Background: Ovine psoroptic mange (sheep scab) is an important disease of sheep worldwide caused by the parasitic mite, Psoroptes ovis. It has a negative impact on animal welfare and leads to significant economic losses for the sheep industry. Effective and targeted management is required to limit its transmission. Methods: A stochastic metapopulation model of sheep scab transmission is used to investigate the contribution of the treatment of sheep prior to movements to sales, gatherings (predominantly markets) and away grazing to the reduction of prevalence of farms with scab in Great Britain. Results: Treatment prior to movement to gatherings resulted in an 86% reduction in the overall prevalence of farms with scab and was more effective at reducing the overall prevalence of farms with scab than treatment before other categories of movements. The relative risk of farms having scab infection was inversely related to the percentage of farms which treated, but this relationship was not linear, with the biggest declines in the prevalence of farms with scab being achieved by small percentages of farms treating; a 50% relative reduction in the farm prevalence was achieved with only 15% of farms treating prior to gathering movements. Conclusions: The results suggest that pre-movement treatment of sheep could make an important contribution to national scab control and, in practice, the approach could be more highly targeted if used in conjunction with known geographic and management risk factors for scab. Small ruminant lentivirus infections in sheep most commonly present with respiratory signs (maedi) and indurative mastitis, while primary neurological signs (visna) have rarely been reported in Great Britain (GB). Most reports of visna describe signs referable to myelitis, although central signs associated with encephalitis may feature. In this case, visna was diagnosed in a 4‐year‐old ewe in a small pedigree sheep flock, recently imported to GB from northern Europe. Initial clinical findings were of a head tilt, circling, facial tremors and a unilaterally reduced menace response. These neurological signs progressed to include hyperaesthesia, ipsilateral hemiparesis and recumbency. Flock level infection had recently been diagnosed by serology, and the diagnosis in this individual case was confirmed by serology and histopathology. The subsequent sale of animals from the flock through a large national auction and at private sales raises significant ethical questions and serves as a reminder of the importance of biosecurity precautions. Sheep scab is endemic in Great Britain with an estimated national herd-level prevalence of 10.9% from a surveyed population of sheep farms. Previous studies have investigated how sheep farmers manage sheep scab on their farms in Great Britain, but there have not been any qualitative studies investigating sheep farmers perceptions on the roles different stakeholders have in the management of sheep scab. This qualitative study aims to explore how sheep farmers perceive their role and the different stakeholders' roles in the management of sheep scab, and how they would like sheep scab to be managed going forward. Semi-structured interviews were conducted with 43 sheep farmers from England, Scotland, and Wales. The data were analysed using reflexive thematic analysis, through the theoretical lens of Foucault's notion of 'biopower'. Two themes were generated: 'The feeling of powerlessness leads to a need of rules and regulations', with sub-themes: 'The need for governmental rules and regulations' and 'The need for rules and regulation at livestock markets', and 'An apparent lack of sheep scab surveillance', with sub-themes: 'The farmers perceive that the veterinarians have control over surveillance' and 'The farmers have control over surveillance on their farms'. In the first theme, the respondents suggested that more rules and regulations to control the management of sheep scab was required. This included reinstating of sheep scab as a notifiable disease in England and Wales, as well as more regulations at livestock markets to prevent the trading of infested sheep; both of which would subject the farmers to regulatory power mechanisms. The second theme centred around who has the control of surveillance on the farms. Most of the respondents perceived that the veterinarians had knowledge and expertise of the local area on sheep scab, which they were able to relay to the farmers. Thus, veterinarians exerted disciplinary power by creating 'docile' bodies. However, it also appeared that veterinarians were not regularly called onto farms. Although disciplinary power flows through the interactions between the farmer and veterinarian, the techniques currently used are not always having their desired effect. The study demonstrated that how sheep farmers want sheep scab to be managed is, at times, conflicting and contradictory, which highlights the complexity of sheep scab as a disease to manage. Research suggests that many sheep farmers continue to carry out traditional antibiotic use practices despite new 'good practice' recommendations. The aim of this study was to group farmers depending on their attitudes around antibiotic use and antibiotic resistance, and determine the behaviours that are associated with the farmers in these groups. In 2017, a flock health survey was sent to British sheep farmers. K-means cluster analysis was used to identify groups of farmers with similar attitudes towards antibiotic use and resistance. A multivariable logistic regression model was built to determine the associations between farmers' past behaviours and their antibiotic attitude group. There were 461 responses. Two groups of farmers were identified based on their antibiotic attitudes. Cluster 1 were defined as the "discordant" group who had positive views of using antibiotics prophylactically and negative views of reducing antibiotic use. Cluster 2 were defined as the "concordant" group who were positive about reducing antibiotic use and had negative views about using antibiotics prophylactically. Using antibiotics in all lambs (OR = 2.689, CI = 1.571, 4.603), using antibiotics in all ewes (OR = 3.388, CI = 1.318, 8.706), always trimming diseased feet over the past three years (OR = 2.487, CI = 1.459, 4.238), not using a computer to record information over the past three years (OR = 1.996, CI = 1.179, 3.381), not changing worming practices over the past three years (OR = 1.879, CI = 1.144, 3.087), and farmers' perceptions that their sheep flock did not make a financial loss in the past three years (OR = 2.088, CI = 1.079, 4.040) were significantly associated with belonging to the discordant group. Talking to their veterinarian about antibiotic use or the frequency of veterinary visits were not associated with antibiotic attitude