Abstract: Wild felids are considered important in maintaining the sylvatic cycle of Toxoplasma gondii. Although, T. gondii antibodies have been reported in several species of wild felids, little is known of the epidemiology and risk factors associated with T. gondii infection in wild cats. The Iberian lynx (Lynx pardinus) is the most endangered felid species in the world. In the present study, seroprevalence and associated risk factors for T. gondii infection in a large population of Iberian lynx in Spain were determined. Serum samples from 129 Iberian lynx collected from 2005 to 2009 and 85 wild rabbits (Oryctolagus cuniculus), sharing the habitat with the Iberian lynx, were tested for antibodies to T. gondii by the modified agglutination test (MAT) using a cut-off value of 1:25. Antibodies to T. gondii were found in 81 of 129 (62.8%) Iberian lynx. Seroprevalence to T. gondii in Iberian lynx significantly increased with age (P <0.001). T. gondii seroprevalences were similar in free-ranging (66.7% of 93) and wild-caught captive lynx (69% of 84) but significantly lower in captive-born lynx (22.5% of 40). Seroprevalence was higher in lynx with concurrent Cytauxzoon felis (88% of 25) but not with concurrent Feline Leukemia Virus (FeLV) infection (53.8% of 13). There were no significant differences in seroprevalence between sexes, geographic region and year of sample collection (2005–2009). Oocysts of T. gondii were not detected microscopically in fecal samples from 58 lynx. Wild rabbits are considered the most important food for the lynx. Antibodies to T. gondii were found in 14 (11.9%) of 85 rabbits tested. The present results indicate that T. gondii infection is widespread in the two areas where Iberian lynx survive in Spain. The fact that four captive-born lynx seroconverted was indication of contact with T. gondii also in the Captive Breeding Centers, hence, control measures to prevent T. gondii infection would be necessary in these centers. Abstract: Serum samples from 251 wild carnivores from different regions of Spain were tested for antibodies to Neospora caninum by the commercial competitive screening enzyme linked immunosorbent assay (c-ELISA) and confirmed by Neospora agglutination test (NAT) and/or by indirect fluorescent antibody test (IFAT). Samples with antibodies detected by at least two serological tests were considered seropositive. Antibodies to N. caninum were found in 3.2% of 95 red foxes (Vulpes vulpes); in 21.4% of 28 wolves (Canis lupus); in 12.0% of 25 Iberian lynx (Lynx pardinus); in 16.7% of 6 European wildcats (Felis silvestris); in 6.4% of 31 Eurasian badgers (Meles meles); in 21.4% of 14 stone martens (Martes foina); in 66.7% of 3 pine martens (M. martes) and in 50% of 2 polecats (Mustela putorius). Antibodies to N. caninum in common genets (Genetta genetta) and Egyptian mongooses (Herpestes ichneumon) were only observed by c-ELISA but were not confirmed by IFAT and/or NAT. No antibodies were detected in 5 Eurasian otters (Lutra lutra) by any technique. Statistically significant differences were observed among species and among geographical areas. The highest seroprevalence of N. caninum infection was observed in the Cantabric Coastal region characterized by high humidity. To our knowledge, this is the first report of antibodies to N. caninum in free ranging wild carnivores, other than wild canids, in Europe. The existence of a possible sylvatic cycle could have important implications in both sylvatic and domestic cycles since they might influence the prevalence of infection in cattle farms in those areas. Abstract: Serum samples from 441 red deer (Cervus elaphus) and 161 other wild ruminant species, collected between 1993 and 2005 from six regions of Spain were tested for antibodies against Toxoplasma gondii by the modified agglutination test (MAT). Antibodies to T. gondii (MAT 1:25 or higher) were detected in 15.6% of red deer. Statistically significant differences were observed among sampling sites with seroprevalence in red deer from Catalonia (42.2%) being significantly higher compared with other Spanish regions (8.7%) (P <0.05). Statistically significant differences were not observed between T. gondii seroprevalence and sex, age or management of hunting estates (open versus fenced). Seroprevalence of T.gondii infection in other ruminants species was 24% of 79 fallow deer (Dama dama), 21.8% of 33 in roe deer (Capreolus capreolus), 33.3% of three Spanish ibex (Capra pyrenaica), 20% of 10 chamois (Rupicapra pyrenaica), 10% of 10 barbary sheep (Ammotragus lervia), and 14.8% of 27 mouflon (Ovis ammon) in areas not including Catalonia, where no samples from these species were available. Serological results indicated a widespread exposure to T. gondii among wildlife in Spain and suggest that consumption of raw or inadequately cooked meat, as well as handling carcasses of wild game, should be taken into account as a source of infection for humans. Abstract: Seroprevalence and associated risk factors for Toxoplasma gondii infection in pigs were analyzed in 1202 sera samples, including sows and pigs of three, seven, 11, 15 and 20weeks of age, from 23 farms in Catalonia, north-eastern Spain. Antibodies were tested by the modified agglutination test (MAT) at titers⩾1:25. Antibodies to T. gondii were found in 228 samples (19.0%; 95% CI: 16.8–21.2). The individual prevalence in animals higher than 7weeks of age was 22.8% (174/762; 95% CI: 16.6–29.0) and the within-farm prevalence ranged from 7.1% to 36.4%. Statistically significant differences were found among age classes. The risk factors significantly associated with T. gondii seroprevalence were the presence of cats, percentage of mortality at weaning and the presence of outdoor facilities in the farms. The seroprevalence observed in the present study indicates widespread exposure to T. gondii among domestic pigs in Catalonia, which may have important implications for public health. Toxoplasmosis is a parasitic zoonosis caused by Toxoplasma gondii which infects warm‐blooded species worldwide. Humans can be infected through ingestion of tissue cysts from raw or undercooked meat, including game meat. A nationwide large‐scale cross‐sectional study was conducted to assess exposure to T. gondii in seven wild ruminant species in Spain. A total of 2,040 serum samples from 77 sampling sites randomly distributed in the five bioregions (BRs) covering mainland Spain were tested for antibodies against T. gondii using the modified agglutination test. The overall seroprevalence was 22.0% (449/2,040). Seroprevalence by species in decreasing order was as follows: 39.6% (141/356) in roe deer (Capreolus capreolus), 37.1% (138/372) in fallow deer (Dama dama), 16.6% (92/553) in red deer (Cervus elaphus), 14.0% (26/186) in Southern chamois (Rupicapra pyrenaica), 11.5% (24/209) in mouflon (Ovis aries musimon), 7.8% (27/346) in Iberian wild goat (Capra pyrenaica) and 5.6% (1/18) in Barbary sheep (Ammotragus lervia). Seropositivity was detected in 74.0% (57/77) of the sampling sites. Results indicate widespread but not homogeneous exposure to T. gondii in wild ruminant populations in Spain during the last two decades and highlight differences related to animal species and spatial distribution of these species in this country; this implies potential consequences of T. gondii for animal health, conservation and public health. Abstract: Serum samples from 2970 (1400 sows, 1570 fattening) pigs, from 100 farms in the 10 main swine production regions in Spain were tested for antibodies against T. gondii by the modified agglutination test (MAT). Antibodies to T. gondii (MAT 1:25 or higher) were detected in 492 pigs (16.6%, 9.7% in fattening pigs and 24.2% in sows). The herd prevalence was 85.0% (95% CI: 78–92) and within-farm prevalence ranged from 2.9% to 92.8% (median=17.6%). Statistically significant differences were observed among sampling regions with seroprevalence significantly higher in pigs from Valencia Community (27.3%), Extremadura (23.3%) and Catalonia (21.2%). A generalized estimating equations model indicated that the risk factors associated with T. gondii seroprevalence were: age, sows compared to fattening pigs (OR=2.9; 95% CI=1.83–4.53), lack of rodent control (OR=1.9; 95% CI=1.04–3.60) and presence of cats (OR=1.6; 95% CI=1.12–2.34). The seroprevalence observed in the present study indicates a widespread, although variable, exposure to T. gondii among domestic pigs in Spain, which might have important implications for public health. Barbary sheep (Ammotragus lervia) is a North African native wild Caprinae, introduced in the 1970s in new territories such as Spain, the USA, and Mexico. Here, we describe Sarcocystis species in Barbary sheep. Sarcocysts were found in 19 out of 56 adult A. lervia in Southern Spain and characterized morphologically and molecularly. By light microscopy, sarcocysts had thin (< 1 μm) or thick (> 2 μm) walls. By transmission electron microscopy, sarcocysts with