School district administrators must address structural racism and inequitable access to advanced learning opportunities in their school districts. District administrators in one large district in the northwestern part of the United States sought research-based advice by asking the authors to provide the answer to the "million-dollar" question, " What is the most effective and equitable way to deliver services to advanced learners? " This paper shares the response provided to the school district and discusses the complexity of the question. Based on a systematic literature review of best practices in the field of gifted education, we share our findings in three categories: (1) administrative structures, (2) pedagogy of gifted education, and (3) social and emotional considerations for advanced learners. This mixed methods study examines the relationship between different components and characteristics of individualized lesson plans and children's social-emotional outcomes to develop a profile of effective planning. Sixty-one individualized SEL lesson plans written and implemented by 20 experienced early childhood educators from a large city in the northeastern United States are examined to understand the types and difficulty level of behavioral challenges teachers addressed, the types and specificity of SEL skills taught and the specificity of teaching strategies implemented. By quantizing qualitative data through a process of descriptive, pattern and then magnitude coding, the study then uses correlational analyses to examine the relationship between successful child outcomes following instruction and the rated components of the lesson plans. Findings reveal that lesson plan alignment is most strongly related to successful social-emotional outcomes (rho coefficient.69), followed by teaching strategy specificity (rho.51). The difficulty level of the children's behavior being addressed by the lesson plans and the teachers' ability to list SEL skills with specificity as learning objectives were not significantly related to later student success. Assigning individualized SEL lesson planning to in-service teachers resulted in at least partially successful child outcomes 59% of the time, showcasing how teachers need more systematic practice and training to refine this important skill. To explore indicators that predict whether patients with extremity pain have a spinal or extremity source of pain. The data were from a prospective cohort study (n = 369). Potential indicators were gathered from a typical Mechanical Diagnosis and Therapy (MDT) history and examination. A stepwise logistic regression with a backward elimination was performed to determine which indicators predict classification into spinal or extremity source groups. A Receiver Operating Characteristic (ROC) curve was constructed to examine the number of significant indicators that could predict group classification. Five indicators were identified to predict group classification. Classification into the spinal group was associated with the presence of paresthesia [odds ratio (OR) 1.984], change in symptoms with sitting/neck or trunk flexion/turning neck/when still (OR 2.642), change in symptoms with posture change (OR 3.956), restrictions in spinal movements (OR 2.633), and no restrictions in extremity movements (OR 2.241). The optimal number of indicators for classification was two (sensitivity = 0.638, specificity = 0.807). This study provides guidance on clinical indicators that predict the source of symptoms for isolated extremity pain. The clinical indicators will allow clinicians to supplement their decision-making process in regard to spinal and extremity differentiation so as to appropriately target their examinations and interventions. This study examined self-reports and instructional videos provided by 25 preservice teachers to demonstrate differentiated instruction in meeting the needs of culturally and linguistically diverse students (CLDS) in the United States. Self-reported journals were thematically analyzed and compared with corresponding instructional videos. The results revealed a mismatch between perceptions and practices of differentiation. Clearly, additional efforts must be taken to prepare preservice teachers to differentiate their instruction for CLDS in the areas of content, process, product, and environment. The beer industry in the United States is in a period of dramatic transformation. Major breweries are acquiring much smaller craft breweries in an attempt to purchase growth, but it is not clear whether these acquisitions are economically viable. In this paper, we study the impact of craft brewery acquisitions on retail beer prices, and firm profitability in a dynamic, Markov‐perfect equilibrium pricing framework. We find that the estimated impact of mergers, or buyouts, is critically dependent upon estimates of the extent of state‐dependence in demand and is, in fact, negatively correlated with the initial shock to demand. That is, if the demand shock is positive, the effect of a buyout will be under‐estimated by not accounting for state‐dependence in demand, while it is over‐estimated if the demand shock is negative. This finding is intuitive as the static model will not properly account for the long‐term positive effects of a demand shock that is initially positive, or the long‐term negative effects that are initially negative. Objective: Building on the original taxonomy of hospital-based health systems from 20 years ago, we develop a new taxonomy to inform emerging public policy and practice developments.Data Sources: The 2016 American Hospital Association's (AHA) Annual Survey; the 2016 IQVIA Healthcare Organizations and Systems (HCOS) database; and the 2017-2018 National Survey of Healthcare Organizations and Systems (NSHOS).Study Design: Cluster analysis of the 2016 AHA Annual Survey data to derive measures of differentiation, centralization, and integration to create categories or types of hospital-based health systems.Data Collection: Principal components factor analysis with varimax rotation generating the factors used in the cluster algorithms.Principal Findings: Among the four cluster types, 54% (N = 202) of systems are decentralized (-0.35) and relatively less differentiated (-0.37); 23% of systems (N = 85) are highly differentiated (1.28) but relatively decentralized (-0.29); 15% (N = 57) are highly centralized (2.04) and highly differentiated (0.65); and approximately 9 percent (N = 33) are least differentiated (-1.35) and most decentralized (-0.64). Despite differences in calculation, the Highly Centralized, Highly Differentiated System Cluster and the Undifferentiated, Decentralized System Cluster were similar to those identified 20 years ago. The other two system clusters contained similarities as well as differences from those 20 years ago. Overall, 82 percent of the systems remain relatively decentralized suggesting they operate largely as holding companies allowing autonomy to individual