group. These results suggest that farmers who had attitudes relating to antibiotic use that did not align with current recommendations carried out more traditional practices, which were strengthened by their positive perceptions of profitability. Psoroptic mange (sheep scab), caused by the parasitic mite, Psoroptes ovis, is an important disease of sheep worldwide. It causes chronic animal welfare issues and economic losses. Eradication of scab has proved impossible in many sheep-rearing areas and recent reports of resistance to macrocyclic lactones, a key class of parasiticide, highlight the importance of improving approaches to scab management. To allow this, the current study aimed to develop a stochastic spatial metapopulation model for sheep scab transmission which can be adapted for use in any geographical region, exhibited here using data for Great Britain. The model uses agricultural survey and sheep movement data to geo-reference farms and capture realistic movement patterns. Reported data on sheep scab outbreaks from 1973 to 1991 were used for model fitting with Sequential Monte Carlo Approximate Bayesian Computation methods. The outbreak incidence predicted by the model was from the same statistical distribution as the reported outbreak data ( χ 2 = 115.3, p = 1) and the spatial location of sheep scab outbreaks predicted was positively correlated with the observed outbreak data by county (τ = 0.55, p < 0.001), confirming that the model developed is able to accurately capture the number of farms infected in a year, the seasonality of scab incidence and the spatial patterns seen in the data. The emergence and spread of tick-borne arboviruses pose an increased challenge to human and animal health. In Europe this is demonstrated by the increasingly wide distribution of tick-borne encephalitis virus (TBEV, Flavivirus, Flaviviridae), which has recently been found in the United Kingdom (UK). However, much less is known about other tick-borne flaviviruses (TBFV), such as the closely related louping ill virus (LIV), an animal pathogen which is endemic to the UK and Ireland, but which has been detected in other parts of Europe including Scandinavia and Russia. The emergence and potential spatial overlap of these viruses necessitates improved understanding of LIV genomic diversity, geographic spread and evolutionary history. We sequenced a virus archive composed of 22 LIV isolates which had been sampled throughout the UK over a period of over 80 years. Combining this dataset with published virus sequences, we detected no sign of recombination and found low diversity and limited evidence for positive selection in the LIV genome. Phylogenetic analysis provided evidence of geographic clustering as well as long-distance movement, including movement events that appear recent. However, despite genomic data and an 80-year time span, we found that the data contained insufficient temporal signal to reliably estimate a molecular clock rate for LIV. Additional analyses revealed that this also applied to TBEV, albeit to a lesser extent, pointing to a general problem with phylogenetic dating for TBFV. The 22 LIV genomes generated during this study provide a more reliable LIV phylogeny, improving our knowledge of the evolution of tick-borne flaviviruses. Our inability to estimate a molecular clock rate for both LIV and TBEV suggests that temporal calibration of tick-borne flavivirus evolution should be interpreted with caution and highlight a unique aspect of these viruses which may be explained by their reliance on tick vectors. Author summary: Tick-borne pathogens represent a major emerging threat to public health and in recent years have been expanding into new areas. LIV is a neglected virus endemic to the UK and Ireland (though it has been detected in Scandinavia and Russia) which is closely related to the major human pathogen TBEV, but predominantly causes disease in sheep and grouse. The recent detection of TBEV in the UK, which has also emerged elsewhere in Europe, requires more detailed understanding of the spread and sequence diversity of LIV. This could be important for diagnosis and vaccination, but also to improve our understanding of the evolution and emergence of these tick-borne viruses. Here we describe the sequencing of 22 LIV isolates which have been sampled from several host species across the past century. We have utilised this dataset to investigate the evolutionary pressures that LIV is subjected to and have explored the evolution of LIV using phylogenetic analysis. Crucially we were unable to estimate a reliable molecular clock rate for LIV and found that this problem also extends to a larger phylogeny of TBEV sequences. This work highlights a previously unknown caveat of tick-borne flavivirus evolutionary analysis which may be important for understanding the evolution of these important pathogens. Introduction: The most abundant and widespread tick species in Great Britain, Ixodes ricinus, is responsible for the transmission of a range of pathogens that cause disease in livestock. Empirical data on tick distribution and prevalence are required to inform farm management strategies. However, such data are largely unavailable; previous surveys have been rare and are usually relatively localised. Methods: A retrospective questionnaire survey of farmers was used to assess the reported prevalence of ticks on livestock across Great Britain. Spatial scan statistics and kernel density maps were used to assess spatial clustering and identify areas of significantly elevated risk, independent of the underlying distribution of respondents. Logistic regression models were used to identify risk factors for tick presence. Results: Tick infection risk to livestock is shown to be spatially aggregated, with areas of significantly elevated risk in north Wales, northwest England and western Scotland. Overall, the prevalence of farms reporting tick presence was 13% for sheep farms and 6% for cattle farms, but in "hot spot" clusters prevalence ranged between 48–100%. The prevalence of farms reporting tick-borne disease overall was 6% for sheep and 2% for cattle, but on farms reporting ticks, prevalence was 44% and 33% for sheep and cattle farms, respectively. Upland farming, larger flock sizes, region and the presence of sheep on cattle farms were all significant risk factors for tick presence. Conclusions: These data have important implications for assessing both the risk of tick-borne disease in livestock and optimising approaches to disease management. In particular, the study highlights the need for effective livestock tick control in upland regions and the southwest, and provides evidence for the importance of sheep as tick maintenance hosts. Background: Footrot and interdigital dermatitis are endemic infectious diseases in all sheep farming