In response to the Howard Hughes Medical Institute/Association of American Medical Colleges Scientific Foundations for Future Physicians (SFFP) report and a concern for better preparing undergraduates for future doctoral programs in the health professions, the deans of the College of Arts and Sciences and Division of Basic Biomedical Sciences of Sanford School of Medicine of the University of South Dakota formed an ad hoc Premedical Curriculum Review Committee with representatives from the science departments and medical school. The Committee began by reviewing the university's suggested premedical curriculum and matching it to the proposed competencies from the SFFP to document duplications and deficiencies. The proposed changes in the Medical College Admission Test for 2015 were also evaluated. The Committee proposed a stronger premedical curriculum, with the development of some new courses, including an inquiry-based physiology course with team-based learning, to more fully address SFFP competencies. These analyses convinced the university that a new major would best help students achieve the competencies and prepare them for admission exams. Thus, a new Medical Biology major was proposed to the South Dakota Board of Regents and accepted for its initial offering in 2012. The new major has been broadly advertised to future students and is successful as a recruiting tool for the university. This article details the process of evaluating the curriculum and designing the new major, describes some of the difficulties in its implementation, and reviews outcomes from the new major to date., (Copyright © 2015 The American Physiological Society.)

Migrant birds require large flight muscles and hearts to enhance aerobic capacity and support sustained flight. A potential mechanism for increasing muscle and heart masses during migration in birds is the muscle growth inhibitor myostatin and its metalloproteinase activators, tolloid-like proteinases (TLL-1 and TLL-2). We hypothesized that myostatin, TLL-1 and TLL-2 are downregulated during migration in pectoralis and hearts of migratory passerines to promote hypertrophy. We measured seasonal variation of tissue masses, mRNA expression of myostatin, TLL-1, and TLL-2, and myostatin protein levels in pectoralis muscle and heart for yellow warblers (Setophaga petechia), warbling vireos (Vireo gilvus), and yellow-rumped warblers (Setophaga coronata). Pectoralis mass was greatest in spring for warbling vireos and yellow warblers, but was stable between spring and fall for yellow-rumped warblers. Heart mass was higher in spring than in fall for yellow-rumped warblers, lowest in fall for warbling vireos, and seasonally stable for yellow warblers. Pectoralis and heart mRNA expression of myostatin and the TLLs did not differ significantly for any of the three species, offering little support for our hypothesis for a prominent role for myostatin in regulating migration-induced variation in pectoralis and heart masses. In contrast, pectoralis myostatin protein levels were lowest in spring for all three species, consistent with our hypothesis. Myostatin protein levels in heart, however, were seasonally stable for warbling vireos and yellow warblers, and increased in spring relative to fall for yellow-rumped warblers. These data offer mixed support for our hypothesis for the pectoralis, but suggest that myostatin is not a prominent regulator of migration-induced heart hypertrophy. Moreover, the different seasonal patterns for pectoralis mRNA and protein expression suggest that post-transcriptional modification of myostatin may contribute to pectoralis mass regulation during migration.

