Unimproved landraces and wild relatives of crops are sources of genetic diversity that were lost post domestication in modern breeding programs. To tap into this rich resource, genome-wide association studies in large plant genomes have enabled the rapid genetic characterization of desired traits from natural landrace and wild populations. Wild barley (Hordeum spontaneum), the progenitor of domesticated barley (Hordeum vulgare), is dispersed across Asia and North Africa, and has co-evolved with the ascomycetous fungal pathogens Pyrenophora teres f. teres and P. teres f. maculata, the causal agents of the diseases net form of net blotch and spot form of net blotch, respectively. Thus, these wild and local adapted barley landraces from the region of origin of both the host and pathogen represent a diverse gene pool to identify new sources of resistance, due to millions of years of co-evolution. The barley—P. teres pathosystem is governed by complex genetic interactions with dominant, recessive, and incomplete resistances and susceptibilities, with many isolate-specific interactions. Here, we provide the first genome-wide association study of wild and landrace barley from the Fertile Crescent for resistance to both forms of P. teres. A total of 14 loci, four against P. teres f. maculata and 10 against P. teres f. teres, were identified in both wild and landrace populations, showing that both are genetic reservoirs for novel sources of resistance. In North Africa, barley (Hordeum vulgare L) is the second most cultivated cereal. In Tunisia, barley is cultivated in mining areas with possible Cd soil contamination. The accumulation of Cd was studied in the 36 most cultivated North African barley cultivars cultured during 6 months on control soil and on soil containing 10 ppm of Cd. Cadmium did not affect germination and morphology in any cultivar. However, Cd induced variable effects on the biomass according to the cultivar. The cultivar Lemsi was the most sensitive one and Gisa 127 the most tolerant to Cd. The spike morphology did not show any differences between control and Cd-treated plants. The number of grains per spike and the weight of kernels were differently affected by Cd. On this basis, we identified Manel, Temassine, Giza 130, and Firdaws as the most tolerant cultivars and Raihane, Giza 123, Adrar, and Amira as the most sensitive ones. Cd accumulated at a higher concentration in straw than in the grains, but for both organs, we observed a significant intraspecific variability. In the straw, Lemsi and Massine showed the highest Cd concentration, while the lowest concentration was recorded in Temassine. In the kernels, Amalou showed the highest Cd concentration, 14 μgg−1 of dry weight (DW), but the lowest Cd concentration was 1.7 μg g−1 DW in Kebelli. Based on the official allowable limit of Cd in the grain, all cultivars represent a potential risk when cultivated on soil contaminated with 10 ppm Cd. Abstract: It was demonstrated that some North Africa barley accessions have diverse tolerance sources for abiotic stresses and a good nutritional quality, but the studies done were incomplete since they were realized separately in each country apart. To implement a more complete analysis, 31 barley accessions originated from North Africa (Algeria, Tunisia and Egypt) were analyzed using 11 SSR markers selected from the seven barley linkage groups for studying the genetic diversity among these chosen barley accessions. Over the 11 SSR markers, a total of 478 reproducible bands were scored with an average of 2.13 alleles/primer and the average polymorphism information content of 0.5. Genetic distance analysis of the 31 accessions showed a large genetic diversity and high number of different groups. The most accessions are clustered according to their eco-geographical origin, according to their pedigree and agronomic characters or according to the caryopsis character (hulled or naked caryopsis). This high number of obtained groups is an invaluable aid in crop improvement strategies and confirms the opinion suggesting that North Africa could be a secondary center of origin of barley. The various growing conditions and the multiple uses of barley in each country may be the cause of the large variability of the barley germplasm in each region. [Copyright &y& Elsevier]

Improved boron (B) tolerance has been an objective of barley breeding programs in regions where B toxicity occurs. Traits associated with B tolerance have been mapped on chromosomes 2H and 4H and it has been proposed that these be used for marker assisted selection for B tolerance. However, there has been little or no improvement in yield using this strategy. This study examined the reasons for the small yield differences among different lines of barley that differ in B tolerance. Experiments used backcross lines derived from crosses between the B-tolerant landrace Sahara 3771 and two adapted recurrent parents, Sloop and VB9104. Lines with different combinations of the Sahara 3771 alleles on chromosomes 2H and 4H were grown over three growing seasons at sites where barley is prone to B toxicity. Grain yields of the backcross lines were similar to or lower than those of the recurrent parents despite showing differences in the expression of B toxicity symptoms and in B concentration in vegetative tissue. There were few significant differences in grain yield among the backcross lines. Variation in dry matter production among the backcross lines in each of the three growing seasons was unrelated to shoot B concentrations while grain yield was correlated with shoot B concentration only among the backcross lines of VB9104 in one season. In this case the yield loss was 4% per 10 mg kg-1 increase in shoot B concentration. Variation in shoot B concentration and yield across seasons was much greater than that observed among the different barley lines. Reduced B accumulation was associated with higher shoot sodium concentration among the Sloop backcross lines. The results suggest that yield gains from selection based largely on B exclusion and symptoms expression may be small and strongly affected by site and seasonal effects. In the regions where other soil constraints, such as soil salinity and micronutrient deficiencies are also important, reducing B uptake alone may have little effect on yield if these other soil properties are also limiting yields. Barley is one of the oldest domesticated crops in the world and is mainly used for feed and malt and to a lesser extent as food. The use of barley as food is a tradition in communities in some countries of North Africa, Europe, and Asia. However, due to the health-promoting properties of barley grain, there is an increasing interest in such use. The International Center for Agricultural Research in the Dry Areas (ICARDA) has a global mandate for barley improvement and holds rich in-trust collections of cultivated and wild Hordeum species. The present study aims to evaluate 117 accessions of Hordeum spontaneum for their contents of β-glucan and microelements for breeding new varieties with enhanced nutritional value. The bulked seed accessions of Hordeum spontaneum were grown over two seasons, and the single plant derived seeds from these accessions were compared to 36 elite lines and varieties of cultivated barley in the second season in Morocco. The results showed large differences in β-glucan and microelements in both the bulk and the single plant seed accessions. The contents of β-glucans ranged from 1.44 to 11.3% in the Hordeum spontaneum accessions and from 1.62 to 7.81% in the cultivated barley lines. Large variations were found for the microelements content, but no differences were noticed between the wild and the cultivated species. However, some accessions of Hordeum spontaneum had higher combined contents of Iron, Zinc, and Selenium.
