Corynebacterium pseudotuberculosis causes disease in several animal species, although distinct biovars exist that appear to be restricted to specific hosts. In order to facilitate a better understanding of the differences between biovars, we report here the complete genome sequence of the equine pathogen Corynebacterium pseudotuberculosis strain 1/06-A.

Mitochondrial DNA (mtDNA) was extracted and analyzed from the skeletal remains of 44 individuals, representing four prehistoric populations, and compared to that from two other prehistoric and several contemporary Native American populations to investigate biological relationships and demographic history in northeastern North America. The mtDNA haplogroup frequencies of ancient human remains from the Morse (Red Ocher tradition, 2,700 BP) and Orendorf (Mississippian tradition, 800 BP) sites from the Central Illinois River Valley, and the Great Western Park (Western Basin tradition, 800 BP) and Glacial Kame (2,900 BP) populations from southwestern Ontario, change over time while maintaining a regional continuity between localities. Haplotype patterns suggest that some ancestors of present day Native Americans in northeastern North America have been in that region for at least 3,000 years but have experienced extensive gene flow throughout time, resulting, at least in part, from a demic expansion of ancestors of modern Algonquian-speaking people. However, genetic drift has also been a significant force, and together with a major population crash after European contact, has altered haplogroup frequencies and caused the loss of many haplotypes.

Previous studies have investigated the human population history of eastern North America by examining mitochondrial DNA (mtDNA) variation among Native Americans, but these studies could only reconstruct maternal population history. To evaluate similarities and differences in the maternal and paternal population histories of this region, we obtained DNA samples from 605 individuals, representing 16 indigenous populations. After amplifying the amelogenin locus to identify males, we genotyped 8 binary polymorphisms and 10 microsatellites in the male-specific region of the Y chromosome. This analysis identified 6 haplogroups and 175 haplotypes. We found that sociocultural factors have played a more important role than language or geography in shaping the patterns of Y chromosome variation in eastern North America. Comparisons with previous mtDNA studies of the same samples demonstrate that male and female demographic histories differ substantially in this region. Postmarital residence patterns have strongly influenced genetic structure, with patrilocal and matrilocal populations showing different patterns of male and female gene flow. European contact also had a significant but sex-specific impact due to a high level of male-mediated European admixture. Finally, this study addresses long-standing questions about the history of Iroquoian populations by suggesting that the ancestral Iroquoian population lived in southeastern North America.

Haplogroup X represents approximately 3% of all modern Native North American mitochondrial lineages. Using RFLP and hypervariable segment I (HVSI) sequence analyses, we identified a prehistoric individual radiocarbon dated to 1,340 +/- 40 years BP that is a member of haplogroup X, found near the Columbia River in Vantage, Washington. The presence of haplogroup X in prehistoric North America, along with recent findings of haplogroup X in southern Siberians, confirms the hypothesis that haplogroup X is a founding lineage., (Copyright 2002 Wiley-Liss, Inc.)

The mitochondrial DNA haplogroups and hypervariable segment I (HVSI) sequences of 1,612 and 395 Native North Americans, respectively, were analyzed to identify major prehistoric population events in North America. Gene maps and spatial autocorrelation analyses suggest that populations with high frequencies of haplogroups A, B, and X experienced prehistoric population expansions in the North, Southwest, and Great Lakes region, respectively. Haplotype networks showing high levels of reticulation and high frequencies of nodal haplotypes support these results. The haplotype networks suggest the existence of additional founding lineages within haplogroups B and C; however, because of the hypervariability exhibited by the HVSI data set, similar haplotypes exhibited in Asia and America could be due to convergence rather than common ancestry. The hypervariability and reticulation preclude the use of estimates of genetic diversity within haplogroups to argue for the number of migrations to the Americas.

Russian olive (Elaeagnus angustifolia L.) was introduced in 1950 onto one site on the Milk River floodplain, northern Montana, 10 km downstream from the Canada/United States border. To analyze dispersal of Russian olive from the point source between 1950 and 1999, we compared distribution, numbers, size structure, and mortality of Russian olive and plains cottonwood (Populus deltoides Marsh:) on an unregulated reach of the Milk River floodplain in southeastern Alberta and north-central Montana. Within 50 years, Russian olive in this reach has moved upriver into Alberta and downriver to the Fresno Reservoir. It is now present on 69 of the 74 meander lobes sampled, comprising 34%, 62%, and 61% of all Russian olive and plains cottonwood seedlings, saplings, and trees, respectively. On some meander lobes, Russian olive has colonized similar elevations on the floodplain as plains cottonwood and is oriented in rows paralleling the river channel, suggesting that recruitment may be related to river processes. Breakup ice had killed 400 Russian olive saplings and trees and damaged >1000 others on 30 of the meander lobes in 1996. Nevertheless, Russian olive now outnumbers cottonwood on many sites on the Milk River floodplain because its seeds can be dispersed by wildlife (particularly birds) and probably by flood water and ice rafts; seeds are viable for up to 3 years and germination can take place on bare and well-vegetated soils; and saplings and trees are less palatable to livestock and beaver than plains cottonwood. Without control, Russian olive could be locally dominant on the Milk River floodplain in all age classes within 10 years and replace plains cottonwood within this century.

The deletion of a 9-bp segment from the intergenic region between the mtDNA cytochrome oxidase II gene and the lysine tRNA gene has been documented mainly in individuals of East Asian ancestry and in individuals from East Asian-derived populations (e.g., Polynesia). Among Native Americans the deletion is absent among Eskimos and northern Na-Dene populations and present among most Amerind populations [sensu Greenberg (1987); i.e., all Native Americans except Eskimo-Aleut and Na-Dene] that have been studied. To better characterize the frequency and distribution of the 9-bp deletion in North America, we surveyed more than 400 individuals from 59 tribes representing a variety of linguistic groups. The absence of the deletion among Eskimo and northern Na-Dene populations is confirmed. Among Amerind groups the deletion is present in all groups represented by more than six individuals. The geographic distribution of the frequencies of the deletion appears to be clinal in North America. The deletion is absent in the Artic and Subartic and reaches its highest frequency in the Southwest. This distribution is consistent with the hypothesis that the ancestors of the Amerinds and Na-Dene arrived in the New World by means of separate migrations. The presence of the 9-bp deletion in high frequencies in all the major linguistic groups in the Southwest suggests that migration among tribes was common.