thick walls had Type 14 villar protrusions corresponding to S. tenella/S. capracanis of domestic sheep (Ovis aries) or goats (Capra hircus). Sarcocysts with thin walls had Type 7b villar protrusions that corresponded to S. arieticanis/S. hircicanis of domestic sheep or goats. Molecular analyses allowed the identification of only thick-walled Sarcocystis species. Six sarcocysts were assigned to S. tenella (99.2–100% and 95.6–100% sequence similarity within 18S rRNA and COI, respectively) and 19 sarcocysts were assigned to S. capracanis (98.5–99.8% and 97.9–99.0% sequence similarity within 18S rRNA and COI, respectively). Further studies are needed for taxonomic identification of sarcocysts in Barbary sheep because Sarcocystis species in sheep and goats are not cross transmissible despite morphological similarities. • We evince a widespread circulation of T. gondii in nonhuman primates in zoos in Spain. • Family Hominidae and sex were risk factors associated with seropositivity to T. gondii. • Results are of conservation concern, as several non-human primate species are highly susceptible to T. gondii. Toxoplasma gondii is a widely distributed zoonotic protozoan parasite, which can affect most warm-blooded species. Some species of non-human primates (NHPs) are highly susceptible to T. gondii infection. The aim of the study was to determine the seroprevalence and risk factors associated with T. gondii infection in NHPs housed in zoos in Spain. Sera from 189 NHPs belonging to 33 species were collected in eight zoos. Additionally, 10 of the 189 animals were longitudinally sampled. Anti- T. gondii antibodies were detected in 48 NHPs (25.4%; confidence interval of 95% (CI 95%): 19.2–31.6) using a modified agglutination test (MAT; cut-off = 25). Seropositive animals had titers of 25 (6.3%), 50 (8.3%), 100 (8.3%) and ≥500 (68.8%). Seropositivity was detected in 15 of the 33 species (45.5%). Of the 10 NHPs sampled more than once, two animals (one Barbary macaque [ Macaca sylvanus ] and one common chimpanzee [ Pan toglodytes ]) seroconverted along the study period, while one seropositive chimpanzee increased antibody titers over time. The Hominidae family (OR = 5.9; CI 95% : 2.7–12.8) and sex (females) (OR = 2.1; CI 95% : 1.1–4.1) were risk factors potentially associated with seropositivity to T. gondii. Our results evince a widespread circulation of T. gondii in NHPs in zoos in Spain, which may be of conservation concern. Toxoplasma gondii is a zoonotic protozoan of worldwide distribution. The present study provides information on risk factors affecting T. gondii infection in domestic and free-ranging wild ungulates sharing habitats in Mediterranean ecosystems in Spain. Serum samples from 482 extensively reared domestic ruminants and 2351 wild ungulates were tested for T. gondii antibodies using the modified agglutination test (MAT, cut-off 1:25). Toxoplasma gondii seroprevalence was 41.2% of 194 sheep, 18.6% of 199 cattle and 5.6% of 89 goats. The main risk factors associated with infection in livestock were the presence of cats, feeding on the ground and at stubble fields. In wild ungulates, T. gondii antibodies were detected in 10.5% of 1063 red deer, 15.6% of 294 fallow deer, 5.6% of 216 European mouflon, 5.6% of 90 Spanish ibex, 13.6% of 22 roe deer and 18.6% of 666 wild boars. The risk factors affecting T. gondii infection in wildlife were species, age and hunting season. Significantly higher seroprevalence was found in domestic ruminants, particularly in sheep, compared to the wild species tested. The present study indicates widespread exposure to T. gondii among domestic and wild ungulates in Southern Spain, with significant differences among species sharing the same ecosystem. Food safety regulations require the control of the presence of protozoa in meats destined for human consumption. Wild boar (Sus scrofa) meat may constitute a source of zoonoses. A 23.8% (688/2881) seroprevalence of anti-Toxoplasma gondii antibodies and 72.2% (662/910) Sarcocystis sarcocysts prevalence were detected among wild boars hunted in Southwestern areas of Spain. Identity of Sarcocystis spp. was performed by RFLP-PCR and sequencing, detecting S. miescheriana (7/8) and the zoonotic S. suihominis (1/8). Abstract: Toxoplasmosis is often fatal in captive wallabies, but the causes of this high susceptibility are not well understood. Here, we report fatal toxoplasmosis in a Bennet's wallaby (Macropus rufogriseus) due to an atypical Toxoplasma gondii