This is the first regional, multi-annual assessment of floating microplastics in Scotland's seas. Sea surface samples were collected from 2014 to 2020, using a catamaran swimmer body/neuston net trawl and evaluated for the presence of microplastics. Microplastics were present in the surface waters of all Scottish Marine Regions (SMR) and Offshore Marine Regions (OMR) though almost 35% of sample sites contained no microplastics. Concentrations ranged from 0 to 91,128 microplastics km -2 sea surface. Potential hotspots were identified in the Clyde (0-77,168 microplastics km -2 ), Forth & Tay (0-83,729 microplastics km -2 ) and the Solway (607-91,128 microplastics km -2 ). Fragmented plastics accounted for almost 50% of the microplastics recovered and this may suggest that the microplastics in Scotland's seas are predominantly from the breakdown of larger items. As temperatures rise, timing of reproduction is changing at different rates across trophic levels, potentially resulting in asynchrony between consumers and their resources. The match-mismatch hypothesis (MMH) suggests that trophic asynchrony will have negative impacts on average productivity of consumers. It is also thought to lead to selection on timing of breeding, as the most asynchronous individuals will show the greatest reductions in fitness. Using a 30-year individual-level dataset of breeding phenology and success from a population of European shags on the Isle of May, Scotland, we tested a series of predictions consistent with the hypothesis that fitness impacts of trophic asynchrony are increasing. These predictions quantified changes in average annual breeding success and strength of selection on timing of breeding, over time and in relation to rising sea surface temperature (SST) and diet composition. Annual average (population) breeding success was negatively correlated with average lay date yet showed no trend over time, or in relation to increasing SST or the proportion of principal prey in the diet, as would be expected if trophic mismatch was increasing. At the individual level, we found evidence for stabilising selection and directional selection for earlier breeding, although the earliest birds were not the most productive. However, selection for earlier laying did not strengthen over time, or in relation to SST or slope of the seasonal shift in diet from principal to secondary prey. We found that the optimum lay date advanced by almost 4 weeks during the study, and that the population mean lay date tracked this shift. Our results indicate that average performance correlates with absolute timing of breeding of the population, and there is selection for earlier laying at the individual level. However, we found no fitness signatures of a change in the impact of climate-induced trophic mismatch, and evidence that shags are tracking long-term shifts in optimum timing. This suggests that if asynchrony is present in this system, breeding success is not impacted. The king scallop (Pecten maximus) is a commercially important species found around the United Kingdom coast. The association of an Apicomplexan-like parasite with mass mortality of Icelandic scallop (Chlamys islandica) in Iceland and the presence of identical parasites in king scallop (Pecten maximus) and queen scallop (Aequipecten opercularis) in Scotland raised serious concerns regarding the health of Scottish king scallops. Marine Scotland Science (MSS) conducted a survey in 2016 to assess the prevalence and the intensity of parasite infection in king scallops. King scallops were collected and sampled during the annual scallop dredge surveys in the Shetland Isles and the east and west coast of Scotland. The king scallop adductor muscle was macroscopically examined and tissue imprints taken to grade the intensity of infection. The parasite was present in the majority of the king scallops sampled in all surveyed areas: Shetland Isles 87.1%, east coast 76.0% and west coast of Scotland 64.1%. However, the parasitic infestations were light in intensity with the majority of the king scallops graded as 1 (≤20 zoites per microscopic field). No macroscopic changes in the adductor muscle were observed and histopathology examination revealed minor localized fiber degeneration of adjacent fibers to parasite clusters. The results suggested the parasite to be widespread around the Scottish coast and it appears to be able to live within the king scallop at low intensity of infection without causing significant downgrade of the adductor muscle (in terms of colour or texture) or mortality. The partial genome sequence of the parasite in king scallops from Scottish waters was identical to the one reported by Kristmundsson and Freeman (2018) in the Icelandic scallop in Icelandic waters., (Crown Copyright © 2020. Foreshore litter data from a UK citizen-science programme, combined with OSPAR data, were analysed for possible spatial patterns around Scottish and North Sea coastlines. Loading distributions were positively skewed, and statistics commensurate with such distributions were used. When considering coast type, litter loadings on Scottish harbour and river foreshores were influenced by local litter sources. When considering exposure to the predominant westerly winds over Scotland, litter loadings on the west coast (i.e. predominant onshore winds) were greater on foreshores on open coasts compared to those within embayments. The opposite was true for the Scottish east coast (i.e. predominantly offshore winds). The north east coast of the UK appeared to have an organised pattern of plastic litter loading, increasing in magnitude (median) and spread (inter quartile range) in the direction of the coastal flow. Oil spills in the marine environment can cause ecosystem-level impacts. Dispersant application as an oil spill response measure leads to the widespread distribution of hydrocarbons in the water column and marine sediments. The North Sea is an area of intense hydrocarbon production and is at risk of oil spills, which are of concern to its benthic ecosystem due to its sediments' high permeability. Here, entrainment of hydrocarbons via pore-water advection into permeable North Sea sands and the associated effect of Superdispersant-25, a commercial oil dispersant, were evaluated in a laboratory. Centrally stirred chambers that induce advective pore-water fluxes in sediments were filled with sediment, seawater and mixtures of oil and Superdispersant-25. Dispersant application had contrasting effects on hydrocarbon interactions with sediment: (1) it reduced accumulation of hydrocarbons in surface sediments and (2) facilitated the entrainment of hydrocarbons up to 8 cm deep into sediments by increasing hydrocarbon solubility in seawater and its subsequent washout or pumping into sediment by pore-water movement. Results here show that dispersant application can have counter-intuitive effects on hydrocarbon interactions with marine sediments and highlight the need for further research in this area to make better informed decision in an oil spill response scenario.

