pig herds in an area of Northern Germany, bordering Southern Denmark, with intensive pig farming. In the first 3 years of the campaign, an intensive compulsory vaccination program, with glycoprotein I (gI)-deleted vaccines, of all pigs in the area was employed. Beginning in June 1990 and for the first 3 years of the project, approximately 200 herds randomly selected from all herds in the area, were serologically tested each quarter. In each farrow-to-feeder (FAFE), feeder-to-finish (FEFI) and farrow-to-finish (FAFI) herd, 20 female breeding pigs, 20 finishing pigs (> or = 50 kgs liveweight) and 10 female breeding pigs and 10 finishing pigs, respectively, were blood sampled. The sera were tested by the Herd-Check Anti-PRV(S) ELISA test (IDDEX Inc., ME). Sera positive to this test were examined by the HerdCheck Anti-ADV gI-ELISA test (IDDEX Inc., ME). Data on potentially confounding management factors were collected through a pilot-tested questionnaire, administered to farmers by 2 veterinarians who blood sampled the pigs. For fattening herds (FEFI and fattening sections of FAFI herds), the association between the odds of > or = 1 gI+ finishing pigs and the time between initiation of the program in the area and sampling date (a surrogate for the effect of the program) was modelled using ordinary logistic regression. The association between the odds of gI+ females in seropositive (> or = gI+ females) FAFE and FAFI herds and time since initiation of the program was investigated with logistic-binomial regression models. Results of the study show that the longer the period from the beginning of compulsory vaccination to the date the herd was sampled the lower the odds of gI+ fattening herds and gI+ female breeding pigs in herds of the area. The beneficial effect of mass vaccination on the reduction of ADV spread was accounted for by this relationship. For fattening herds this relationship appeared curvilinear, with the reduction in the log-odds being more rapid in the 1st year of the program. This non-linear pattern indicates that for the elimination of the risk of ADV-infection from fattening herds of the area, the mass vaccination program should be complemented with additional measures such as test-and-slaughter of infected breeding pigs. A computerized economical model to estimate the effects of ADV-infection at the herd and area level has been developed. The analytical structure consists of a basic epidemiological model linked to an economic estimation framework. The economic model predictions allow priorities to be given to alternative control strategies. Mass vaccination of all pigs in regions with endemically infected herds followed by test-and-removal of seropositive animals is the most cost-effective way to control the spread of ADV within the swine population. Other possible control strategies such as intensive vaccination or complete test-and-removal all had higher overall costs, either because of the less efficient production, or because of the high costs of straight test-and-removal.

In the European Union, vaccination campaigns against Pseudorabies virus (PRV) in swine have been started to eradicate PRV. Specific sampling designs are needed to monitor PRV seroprevalence at a regional level. This paper demonstrates how sampling theory can be applied to design a disease seroprevalence survey, using PRV as an example. In the spring of 1994, the four regions in the Netherlands covered by the regional Animal Health Services were monitored with respect to PRV seroprevalence. Per region, blood samples from approximately 1400 herds, with two animals per herd, were collected. The sampling design accounted for stratification by fattening pig and sow population within each region. The regional PRV seroprevalence of swine in the Southern region was the highest (24.9%), closely followed by the PRV seroprevalence of swine in the Eastern region (20.5%). These regions have the highest density of swine in the Netherlands. The PRV seroprevalence in the Western and Central region (11.7%) was about half of the seroprevalence in the Southern and Eastern regions; the lowest regional PRV seroprevalence was observed in the Northern region (3.5%). The Northern part also has the lowest pig density. The PRV seroprevalence was approximately two times higher in sows than in fattening pigs.

Whereas the clinical efficacy of vaccination against pseudorabies has been studied extensively, methods to evaluate the influence of vaccination on pseudorabies virus (PRV) transmission have only recently become available. In this study, PRV transmission and growth performance in finishing pigs vaccinated either once or twice were compared. The incidence of PRV infections was significantly (P = 0.039) higher in the group vaccinated once (38%) than in the group vaccinated twice (10%). The reproduction ratio R, which is defined as the average number of new infections caused by 1 infectious individual, was estimated in both groups. This ratio was also significantly (P = 0.025) higher among single vaccinated pigs (R = 3.4) than among pigs that had received double vaccination (R = 1.5). In compartments where serologic evidence of PRV introduction was observed, the mean daily weight gain was significantly (P = 0.029) lower in pigs vaccinated once (698 g/d) than in pigs vaccinated twice (721 g/d). Results of this study document the possibility to objectively evaluate the effect of vaccination on PRV transmission under field conditions. From the results, we concluded that double vaccination is advantageous in populations of finishing pigs at risk for PRV introduction. However, even among pigs vaccinated twice, extensive spread of PRV can occur.

Aujeszky's disease is caused by a herpesvirus. The pig is the natural host and sole source of dissemination of the virus. After a primary infection, during which the pig excretes virus for 10-14 days, the virus remains latent in the host. Eradication has been achieved in individual herds, areas and countries with a low prevalence. The crucial question is whether it is feasible to eradicate the virus in heavily infected areas. Using so-called marker vaccines in conjunction with companion diagnostic test kits the virus was eliminated in individual herds. The preliminary results of extensive field studies to investigate whether intensive vaccination curtails the circulation of virus are encouraging.