strain for the first time in Europe. The wallaby was from a colony of 7 Bennet's wallabies that died over a 17-month period at a safari-zoological park in northeastern Spain. Only one of these wallabies was examined at necropsy. T. gondii-like organisms were detected by histological examination in several tissues and the diagnosis was confirmed through detection of T. gondii DNA by PCR. A nested PCR-based assay detected the 200- to 300-fold repetitive 529bp DNA fragment of T. gondii in a sample of brain tissue. Genotyping analysis with 15 single-copy microsatellite markers was performed on this positive DNA sample and revealed an atypical genotype. Atypical genotypes are frequently associated with severe forms of toxoplasmosis in humans. A cross-sectional study was carried out on Spanish ibex populations in Southern Spain to assess the seroprevalence of Toxoplasma gondii and Neospora caninum and to investigate the risk factors associated with these infections. Using the modified agglutination test, the seroprevalence to T. gondii was 27.5% (146/531; CI95%, 23.7-31.3), and this seropositivity significantly increased with age. Among adults, statistically significant differences were observed between geographical locations and over different sampling years. Thirty of 531 (5.6%) ibex had antibodies to N. caninum using a competitive ELISA, of which 27/30 (5.1%; CI95%, 3.1-7.1) were confirmed as seropositive by the indirect fluorescent antibody test. This study is the first to report the presence of N. caninum antibodies in Spanish ibex and also indicates widespread exposure of this species to T. gondii. The findings indicate that ibex are more exposed to T. gondii than to N. caninum in their natural environment and there is little evidence of co-infection with both parasites. Kennel dogs can serve as sentinels and/or reservoirs of diseases of veterinary and zoonotic interest because they have often roamed free and lived outdoors, thus being exposed to pathogens. We tested dogs from the kennel of Inca (Majorca/Mallorca, Balearic Islands, Spain) for evidence of infection with three protozoan parasites: Leishmania infantum, Toxoplasma gondii, and Neospora caninum. Exposure to L. infantum was found in 56.3% of 48 dogs (37.5% by Western blot, 43.8% by PCR). Only 30% of infected dogs had leishmaniosis-like lesions. Seroprevalence to T. gondii was 58.7% of 46 dogs using the modified agglutination test (MAT, titer 1:25). None of the 44 dogs tested had N. caninum antibodies using a commercial competitive ELISA, probably because the surveyed dogs did not roam in the proximity of cattle farms. Abstract: Felids are important in the epidemiology of Toxoplasma gondii infection because they are the only hosts that can excrete environmentally resistant oocysts. Antibodies to T. gondii and Neospora caninum were determined in serum samples from 59 feral cats (Felis silvestris catus) captured in baited traps during authorized predator control campaigns in Majorca, Balearic Islands, Spain. Antibodies to T. gondii assayed by the modified agglutination test (MAT) were found in 50 (84.7%) of 59 cats with MAT titers of 1:25 in one cat, 1:200 in four cats, 1:500 in seven cats, 1:1000 in nine cats, and ≥1:2000 in 29 cats. Seroprevalence (MAT, 1:25 or more) was significantly higher in adults (94.6%) than in juveniles (<6 months old; 40.0%). Seroprevalence to N. caninum, assayed by cELISA (VMRD) and confirmed by an indirect fluorescent antibody test, was low (6.8%, 4 of 59). The prevalence of T. gondii observed in wild cats in Majorca is one of the highest reported worldwide in this species and the highest observed in Europe to date. The results suggest that feral cats in Majorca have a high rate of T. gondii infection with important implications for public health on the island since the seropositive cats are likely to have already shed T. gondii oocysts in the environment. Abstract: Serum samples from 1034 non-carnivorous wildlife from Spain were tested for antibodies to Neospora caninum by competitive screening enzyme linked immunosorbent assay (ELISA) and confirmed by an indirect fluorescent antibody test (IFAT). High agreement was observed between results in both techniques (kappa value higher than 0.9). Prevalences of N. caninum antibodies positive by both techniques were 11.8% of 237 red deer (Cervus elaphus), 7.7% of 13 barbary sheep (Ammotragus lervia), 6.1% of 33 roe deer (Capreolus capreolus) and 0.3% of 298 wild boar (Sus scrofa). In one of 53 