Diarrhetic shellfish toxins produced by the dinoflagellate genus Dinophysis are a major problem for the shellfish industry worldwide. Separate species of the genus have been associated with the production of different analogues of the okadaic acid group of toxins. To evaluate the spatial and temporal variability of Dinophysis species and toxins in the important shellfish-harvesting region of the Scottish west coast, we analysed data collected from 1996 to 2017 in two contrasting locations: Loch Ewe and the Clyde Sea. Seasonal studies were also undertaken, in Loch Ewe in both 2001 and 2002, and in the Clyde in 2015. Dinophysis acuminata was present throughout the growing season during every year of the study, with blooms typically occurring between May and September at both locations. The appearance of D. acuta was interannually sporadic and, when present, was most abundant in the late summer and autumn. The Clyde field study in 2015 indicated the importance of a temperature front in the formation of a D. acuta bloom. A shift in toxin profiles of common mussels ( Mytilus edulis ) tested during regulatory monitoring was evident, with a proportional decrease in okadaic acid (OA) and dinophysistoxin-1 (DTX1) and an increase in dinophysistoxin-2 (DTX2) occurring when D. acuta became dominant. Routine enumeration of Dinophysis to species level could provide early warning of potential contamination of shellfish with DTX2 and thus determine the choice of the most suitable kit for effective end-product testing.

Fjordic coastlines provide an ideal protected environment for both finfish and shellfish aquaculture operations. This study reports the results of a cruise to the Scottish Clyde Sea, and associated fjordic sea lochs, that coincided with blooms of the diarrhetic shellfish toxin producing dinoflagellate Dinophysis acuta and the diatom genus Chaetoceros, that can generate finfish mortalities. Unusually, D. acuta reached one order of magnitude higher cell abundance in the water column (2840cellsL -1 ) than the more common Dinophysis acuminata (200cellsL -1 ) and was linked with elevated shellfish toxicity (maximum 601±237μg OA eq/kg shellfish flesh) which caused shellfish harvesting closures in the region. Significant correlations between D. acuta abundance and that of Mesodinium rubrum were also observed across the cruise transect potentially supporting bloom formation of the mixotrophic D. acuta. Significant spatial variability in phytoplankton that was related to physical characteristics of the water column was observed, with a temperature-driven frontal region at the mouth of Loch Fyne being important in the development of the D. acuta, but not the Chaetoceros bloom. The front also provided important protection to the aquaculture located within the loch, with neither of the blooms encroaching within it. Analysis based on a particle-tracking model confirms the importance of the front to cell transport and shows significant inter-annual differences in advection within the region, that are important to the harmful algal bloom risk therein., (Copyright © 2017 Elsevier B.V. The diatom genus Pseudo-nitzschia (Peragallo) associated with the production of domoic acid (DA), the toxin reposnsible for amnesic shellfish poisoning, is abundant in Scottish waters. A two year study examined the relationship between Pseudo-nitzschia cells in the water column and DA concentration in blue mussels (Mytilus edulis) at two sites, and king scallops (Pecten maximus) at one site. The rate of DA uptake and depuration differed greatly between the two species with M. edulis whole tissue accumulating and depurating 7μgg -1 (now expressed as mgkg -1 ) per week. In contrast, it took 12 weeks for DA to depurate from P. maximus gonad tissue from a concentration of 68μgg -1 (now mgkg -1 ) to <20μgg -1 (now mgkg -1 ). The DA depuration rate from P. maximus whole tissue was <5% per week during both years of the study. Correlations between the Pseudo-nitzschia cell densities and toxin concentrations were weak to moderate for M. edulis and weak for P. maximus. Seasonal diversity on a species level was observed within the Pseudo-nitzschia genus at both sites with more DA toxicity associated with summer/autumn Pseudo-nitzschia blooms when P. australis was observed in phytoplankton samples. This study reveals the marked difference in DA uptake and depuration in two shellfish species of commercial importance in Scotland. Viral haemorrhagic septicaemia virus (VHSV) was isolated from five species of wrasse (Labridae) used as biological controls for parasitic sea lice predominantly, Lepeophtheirus salmonis (Krøyer, 1837), on marine Atlantic salmon, Salmo salar L., farms in Shetland. As part of the epidemiological investigation, 1400 wild marine fish were caught and screened in pools of 10 for VHSV using virus isolation. Eleven pools (8%) were confirmed VHSV positive from: grey gurnard, Eutrigla gurnardus L.; Atlantic herring, Clupea harengus L.; Norway pout, Trisopterus esmarkii (Nilsson); plaice, Pleuronectes platessa L.; sprat, Sprattus sprattus L. and whiting, Merlangius merlangus L. The isolation of VHSV from grey gurnard is the first documented report in this species. Nucleic acid sequencing of the partial nucleocapsid (N) and glycoprotein (G) genes was carried out for viral characterization. Sequence analysis confirmed that all wild isolates were genotype III the same as the wrasse and there was a close genetic similarity between the isolates from wild fish and wrasse on the farms. Movements of water that transport pathogens mean that in net-pen aquaculture diseases are often most effectively managed collaboratively among neighbours. Such area management is widely and explicitly applied for pathogen management in marine salmon farms. Effective area management requires the active support of farm managers and a simple game-theory based framework was developed to identify the conditions required under which collaboration is perceived to be in their own best interest. The model applied is based on area management as practiced for Scottish salmon farms, but its simplicity allows it to be generalised to other area-managed net-pen aquaculture systems. In this model managers choose between purchasing tested pathogen-free fish or cheaper, untested fish that might carry pathogens. Perceived pay-off depends on degree of confidence that neighbours will not buy untested fish, risking input of pathogens that spread between farms. For a given level of risk, confidence in neighbours is most important in control of moderate-impact moderate-probability diseases. Common low-impact diseases require high confidence since there is a high probability a neighbour will import, while testing for rare high-impact diseases may be cost-effective regardless of neighbours actions. In some cases testing may be beneficial at an area level, even if all individual farms are better off not testing. Higher confidence is required for areas with many farms and so focusing management on smaller, epidemiologically imperfect, areas may be more effective. The confidence required for collaboration can be enhanced by the development of formal agreements and the involvement of outside disinterested parties such as trade bodies or government., (Copyright © 2014. Scotland is the largest Atlantic salmon (Salmo salar) producer in the EU with an output of over 150,000 t, contributing over £500 million annually towards the economy. Production continues to increase, predominantly due to the increase in output per farm and reduction in losses due to infectious diseases. Farms are grouped within disease management areas whose boundaries are defined by where the closest pair of farms is separated by more than twice the tidal excursion distance (TE) Tidal excursion is defined as 7.2 km in mainland Scotland, or 3.6 km in the Shetland Islands). The majority of salmon farms are located within relatively sheltered inshore areas where non-tidal advective current speed is minimal. However there is an aspiration for offshore production where it might be possible to increase stocking levels and where current speeds will be greater so TE models could break down. Separation distances whereby farms would avoid infection risk were obtained using an analytical, discrete-time Susceptible-Exposed-Infectious-Recovered (SEIR) model coupled with a hydrodynamic transport expression representing transmission of pathogenic agents between fish farms. The model incorporated transmission, expression and recovery parameters as well as pathogen shedding and decay. The simplified hydrodynamic model incorporated residual advection, tidal advection and turbulent diffusion elements. The obtained separation distances were compared to a computationally intensive, numerical model and were demonstrated to be comparable, although the analytical model underestimated the variation within the transmission distances. Applying characteristics for a robust pathogen, infectious pancreatic necrosis virus type (IPNV-type), and less robust pathogens such as infectious salmon anaemia virus type (ISAV-type) and Aeromonas salmonicida type (AS-type) pathogens, it was possible to obtain separation distances whereby farms avoided infection. Simulation outputs indicated that separation distances should increase to avoid disease as farm size and current speed increase. The more conserved IPNV-type pathogen required separation distances of hundreds of kilometres, AS-type required tens of kilometres, whilst the distances for ISAV-type were within the scale of the current DMAs, that were developed for ISAV control. Salmon aquaculture in Scotland continues to increase; however, one of the potential limitations to its further sustainable growth is the ectoparasitic sea louse Lepeophtheirus salmonis. The industry in Scotland undertakes coordinated management procedures to control the levels of sea lice on farms in designated production areas. We developed a biophysical sea lice dispersal model for Loch Linnhe, one of the