hospitals operating within the system.Conclusions: The new taxonomy of hospital-based health systems bears similarities as well as differences from 20 years ago. Brands and word of mouth (WOM) are cornerstones of the marketing field, and yet their relationship has received relatively little attention. This study aims to enhance understanding of brand characteristics as antecedents of WOM by executing a comprehensive empirical analysis. For this purpose, the authors constructed a unique data set on online and offline WOM and characteristics for more than 600 of the most talked-about U.S. brands. To guide this empirical analysis, they present a theoretical framework arguing that consumers spread WOM on brands as a result of social, emotional, and functional drivers. Using these drivers, the authors identify a set of 13 brand characteristics that stimulate WOM, including three (level of differentiation, excitement, and complexity) that have not been studied to date as WOM antecedents. The authors find that whereas the social and functional drivers are the most important for online WOM, the emotional driver is the most important for offline WOM. This article focuses on research findings from multiple evaluation studies on the teacher use of differentiated practices for gifted learners that have implications for best practice in gifted education internationally. Research questions centered on what specific differentiation practices were employed with gifted learners and distinctions found between levels of teaching and content areas. Using the Classroom Observation Scale-Revised (COS-R) with 329 teachers in six different school districts in four states in the Eastern United States, we report data on differentiation practices that suggest that teachers of the gifted as well as regular classroom teachers underutilized these practices, did not match them to instructional purposes, and failed to implement them as effectively as possible to have an impact on gifted student learning. Moreover, the lack of consistent use of flexible grouping practices to support instructional goals was apparent in the majority of regular classroom contexts. Findings also suggested that teachers who were using differentiated practices were using them at least somewhat effectively, suggesting that some teachers were successful in implementing best-practice strategies. The results of these multiple studies also revealed that middle school classrooms were less effective in the use of differentiation for the gifted than either elementary or high schools. A comparison of results by subject area showed that math classrooms were the most successful in implementing differentiation practices for the gifted. Overall, the study suggests that differentiated practices are underutilized in the majority of gifted program sites studied. Implications from the study suggest that more careful attention needs to be paid to the use of specific differentiation practices, found in the research literature and taught in model university gifted education training programs. Purpose: Intimate partner violence (IPV) is prevalent in the U.S., and has many physical and mental health implications. While several studies have explored the impact of IPV experience on individuals, less is known about which factors are predictive of IPV victimization. Method: The current study examined the extent to which substance use, self-efficacy, and differentiation each predicted experiences of IPV among women seeking treatment for a substance use disorder using a multinomial logistic regression analysis. Results: Results showed that women who reported lower levels of differentiation were significantly more likely to experience IPV. However, no other significant IPV predictors were found. This study explored the role of sibling modeling and differentiation in young adults' beliefs about marriage and expectation of marriage in reference to their perception of their closest aged sibling's marital centrality. Siblings play an important role in socialization, yet their role in young adulthood, and in relation to attitudes about marriage, has received limited attention. Marriage is an important aspect of development for young adults; therefore, this study specifically examined the role of sibling modeling and differentiation in young adults' formation of expectation of marriage and marital centrality. Data came from 1,258 unmarried young adults (age 18–29 years) from across the U.S. (M = 25.02; SD = 2.60; 47% female). Models for marital centrality were tested using hierarchical ordinary least squares regression, and models for the expectation of marriage were examined using binary logistic regression. Findings revealed that siblings' beliefs and expectations to marry were more closely related in conditions of high modeling. For those with only one sibling, greater differentiation was linked to less similarity between siblings' marital centrality. The level of diversity present in a national higher education system plays an important role in enabling the system to respond to a variety of demands from society. Within the US context, diversity is often hailed as a strength although research has found a consistent decline in institutional diversity for the last 40 years. This article uses data from 1989 to 2014 to compare the level of diversity present across American higher education. The article draws on institutional theory to understand how institutions change in respond to environmental pressures impacting institutional diversity. The findings demonstrate that institutional diversity has continued to decrease and demonstrates how institutional decisions impact overall diversity present in the system. To conclude, the article discusses the causes and potential concerns for national systems that experience decreased institutional diversity. An improved understanding of the current state of institutional diversity provides useful measures for evaluating national higher education systems and raises critical questions for the mission and activities of higher education institutions themselves. Specialty hospitals tend to negotiate higher commercial insurance payments, even for relatively routine procedures with comparable clinical quality across hospital types. How specialty hospitals can maintain such a price premium remains an open question. In this paper, we examine a potential (horizontal) differentiation effect in which patients perceive specialty hospitals as sufficiently distinct from other hospitals, so that specialty hospitals effectively compete in a separate market from general acute care hospitals. We estimate this effect in the context of routine pediatric procedures offered by both specialty children's hospitals as well as general acute care hospitals, and we find strong empirical evidence of a differentiation effect in which specialty children's hospitals appear largely immune to competitive forces from non-children's hospitals., (© 2023 John Wiley & Sons Ltd.)