hares (Lepus granatensis), antibodies were found in the ELISA but could not be confirmed by IFAT due to lack of sample. Antibodies to N. caninum were not found in any of 251 wild rabbits (Oryctolagus cuniculus), 79 fallow deer (Dama dama), 27 mouflon (Ovis ammon), 40 chamois (Rupicapra pyrenaica) and three Spanish ibex (Capra pyrenaica). Statistically significant differences were observed between N. caninum seroprevalence in red deer and management of hunting estates (open versus fenced) with higher prevalence in fenced estates, and among sampling sites. Seroprevalence was particularly high in some areas (MO estate in South-Central Spain or some estates of Catalonia, North-East Spain), while no contact with N. caninum was observed in others. Results indicate that in certain areas of Spain, N. caninum is present in wildlife, especially in red deer. These results have important implications in both sylvatic cycles and may influence the prevalence of infection in cattle farms in those areas. Abstract: Four Korean squirrels (Tanias siberius) imported in Spain from People''s Republic of China died 2 days after their arrival at a pet shop. They had neurological signs associated with generalized toxoplasmosis involving brain, lungs, liver, and the heart. Toxoplasma gondii-like tachyzoites and tissue cysts were found in organs of all four squirrels. The protozoa stained positively with T. gondii polyclonal antibodies and were ultrastructurally similar to T. gondii. Calodium (Capillaria) hepaticum infection was found in the liver of one squirrel. Sera from 456 wild rabbits (Oryctolagus cuniculus) collected between 1992 and 2003 from five geographical regions of Spain were examined for antibodies to Toxoplasma gondii by the modified agglutination test. Antibodies to T. gondii were found in 65 (14.2%) wild rabbits. Prevalence of infection was significantly higher in samples collected from wild rabbits from Catalonia, northeast Spain (53.8%), where rabbits lived in forest, compared to other areas (Huelva and Cádiz, Andalucía, south Spain; Toledo, Castilla-La Mancha, central Spain; and Zaragoza, Aragón, northeast Spain) with more dry conditions, where prevalence ranged from 6.1 to 14.6%. No differences were observed on prevalence and age (young animals <7 months of age compared to older animals), sex, date of samples collection or season of samples collection. The results indicate that prevalence of T. gondii in some areas of Spain is high, and this finding could have environmental and/or public health implications if wild rabbits are to be used as a source of food. Simple Summary: Toxoplasma gondii is a zoonotic intracellular parasite which infects a wide range of warm-blooded animals. Long-term studies provide the necessary perspective required to understand those processes which took place over many years in order to address epidemiology and ecology in complex host communities. This study is focused on evaluating what the main long-term determinants of the seroprevalence of T. gondii are in the wild ungulate community from Doñana National Park (southwestern Spain). With this purpose, we assayed sera from 1573 wild ungulates (wild boar, red deer, and fallow deer), collected for 13 years (from 2005 to 2018). We found high seroprevalence values of T. gondii (% ± CI 95%; wild boar 39 ± 3.3; red deer 30.7 ± 4.4; and fallow deer 29.7 ± 4.2. Several factors operating in the medium and long-term (individual, environmental, population and stochastic) explained the risk of T. gondii in wild boar and deer, some of them operating at the community level. Toxoplasma gondii is an obligate intracellular protozoan which infects warm-blooded vertebrates, including humans, worldwide. In the present study, the epidemiology of T. gondii was studied in the wild ungulate host community (wild boar, red deer, and fallow deer) of Doñana National Park (DNP, south-western Spain) for 13 years (2005–2018). We assessed several variables which potentially operate in the medium and long-term (environmental features, population, and stochastic factors). Overall, the wild ungulate host community of DNP had high seroprevalence values of T. gondii (STG; % ± confidence interval (CI) 95%; wild boar (Sus scrofa) 39 ± 3.3, n = 698; red deer (Cervus elaphus) 30.7 ± 4.4, n = 423; fallow deer (Dama dama) 29.7 ± 4.2, n = 452). The complex interplay of hosts and ecological/epidemiological niches, together with the optimal climatic conditions for the survival of oocysts that converge