regions, impairing welfare and production. The development of efficacious vaccines against the primary causative pathogen has been hampered by the extensive antigenic diversity of Dichelobacter nodosus. Understanding the heterogeneity of the pathogen within and between flocks is essential if the feasibility of bespoke vaccine production is to be assessed for use in the U.K. Results: In this study 56 ewe and lamb isolates from 9 flocks were compared by D. nodosus serogroup and Multi Locus Sequence Type which provides significantly enhanced discriminatory power for molecular epidemiology. Serogroup heterogeneity between flocks ranged from two to five unique serogroups per flock. Three flocks contained isolates of two serogroups, two flocks contained isolates of three serogroups and one flock included isolates of five serogroups. Analysis of 25 isolates from one flock with high prevalence of lameness, identified that serogroup and sequence type was significantly correlated with age. Significantly higher proportion of lambs were infected with serogroup B (principally ST85) as opposed to serogroup H (principally ST86), which predominated amongst adult sheep. Conclusions: Genomic heterogeneity of the pathogen was significantly lower within flock compared to heterogenicity observed between flocks. Furthermore, this study indicates that within a flock, the host-pathogen dynamics and susceptibility to particular D. nodosus strains may be age dependent. [ABSTRACT FROM AUTHOR]

Abstract Certain genotypes of sheep have been identified to increase their susceptibility (the VRQ allele) or resistance (the ARR allele) to classical scrapie. This study's aim was to assess the spatio-temporal pattern of the ARR and VRQ alleles in Great Britain (GB) and to explore the risk factors associated to their presence. Data was collected from the GB scrapie active surveillance program, the sheep and goat inventory survey (GB census survey) and the agricultural survey for the period 2002-2015. Spatio-temporal trends of genotypes were assessed through the use of choropleth maps, spatial cluster and linear regression analyses. Multivariable mixed effect logistic regression analyses were performed to investigate the association between the resistant or susceptible genotypes, and breeds, farm purpose, animal purpose, surveillance stream, country location and herd size. The results show a significant upward trend in the frequency of most resistant ARR alleles (1.15% per year, 95%CI: 0.76-1.53) and significant downward trend of most susceptible VRQ alleles (-0.40% per year; 95%CI: -0.69 to -0.10]. The trend continues after the termination of the national scrapie plan in 2009. Breeds such as Herdwick (OR = 0,26; 95%CI: 0.14-0.46), Shetland (OR = 0.22; 95%CI: 0.13-0.39), Swaledale (OR = 0.58; 95%CI: 0.47-0.73), Scottish blackface (OR = 0.54; 95%CI: 0.41-0.71) and Welsh Montain (OR: 0.59; 95%CI: 0.44-0.79) were identified with lower odds ratios of having the resistant ARR allele, while Beulah speckled face (OR = 1.58; 95%CI: 1.04-2.41), Jacob (OR = 2.91; 95%CI: 1.33-6.40), Lleyn (OR = 2.94; 95%CI: 1.28-6.74) and Suffolk (OR = 2.19; 95%CI: 1.69-2.84) had higher odds ratios of having the ARR allele. Other risk factors associated to presence of ARR allele were finishing farms (OR = 1.15; 95%CI: 1.06-1.24) and farms in Scotland (OR = 0,78; 95%CI: 0.73-0.83) and in Lowland grazing areas (OR = 1.53; 95%CI: 1.39-1.67). Factors associated with presence the VRQ genotype were farms in Scotland (OR = 0,85; 95%CI: 0.77-0.93) and breeds such as Herdwick (OR = 2.2; 95%CI: 1.08-4.97), Shetland (OR = 4.12; 95%CI: 2.20-7.73) and Sweledale (OR = 1.51; 95%CI: 1.10-2.09). For the most resistant genotype, two significant spatial clusters were identified: a high-risk cluster in the south-west of GB (RR = 1.51, p < 0.001) and a low-risk cluster in northern GB (RR = 0.65, p < 0.001). For the most susceptible genotypes, one significant high-risk cluster was identified in Wales (RR = 2.89 and p = 0.013). Surveillance for classical scrapie could be improved with a risk-based approach by focussing on those areas and farm types identified to have higher frequency of VRQ alleles and less frequency of ARR alleles. Anthelmintic treatments to ewes around lambing time, often with long acting products, have become common practice on UK sheep farms, but these treatments have also been shown to be highly selective for anthelmintic resistance in New Zealand and Australia, with field data supported by modelling results. The purpose of this study was to determine (1) the effect of treating or withholding anthelmintic treatments and (2) the effect of treatment of ewes with a persistent or non-persistent anthelmintic, on early infection in lambs in the UK. Faecal egg count data for 10–16 weeks old lambs collected over a three year period (2012–2014) was analysed. Samples were grouped according to whether the ewes on the farm had or had not been treated with an anthelmintic at lambing. For both analyses, data for early infection were analysed by generalised linear mixed model. There was no effect of withholding or treating ewes on subsequent early infection in lambs. In addition, there was no effect of region, farm type or management type on the faecal egg counts. There was, however an effect of year, with lambs having lower counts in 2014 than in 2012 and an interaction between year and ewe treatment, with data suggesting lower infection levels over time for those farms withholding anthelmintic treatments altogether. There was no effect of drug type on early infection in lambs nor region, farm type or treatment on the lamb faecal egg counts. However, there was an effect of year and an interaction between year and drug type with lower egg count over time with the short acting drugs. Our study supports data generated by other researchers suggesting that the practice of treating ewes at lambing to reduce contamination on pasture and minimise subsequent disease may not in fact always result in lower levels of infection in lambs. The study also demonstrated no significant benefit in early infection in lambs when ewes were treated with long acting compared to short acting anthelmintics. Ovine psoroptic mange (commonly known as sheep scab) is, arguably, one of the most important ectoparasitic diseases of sheep in the UK. Although research has been conducted on the practices and knowledge of British sheep farmers regarding the management of sheep scab, recent data is limited. During October 2021 an online survey was completed by 725 British sheep farmers. Most respondents farmed in England (63.0%, 457/725), followed by Wales (20.6%, 149/725) and Scotland (16.4%, 119/725). Over half (53.0%, 384/725) of the respondents had experienced sheep scab in their flocks before, and 38.3% (147/384) of respondents experienced sheep scab