Ovarian cancer is the most fatal gynecological cancer in the USA and the fifth most common cancer-related cause of death in women. Inflammation has been shown to play many roles in ovarian cancer tumor growth, with the proinflammatory cytokine interleukin-6 (IL-6) having been established as a key immunoregulatory cytokine. Ovarian cancer cells continuously secrete cytokines that promote tumorigenicity in both autocrine and paracrine fashions while also receiving signals from the tumor microenvironment (TME). The TME contains many cells including leukocytes and fibroblasts, which respond to proinflammatory cytokines and secrete their own cytokines, which can produce many effects including promotion of chemoresistance, resistance to apoptosis, invasion, angiogenesis by way of overexpression of vascular endothelial growth factor, and promotion of metastatic growth at distant sites. IL-6 and its proinflammatory family members, including oncostatin M, have been found to directly stimulate enhanced invasion of cancer cells through basement membrane degradation caused by the overexpression of matrix metalloproteinases, stimulate promotion of cell cycle, enhance resistance to chemotherapy, and cause epithelial-to-mesenchymal transition (EMT). IL-6 has been shown to activate signaling pathways that lead to tumor proliferation, the most studied of which being the Janus kinase (JAK) and STAT3 pathway. IL-6-induced JAK/STAT activation leads to constitutive activation of STAT3, which has been correlated with enhanced tumor cell growth and resistance to chemotherapy. IL-6 has also been shown to act as a trigger of the EMT, the hypothesized first step in the metastatic cascade. Understanding the important role of IL-6 and its family members' effects on the pathogenesis of ovarian cancer tumor growth and metastasis may lead to more novel treatments, detection methods, and improvement of overall clinical outcomes. Adipose-derived stem cells (ADSCs) are isolated from abundant adipose tissue and have the capacity to differentiate into multiple cell lineages. ADSCs have raised big interest in therapeutic applications in regenerative medicine and demonstrated to fulfill the criteria for a successful cell therapy. There are several methods for isolation of ADSCs from adipose tissue and cryopreservation of ADSCs. Here, novel methods for the isolation and cryopreservation of ADSCs are presented and focused. Microscopic pieces of adipose tissue were placed on transwell inserts, and the ADSCs were induced to migrate to the lower wells for 1 week. We compared the properties of our ADSCs with those isolated by enzymatic digestion and enzyme-free method of culture plate, and our ADSCs were found to be more stable and healthier. In addition, we proposed a novel cryoprotectant solution (FNCP) containing pectin and L-alanine, which was compared with standard cryoprotectant solution. Overall, our methods proved more useful for ADSCs isolation than other methods and did not require consideration of "minimal manipulation" by the U.S. Food and Drug Administration (FDA). Furthermore, our FNCP did not contain dimethyl sulfoxide and fetal bovine serum, therefore stable storage is possible in xeno-free and animal-free cryopreservation solutions.

Billions of US dollars are invested every year by the pharmaceutical industry in drug development, with the aim of introducing new drugs that are effective and have minimal side effects. Thirty percent of in-pipeline drugs are excluded in an early phase of preclinical and clinical screening owing to cardiovascular safety concerns, and several lead molecules that pass the early safety screening make it to market but are later withdrawn owing to severe cardiac side effects. Although the current drug safety screening methodologies can identify some cardiotoxic drug candidates, they cannot accurately represent the human heart in many aspects, including genomics, transcriptomics, and patient- or population-specific cardiotoxicity. Despite some limitations, human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) are a powerful and evolving technology that has been shown to recapitulate many attributes of human cardiomyocytes and their drug responses. Despite their prevalence and power in markets throughout the United States, local multihospital systems (LMSs)-also referred to as hospital-based "clusters"-remain an understudied organizational form, with studies instead primarily focusing either upon individual hospitals or viewing hospital systems collectively without distinguishing the local "sub-systems" that comprise larger regional or national hospital chains. To better understand these organizational forms, we develop a taxonomy specifically devoted to LMSs, applying taxonomic analysis methods to a sample of LMSs in six U.S. states while accounting for LMSs' geographic arrangements and non-hospital-based service locations. Our analysis identifies five distinct LMS categories, with forms clearly distinguished according to their varying degrees of differentiation and integration. The study's results accentuate the importance of accounting for hospital systems' activities and arrangements in local markets-including their non-hospital-based sites-and highlight differences in systems' achievement of integration and coordination across services and locations, providing considerations in light of U.S. health system reform as well as international patterns of regional system formation. Black dot disease of potato, caused by Colletotrichum coccodes (Wallr.) Hughes, is widely distributed in the United States. However, little is known regarding the population biology of this fungus. A total of 370 single-spore isolates of C. coccodes were collected from naturally infected potato plants in nine states and analyzed using amplified fragment length polymorphism (AFLP) markers and three primer pairs, yielding 190 polymorphic bands with 90.7 % polymorphism. The isolates were assigned to four vegetative compatibility groups (AFLP/VCG): AFLP/VCG1, AFLP/VCG2, AFLP/VCG4/5, and AFLP/VCG6/7. No isolates tested belonged to AFLP/VCG3. The United States C. coccodes population structure was confirmed with a high differentiation value ( G = 0.30) among VCGs. AFLP/VCG2 was the dominant group in the population ( n = 262) and was the most frequent AFLP/VCG among states, fields, farms, and plants. However, in several instances, more than one AFLP/VCG was isolated from the same plant, field, farm, and state, indicating variability within the C. coccodes population in the United States. A geographic pattern was found for isolates originating from Texas, Montana, North Dakota, and Wisconsin. Diversity within states accounted for 73 % of the total genetic diversity, and among populations accounted for 27 %. These results suggest that several AFLP/VCGs are widely distributed in the United States and that they form a single large population of C. coccodes. [ABSTRACT FROM AUTHOR]