largest fjords on the west coast of Scotland, to provide further information to help establish more effective farm management areas. We successfully extend modelling principles previously applied to a small Scottish fjordic system. Modelling scenarios demonstrate heterogeneity in the distribution of sea lice within the system and simulations, suggesting that lice could be transmitted up to 30 km. The scenarios are assessed by comparing model predictions against lice sampled by both planktonic trawls and settlement on sentinel caged fish. The model predicts the ranked abundance of both planktonic and settled lice assuming that the lice input to the system is relative to host biomass. This study evaluates the potential of silicone rubber passive sampling devices (SR-PSDs) as a suitable alternative to automatic water samplers (autosamplers) for the preliminary identification of a wide range of organic contaminants in freshwater systems. The field performance of SR-PSDs deployed at three sites on two streams of an agricultural catchment area in North East (NE) Scotland, United Kingdom (UK) was assessed concurrently with composite water samples collected from two of the sites using autosamplers. The analytical suite consisted of selected plant protection products (PPPs; commonly referred to collectively as 'pesticides'), including 47 pesticides and a separate sub-category of 22 acid/urea herbicides. Of these, a total of 54 substances, comprising 46 pesticides and 8 urea herbicides were detected in at least one of the SR samplers. All but 6 of these SR-PSD detected substances were quantifiable. By comparison, a total of 25 substances comprising 3 pesticides and 22 acid/urea herbicides were detected in the composite water samples, of which only 8 acid/urea herbicides were quantifiable. The larger number and chemical classes of compounds detected and quantified via passive sampling reflect the lower limits of detection achieved by this device when compared to autosamplers. The determination of dissolved concentrations of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) added to the information on contaminant pressures at each site, allowing assessment of the reliability of SR-PSDs in freshwater systems and the identification of possible contaminant sources. The study demonstrated the utility of SR-PSDs for detecting and semi-quantifying low concentrations of analytes, including those which hitherto have not been measured in the catchment area and also some pesticides that are no longer approved for agricultural use in the UK and EU. To investigate the influence of towed demersal fishing gears on dinoflagellate cyst resuspension, towing trials with four gear components were carried out at three sites of differing sediment type in the Moray Firth, Scotland. Samples of sediment plumes were collected using plankton nets mounted on a towed sledge. Diversity of resuspended dinoflagellate cysts was similar at all sites and included Protoperidinium and Gonyaulax spp., Proroceratium reticulatum and unidentified 'round brown' cysts. Cyst concentrations per gram of resuspended sediment varied by gear component and sediment particle size distribution. Gear components with lower hydrodynamic drag generated wakes with smaller shear stresses, mobilising fewer larger sand particles, giving larger concentrations of cysts. Muddy sediments contained higher cyst concentrations which declined with increasing grain size. This study has shown that fishing gear and sediment type can influence the redistribution of dinoflagellate cysts and highlights the importance this may have in relation to dinoflagellate blooms., (Crown Copyright © 2012. Disease is a major constraint on animal production and welfare in agriculture and aquaculture. Movement of animals between farms is one of the most significant routes of disease transmission and is particularly hard to control for pathogens with subclinical infection. Renibacterium salmoninarum causes bacterial kidney disease (BKD) in salmonid fish, but infection is often sub-clinical and may go undetected with major potential implications for disease control programmes. A Susceptible-Infected model of R. salmoninarum in Scottish aquaculture has been developed that subdivides the infected phase between known and undetected sub-clinically infected farms and diseased farms whose status is assumed to be known. Farms officially known to be infected are subject to movement controls restricting spread of infection. Model results are sensitive to prevalence of undetected infection, which is unknown. However, the modelling suggests that controls that reduce BKD prevalence include improve biosecurity on farms, including those not known to be infected, and improved detection of infection. Culling appears of little value for BKD control. BKD prevalence for rainbow trout farms is less sensitive to controls than it is for Atlantic salmon farms and so different management strategies may be required for the sectors., (Crown Copyright © 2011.