twice or more in the previous ten years. The estimated prevalence of sheep scab from these surveyed farmers was 10.9% (79/725). Of the farmers who had experienced sheep scab before, only 36.2% (139/384) reported using skin scrapes and 12.5% (48/384) reported using blood tests for diagnosis. Over three-quarters of farmers quarantined all new sheep (76.6%; 498/650), but fewer quarantined returning sheep (28.6%; 83/290). Many of the respondents (43.7%, 252/577) did not quarantine new sheep for the suggested minimum 21 days. In addition, only 53.0% (384/725) of respondents answered all three of the knowledge questions regarding sheep scab correctly. The analysis illustrates that there is scope to improve the use of diagnostics, which suggests that misdiagnosis of sheep scab may be high and thus inappropriate treatments may be used, or treatments may not be used at all. It also demonstrated that there is scope for improvement in farmers' knowledge regarding sheep scab, particularly that sheep scab could be detected via a blood test and that the disease could present without visible signs. Our results further support that sheep scab is a large-scale endemic issue, potentially with an increasing prevalence in Great Britain. Future control interventions need to focus on improving knowledge exchange, raising awareness of best practice controls to ensure that better management strategies are employed by farmers, including improved control at livestock markets to reduce the national transmission and thus focus on coordinated control interventions. Anthelmintics are commonly used on the majority of UK commercial sheep farms to reduce major economic losses associated with parasitic diseases. With increasing anthelmintic resistance worldwide, several countries have produced evidence-based, best practice guidelines with an example being the UK’s Sustainable Control of Parasites in Sheep (SCOPS) initiative. In 2012, a pilot study demonstrated that SCOPS-managed farms used fewer anthelmintic treatments than traditionally managed farms, with no impact on lamb productivity and worm burden. Building on these results, we collected data for three consecutive years (2012–2014) with the following aims: (1) To compare the effects of traditional and SCOPS-based parasite management on lamb productivity and worm burden; (2) To evaluate the effect of region and farm type on lamb productivity and worm burden; (3) To compare the frequency and patterns of use of anthelmintic treatment on traditional and SCOPS-managed farms. The study was carried out on 16 farms located in the North east and the South west of England and Wales. Lamb productivity was assessed by quantifying birth, mid-season and finish weights and calculating daily live-weight gains and time to finish in a cohort of 40–50 lambs on each farm. Five annual faecal egg counts were carried out on each farm to assess worm burden. No differences in lamb productivity and worm burdens were found between farms that adopted SCOPS guidelines and traditional farms across the three years. However, mean infection levels increased for both the SCOPS and the traditional groups. Lamb production was not significantly different for farm type and region but the effect of region on infection was significant. For both ewes and lambs, SCOPS farms carried out significantly fewer anthelmintic treatments per year, and used fewer anthelmintic doses/animal than traditional farms. The data suggest a trend to increasing use of anthelmintics in ewes on traditional but not on the SCOPS farms and a decreasing use of anthelmintics in lambs on both SCOPS and traditional farms. Across time, an increasing number of SCOPS farmers left their ewes and lambs untreated and the reverse was true for traditional farmers. Overall, farms implementing SCOPS guidelines used less anthelmintic treatments and less frequently than traditionally managed farms, without loss of animal performance or increased worm burden. Implementing SCOPS guidelines might have economic benefits for farmers, help reduce development of anthelmintic resistance on farms and decrease any environmental impact of anthelmintics. Endemic sheep and cattle diseases represent a constant strain and impact to animal health and welfare, the environment, public health, and the economy. Quantifying this impact helps to inform decisions on surveillance and control of livestock diseases. This systematic literature review had two objectives; to describe the economic impact of endemic sheep and cattle diseases in the United Kingdom using a broad conceptualisation of impact, and to investigate what variables (e.g. medication costs, loss of production) and methods are included in these calculations. The Prisma protocol for systematic literature reviews was followed. Searches were performed in Pubmed, Scopus and Web of Science using selected and trialled search terms. A total of 1129 papers were identified and screened for relevancy; 38 papers were selected for full review extracting and analysing data on disease, impact and methods. From this final selection of papers it was found that; 1) research in this area is mainly focused on the dairy sub-sector, 2) the most mentioned diseases were mastitis and lameness in dairy cattle; bovine viral diarrhoea in beef cattle; and ectoparasites in sheep with reported animal-level costs of £ 77-£ 548/cow/year, £ 26-£ 185/bovine animal/year and £ 40-£ 47/ewe/year, respectively, 3) numerous methods and variables were used to calculate or estimate the economic impact with most studies focussing on the direct producer impacts and less on the wider implications and affected stakeholders; and 4) it was common for studies to look at one disease in isolation rather than consider disease impact on the farm as a whole. It is recommended that future economic impact calculations on livestock disease include wider implications to estimate the true cost of disease. To generate the necessary data, a wider, more inclusive conceptualisation of impact will be needed to support the collection of data and facilitate communication between stakeholders. Systematic health data recording combined with assessment calculations and metrics that allow comparability within or across livestock sub-sectors will increase the informative value of these impact calculations. • Research is heavily weighted towards the dairy industry. • The range in costs demonstrates work is still needed to reduce the avoidable costs. • Few externalities are captured thereby underestimating the societal impact. • Disease interactions and total disease burden on farm needs to be considered. In recent years whole flock antibiotic treatment has been applied in practice in order to eradicate footrot from individual flocks. If footrot can be eradicated by a one-off whole flock treatment, this could result in lower