Methyl benzimidazole carbamate (MBC) fungicides were once widely used for brown rot (Monilinia fructicola) control of peach (Prunus persica (L.) Batsch) in the southeastern US, but their use was substantially reduced and often eliminated due to widespread resistance. In this study, 233 M. fructicola isolates were collected from major peach production areas in Alabama, Georgia, and South Carolina, and sensitivity to thiophanate-methyl was examined. Isolates were also collected from one organic and two experimental peach orchards. A discriminatory dose of 1 μg/ml was used to distinguish sensitive (S) and moderately sensitive (S-LR) isolates from low resistant phenotypes, while 50 and 500 μg/ml thiophanate-methyl concentrations were used to determine high resistant (HR) phenotypes. Sequence analyses were performed to identify mutations in the β-tubulin target gene and detached fruit assays were performed to determine the efficacy of a commercial product against isolates representing each phenotype. Results indicated 55.7%, 63.5%, and 75.9% of isolates from Alabama, Georgia, and South Carolina, respectively, were S to thiophanate-methyl; 44.3%, 36.5%, and 21.4% were S-LR; no isolates were LR; and only 3 isolates (1.3%) from South Carolina were HR. No mutations in S or S-LR isolates were found, but HR isolates revealed the E198A mutation, an amino acid change of glutamic acid to alanine conferring high resistance. The high label rate of a commercial product containing thiophanate-methyl controlled brown rot caused by S and S-LR isolates in detached fruit studies but was ineffective against HR isolates. The combinations of thiophanate-methyl with azoxystrobin or isofetamid, when mixed together and applied in an experimental orchard 14 days preharvest, significantly reduced brown rot incidence on pre and postharvest commercially ripe fruit and efficacy was comparable to that of a grower standard fungicide. These results indicate that thiophanate-methyl may again be useful to peach growers in the southeastern US for brown rot and fungicide resistance management., (Copyright © 2023 Elsevier Inc. In the southeastern United States, Armillaria root rot (ARR) is caused by Desarmillaria tabescens and has become the primary cause of premature mortality in peach orchards. Most rootstocks used in commercial orchards are susceptible and management options are limited. A postinfection practice known as root-collar excavation (RCE), which involves permanent removal of the soil from the base of the trunk, has been shown to improve yields and prolong the productive life of symptomatic trees. However, symptomatic trees already have an advanced infection at the base of the trunk. This study evaluated the efficacy of preventative RCE on the progression of tree mortality in two orchards that were planted in infested replant sites. To provide convincing data for growers, the study was carried out in a commercial orchard and an experimental orchard for 8 years. Furthermore, representative enterprise budgets and net present value (NPV) analysis were utilized to compare the profitability of the two approaches. Trees were planted shallow on berms (45 by 90 cm) to facilitate RCE with hoes and AirSpade 2 years later. Tree mortality in the RCE treatment of the experimental orchard was first observed in year 6 and increased 8% on average per year thereafter. In contrast, tree mortality in the "Grower Standard" treatment was first observed in year 4 and increased 12.7% on average per year thereafter. At the commercial orchard, tree mortality in the RCE treatment was first observed in year 7 and increased 1.9% on average thereafter, while tree mortality in the Grower Standard treatment first appeared in year 5 and increased 4.3% on average thereafter. The delayed onset of ARR-associated tree mortality and the lower annual tree mortality rate in the RCE treatment led to higher NPVs in both locations. There were no negative effects on yield or fruit quality. However, the new planting system can create horticultural challenges, including the formation of a proper berm, uneven ground around the tree interfering with tree care and harvest, increased erosion due to channeling of rainwater, and increased rootstock suckering. The RCE is a valid option for southeastern growers needing to manage high ARR disease pressure on replant sites or on sites only recently cleared from ARR-infected forest land.

Strawberry production in Florida and South Carolina is affected by two major diseases, anthracnose fruit rot (AFR) and Botrytis fruit rot (BFR), caused by Colletotrichum acutatum and Botrytis cinerea, respectively. The effective management of both diseases traditionally relied on weekly fungicide applications. However, to improve timing and reduce the number of fungicide sprays, many growers follow the Strawberry Advisory System (StAS), a decision support system for forecasting fungicide applications based on environmental conditions and previously developed models. The objective of this study was to perform a meta-analysis to determine the effectiveness of the StAS for AFR and BFR management compared with a calendar-based spray program. Thirty-nine trials were conducted from 2009 to 2014 in Florida and South Carolina commercial strawberry fields. Meta-analysis was conducted to quantify the treatment effects on four effect sizes, all based on the difference in response variables for StAS and the calendar-based treatments in each trial. The mean difference in BFR incidence, AFR incidence, yield, and number of marketable fruit between the two treatments was not significantly different from 0 (P < 0.05). However, the number of fungicide applications per season was reduced by a median of seven when using the StAS, a 50% reduction in sprays compared with the calendar-based approach. Effect sizes were not influenced by location or the favorability of the environment for disease development. These findings indicate that use of StAS in commercial fields is effective in controlling fruit rot diseases with no reduction in yield while substantially reducing fungicide applications.

In the southeastern USA, Armillaria root rot disease on peach (Prunus persica) is caused by Armillaria tabescens, and to a lesser degree by A. mellea. Recent attempts to genetically characterize A. tabescens isolates using rDNA indicated the existence of heterozygosity in diploid isolates. In order to clarify this heterozygosity, DNA from stipe and single spore cultures of A. tabescens isolate SC.MF-1.01 was characterized using RFLP and sequence analysis. Direct sequencing of rDNA amplicons from diploid stipe tissue indicated heterozygosity in the IGS1 and ITS2 regions. IGS1 copies A and B, and ITS copies A and B segregated in a 1:1 ratio in single spore progeny, whereby IGS1 copy A always segregated with ITS copy A and IGS1 copy B always segregated with ITS copy B. The results indicate the existence of divergent haplotypes in the two nuclei of SC.MF-1.01 diploid mycelium and a single locus for rDNA tandem repeats. Additional IGS1 copies, designated IGS1 copy C, D, and E, with variable AluI restriction sites were cloned from SC.MF-1.01 diploid mycelium indicating polymorphism within ribosomal tandem repeats in A. tabescens. IGS1 copies C, D, and E were found once each in 100 clones analyzed, and therefore seemed to be rare compared to copies A (44 clones) and B (53 clones). RFLP and sequence analysis of the ribosomal IGS1 and ITS1 regions in North American A. tabescens isolates indicated that heterozygosity is common in A. tabescens diploid mycelium. A PCR-based molecular technique was developed to distinguish A. tabescens from many other North American Armillaria species, including A. mellea.
