Anopheles culicifacies is the major vector of malaria in Sri Lanka and the Indian subcontinent which is characterized as a species complex with five sibling species provisionally designated as A, B, C, D and E. The current study was carried out to understand the phylogenetic and phylogeographic relationships between the sibling species of the species complex while observing their genetic diversity and genetic differentiation. Thirty-five ITS2 and seventy-seven COI sequences of An. culicifacies species complex reported from different geographical locations of Asia and China at the NCBI public database were used for the analysis. Bayesian likelihood trees were generated for the phylogenetic analysis. The divergence of the species complex was obtained from the Bayesian phylogeographic model in BEAST. There were two clades of the sibling species of An. culicifacies species complex as A, D and B, C and E in both phylogenetic and phylogeographic analysis using ITS2 sequences. Based on the highly divergent COI sequences and the high mutation rate of the mitochondrial genome, there were four and three clades in both phylogenetic and phylogeographic analysis using COI sequences. The diversification of An. culicifacies species complex was obtained as ranging from 20.25 to 24.12 Mya and 22.37 to 26.22 Mya based on ITS2 and COI phylogeographic analysis respectively. There was a recent diversification of the sibling species A and D than the sibling species B, C and E. Low haplotype diversity was observed in the sequences reported from Sri Lanka in both ITS2 and COI analysis that can be due to bottlenecks resulting from the intense malaria control efforts. A high genetic differentiation was achieved for some populations due to the large geographical distance. The high genetic diversity based on the five sibling species implies the possibility of maintaining a relatively high effective population size despite the vector control efforts. Climate change is a serious and complex crisis that impacts humankind in different ways. It affects the availability of water resources, especially in the tropical regions of South Asia to a greater extent. However, the impact of climate change on water resources in Sri Lanka has been the least explored. Noteworthy, this is the first study in Sri Lanka that attempts to evaluate the impact of climate change in streamflow in a watershed located in the southern coastal belt of the island. The objective of this paper is to evaluate the climate change impact on streamflow of the Upper Nilwala River Basin (UNRB), Sri Lanka. In this study, the bias-corrected rainfall data from three Regional Climate Models (RCMs) under two Representative Concentration Pathways (RCPs): RCP4.5 and RCP8.5 were fed into the Hydrologic Engineering Center-Hydrologic Modeling System (HEC-HMS) model to obtain future streamflow. Bias correction of future rainfall data in the Nilwala River Basin (NRB) was conducted using the Linear Scaling Method (LSM). Future precipitation was projected under three timelines: 2020s (2021–2047), 2050s (2048–2073), and 2080s (2074–2099) and was compared against the baseline period from 1980 to 2020. The ensemble mean annual precipitation in the NRB is expected to rise by 3.63%, 16.49%, and 12.82% under the RCP 4.5 emission scenario during the 2020s, 2050s, and 2080s, and 4.26%, 8.94%, and 18.04% under RCP 8.5 emission scenario during 2020s, 2050s and 2080s, respectively. The future annual streamflow of the UNRB is projected to increase by 59.30% and 65.79% under the ensemble RCP4.5 and RCP8.5 climate scenarios, respectively, when compared to the baseline scenario. In addition, the seasonal flows are also expected to increase for both RCPs for all seasons with an exception during the southwest monsoon season in the 2015–2042 period under the RCP4.5 emission scenario. In general, the results of the present study demonstrate that climate and streamflow of the NRB are expected to experience changes when compared to current climatic conditions. The results of the present study will be of major importance for river basin planners and government agencies to develop sustainable water management strategies and adaptation options to offset the negative impacts of future changes in climate. Vibrio cholerae is the etiological agent of cholera, a severe diarrheal disease, which can occur as either an epidemic or sporadic disease. Cholera pandemic-causing V. cholerae O1 and O139 serogroups originated from the Indian subcontinent and spread globally and millions of lives are lost each year, mainly in developing and underdeveloped countries due to this disease. V. cholerae O1 is further classified as classical and El Tor biotype which can produce biotype specific cholera toxin (CT). Since 1961, the current seventh pandemic El Tor strains replaced the sixth pandemic strains resulting in the classical biotype strain that produces classical CT. The ongoing evolution of Atypical El Tor V. cholerae srains encoding classical CT is of global concern. The severity in the pathophysiology of these Atypical El Tor strains is significantly higher than El Tor or classical strains. Pathogenesis of V. cholerae is a complex process that involves coordinated expression of different sets of virulence-associated genes to cause disease. We are yet to understand the complete virulence profile of V. cholerae, including direct and indirect expression of genes involved in its survival and stress adaptation in the host. In recent years, whole genome sequencing has paved the way for better understanding of the evolution and strain distribution, outbreak identification and pathogen surveillance for the implementation of direct infection control measures in the clinic against many infectious pathogens including V. cholerae. This review provides a synopsis of recent studies that have contributed to the understanding of the evolution, distribution and genetics of the seventh pandemic Atypical El Tor V. cholerae strains. • Cholera is endemic in more than 47 countries around the world. • Climate change is influencing shifts in genetic diversity of V. cholerae. • Global increases of non-toxigenic V. cholerae disease occurrence is of public health concern.