usages of antibiotics overall and would improve welfare on these farms. Examination of the lungs of adult sheep with chronic respiratory diseases was readily achieved using both 5MHz linear and sector scanners. Superficial lung abscesses in eight sheep appeared as anechoic areas containing multiple hyperechoic dots bordered distally by a broad hyperechoic capsule. Unilateral fibrinous pleurisy (2 sheep) appeared as an anechoic area containing a hyperechoic latticework. Ovine pulmonary adenocarcinoma (OPA) lesions appeared as sharply demarcated hypoechoic areas in the lung parenchyma initially in the cranioventral lung lobes (21 sheep) with lesions also present in the caudodorsal diaphragmatic lobe (11 sheep); abscesses and areas of calcification within the OPA tumour mass were also identified. Daily treatment with procaine penicillin for 30 consecutive days was successful in both sheep with unilateral fibrinous pleurisy and six sheep identified with superficial lung abscesses measuring 2-8 cmin diameter; only one of two sheep with more extensive lesions recovered. Auscultation of the chest failed to detect adventitious sounds in any of the ten sheep with lung abscesses; normal breath sounds were reduced over the area of fibrinous pleurisy; no pleuritic rubs were heard. The Cattle Health and Welfare Group of Great Britain report (CHAWG; 2012) lists the most important cattle diseases and disorders but fails to fully acknowledge the importance of animal mental health and; in so doing; misses the opportunity to further promote animal welfare. There are effective prevention regimens; including vaccination; husbandry and management strategies for all ten listed animal health concerns in the CHAWG report; however control measures are infrequently implemented because of perceived costs and unwillingness of many farmers to commit adequate time and resources to basic farm management tasks such as biosecurity; and biocontainment. Reducing disease prevalence rates by active veterinary herd and flock health planning; and veterinary care of many individual animal problems presently "treated" by farmers; would greatly improve animal welfare. Published studies have highlighted that treatments for lame sheep are not implemented early enough with many farmers delaying treatment for weeks; and sometimes even months; which adversely affects prognosis. Disease and welfare concerns as a consequence of sheep ectoparasites could be greatly reduced if farmers applied proven control strategies detailed in either veterinary flock health plans or advice available from expert veterinary websites. Recent studies have concluded that there is also an urgent need for veterinarians to better manage pain in livestock. Where proven treatments are available; such as blockage of pain arising from ovine obstetrical problems by combined low extradural injection of lignocaine and xylazine; these are seldom requested by farmers because the technique is a veterinary procedure and incurs a professional fee which highlights many farmers' focus on economics rather than individual animal welfare. Abstract: Protozoal merozoites were identified in the cerebrospinal fluid of two sheep with neurological disease in the UK. Polymerase chain reaction (PCR) identified the merozoites as Sarcocystis capracanis, a common protozoal pathogen of goats. This is the first report of this species infecting sheep and may represent an aberrant infection with sheep acting as dead end hosts, or alternatively could indicate that sheep are able to act as intermediate hosts for S. capracanis, widening the previously reported host range of this pathogen. It is possible that S. capracanis is a previously unrecognised cause of ovine protozoal meningoencephalitis (OPM) in the UK. [Copyright &y& Elsevier]

Abstract: The objective of the present studies was to evaluate the efficacy of a combined formulation (Startect® Dual Active Oral Solution for Sheep, Pfizer Animal Health) of derquantel (DQL) and abamectin (ABA) for the treatment of: (1) sheep experimentally infected with a moxidectin (MOX)-resistant isolate of Teladorsagia circumcincta, and (2) multi-drug resistant gastrointestinal nematode parasites under UK field conditions. In the first study, a total of 40 animals were allocated into 4 treatment groups, and were either left untreated or treated with DQL+ABA, MOX or ABA. Faecal samples were collected on days 1–5 and on day 7 after treatment to examine the reduction in faecal egg excretion and to evaluate the egg viability. On day 14 post treatment all animals were euthanised for abomasal worm counts. There was a 100% reduction in geometric mean worm counts for the DQL+ABA treated animals compared to the untreated control animals (P <0.0001), whereas the percentage reduction in worm counts for the MOX- (P >0.05) and ABA-treated (P =0.0004) animals was 12.4% and 71.8%, respectively. The data from the egg hatch assay (EHA) indicated that in the MOX-treated and the ABA-treated animals, the majority of the eggs hatched after treatment. In the field study, performed on four farms, animals were allocated into 6 groups of 11–15 animals each in order to conduct a faecal egg count reduction test (FECRT), based on arithmetic mean egg counts. One group of animals remained untreated, whereas the other animals were treated with DQL+ABA, MOX, fenbendazole (FBZ), levamisole (LV) or ivermectin (IVM). On each of the farms the reduction in egg excretion after treatment with FBZ, LV or IVM was below 95.0%, indicating anthelmintic resistance. The efficacy of DQL+ABA ranged from 99.1 to 100%, yielding significantly lower egg counts compared to the untreated control group (P ≤0.003). For MOX the egg counts were significantly (P ≤0.003) lower compared to the untreated group at each farm, with reductions varying from 98.2 to 100%. The post-treatment copro-cultures for larva identification indicated that T. circumcincta was the most abundant worm species after treatment (52–99% of the larvae). A telephone survey of 600 farmers throughout Great Britain and Ireland was conducted in order to characterise helminth control practices, and identify factors correlated with perceived anthelmintic failure. Overall, 93% of surveyed farmers routinely treated their sheep against nematodes, 67% against liver fluke and 58% against tapeworms. Anthelmintic resistance in nematodes was perceived by farmers to be present on 10% of farms. Farmers who dosed ewes at mating were more likely to have observed anthelmintic failure, than those who were aware of national guidelines on parasite control. However, objective assessment of anthelmintic efficacy had only been undertaken on 19% of farms. Ewes were treated at mating and lambing on 63% and 62% of farms, respectively. On average, lambs were treated 3.6 times annually, depending on geographical region