The gut microbiota in animals is a dynamic ecosystem influenced by both the host itself and the environment it inhabits. It is known that short-term captivity can significantly impact the gut microbiota of plateau zokors, leading to substantial inter-individual variation. However, the specific changes in the assembly process of the gut microbiota in plateau zokors during captivity remain unclear. In this study, we conducted a comparative analysis on the assembly process of the gut microbiota in 22 male plateau zokors from the same location in Qinglin Township, Datong County, Qinghai Province, before (W) and after (L) laboratory rearing. We performed a single-factor correlation network analysis on the top 50 genera with relative abundance in each group. The results revealed that captivity increased the complexity of the gut microbiota in plateau zokors, indicating a higher number of interactions between different microbial species. However, this increase in complexity was accompanied by a decrease in stability, suggesting a higher degree of variability and potential disruption in the microbial community. According to the results of the neutral community model, the gut microbiota of plateau zokors in the W had a higher Nm value (Nm = 48,135) compared to the L (Nm = 39,671), indicating that species dispersal of the gut microbiota was greater in the wild than in captivity. In the wild, the modified stochasticity ratio (MST) was less than 0.5, suggesting that deterministic processes dominated. However, after 15 days of laboratory rearing, the MST became greater than 0.5, indicating a shift toward stochastic processes, and this difference was highly significant (p < 0.001). Wild-caught animals must cope with drastic lifestyle and dietary changes after being induced to captivity. How the gut microbiome structure of these animals will change in response receives increasing attention. The plateau zokor (Eospalax baileyi), a typic subterranean rodent endemic to the Qinghai-Tibet plateau, spends almost the whole life underground and is well adapted to the environmental pressures of both plateau and underground. However, how the gut microbiome of the plateau zokor will change in response to captivity has not been reported to date. This study compared the microbial community structure and functions of 22 plateau zokors before (the WS group) and after being kept in captivity for 15 days (the LS group, fed on carrots) using the 16S rRNA gene via high-throughput sequencing technology. The results showed that the LS group retained 973 of the 977 operational taxonomic units (OTUs) in the WS group, and no new OTUs were found in the LS group. The dominant bacterial phyla were Bacteroides and Firmicutes in both groups. In alpha diversity analysis, the Shannon, Sobs, and ACE indexes of the LS group were significantly lower than those of the WS group. A remarkable difference (P < 0.01) between groups was also detected in beta diversity analysis. The UPGMA clustering, NMDS, PCoA, and Anosim results all showed that the intergroup difference was significantly greater than the intragroup difference. And compared with the WS group, the intragroup difference of the gut microbiota in the LS group was much larger, which failed to support the assumption that similar diets should drive convergence of gut microbial communities. PICRUSt revealed that although some functional categories displayed significant differences between groups, the relative abundances of these categories were very close in both groups. Based on all the results, we conclude that as plateau zokors enter captivity for a short time, although the relative abundances of different gut microbiota categories shifted significantly, they can maintain almost all the OTUs and the functions of the gut microbiota in the wild. So, the use of wild-caught plateau zokors in gut microbial studies is acceptable if the time in captivity is short. [ABSTRACT FROM AUTHOR]

The Qinghai-Tibetan Plateau (QTP, in western China), which is the largest and highest plateau on Earth, is a highly epidemic region for Echinococcus spp. We collected 70 Echinococcus samples from humans, dogs, sheep, yaks, plateau pikas, and voles in eastern and southern Qinghai and genotyped them using the mitochondrial DNA marker cytochrome oxidase subunit I gene and maximum parsimony and Bayesian reconstruction methods. Based on the 792-bp sequence matrix, we recorded 124 variable sites, of which, 115 were parsimony-informative. Thirty-four haplotypes (H1-H34) were detected, of which H1-H15, H16-H17, and H18-H34 belonged to Echinococcus shiquicus, Echinococcus multilocularis, and Echinococcus granulosus, respectively. Within 26 human isolates, three were identified as E. multilocularis and 23 were E. granulosus. We also detected a dual infection case in a dog with E. multilocularis and E. granulosus. The intraspecific haplotype (Hd ± SD) and nucleotide (Nd ± SD) diversity of E. shiquicus (0.947 ± 0.021; 0.00441 ± 0.00062) was higher than that for E. granulosus (0.896 ± 0.038; 0.00221 ± 0.00031) and E. multilocularis (0.286 ± 0.196; 0.00036 ± 0.00025). Moreover, the haplotype network of E. shiquicus showed a radial feature rather than a divergent feature in a previous study, indicating this species in the QTP has also evolved with bottleneck effects. National parks in China play a vital role in preserving biodiversity, protecting endangered species, maintaining ecological balance, and promoting sustainable development amid global biodiversity decline. Boundary delimitation and functional zoning within these parks are crucial for preserving ecosystem integrity and species diversity. In the case study of Kunlun Mountains National Park (KMNP, Qinghai section), an extensive wildlife background survey yielded 4,523 occurrence records (3,921 after optimization) for 16 nationally protected wildlife species. Using the ecological niche model, this study aimed to explore the boundary range, functional zoning, and protection gap areas within KMNP by simulating the suitable spatial distribution of wildlife species, taking into account their protection and endangered statuses. The findings indicated that the primary habitat for key protected species in KMNP mainly occupied meadow and steppe in terms of vegetation types, along with grassland and unused land in terms of land use types, with average annual precipitation from 200 mm to 400 mm, elevations ranging from 4,100 m to 4,700 m, and average annual temperatures from −4.5℃ to −2℃. The suitable distribution of different wildlife species in the Kunlun Mountains area showed variations, mainly concentrated in the