The RELIANCE study has demonstrated the activity and safety of relmacabtagene autoleucel (relma-cel) (JW Therapeutics [Shanghai] Co, Ltd, Shanghai, China), a CD19-targeted chimeric antigen receptor T-cell product, in patients with heavily pre-treated relapsed/refractory large B-cell lymphoma (r/r LBCL). This study aimed to report the updated 2-year data of the RELIANCE study. The RELIANCE study (NCT04089215) was an open-label, multi-center, randomized, phase 1/2 registrational clinical trial conducted at 10 clinical sites in China. Adult patients with heavily pre-treated r/r LBCL were enrolled and received lymphodepletion chemotherapy followed by infusion of 100 × 106 or 150 × 106 relma-cel. The primary endpoint was objective response rate (ORR) at 3 months, as assessed by investigators. Secondary endpoints were duration of response (DoR), progression-free survival (PFS), overall survival (OS) and safety profiles. From November 2017 to January 2022, a total of 68 patients were enrolled, and 59 patients received relma-cel infusion. As of March 29, 2022, a total of 59 patients had a median follow-up of 17.9 months (range, 0.3–25.6). ORR was 77.59% (95% confidence interval [CI], 64.73–87.49) and complete response rate was 53.45% (95% CI, 39.87–66.66). Median DoR was 20.3 months (95% CI, 4.86–not reached [NR]) and median PFS was 7.0 months (95% CI, 4.76–24.15). Median OS was NR and 1-year and 2-year OS rates were 75.0% and 69.3%, respectively. Three (5.1%) patients experienced grade ≥3 cytokine release syndrome and two (3.4%) patients had grade ≥3 neurotoxicity. The characteristics of distribution of Pseudomonas aeruginosa in patients with agranulocytosis and fever in 12 hospitals in Shanghai from 2012 to 2014 were retrospectively analyzed. WHONET 5.6 software was used to analyze the results of drug sensitivity test. Data from different diseases, different sample sources, and drug sensitivity tests were statistically analyzed to investigate the clinical distribution characteristics and drug resistance of Pseudomonas aeruginosa in patients with agranulocytosis and fever in Shanghai, China. This study revealed that, among these 109 strains of P. aeruginosa, they were mainly found in patients with acute myelocytic leukemia (AML; 48 strains, 44.04%) and patients with acute lymphocytic leukemia (ALL; 36 strains, 33.03%). The specimen sources were mainly respiratory tract secretions (58 strains, 53.21%) and blood (21 strains, 19.26%). The P. aeruginosa isolates from neutropenic sepsis patients showed high sensitivity to the following antibiotics: piperacillin/tazobactam, cefepime, ciprofloxacin, ceftazidime, and amikacin with 91.1%, 89%, 89%, 87.9%, and 85.7% of isolates being sensitive, respectively. Furthermore, for P. aeruginosa isolates from the AML group of patients, the lowest antibiotic resistance rates were seen for ciprofloxacin (0%), cefoperazone/sulbactam (2.1%), and cefepime (7.1%), while for the ALL group the lowest antibiotic resistance rates were seen for piperacillin (2.8%), ceftazidime (2.8%), and cefepime (2.8%). Isolates from AML patients (21.3%) were significantly more likely to be piperacillin resistant than those from the ALL patients (2.8%). Therefore, P. aeruginosa infection is relatively common in patients with agranulocytosis and fever. The strains had a certain degree of resistance to commonly used antibiotics. Dianshan Lake (DSL) as an important water source protection area in Shanghai is facing serious water quality deterioration. A detailed investigation of persistent organic pollutants (POPs) including polycyclic aromatic hydrocarbons (PAHs), polychlorinated biphenyls (PCBs), and organochlorine pesticides (OCPs) in the multiphase system of DSL is very important for understanding their occurrence, sources, partition, and health risks. Total 11 sampling sites were selected including 3 inflowing rivers, 6 lake body sites, and 2 outflowing rivers. Total OCPs (Σ 20 OCPs) showed the highest concentration in water dissolved phase with a range of 47.8–143 ng/L. Total PAHs (Σ 16 PAHs) were the dominant compounds for suspended particle matter (SPM) and sediment phase, with a range of 2805–4130 and 206–4262 ng/g, respectively. The nonparametric test revealed the significantly higher concentrations of Σ 16 PAHs in sediment, Σ 18 PCBs in SPM, and Σ 20 OCPs in dissolved water from inflowing rivers than lake body (p < 0.05), indicating the strong effect of riverine pollution input. The different seasonal distribution between Σ 16 PAHs with Σ 18 PCBs/Σ 20 OCPs in SPM phase of lake body was closely attributed to their source disparities and the changes of rainfall and hydrodynamic conditions. Homologue compositions and isomer ratios analysis showed in DSL watershed PAHs were mainly derived from pyrogenic sources, PCBs from the leakage of capacitor equipment, sewage drainage of industries, and exfoliation of ship paints, and DDT from historical and recent application. The bioaccumulations of POPs in six fish species (n = 20) of DSL had high detection frequencies and a descending concentration order: Σ 16 PAHs (353 ± 106 ng/g) > Σ 20 OCPs (67.4 ± 27.0 ng/g) > Σ 18 PCBs (32.0 ± 13.7 ng/g), with obvious species-specific characteristics. The partitioning behaviors of PAH congeners in the water-SPM-sediment-fish system of DSL were greatly influenced by log K ow values. However, the feeding habits (e.g., sediment-feeding behavior) of fish species played a predominant role in the bioaccumulation of PCB and OCP congeners compared to two-phase partition. Health risk assessments of POPs in dissolved water and fish samples revealed that only Σ 16 PAHs in dissolved phase have potential carcinogenic risk.