In this small essay, I will reflect on Michael Bérubé and Jennifer Ruth's arguments on the decline of educational professionalism in the United States. The purpose is to consider this loss of professionalism, and I will consider it in light of the arts and humanities in the Danish educational debate. Two reflections are presented: first, the customer relation is reversed in a Scandinavian context where students are politically demanded products rather than informed customers. This implies that universities' output serve political agendas. Second, I suggest that the Scandinavian conception of equality in welfare might entail an inexpedient side-effect when it comes to education. Equality becomes alignment rather than educative edification of individual and, in consequence, communal autonomy. This jeopardises the historically fruitful role of the Scandinavian university – and ultimately caters to a shift from edification to serving interests that are beyond the professional heart of arts and humanities in higher education. In this paper, we study spatial competition in the U.S. audit market while accounting for its two-tiered nature. We provide evidence on the differential impact that market share distances within and between the players in the large and small audit markets have on competition. We find that the market share distance from small audit firm competitors has a greater effect on the Big 4's audit fees than distances from other Big 4 competitors. This finding suggests that small audit firms play a significant part in the competitive landscape in local markets. Further, we find that audit fees are increasing with the distance between a small audit firm and its closest competing small audit firm while audit fees are decreasing with the distance between a small audit firm and its closest competing large audit firm. This suggests that while obtaining separation in market space from competing small audit firms reduces competitive pressure from other small audit firms, as a small audit firm gets closer to the market space of a large audit firm it is perceived as being more like the larger audit firm and is able to obtain a fee premium like that attained by the larger audit firms. This case study of 21c Museum Hotels introduces a new concept in hotel branding, that of combining three strong elements not formerly developed by boutique hotels: a first class restaurant, an art museum with large gallery spaces, and a deluxe hotel. The case discusses how the owners have branded this unconventional business, their differentiation strategies, their engagement with local communities, and how they have used a wide variety of marketing communication methods to engage potential customers. A comparison of four elements that constitute commoditization as viewed by U.S. hotel industry executives and by customers from a national sample found that the two groups ascribe different weights to the four factors in the commoditization model. In a test of these four factors, the seventy industry executives in the survey thought that product homogeneity contributed the most to commoditization, followed by industry stability, switching costs, and price sensitivity. However, from the customers’ point of view, price sensitivity was far and away the key element of commoditization, with product homogeneity having a much smaller effect. Stability and switching costs were not considered as part of the consumers’ perception of commoditization. Although the study is based on the views of hotel industry executives and customers, it makes no attempt to gauge the level of industry commoditization, because its goal is to establish the nature of commoditization as framed in the hotel industry. In this investigation, we examined the effects of a differentiated reading approach on fourth grade students’ reading comprehension and attitudes toward reading. Eight Title I schools within one urban district were randomly assigned to treatment (Schoolwide Enrichment Model–Reading [SEM-R]) or control (district reading curriculum) conditions. Treatment teachers implemented SEM-R as a supplement to the district curriculum for 1 academic year, whereas control teachers used the district curriculum. Based on multilevel analyses of students’ posttest reading comprehension scores on the Iowa Test of Basic Skills (n = 358) and attitudes toward reading as measured by the Elementary Reading Attitudes Survey (n = 429), no statistically significant differences in students’ attitudes toward reading were found, but SEM-R students had significantly higher scores on the comprehension posttest compared with the students in the control schools. Porcine epidemic diarrhea virus (PEDV) has caused significant economic losses in the US swine industry since May 2013. A new variant strain of PEDV emerged in the US in the late December, 2013. This variant strain of PEDV differs from the virulent strain of PEDV currently circulating in the US in 1170 nt of the 5'end of the S1 domain in the spike gene. Importantly, the variant PEDV caused significantly less mortality in piglets than the virulent PEDV, based on clinical observations. This suggests it may be a potential vaccine candidate for PED. Variant PEDV has been detected in samples from multiple states by our laboratory as well as other laboratories in the US. It is critical to detect and differentiate variant PEDV from the virulent PEDV during outbreaks to enhance control and to prevent PED associated disease. In this study, the development and validation of a duplex real-time RT-PCR assay for detection and differentiation of the variant and the virulent strains of PEDV currently circulating in the US was reported. The United States is a world leader in poultry production, which is the reason why achieving better performance and muscle growth each year is a necessity. Reducing accretion of adipose tissue is another important factor for poultry producers because this allows more nutrients to be directed toward muscle growth, but the effect of embryonic adipose growth on posthatch development has not been fully understood. The purpose of this study was to investigate the total DNA mass, morphological characteristics, differentiation markers, and triglyceride breakdown factors of embryonic adipose tissue, and