southwestern, central, and southeastern regions. The optimized KMNP covered around 49,800 km2, a 16.3 % reduction from the selected area, with the core protection zone at 16,300 km2 and the general control zone at 33,200 km2. The suitability of 93.75 % (15/16) species within the core protection area of KMNP was higher comparing to the optimized area, selected area, and Mangya-Golmud-Dulan area, indicating that the boundary delimitation and functional zoning of KMNP were highly rational. The boundary range and functional zoning method developed in this study provide a crucial scientific foundation for KMNP's conservation and management, and valuable guidance for national park construction, ecosystem preservation, biodiversity conservation, and the promotion of key species' reproduction and survival on the Qinghai-Tibet Plateau, with promising future applications. • Both wildlife and livestock were considered to assess grazing stress to obtain accurate results. • Domestic livestock contributed more to grazing pressure than wildlife in Three-River Headwaters. • Edible forage carrying capacity is more beneficial to achieve grassland and livestock balance than crude protein carrying capacity. • Analysis of grazing pressure in Qinghai-Tibet Plateau should focus more on the selective feeding of animals and seasonal factors. Grassland is not only the natural habitats for the survival of wild animals, but also the material guarantees for livestock husbandry. However, the previous studies on grassland carrying capacity are mostly only considering livestock but wild herbivores were ignored. In this study, the population of large wild herbivores, Tibetan wild ass, Tibetan gazelle and Tibetan antelope, and livestock were surveyed, and the grassland forage yield was estimated by a remote sensing-process coupled model in Qumahe Township in Three-River Headwaters, Qinghai-Tibetan in 2020. The actual carrying capacity (AC) was calculated from the both wildlife and livestock and the theoretical carrying capacity (TC) in the term on edible herbage and crude protein was estimated. Grazing pressure index was measured by the ratio of AC to TC in this region. The results showed the AC was contributed by about 20,700 standard sheep units (SHU) from wild large herbivores, and 256,500 SHU from the livestock. The theoretical carrying capacity of edible herbage, which is 274,800 SHU, was lower than the theoretical carrying capacity of crude protein in the region. If only considering livestock, the grazing capacity pressure index is 0.93, but the index will be 1.01 if the both wildlife and livestock were considered, and wildlife contributes 7.5% to the grazing pressure. This study suggested the wildlife population should be calculated in grassland ecological carrying capacity estimation though domestic livestock contributed much more to grazing pressure than wild herbivores. Echinococcosis is highly endemic over large parts of the Qinghai–Tibet Plateau (QTP), China. Based on a large number of samples, we present data on the current presence, host distribution, and genetic diversity of Echinococcus in the Qinghai Province, located in the northeastern corner of the QTP and constituting >25% of the area of the plateau. We used 521 samples (including 451 newly collected samples and 70 samples from our previous study) from humans, yaks, sheep, goats, dogs, fox, plateau pikas, and voles in 36 counties, and genotyped them using the mitochondrial DNA marker cytochrome oxidase subunit I (cox1) gene and the maximum parsimony and Bayesian reconstruction methods. Based on the 792 bp sequence matrix, we recorded 177 variable sites; 157 were parsimony-informative. A total of 105 haplotypes (H1–H105) were detected, of which H1–H15 and H90–H104, H16–H17, H18–H89, and H105 belonged to Echinococcus shiquicus , Echinococcus multilocularis , Echinococcus granulosus , and Echinococcus canadensis , respectively. Our results showed that, (i) the Qinghai Province was under a high burden of Echinococcus epidemiology; (ii) E. granulosus was the main echinococcosis threat to the local people, and the followed is E. multilocularis ; (iii) there are a considerable number of haplotypes shared by domestic animals (sheep, yaks, and dogs) and humans, demonstrating the close relationship between human and domestic animals epidemiology; (iv) the threat of E. shiquicus on humans and livestock can be mostly ignored, while the infection risk of E. canadensis echinococcosis should not be neglected. • The habitat of four antelopes showed a decreasing trend under climate change. • The four species were predicted to migrate to higher latitudes in the future. • The migration paths provide reference for the establishment of corridors. Climate change is predicted to directly or indirectly affect species distribution and abundance, especially the species that live on the Qinghai-Tibet plateau that is highly sensitive to climate change. The Przewalski's gazelle (Procapra przewalskii), the Goitered gazelle (Gazella subgutturosa), the Tibetan gazelle (Procapra picticaudata) and the Tibetan antelope (Pantholops hodgsonii) are the only four existing antelopes living on the plateau. They play indispensable roles in regulating the structure and function of the plateau ecosystem. To understand how climate change affects the spatial distribution and migration direction of these ungulates, we applied the maximum entropy (MaxEnt) model and used 82, 57, 397 and 324 GPS points of Przewalski's gazelle, Goitered gazelle, Tibetan gazelle and Tibetan antelope, respectively. These points were mainly obtained through the survey of the line transect method and a small part from the database, and then we combined with the related environmental variables, and afterwards evaluated to predict the habitat change and shift of species geographic range under three climate scenarios in the 2050s and 2070s. Additionally, the potential migration paths in the future were simulated by the Minimal Cumulative Resistance (MCR) model. The results showed that climate change would cause habitat loss for all four species. The Tibetan antelope was predicted to lose over 50% of its current inhabited area under the most severe climate scenario. Also, the suitable habitat of all species would shift to higher latitudes. In particular, the Przewalski's gazelle as an endangered species that occupies narrow habitat area would face more severe challenges in the future. Therefore, all suitable habitats should be considered as important protection areas, and our results also provide a reference for designing the optimal migration corridors for the investigated species.
