In this study, we investigated the compositions of vitamin E tocochromanol [tocopherol (Toc) and tocotrienol (T3)] in crude and refined rice bran oil (RBO) produced in Japan and other countries, including Brazil, Thailand, and Vietnam, based on high-performance liquid chromatography analysis. All RBO analyzed contained α-, β- and γ-Toc and α-, γ- and δ-T3. Japanese crude RBO, although not refined RBO, also contained β-T3. Furthermore, total Toc contents in both Japanese crude and refined oils were found to be higher than those in the crude and refined RBO from other countries. Total T3 contents in Japanese crude RBO were similar to those in the crude RBO from Brazil and Vietnam. The α-Toc and α-T3 contents in Japanese crude and refined RBO were considerably higher than those in the crude and refined RBO produced in other countries, whereas in contrast, γ-Toc and γ-T3 contents in Japanese crude and refined RBO were lower. Consequently, the ratios of total α-Toc and α-T3 contents to total γ-Toc and γ-T3 contents in Japanese crude and refined RBO (1.75 and 1.91, respectively) were notably higher than those in the crude and refined RBO produced in other countries. Similarly, the ratios of total Toc to total T3 in Japanese crude and refined RBO were higher than those in the crude and refined RBO produced in other countries. These results accordingly indicate that the ratio of total α-Toc and α-T3 contents to γ-Toc and γ-T3 contents could be used as an effective index to discriminate between the RBO produced in Japan and that produced in other countries.

Background: Piper chaba Hunt. is used as an ingredient in Thai traditional preparation for arthritis. Its isolated compound is piperine which shows anti-inflammatory activity. Piperine produces a burning sensation because it activates TRPV1 receptor. The TRPV1 activation involved with the analgesic and adjuvant effect. P. chaba Hunt. has not been reported about TRPV1 activation and adjuvant effect. The aim of this study was to investigate the effect of P. chaba extract and piperine on TRPV1 receptor, which is considered as a target for analgesic and their adjuvant effects to support the development of an analgesic drug from herbal medicine., Methods: The effect of P. chaba extract and piperine on HEK cells expressing TRPV1 channel was examined by calcium imaging assay. Adjuvant effects of P. chaba extract and piperine were investigated by a fluorescein isothiocyanate (FITC)-induced contact hypersensitivity (CHS) model in mice., Results: P. chaba extract induced calcium influx with EC 50 value of 0.67 μg/ml. Piperine induced calcium influx with EC 50 value of 0.31 μg/ml or 1.08 μM. For mouse CHS model, we found that 1% piperine, 5% piperine, 1% P. chaba extract and 5% P. chaba extract significantly enhanced sensitization to FITC as revealed by ear swelling responses., Conclusion: P. chaba extract and piperine activated TRPV1 channel and enhanced contact sensitization to FITC.

Opisthorchis viverrini can develop mitogenic substances into the excretory/secretory product (ESP) that may play an important role in promoting the genesis of cholangiocarcinoma (CCA). In the present study, glutathione S-transferase (GST) is identified as being secreted into Ov-ESP and acting as one of the parasitic mitogens. Its proliferative effect and possible mechanism were explored and its association with the tumor development is proposed. Ov-ESP was concentrated and purified by gel filtration chromatography. SDS-PAGE, 2-DE, and LC-MS/MS identified GST predominantly expressed in the proliferative ESP fraction. The recombinant OvGST (rOvGST) was produced by wheat germ cell-free expression and confirmed by an MTS assay to have a proliferative function on NIH-3T3 murine fibroblasts and MMNK1 non-tumorigenic human bile duct epithelial cells in a dose dependent manner with different optimal doses. The cell surface binding of rOvGST was confirmed in vitro and the activation of both pAKT and pERK was revealed as the mechanism of OvGST-mediated cell proliferation. With support from the observation of secreted OvGST on the biliary cells surrounding the parasites, it is suggested that OvGST can promote cell proliferation that consequently may accelerate the genesis of CCA., (Copyright © 2011 Elsevier Ireland Ltd.