their relation to hyperplastic and hypertrophic growth within layers (Leghorn) and meat-type chickens (broilers). After embryonic day (E) 12, broiler weight was significantly higher than Leghorn, and this trend continued throughout the rest of incubation and posthatch ( P < 0.05). Neck and leg fat pad weights between the 2 breeds did not differ at most of the time points. A remarkable increase in total DNA mass was observed between E12 and E14 in both Leghorn and broilers (P < 0.05), indicating a high potential for hyperplastic growth during this time. Histological analysis revealed clusters of preadipocytes at E12; however, the majority of these cells differentiated by E14 and continued to grow until the time of hatch. The adipocyte sizes between both breeds did not generally differ, even though broilers are known to have larger adipocytes posthatch. Fatty acid-binding protein 4 expression levels in Leghorn and broilers continued to rise with each time point, which paralleled the expansion of mature adipocytes. Adipose triglyceride lipase was highly expressed at E20 and d 1 posthatch to mobilize triglyceride degradation for energy during hatching. Thus, embryonic chicken adipose tissue was found to develop by hyperplastic mechanisms followed by hypertrophy. This case study examines two 10th-grade US History teachers who collaborated to create and implement an integrated, thematic eight-week unit on war with an emphasis differentiated instruction. Drawing on the National Council for the Social Studies (2010) framework for powerful and purposeful social studies instruction, the case study uses multiple sources of data, including 38 lesson observations, analyses of the teachers' lesson plans and student work, and interviews of teachers. Initially, the teachers were successful at engaging students in simulations, small-group discussions, and higher-order thinking. As the unit progressed, however, the teachers reverted to transmission-style teaching with an emphasis on breadth over depth. The technique of nuclear transplantation - popularly known as cloning - has been integrated into several different histories of twentieth century biology. Historians and science scholars have situated nuclear transplantation within narratives of scientific practice, biotechnology, bioethics, biomedicine, and changing views of life. However, nuclear transplantation has never been the focus of analysis. In this article, I examine the development of nuclear transplantation techniques, focusing on the people, motivations, and institutions associated with the first successful nuclear transfer in metazoans in 1952. The conflict between embryologists and geneticists over the mechanisms of differentiation motivated Robert Briggs to pursue nuclear transplantation experiments as a way to resolve the debate. Briggs worked at the Lankenau Hospital Research Institute, a research facility devoted to the study of cancer. The goal of understanding cancer would play a role in the development of the technique, and the story of nuclear transplantation sheds light on the role that biomedical contexts play in biological research in the second half of the twentieth century. Sampling selectivity in acoustic-trawl surveys of Pacific sardine (Sardinops sagax) biomass and length distribution. – ICES Journal of Marine Science, 70: .To annually assess the northern stock of Pacific sardine (Sardinops sagax) in the California Current and set harvest quotas for the US fishery, managers have used an age-structured stock synthesis model fitted with results from acoustic-trawl (ATM), daily-egg-production, and aerial-photogrammetric survey methods, fishery landing and individual-length data, and many assumed or empirically derived parameters. In these assessments, sardine landed at ports spanning from Ensenada, México to Vancouver Island, Canada were assumed to be solely from the northern stock. It was also assumed that the ATM estimates of sardine biomass were negligibly biased for the sizes of fish sampled by the survey trawls (i.e., catchability q = 1 for sardine standard length (SL) values greater than ∼17 cm). Due to these catchability and length-selectivity assumptions, the ATM- and assessment-estimated abundances are mostly similar for larger sardine. However, the assessment estimates include large abundances of small sardine (SL values less than ∼15 cm) that are not represented in either the ATM-survey results or the fishery landings, and generally did not recruit to the migrating northern stock sampled by the ATM surveys. We considered four explanations for this disparity: (i) the ATM length-selectivity assumption is correct; (ii) the non-recruiting small fish may comprise a smaller portion of the stock than indicated by the assessments; (iii) during years of low recruitment success, those size classes may be virtually completely fished by the Ensenada and San Pedro fisheries; or (iv) they may belong to the southern sardine stock. Creole sheep represent a strategic genetic resource for populations living in marginal areas under financial restrictions on the American continent. Six Colombian sheep breeds (two wool (BCL-Boyacá and NCL-Nariño, 12 and 14 samples) and four hair (OPCE-Ethiopian, 54 samples; OPCS-Sudan, 74 samples; OPCP-Pelibeuy, 59 samples; OPCW-Wayúu, 24 samples) were genotyped using the Illumina Ovine SNP50 BeadChip. Data was also included from international 44 breeds from International Sheep Genomics Consortium (ISGC) and from data published in previous a previous work on the Caribbean and African breeds. Although geographically separated, wool (NCL, BCL) and hair types (OPCE, OPCS, OPCW) presented little genetic differentiation (FST 0.05) at a global level but several groups of animals separated suggesting local clustering due to geographical isolation. The OPCP underwent a recent crossing with Mexican Pelibuey, explaining its differentiation. Findings in this work such as the proximity to West African Djallonké (WAD) and Barbados Black Belly (BBB), suggest different introductions of African type animals from the Caribbean region on a pre-existing genetic basis formed by animals deriving from the first importations coming from Europe in colonial times. As expected, Colombian wool breeds showed, in particular in Admixture software results, a greater genomic component in common with European breeds and in particular with Iberian ones (Churra). This study provides a basis for future research into the genetic diversity within and between the Colombian sheep breeds analysed, and scientific data for policy decisions on Farm Animal Genetic Resources (FAnGR).