and on dates of lambing and finishing. Although 'quarantine' treatments were widely administered to bought-in stock, these were appropriately applied in only 3% of cases. This study provides baseline data against which the impact of future anthelmintic information campaigns can be assessed; it will facilitate the development of rational, farm-level mathematical models in support of sustainable parasite control, and will aid in the design of farm management practices that prolong the effective lifespan of novel classes of anthelmintic. Abstract: The inhibition of Teladorsagia and other nematode genera at the early fourth-stage is a biological process that allows the parasites to survive in their host in a dormant state when prevailing conditions may otherwise kill them or prevent their progeny from surviving in the external environment. A study was conducted in Scotland to evaluate the efficacy of monepantel, an amino-acetonitrile derivative, against natural infections of inhibited fourth-stage Teladorsagia spp. larvae. At necropsy it was determined that the untreated control sheep were additionally infected with developing fourth-stage Teladorsagia spp. larvae and this is the first published evidence on the efficacy of monepantel against natural infections of this parasite and stage. The study sheep, which had grazed on naturally contaminated pastures since birth, were transferred to indoor housing after a subset of animals was examined to confirm the presence of inhibited larvae within the study population prior to the experiment. After 14 days of housing, monepantel was orally administered at 2.5mg/kg to half of the animals. The sheep were necropsied seven days later and their parasite burdens recovered for the determination of efficacy, which was 99.7% for the inhibited stages and 99.3% for the developing fourth-stages. In conclusion, monepantel dosed orally at 2.5mg/kg is a highly effective treatment against naturally acquired infections of inhibited and developing fourth-stage larvae of Teladorsagia spp. [Copyright &y& Elsevier]

Campylobacter jejuni and C. coli were quantified and typed, using multilocus sequence typing (MLST), from fecal samples collected from a mixed cattle and sheep farm during summer. Cattle had a significantly higher prevalence than sheep (21.9% [74/338] and 14.0% [30/214], respectively), but both decreased over time. There were no differences in the average Campylobacter concentrations shed by cattle (600 CFU g-1) and sheep (820 CFU g-1), although sheep did show a significant temporal reduction in the number of Campylobacter organisms shed in their feces. A total of 21 different sequence types (STs) (97.7% C. jejuni, 2.3% C. coli) were isolated from cattle, and 9 different STs were isolated from sheep (40.6% C. jejuni, 59.4% C. coli). The Campylobacter population in cattle was relatively stable, and the frequencies of genotypes isolated showed little temporal variation. However, the composition of subtypes isolated from sheep did show significant temporal differences. The cattle and sheep consistently showed significant differences in their carriage of Campylobacter species, STs, and CCs despite the fact that both were exposed to the same farming environment. Abstract: Polymorphisms in the isotype I β-tubulin gene are important genetic determinants of benzimidazole (BZ) resistance in a number of parasitic nematode species including Teladorsagia circumcincta, a major gastrointestinal nematode of sheep. This study investigates the genetic diversity at this locus in a BZ-resistant isolate of T. circumcincta (MTci5) derived from a sheep farm in the United Kingdom (UK) that was open to animal, and therefore parasite, migration. Pyrosequencing was used to determine the frequency of single nucleotide polymorphisms (SNPs) known to be associated with BZ resistance. This was followed by a combination of single strand conformation polymorphism (SSCP) analysis and nucleotide sequencing to sample allelic diversity in a 276bp fragment immediately surrounding the isotype I β-tubulin F200Y mutation. The genetic diversity at this locus was extremely high, with seven different haplotypes found to contain the resistant F200Y polymorphism in this single resistant isolate. Genotyping by SSCP interfaced with pyrosequencing demonstrated that the P200Y mutation is also present on multiple haplotypes in two other BZ-resistant T. circumcincta isolates from the UK. This contrasts with much lower levels of haplotype diversity in BZ-resistant alleles present in T. circumcincta isolates from French goat farms that are closed to parasite migration. Taken together with our knowledge of T. circumcincta population genetic structure, these results are most consistent with multiple independent origins of resistance and mixing of alleles due to the large amount of livestock movement in the UK. HARD ticks are distributed throughout the UK and there are concerns that their numbers are increasing and that their distribution and competency to transmit disease are changing, due partly to changes in climate and in land management. Ticks can transmit a variety of diseases, such as tickborne fever and louping ill, resulting in significant production losses and welfare problems in sheep flocks. The louping ill virus is also an important cause of high mortality in red grouse, which underpin the economy of many hill and upland areas of the UK. Control of louping ill virus in these birds often depends on control of the disease in sheep, which is becoming increasingly difficult and complex as a result of conflicts between sheep and grouse management and due to a decreasing number of acaricides available for tick control. Ticks can also transmit potentially serious diseases to humans and there is concern that the risks may be increased because of increased public access to tick-infested countryside. The recent emergence of bluetongue virus (Reoviridae: Orbivirus) (BTV) in northern Europe, for the first time in recorded history, has led to an urgent need for methods to control the disease caused by this virus and the midges that spread it. This paper reviews various methods of vector control that have been employed elsewhere and assesses their likely efficacy for controlling vectors of BTV in northern Europe. Methods of controlling Culicoides spp. (Diptera: Ceratopogonidae) have included: (a) application of insecticides and pathogens to habitats where larvae develop; (b) environmental interventions to remove larval breeding sites; (c) controlling adult midges by treating either resting sites, such as animal housing, or host animals with insecticides; (d) housing livestock in screened buildings, and (e) using repellents or host kairomones to lure and kill adult midges. The major vectors of BTV in northern Europe are species from the Culicoides obsoletus (Meigen) and Culicoides pulicaris (L.) groups, for which there are scant data on breeding habits, resting behaviour and host-oriented responses. Consequently, there is little information on which to base a rational strategy for controlling midges or for predicting the likely impact of interventions. However, data extrapolated from the results of vector control operations conducted elsewhere, combined with some assessment of how acceptable or not different methods may be within northern Europe, indicate that the treatment of livestock and animal housing with pyrethroids, the use of midge-proofed stabling for viraemic or high-value animals and the promotion of good farm practice to at least partially eliminate local breeding sites are the best options currently available. Research to assess and improve the efficacy of these methods is required and, in the longer term, efforts should be made to develop better bait systems for monitoring and, possibly, controlling midges. All these studies will need better methods of analysing the ecology and behaviour of midges in the field than are currently in use. The paucity of control options and basic knowledge serve to warn us that we must be better prepared for the possible emergence of other midge-borne diseases, particularly African horse sickness. Abstract: The patterns of insecticide use for the prevention and control of sheep lice and Psoroptes mites, in England, Wales and Scotland, in 2003–2004, were examined using data from a retrospective questionnaire. Overall, 375 of the 966 (39%) farmers who responded said that they treated at least once to prevent louse infestation. For scab, 426 of the 966 (44%) farmers who responded said that they had treated prophylactically to prevent the disease. There were no significant differences between regions in the number of times farmers treated to prevent lice (χ 2 =0.54, P >0.1) or scab (χ 2 =0.16, P >0.1). Among the farmers who applied prophylactic treatment 23% had a subsequent outbreak of lice and 17% had an outbreak of scab. The majority of sheep farmers who used preventative treatment did so in June and July. Where a second treatment was given it was largely in October. For lice, 52% of farmers used pyrethroids to prevent and 70% to treat louse infestation. For scab prophylaxis, most farmers said that they used either the macrocyclic lactones (41%) or organophosphates (36%) as a preventative. For the therapeutic treatment of scab outbreaks, macrocyclic lactones (largely doramectin) were used on 58% of sheep farms. The high national prevalence of sheep scab and pediculosis indicates that current control methods are failing to contain these ectoparasites. The results of this study suggest that a large proportion of insecticide applications take place at times of year when they might expect to be relatively ineffective, and the use of inappropriate compounds for the parasites in question is common. The widespread use of macrocyclic lactones in particular for scab prevention and control is of concern, because of the additional use of these compounds in parasitic worm control and the potential that their overuse will hasten the development of resistance. The prevalence of psoroptic mange (sheep scab), louse infestation and blowfly strike in sheep in Great Britain between March 2003 and February 2004 was investigated by means of a retrospective postal survey, stratified by region. Of the 3530 questionnaires sent out, 1067 were returned completed, a response rate of 30.2 per cent. Overall, 9 per cent of the farmers reported at least one outbreak of scab, 10.7 per cent reported an outbreak of lice and 75 per cent reported at least one case of blowfly strike. A mean of 1.4 per cent of ewes were struck by blowfly and 2.8 per cent of lambs. There were strong regional variations in disease prevalence, with scab and lice infestation being highest in Wales, Scotland and the north of England and blowfly strike highest in the south west of England. As a consequence of large-scale outdoor slaughter of sheep during the 2001 foot and mouth disease (FMD) outbreak in the United Kingdom and the possibility of increased risk for transmission of Echinococcus granulosus between sheep and dogs, a large survey of canine echinococcosis was undertaken in mid-Wales in 2002. An Echinococcus coproantigen-positive rate of 8.1% (94/1,164) was recorded on 22% of farms surveyed, which compares to a rate of 3.4% obtained in the same region in 1993. Positivity rates between FMD-affected properties and unaffected ones did not differ significantly. Significant risk factors for positive results in farm dogs were allowing dogs to roam free and the infrequent dosing (>4-month intervals) of dogs with praziquantel. The entire sheep flock in the UK has been threatened with slaughter if BSE is found in farmed sheep, largely on the grounds that an epidemic of BSE in sheep could be harder to contain than was the case for cattle, and that lamb could present a greater risk to consumers than beef. However, identifying BSE in a sheep is not straightforward, because of its similarities to the related disease, scrapie. Here, we review the likelihood that any UK sheep have BSE, how they might have got it, how a case could be identified and what the Government is doing in terms of surveillance and possible control methods. Because there is a theoretical possibility that the British national sheep flock is infected with bovine spongiform encephalopathy (BSE), we examined the extent of a putative epidemic. An age cohort analysis based on numbers of infected cattle, dose responses of cattle and sheep to BSE, levels of exposure to infected feed, and number of BSE-susceptible sheep in the United Kingdom showed that at the putative epidemic peak in 1990, the number of cases of BSE-infected sheep would have ranged from fewer than 10 to about 1500. The model predicts that fewer than 20 clinical cases of BSE in sheep would be expected in 2001 if maternal transmission occurred at a rate of 10%. Although there are large uncertainties in the parameter estimates, all indications are that current prevalence is low; however, a simple model of flock-to-flock BSE transmission shows that horizontal transmission, if it has occurred, could eventually cause a large epidemic. Background: Transthoracic ultrasonography (TTUS) is currently the only widely used method to diagnose preclinical or subclinical ovine pulmonary adenocarcinoma (OPA) in the live sheep. However, little is known about the test characteristics of TTUS., Methods: One thousand and seventy-four breeding ewes in a flock with evidence of low OPA prevalence underwent TTUS by an experienced operator. Fifty-one sheep were diagnosed with OPA and underwent gross postmortem examination (PME)., Results: Lesions consistent with OPA were found in only 24% (12/51) of the culled