Winter acclimatization in small birds living in cold climates produces a winter phenotype characterized by upregulation of metabolic rates to meet enhanced thermoregulatory demands. We measured several key aspects of fuel storage, mobilization and transport in summer and winter to determine whether black-capped chickadees (Poecile atricapillus), white-breasted nuthatches (Sitta carolinensis), and house sparrows (Passer domesticus) seasonally modulate these attributes to meet enhanced winter thermoregulatory demands. In addition, we exposed birds to thermoneutral (control) and severe cold exposure treatments to determine whether acute cold exposure influenced fuel storage, mobilization or transport. Carcass lipid mass and pectoralis intramuscular lipid did not vary significantly between seasons or temperature treatments for any of the study species. Muscle glycogen varied significantly seasonally only for chickadee supracoracoideus and leg muscles, and did not vary among warm or cold treatments for any species. Pectoralis fatty acid binding protein (FABPc) was significantly elevated in winter for chickadees and nuthatches, but not for sparrows. Plasma metabolites showed little consistent variation in response to season or acute cold exposure. Thus, fuel storage and mobilization do not appear to be major targets of adjustment associated with seasonal metabolic flexibility in these species, but modulation of intracellular lipid transport by FABPc may be an important contributor to seasonal phenotypes in some species of small birds., (Copyright © 2014 Elsevier Inc. The ability to find sufficient high-quality stopover habitat is a crucial factor for successful migration for woodland migrant birds. Woodland habitats are scarce in the Northern Prairie region of North America, and natural woodlands have been greatly reduced concurrent with the appearance of small anthropogenic woodlands on the landscape. Landbird migrants use both natural and anthropogenic woodlands in this region as stopover habitats, but the relative quality of these two habitats is unknown. We assessed the relative habitat quality of the two habitats by comparing body mass (Mb) and plasma metabolites associated with fattening (triglycerides [TRIG]) or fat catabolism (β-hydroxybutyrate [BUTY], glycerol [GLYC]) in individual species, taxa, and foraging guilds of migrating woodland birds during both spring and fall migrations. The only significant difference in Mb between birds in the two habitats occurred for fall yellow-rumped warblers (Setophaga coronata), where Mb was 8% greater in corridors than in woodlots. No significant between-habitat differences occurred for plasma TRIG at either season. Significant between-habitat differences for plasma BUTY occurred only for ruby-crowned kinglets (Regulus calendula; 61% higher in corridors) in fall. Plasma GLYC differed significantly between habitats for a few groups, including vireos (190% higher in woodlots), warbling vireos (Vireo gilvus; 263% higher in woodlots), and Nashville warblers (Oreothlypis ruficapilla; 226% higher in woodlots) in fall. The few significant differences and absence of a consistent direction of variation in Mb and plasma metabolites suggests similar stopover habitat quality in these two habitat types. Thus, during migration through the Northern Prairie region anthropogenic woodlots can, at least partially, substitute as stopover habitat for lost and degraded native riparian habitats for woodland birds.

Abstract Cold tolerance and overwinter survival are positively correlated with organismal thermogenic capacity (=summit metabolic rate [Msum]) in endotherms. Msum varies seasonally in small-bird populations and may be mechanistically associated with variation in flight muscle size or cellular metabolic capacity, but the relative roles of these traits as drivers of individual variation in thermogenic performance are poorly known. We measured flight muscle size by ultrasonography, pectoralis and supracoracoideus muscle masses, and muscular activities of key aerobic enzymes (citrate synthase, carnitine palmitoyl transferase, and β-hydroxyacyl-CoA dehydrogenase) and correlated these measurements with Msum for individual American goldfinches (Spinus tristis) to test the hypotheses that muscle size and/or cellular metabolic capacity serve as prominent drivers of individual variation in organismal metabolic capacity. Ultrasonographic flight muscle size was weakly positively correlated with Msum ([Formula: see text]). Both log10-transformed Msum and flight muscle mass were significantly correlated with log10 body mass, so we calculated allometric residuals for log Msum and for log flight muscle mass to test their correlation independent of body mass. Flight muscle mass residuals were significantly positively correlated with Msum residuals, and this correlation was primarily driven by variation in pectoralis muscle mass. In contrast, none of the mass-specific activities of any enzyme in any muscle were significantly correlated with Msum. These data suggest that flight muscle size, not cellular metabolic capacity, is the primary driver of individual variation in thermogenic performance in goldfinches. This is consistent with the idea that phenotypic flexibility of flight muscle mass is a general mechanism mediating variation in metabolic performance in response to changing energy demands in birds.

Winter acclimatization in small birds overwintering in cold climates, including house sparrows (Passer domesticus), is associated with improved cold tolerance, elevated summit metabolic rates (M(sum) = maximum cold-induced metabolic rate), and increased pectoralis muscle mass compared to summer birds. Myostatin is a potent autocrine/paracrine inhibitor of skeletal muscle growth in mammals and birds and is a potential candidate for regulation of seasonal phenotypic flexibility in birds. As a first step toward examining such a role for myostatin in small birds, we measured summer and winter gene expression of myostatin and its potential metalloproteinase activators TLL-1 and TLL-2 in house sparrows from southeastern South Dakota. Gene expression of myostatin decreased significantly in winter, with summer values exceeding winter values by 1.52-fold. Moreover, gene expression of TLL-1 was also significantly reduced in winter, with summer values exceeding winter values by 1.55-fold. These data are consistent with the hypothesis that the winter increases in pectoralis muscle mass, M(sum), and cold tolerance in house sparrows are mediated by reduced levels of myostatin and its activator TLL-1, and they suggest the possibility that myostatin may be a common mediator of phenotypic flexibility of muscle mass in birds.