Background: Hair follicle stem cells (HFSCs) are derived from the bulge region and are important autologous stem cell sources. Bibliometric is a statistical method that quantitatively analyses the research papers concerned about one special topic. This study aims to estimate the research status and trends of HFSCs worldwide by bibliometric analyses., Methods: Data were obtained from the Web of Science by searching keywords related to HFSCs. Publication distributions stratified by countries/regions, institutions, journals, and authors were systematically assessed. The frequency of keywords was assessed, and bibliometric mapping was employed to describe the development of HFSC research., Results: A total of 458 publications that met our screening criteria were included in this study, consisting of 423 (92.4%) articles and 35 (7.6%) reviews. The United States of America (USA) ranked first in the number of publications at 146 (31.9%), followed by China at 130 (28.4%), which is consistent with the rank of the H-index. Author keywords were classified into three clusters, namely, basic study, applied study, and biomarker; average publication time of keywords in applied study cluster is later than basic study cluster. The keywords "bulge", "nestin", and "skin" are the top three most frequent keywords in basic studies; "differentiation", "proliferation", and "alopecia" are the top three most frequent keywords in applied studies. With respect to the latest research hotspots, "apoptosis" and "tissue engineering" are relatively new keywords., Conclusions: The USA and China were the most productive countries for research on HFSCs. The focus of keywords gradually shifted from basic study to applied study. Research on the differentiation/proliferation of HFSCs and the role of HFSCs in alopecia have been recent research focuses. Apoptosis and tissue engineering are recommended as promising research hotspots. Our study provides profound insights into the research history, current status, and future trend of HFSCs., (© 2022. CS055 (Chidamide/HBI-8000) is a novel benzamide-typeHDACi (histone deacetylase inhibitor), which has entered Phase I clinical trials in the U.S. and Phase II/III in China. In the present study, we investigated the effects of CS055 on proliferation, differentiation and apoptosis in human leukaemia cell lines and primary myeloid leukaemia cells. The results showed that at low concentrations (<1 μM), CS055 induced G1 arrest. At moderate concentrations (0.5 μM-2 μM), CS055 induced differentiation, as determined by the increased expression of the myeloid differentiation marker CD11b. At relatively high concentrations (2 μM-4 μM), CS055 potently induced caspasedependent apoptosis. Co-treatment with the ROS (reactive oxygen species) scavengers N-acetyl-L-cysteine or Tiron blocked CS055- induced cell differentiation and apoptosis, suggesting an essential role for ROS in these effects. Cytochrome c release and ROSmediated mitochondrial dysfunction are involved in CS055- induced apoptosis of leukaemia. In addition to cell lines, CS055 also exhibits therapeutic effects in human primary leukaemia cells.Moreover, daily oral CS055 treatment of nude mice bearing HL60 cell xenografts suppressed tumour growth, induced tumour cell apoptosis and prolonged the survival of tumour-bearing mice. Purpose: Colorectal cancer (CRC) is a major cause of death in the western world and a leading cause of cancer-related death. It is one of the most common human malignancies with >300,000 cases both in the United States and in the European Union each year. The present study was conducted to assess differences in various variables of CRC, such as location of the tumor, differentiation, Dukes classification, 5-year survival and possible changes in these patterns during the examined period. Methods: We collected data on 2000 patients with colorectal cancer, diagnosed and treated from 1960 to 2008 in 1st Propedeutic Surgical Clinic of Aristotle's University, Thessaloniki. Results: Of 2000 cases reviewed, cancer was almost equal presented to both sexes, for all groups. Rectum was the most common tumor location in all analyzed groups (40.1%). The most common tumor differentiation was the moderate one (68.5%). Concerning tumor staging, Dukes' B tumors were most common (42.5%), and the cancer-related 5-year survival was increased by the time from 42 to 71%. Conclusion: In the past 20 years, considerable improvements have been made in colorectal cancer therapy, and patients had received more sophisticated and multidisciplinary treatments, resulting in a better 5-year survival rate. This article studies the nature of differentiation in the market for medical tourism facilitators in the USA. Medical tourism facilitators in the USA resemble a monopolistically competitive industry. They choose to differentiate their services in several ways, including by the scope of countries and hospitals which they use, the scope of the treatments in which they specialize, the extent of involvement of medical professionals in the company, and the kinds of ancillary services they offer. The authors perform an exploratory study of the differentiation among 46 such firms. Using principal-components analysis, they detect three components: an emphasis on providing a broad selection of destinations, an emphasis on physician’s concerns, and an emphasis on the travel aspects of medical tourism. While much stratification research has focused on understanding the patterns and consequences of differentiation, previous studies have not considered similarly important variation in