Over a period of six months, approximately 4700 blood samples were collected from 97 pig-finishing farms in the provinces of Noord-Brabant and Gelderland and screened for antibodies with respect to Aujeszky's disease virus (ADV), porcine influenza virus (PI) and Actinobacillus (Haemophilus) pleuropneumoniae (App). There were significant differences in the percentages of seropositive pigs between the two provinces, which may be related to the difference in the density of the pig population in the two provinces. In practice, it was possible to perform a reliable sera collecting procedure at the slaughterhouse. No farms remained seronegative with respect to most of the disease agents during the sampling period. There was a high degree of variation in the percentages of seropositive pigs per farm as to most of the disease agents. Evidence was found that animals that were seropositive with respect to ADV were significantly more susceptible to becoming seropositive with respect to App. serotype 2, and vice versa. The same connection was observed for PI serotype H1N1 and PI serotype H3N2. Furthermore, evidence was found that pigs seropositive with respect to PI serotype H1Ni only, or to PI serotype H1N1 and ADV or PI serotype H3N2 show a significant decrease in average daily weight gain compared to pigs that were seronegative.

Pigs are either directly or indirectly the source of infection with Herpes virus suis in dogs and cats. On oral or oronasal infection, the virus is spread by way of the cranial nerves to the central nervous system. The incubation period is short: from one to six days. The disease is mainly characterised by nervous disorders. Pruritus is a characteristic clinical sign but is not present in every case. Mortality in dogs and cats is a hundred per cent. In rural areas, infection is caused by consumption of uncooked pork or offal from pigs. Prevention is possible by avoiding exposure to pigs or fresh pig products. It is possible to vaccinate dogs and cats with an inactivated vaccine. Results of vaccination studies show some variation, however.

On seven fattening farms and seven breeding farms, investigations were carried out to examine whether Aujeszky virus was capable of persisting in particular animal groups or units of the farm as a productive infection following a virologically verified outbreak. After an outbreak on fattening farms, virus persisted temporarily as newly arrived piglets became systematically infected. On the farms on which this could be accurately followed, virus persistence continued for at least 1 1/2 to 2 months. However, this cycle of infection was found to be interrupted on each farm within approximately three months after the outbreak, which resulted in a gradual return to a totally sensitive animal population, so that new outbreaks could occur. On the seven swine breeding farms, virus persistence in the farrowing house was examined by placing seronegative sentinel piglets with sows during the immediate post-partum period. Twelve sentinel piglets were placed in the various farrowing houses for a total period of sixty-seven weeks, during which time 172 sows farrowed. None of these piglets became seropositive during their stay on the breeding farms. The results of this study show that Aujeszky's disease virus did not persist on breeding farms and that the productive infection on fattening farms in only transient following an outbreak. Therefore, it is very likely that new outbreaks of the disease are due to reintroduction of Aujeszky's disease virus on the farm. Though the role of latent carrier animals cannot be totally disregarded, natural reactivation of latent virus is believed an exception rather than the general rule.

The small number of cases of Aujeszky's disease in goats referred to in the literature is stressed, and an outbreak in goats causing many deaths is reported. The flock of goats concerned was occasionally housed in a barn during the night in which there also were fattening pigs. The first deaths occurred without previous symptoms, the animals being found dead in the morning. In subsequent cases, symptoms were observed which continued for a few or several hours and consisted in agitation, lying down and rising, screaming plaintively, profuse sweating and, in the terminal stage, spasms and paralysis. Pruritus was not observed in any of the animals. Of the fifteen goats which had been housed in the pig-sty, thirteen died during ten days. In the same period, only one out of forty pigs died, and the others showed anorexia and somnolence for a couple of days but soon recovered. In two goats studied, the virus of Aujeszky's disease was isolated from the central nervous system. It is suggested that, in cases of disease or death in ruminants occurring in the proximity of pigs, there should primarily be a strong suspicion of Aujeszky's disease, regardless of the symptoms observed.

When Aujeszky's disease was detected on a large pig-breeding farm, all normal sows and boars were vaccinated with an ethyl-ethylene-imine (EEI) vaccine produced in the Bundesforschungsanstalt für Viruskrankheiten der Tiere (Federal Research Institute for Viruses Diseases in Animals) in Tübbingen, Western Germany. The vaccine contained DEAE dextran as an adjuvant. The vaccine was inoculated twice by intramuscular route at a three weeks' interval, 5 ml. being inoculated at one time. Hyperimmune serum was inoculated in young pigs. In addition, hygienic procedures were adopted, consisting in disinfection and isolation. The disease was successfully controlled within eight weeks. Sows which developed Aujeszky's disease showed hardly any symptoms suggesting lesions of the central nervous system. Pulmonary lesions were the outstanding clinical feature. The semen quality of the affected male swine had markedly deteriorated. This was resorted to normal, however, within from five to ten weeks. The incidence of abortion and piglet mortality were low. In view of the results of virological and serological studies, it was concluded that the persistence of this strain of virus (causing Aujeszky's disease) was very slight.
