The DNA copy number variations (CNVs) are widely involved in affecting various kinds of biological functions, such as environmental adaptation. Tibetan sheep and White Suffolk sheep are two representative indigenous and exotic breeds raised in Sichuan, China, and both of them have many contrasting biological characteristics. In this study, we employed high-throughput sequencing approach to investigate genome-wide CNVs between the two sheep breeds. A total of 11,135 CNV regions (CNVRs) consisting of 6,488 deletions and 4,647 duplications were detected, whose length ranged from 1,599 bp to 0.56 Mb with the mean of 4,658 bp. There were 281 CNVRs segregated between Tibetan sheep and White Suffolk sheep, and 18 of them have been fixed within both breeds. Functional analyses of candidate genes within the segregating CNVRs revealed the thyroid hormone signaling pathway and CTNNB1 gene that would be responsible for differential biological characteristics of breeds, such as energy metabolism, seasonal reproduction, and litter size. Furthermore, the segregating CNVRs identified in this study were overlapped with many known quantitative trait loci that are associated with growth, testis weight, and reproductive seasonality. In conclusion, these results help us better understanding differential biological characteristics between Tibetan sheep and White Suffolk sheep.

Lakes on the Qinghai-Tibet Plateau (QTP) have notably expanded over the past 20 years. Due to lake water level rise and lake area expansion, the permafrost surrounding these lakes is increasingly becoming submerged by lake water. However, the change process of submerged permafrost remains unclear, which is not conducive to further analyzing the environmental effects of permafrost change. Yanhu Lake, a tectonic lake on the QTP, has experienced significant expansion and water level rise. Field measurement results indicate that the water level of Yanhu Lake increased by 2.87 m per year on average from 2016 to 2019. Cold permafrost, developed in the lake basin, was partially submerged by lake water at the end of 2017. Based on the water level change and permafrost thermal regime, a numerical heat conduction permafrost model was employed to predict future changes in permafrost beneath the lake bottom. The simulated results indicate that the submerged permafrost would continuously degrade because of the significant thermal impact of lake water. By 2100, the maximum talik thicknesses could reach approximately 7, 12, 16, and 19 m under lake-bottom temperatures of +2.0, +4.0, +6.0, and +8.0 °C, respectively. Approximately 291 years would be required to completely melt 47 m of submerged permafrost under the lake-bottom temperature of +4 °C. Note that the permafrost table begins to melt earlier than does the permafrost base, and the decline in the permafrost table occurs relatively fast at first, but then the process is attenuated, after which the permafrost table again rapidly declines. Soils, especially permafrost in the Arctic and the Tibetan Plateau, are one of the largest reservoirs of mercury (Hg) in the global environment. The Hg concentration in the grassland soils over the Tibetan Plateau and its driving factors have been less studied. This study analyzes soil total mercury (STHg) concentrations and its vertical distribution in grassland soil samples collected from the Tibetan Plateau. We adopt a nested-grid high-resolution GEOS-Chem model to simulate atmospheric Hg deposition. The relationship between STHg and soil organic carbon (SOC), as well as atmospheric deposition, are explored. Our results show that the STHg concentrations in the Tibetan Plateau are 19.8 ± 12.2 ng/g. The concentrations are higher in the south and lower in the north in the Tibetan Plateau, consistent with the previous results. Our model shows that the average deposition flux of Hg is 3.3 μg m -2 yr -1 , with 57% contributed by dry deposition of elemental mercury (Hg 0 ), followed by dry (19%) and wet (24%) deposition of divalent mercury. We calculate the Hg to carbon ratio (R Hg:C ) as 5.6 ± 6.5 μg Hg/g C, and the estimated STHg is 86.6 ± 101.2 Gg in alpine grasslands in the Tibetan Plateau. We find a positive relationship between STHg and SOC in the Tibetan Plateau (r 2  = 0.36) and a similar positive relationship between STHg and atmospheric total Hg deposition (r 2  = 0.24). A multiple linear regression involving both variables better model the observed STHg (r 2  = 0.42). We conclude that SOC and atmospheric deposition influence STHg simultaneously in this region.