in this area may favor the spread of the parasite in its host community. The temporal evolution of STG oscillated considerably, mostly in deer species. The relationships shown by statistical models indicated that several factors determined species patterns. Concomitance of effects among species, indicated that relevant drivers of risk operated at the community level. Our focus, addressing factors operating at broad temporal scale, allows showing their impacts on the epidemiology of T. gondii and its trends. This approach is key to understanding the epidemiology and ecology to T. gondii infection in wild host communities in a context where the decline in seroprevalence leads to loss of immunity in humans. Toxoplasmosis, caused by the protozoan Toxoplasma gondii , is a worldwide zoonotic disease, which affects most warm-blooded species. Besides the zoonotic relevance, toxoplasmosis is one of the major causes of reproductive disorders in small ruminants. A cross-sectional study was carried out to determine the seroprevalence, spatial distribution and risk factors associated with T. gondii infection in sheep and goats in southern Spain. During 2015–2017, a total of 1,943 small ruminants (998 sheep and 945 goats) from 127 flocks were tested for antibodies against T. gondii using the modified agglutination test (MAT, cut-off 1:25). Antibodies against T. gondii were detected in 464 of the 998 sheep (46.5 %; CI95 %: 43.4–49.6%) and 362 of the 945 goats (38.3 %; CI95 %: 35.2–41.4%) tested. The farm prevalence was 98.4 % (CI95 %: 95.4–100%) for sheep and 93.7 % (CI95 %: 87.6–99.7%) for goats. The generalized estimating equation analysis showed that presence of cats and existence of previous reproductive disorders were risk factors potentially associated with T. gondii seropositivity in small ruminants. Abstract: In recent years, multiple cases of aggressive behavior of Common ravens (Corvus corax) have been reported by farmers in Catalonia (NE Spain), including attacking of newborn animals and consumption of dead foetuses. In the present study, seroprevalence of Toxoplasma gondii and Neospora caninum was determined from 113 legally trapped and released Common ravens. T. gondii antibodies were found in 91 (80.5%; CI 95%:72–87) of 113 sera tested by the modified agglutination test. Antibodies to N. caninum were found in 24 (35.8%; IC 95%: 24.5–48.5) of 67 Common ravens tested by an indirect fluorescence antibody test with titers ranging from 1:50 (n =18) to ⩾1:100 (n =6). To the author’s knowledge, this is the first report of the presence of T. gondii and N. caninum antibodies in C. corax. The seroprevalence detected is one of the highest reported worldwide in wild birds, suggesting an important role for this species in the epidemiology of both parasites. [Copyright &y& Elsevier]

• Toxoplasma gondii is widespread in zoo animals in Spain, which could be of public health and conservation concerns. • The feeding habit (carnivorous) was a risk factor associated with T. gondii in captive animals. • Captive zoo animals can be useful sentinel species to monitor T. gondii. A serosurvey was performed to assess exposure to Toxoplasma gondii in zoo animals in Spain and to determine the dynamics of seropositivity in some zoo species over time. Sera from 393 zoo animals belonging to 91 species were collected in eight zoos in Spain between 2007 and 2019. Additionally, 39 of the 393 animals from five of the analyzed zoos were longitudinally sampled during the same study period. Antibodies to T. gondii were detected in 165 (42.0 %; CI95 %: 37.1–46.9) of 393 animals by the modified agglutination test (MAT, cut-off = 25). Antibody titers of 25, 50, 100 and ≥500 were found in 25 (15.2 %), 65 (39.4 %), 26 (15.8 %), and 49 (29.7 %) of the seropositive animals, respectively. Feeding habits (carnivorous species) was a risk factor (OR = 5.6; P < 0.001; CI95 %: 3.8–9.0) potentially associated with T. gondii exposure. Eighteen (46.2 %) of the 39 animals longitudinally sampled were always seropositive and five animals (12.8 %) (two spotted hyaena [ Crocuta crocuta ], one blesbok [ Damaliscus pygargus ], one white rhinoceros [ Ceratotherium simum ] and one mouflon [ Ovis aries musimon ]) seroconverted during the study period. Our results evince a wide circulation of T. gondii in zoos in Spain, including wild felids. Zoo animals can be useful sentinel species to monitor T. gondii circulation in epidemiological scenarios with a close human-wildlife interface.