ewes. Thirty-five percent (18/51) of culled ewes had gross lesions consistent with other pulmonary disease and 41% (21/51) had no detectable gross lesions on PME. Histopathology and immunohistochemistry confirmed OPA in only the 12 animals identified with OPA lesions from PME., Conclusion: Great caution should be exercised when deciding if TTUS is an appropriate screening test in groups of sheep where OPA prevalence may be anticipated to be low. TTUS is a subjective test and thus individual operator ability will influence the sensitivity and specificity of TTUS for OPA diagnosis while the underlying prevalence influences the eventual positive predictive value., (© 2022 The Authors. Up-to-date data on their prevalence and spatial distribution on farms in Great Britain is important given that prevalence is believed to be increasing as a result of insecticide resistance. Here the prevalence of farms reporting lice, and factors associated with louse presence, were assessed using a retrospective questionnaire. For sheep and cattle farms, 16.1 % and 15.8 % reported lice on their livestock, respectively. Beef farms were more likely to report lice than dairy farms, with a prevalence of 18.0 % and 7.8 %, respectively. For sheep farms, prevalence was highest in Wales (27.7 %) and Scotland (22.4 %). For cattle farms, prevalence was highest in Scotland (27.6 %), Wales (18.5 %) and SW England (18.5 %). For sheep farms, statistical hotspot clusters were identified in Wales, NW England and SW Scotland, with prevalence in these areas ranging from 30.7 to 40.0%. For cattle farms clustering of cases was less evident. Multivariable analysis showed that significant factors associated with lice on sheep farms were larger flock sizes and geographic location (Scotland or Wales). Simple Summary: The bacterium Dichelobacter nodosus is the cause of footrot in sheep: a painful, contagious foot disease. Some sheep may be more susceptible to carrying D. nodosus on their feet, despite showing no signs of disease, and pose a risk to flock health and welfare. This study investigated whether misshapen or damaged feet (poor hoof conformation) were more likely to have D. nodosus present and in greater quantities than feet in good condition. Eighty-five ewes from four flocks were examined three times, on average, across a 12-month period. Poor hoof conformation traits were observed in over 92% of foot observations. Feet with poor sole and heel conformation were more likely to have higher quantities of D. nodosus than those in good conformation. Furthermore, on feet positive for D. nodosus, wall overgrowth contributed towards higher D. nodosus load. We highlight feet with aspects of poor conformation traits to covertly harbour D. nodosus. These feet could transmit D. nodosus to other feet of sheep. Increasing our understanding of D. nodosus infection is crucial in helping farmers improve management practices to reduce footrot. Dichelobacter nodosus is the causal agent of ovine footrot, a contagious disease of welfare and economic concern worldwide. Damaged feet may be subclinical carriers of D. nodosus and covertly spread infection. Accordingly, we evaluated the risk of misshapen and damaged feet on D. nodosus presence and load in four commercial UK sheep flocks. Foot-level observations and swabs (n = 972) were collected from ewes (n = 85) over 12 months. On average, ewes were sampled three times. Feet were inspected for disease and scored (good/poor) for three hoof conformation traits (sole and heel, wall, and wall overgrowth). Swabs were analysed for presence and load of D. nodosus, and mixed models were constructed. Poor hoof conformation traits were present in 92.5% of foot-level observations. Feet with poor sole and heel conformation were more likely to have higher D. nodosus loads (β = 0.19, 95% CI: 0.04–0.35) than those with good conformation. Furthermore, on feet positive for D. nodosus, wall overgrowth was associated with higher D. nodosus loads (β = 0.27, 95% CI: 0.01–0.52). Feet with aspects of poor conformation covertly harbour D. nodosus and are a source of infection. Flock management should be guided by hoof conformation to reduce disease challenge. Their remarkable popularity and further sustainable breeding on grass pastures of British Isles and elsewhere can benefit from genomic applications. Sheep farming has been an important sector of the UK's economy and rural life for many centuries. It is the favored source of wool, meat and milk products. In the era of exponential progress in genomic technologies, we can now address the questions of what is special about UK sheep breed genotypes and how they differ genetically form one another and from other countries. We can reflect how their natural history has been determined at the level of their genetic code and what traces have been left in their genomes because of selection for phenotypic traits. These include adaptability to certain environmental conditions and management, as well as resistance to disease. Application of these advancements in genetics and genomics to study sheep breeds of British domestic selection has begun and will continue in order to facilitate conservation solutions and production improvement. ABSTRACT: The incidence of lameness in sheep in the UK is high in many flocks, often as a permanent and grumbling problem. This article describes a sudden, severe outbreak of lameness in finishing lambs that affected many animals over a few days. Abstract: In Great Britain changes in the proportion of samples submitted to diagnostic laboratories, for which no diagnosis is confirmed, are monitored as a possible indicator of new or emerging diseases. Herewith, perceived reasons for failing to reach a diagnosis were provided for the year 2008. No diagnosis could be confirmed in 39% of these; however, if those subjected to limited testing were excluded, then that proportion was 20%. Most frequently recorded reasons that no diagnosis was confirmed were “incomplete sample range submitted” (8% of total samples submitted), “full diagnostic package not requested” (6%), “post-mortem material required but unavailable” (4%) and “inconclusive test results” (2%). No reason could be identified in 8% of samples submitted. Selection of these samples for further analysis may enhance surveillance for emerging/re-emerging diseases. [Copyright &y& Elsevier]

The article describes a case of atypical scrapie in the Neuropathogenesis Unit (NPU) Cheviot flock, which has also endemic natural scrapie in Great Britain. According to the authors, immunohistological analysis of brain sections with the antibody BG4 showed pronounced immunostaining in the granular and molecular layer of the cerebellum. They add that the overall pattern of immunostaining was a widespread, extracellular and slightly diffuse puncture decoration.