students' trajectories through higher education, and particularly their participation in the labor market. Results from the National Longitudinal Survey of Youth of 1997 (NLSY97) indicate that degree completion in a differentiated system of higher education is related to students' employment patterns. Students who begin their educational journeys in community colleges as well as students from less advantaged family backgrounds are more likely to dedicate longer hours to paid employment, which has negative consequences for degree attainment. Employment patterns contribute to gaps in degree completion among students from different family backgrounds and to a lesser extent to inequality in degree completion between students beginning postsecondary education in community colleges vs. 4-year institutions. A more complex set of patterns is revealed when examining the relationship between employment, family background, and degree attainment across different institutional types and educational credentials. These findings highlight the importance of developing a more comprehensive understanding of inequality in educational attainment by considering the relationship between differentiation and work. In this article we explore the role of differentiation of self in facilitating forgiveness in the context of couple and family relationships. Differentiation is defined from the Bowen perspective as the ability to connect with others without being excessively emotionally reactive to the ebb and flow inherent in all significant relationships (being able to connect to others yet also being able to self-regulate). Forgiveness is described as the releasing of an emotional injury via a complex psychological and relational process that is less an act of will than a discovery or possibility through understanding and empathy. Differentiation of self is related to emotional intelligence and empathy The developmental and relational benefits of such are illustrated and discussed. A rationale for viewing differentiation and forgiveness in a contextual, historical, and relational attachment paradigm is suggested. This study assesses trends in leisure time by life course and family characteristics in Canada, the United Kingdom, and the United States. Using national time-use data collected since the mid-1960s, it is hypothesized that important family characteristics are responsible for substantial variation in leisure time that is not recognized in accounts of leisure time among working adults or within national populations. An important finding indicates that leisure is either stable or has increased somewhat in the three Western democracies studied. Social characteristics, family, and employment contexts account for considerable variation in leisure time. Findings demonstrate an increased disadvantage in leisure time among parents of young children after controlling for social background characteristics. Analyses demonstrate the need to qualify accounts of over-work and the double-burden on available leisure time. The perspective of structural constraints integrates neoclassical economic approaches to marginal utility maximization and sociological approaches to gender, and clarifies the impact of changed economic relations that produce less gendered though more differentiated patterns of leisure when parenting. Abstract: Obesity is a public health crisis in the United States. Targeting preadipocyte to adipocyte conversion may be an effective approach to regulate adipose mass. Using differential screening we identified Fstl1, a secreted glycoprotein with roles in immunomodulation, cell growth, cardioprotection, and vascularization, as a “preadipokine”. Fstl1 is highly expressed in 3T3-L1 preadipocytes and dramatically downregulated early in their differentiation to adipocytes. Northern blot analysis of murine tissues reveals white adipose tissue (WAT), lung and heart as primary sites of Fstl1 transcript expression. In WAT, Fstl1 transcript is restricted to the preadipocyte-containing stromal-vascular cell population. Time course studies in multiple adipogenesis models reveal downregulation of Fstl1 is a hallmark of white and brown adipocyte conversion. By Western blot, we show culture media of 3T3-L1 preadipocytes contains high levels of Fstl1 protein that rapidly decline in adipocyte conversion. Moreover, we observe a correlation between preadipocyte phenotype and Fstl1 expression in that TNFα-mediated de-differentiation of 3T3-L1 adipocytes is accompanied by re-expression of Fstl1 transcript and protein. Treatment of 3T3-L1 preadipocytes with a panel of 18 hormones and other agents revealed the demethylating agent 5-aza-cytidine decreases Fstl1 transcript and protein levels by ∼90%. Furthermore, of 10 additional preadipocyte-expressed genes analyzed we find Pref-1, Col1A1, Sca-1/Ly6a, Lox and Thbs2, are also downregulated by 5-aza-cytidine. Using luciferase reporter constructs containing 791 or 3922bp of the Fstl1 5′ flanking region, we determine negative transcriptional regulation by Kruppel-like factor 15. Together, our data suggest downregulation of Fstl1 expression may be an important feature of preadipocyte to adipocyte conversion. [Copyright &y& Elsevier]

The article presents a study on the capability of the prion protein (PrP) to separate the cardiomyogenic and noncardiomyogenic cellular fractions in the U.S. The study intends to show the efficiency of the cellular PrP to serve as a surface marker to isolate nascent cardiomyocytes. The flow cytometry was used to analyze the embryonic stem cells to be able to detect the surface expression of PrP.