It has been hypothesized that freeze-tolerance in anurans evolved from a predisposition for dehydration tolerance. To test this hypothesis, we dehydrated summer/fall-collected and winter acclimated freeze-tolerant chorus frogs and dehydration-tolerant, but freeze-intolerant, Woodhouse's and Great Plains toads to 25% and 50% body water loss (BWL). Following treatments, we measured glucose, glycogen, and glycogen phosphorylase and glycogen synthetase (summer/fall only) activities in liver and leg muscle. Hepatic glucose levels were not significantly altered by dehydration in either summer/fall-collected frogs or toads. Conversely, winter acclimated frogs did show an increment (2.9-fold) in hepatic glucose with dehydration, accompanied by a reduction in hepatic glycogen levels. Winter acclimated toads did not mobilize hepatic glucose in response to dehydration. Further, hepatic glycogen and phosphorylase activities did not vary in any consistent manner with dehydration in winter toads. Mean leg muscle glucose values were elevated at 50% BWL relative to other treatments, significantly so compared to 25% BWL for summer/fall-collected frogs. The pattern of hepatic glucose mobilization with dehydration in winter frogs is consistent with that in other freeze-tolerant frog species, and provides additional support for the hypothesis that freezing tolerance evolved from a capacity for dehydration tolerance. However, the lack of hepatic glucose mobilization in response to dehydration in fall frogs suggests that a seasonal component to dehydration-induced regulation of glucose metabolism exists in chorus frogs. Furthermore, the absence of a dehydration-induced mobilization of hepatic glucose at both seasons in toads suggests that this dehydration response is not universal for terrestrial anurans., (Copyright 2004 Wiley-Liss, Inc.)

The use of heterothermy is well documented in the order Caprimulgiformes, but there is conflicting information regarding whether whip-poor-wills are heterothermic. Consequently, we sought to rigorously examine the thermoregulatory abilities of this species. Our study was conducted in southeast South Dakota (42 degrees 47'N, 97 degrees 0'W), where 35 individuals were captured and outfitted with external, temperature-sensitive radio transmitters. We found evidence that whip-poor-wills used daily torpor during the autumn of 2000 and the spring of 2001 (n=12 torpor bouts, based on 346 bird-nights of observation). The average minimum skin temperature of two torpid whip-poor-wills (n=5 torpor bouts) in spring 2001 was 20.1 degrees +/-2.6 degrees C, and bouts of reduced skin temperature lasted an average of 360.0+/-93.7 min. The distribution of heterothermy within the Caprimulgiform phylogeny suggests that the trait is ancestral in the order. Specific heterothermic parameters, however, differ among the different species. In particular, the frequency of torpor use in whip-poor-wills is lower than for other species. These data suggest that several factors, including weather conditions and gender-specific reproductive ecology, influence the propensity of whip-poor-wills and other Caprimulgiformes to enter torpor.

Mechanistic bases for freezing tolerance in anurans have been well-studied only in wood frogs, Rana sylvatica, so comprehensive explanations for the mechanisms and evolution of freezing tolerance in anurans are lacking. We measured crystallization temperatures, freezing tolerance/intolerance, and tissue glucose and glycogen phosphorylase activities in frozen and unfrozen winter-acclimated Pseudacris triseriata, Bufo cognatus and B. woodhousei. Freezing occurred at higher subzero temperatures on wet substrate than on dry substrate in all species, indicating susceptibility to inoculative freezing. P. triseriata was freeze-tolerant, but survival was dependent on the level of supercooling prior to freezing. All Bufo were freezing intolerant, regardless of crystallization temperature. Glucose was significantly elevated by freezing in both liver (35-fold) and leg muscle (22-fold) in winter P. triseriata, but only liver glucose was significantly elevated in B. cognatus. However, freezing did not alter glycogen phosphorylase activity in either species. Liver phosphorylase activity was significantly higher in P. triseriata than in B. cognatus, suggesting that capacity for mobilizing glucose from liver glycogen is associated with freezing tolerance. Summer measurements of liver phosphorylase activity, however, did not differ between species. Thus, P. triseriata, but not B. cognatus, exhibited winter increment of liver phosphorylase activity that is correlated with the development of freezing tolerance.