Alcohol affects approximately 1% (40,000) of new born infants each year and is the main preventable cause of mental retardation in the US. Ethanol alters cell signaling and promotes apoptosis and differentiation. Heparin-binding epidermal growth factor-like growth factor (HB-EGF), a member of the EGF family of growth factors, has been reported to prevent apoptosis and differentiation. We treated human embryonic stem cells (hESCs) with ethanol (20 mM) to reflect casual drinking, with and without HB-EGF to measure its ability to prevent ethanol-induced apoptosis and differentiation. Apoptosis was measured by DNA fragmentation (terminal dUTP nick-end labeling assays) and activated caspase-3, while differentiation was accessed by SSEA-1 and OCT-3/4; western blotting assessed MAPK signaling. HB-EGF reduced SSEA-1 and elevated OCT-3/4, while reducing the amount of activated caspase-3 and DNA fragmentation. Western blot analysis showed HB-EGF prevents ethanol from altering MAPK phosphorylation. This data suggests that ethanol-induced apoptosis was reduced by HB-EGF, while hESC pluripotency was maintained. [ABSTRACT FROM AUTHOR]

Existing representations of cognitive ability structure are exclusively based on linear patterns of interrelations. However, a number of developmental and cognitive theories predict that abilities are differentially related across ages (age differentiation-dedifferentiation) and across levels of functioning (ability differentiation). Nonlinear factor analytic models were applied to multivariate cognitive ability data from 6,273 individuals, ages 4 to 101 years, who were selected to be nationally representative of the U.S. population. Results consistently supported ability differentiation but were less clear with respect to age differentiation-dedifferentiation. Little evidence for age modification of ability differentiation was found. These findings are particularly informative about the nature of individual differences in cognition and about the developmental course of cognitive ability level and structure. Summary: Abnormalities of kidney and urinary tract development are the most common cause of end-stage kidney failure in childhood in the United States. Over the past 20 years, the advent of mutant and transgenic mice and the manipulation of gene expression in other animal models has resulted in major advances in identification of the cellular and molecular mechanisms that direct kidney morphogenesis, providing insights into the pathophysiology of renal and urologic anomalies. This review focuses on the molecular mechanisms that define kidney progenitor cell populations, induce nephron formation within the metanephric mesenchyme, initiate and organize ureteric bud branching, and participate in terminal differentiation of the nephron. Highlighted are common signaling pathways that function at multiple stages during kidney development, including signaling via Wnts, bone morphogenic proteins, fibroblast growth factor, sonic hedgehog, RET/glial cell–derived neurotrophic factor, and notch pathways. Also emphasized are the roles of transcription factors Odd1, Eya1, Pax2, Lim1, and WT-1 in directing renal development. Areas requiring future investigation include the factors that modulate signaling pathways to provide temporal and site-specific effects. The evolution of our understanding of the cellular and molecular mechanisms of kidney development may provide methods for improved diagnosis of renal anomalies and, hopefully, targets for intervention for this common cause of childhood end-stage kidney disease. Human cytomegalovirus (HCMV) is the leading cause of congenital viral infection in the United States and intrauterine transmission of HCMV occurs in approximately 40% of pregnant women with primary HCMV infection. Although HCMV infection during pregnancy clearly may be detrimental to fetal development, its consequences on placentation remain largely unexplored. In this study, the effects of HCMV infection on cytotrophoblast (CTB) invasion were investigated utilizing the first trimester extravillous CTB cell line SGHPL-4. HCMV infection significantly inhibited SGHPL-4 proliferation, epidermal growth factor (EGF)- and hepatocyte growth factor (HGF)-induced migration and invasion, as well as the secretion of matrix metalloproteinase (MMP)-2 and MMP-9. Both HCMV and EGF activated the EGF receptor (EGFR), inducing receptor tyrosine phosphorylation at specific residues. Of interest, EGFR was differentially activated by HCMV, and viral gene transcription was not required for the observed inhibitory effect on CTB invasiveness. These findings demonstrate that HCMV infection impairs CTB differentiation along the invasive pathway and that the differential regulation of EGFR by HCMV may contribute to impaired CTB function. Elucidating the mechanisms by which HCMV impairs placentation may be key in understanding fetal and maternal pathologies associated with intrauterine HCMV infection. [Copyright &y& Elsevier]
