Giant cell tumor of bone is a highly invasive benign tumor with a high postoperative recurrence rate. Objective: This study aims to analyze the research hotspots and trends of global research on giant cell tumor of bone in the past 20 years, to provide a reference for relevant personnel in this field to carry out academic research. Methods: The literature related to giant cell tumor of bone from 2001 to 2021 was retrieved from the Web of Science. The bibliometrics research method and VOS Viewer were used to extract and analyze the keywords of the journal authors' research institutions, and the research status and development direction in the recent 20 years were visualized. Results: A total of 2063 articles were included. The number of global publications is increasing every year. The United States contributes the most to global research, with the most citations and the highest H-index. The journal Clinical Orthopaedics and Related Research published the most articles on this issue. "Denosumab" and "h3f3a" will get more attention and be the next popular hotspot in the future. One popular intelligent transportation systems (ITS) element that has been deployed in various areas of the United States is variable-message signs (VMSs). Such signs are part of advanced traveler information systems, and they can help drivers make better en route decisions regarding their trips. Unlike most other ITS elements (e.g., ramp metering, advanced signals), VMSs are not supported by mandatory regulations. Consequently, the expected benefit from implementing VMSs largely depends on the response of drivers. This study explored the factors affecting alternative route choices of car drivers with VMSs on the interstate highway system. This study adopted a method called LOTUS to analyze driver-compliance behavior with VMSs. Driver behavior under information provision through VMS data has been collected using the stated-preference approach. The study showed that travel-time saving is not the single dominant factor for driver route choice under information provision. Background: Breast cancer is a leading cause of cancer-related death for women in the USA. Thus, there is an increasing need to investigate novel prognostic markers and therapeutic methods. Inflammation raises challenges in treating and preventing the spread of breast cancer. Specifically, the nuclear factor kappa b (NFκB) pathway contributes to cancer progression by stimulating proliferation and preventing apoptosis. One target gene of this pathway is PTGS2, which encodes for cyclooxygenase 2 (COX-2) and is upregulated in 40% of human breast carcinomas. COX-2 is an enzyme involved in the production of prostaglandins, which mediate inflammation. Here, we investigate the effect of Singleminded-2s (SIM2s), a transcriptional tumor suppressor that is implicated in inhibition of tumor growth and metastasis, in regulating NFκB signaling and COX-2.Methods: For in vitro experiments, reporter luciferase assays were utilized in MCF7 cells to investigate promoter activity of NFκB and SIM2. Real-time PCR, immunoblotting, immunohistochemistry, and chromatin immunoprecipitation assays were performed in SUM159 and MCF7 cells. For in vivo experiments, MCF10DCIS.COM cells stably expressing SIM2s-FLAG or shPTGS2 were injected into SCID mice and subsequent tumors harvested for immunostaining and analysis.Results: Our results reveal that SIM2 attenuates the activation of NFκB as measured using NFκB-luciferase reporter assay. Furthermore, immunostaining of lysates from breast cancer cells overexpressing SIM2s showed reduction in various NFκB signaling proteins, as well as pAkt, whereas knockdown of SIM2 revealed increases in NFκB signaling proteins and pAkt. Additionally, we show that NFκB signaling can act in a reciprocal manner to decrease expression of SIM2s. Likewise, suppressing NFκB translocation in DCIS.COM cells increased SIM2s expression. We also found that NFκB/p65 represses SIM2 in a dose-dependent manner, and when NFκB is suppressed, the effect on the SIM2 is negated. Additionally, our ChIP analysis confirms that NFκB/p65 binds directly to SIM2 promoter site and that the NFκB sites in the SIM2 promoter are required for NFκB-mediated suppression of SIM2s. Finally, overexpression of SIM2s decreases PTGS2 in vitro, and COX-2 staining in vivo while decreasing PTGS2 and/or COX-2 activity results in re-expression of SIM2.Conclusion: Our findings identify a novel role for SIM2s in NFκB signaling and COX-2 expression. Abstract: In March and April 2009, an entirely novel influenza A (H1N1) virus (NIAV) emerged in Mexico and the USA. During the subsequent months, the virus rapidly spread all over the world by person-to-person transmission. In this report, RNA interference (RNAi) was used as an antiviral agent to inhibit NIAV replication in A549 cells. Ten small interfering RNAs (siRNAs) targeting extremely conserved regions among multiple NIAV genomes could effectively block the replication of NIAV strain A/Beijing/01/2009 (H1N1) in A549 cells. Epidemiologic studies evaluating the association of whole-grain intake with risk for coronary heart disease (CHD) have produced inconsistent results. The aim of this metaanalysis was to summarize the evidence from observed studies regarding the association between whole-grain intake and risk for CHD. Pertinent studies were identified by searching the Web of Knowledge and PubMed up to July 2014. A random-effects model was used to combine the results. Publication bias was estimated using Begg's funnel plot and Egger's regression asymmetry test. Ultimately, fourteen reports of 18 studies (15 cohort studies and 3 case-control studies) involving 14,427 patients with CHD and 400,492 participants were used in this meta-analysis. Pooled results suggested that highest whole-grain intake amount compared with the lowest amount was significantly associated with reduced risk for CHD (summary relative risk 0.787, 95% confidence interval 0.743 to 0.833), with no between-study heterogeneity observed (I² = 0%, p = 0.537). The association was significant in cohort studies but not in case-control studies. Inverse associations were also found in the United States and Europe. No publication bias was found. Background: Arterial stiffness, typically evaluated via estimated pulse wave velocity (ePWV), is believed to have a significant association with cardiovascular diseases. The objective of this study was to investigate the correlation between Life's Essential 8 (LE8), a newly revised metric of cardiovascular health, and ePWV among adult population in the United States., Methods: This research employed a cross-sectional methodology, drawing upon data from the National Health and Nutrition Examination Survey (NHANES) spanning from 2011 to 2018. To explore the relationship between LE8 and ePWV among adults in the US, both univariate and multivariate linear regression analyses were carried out. Additionally, the restricted cubic splines method was utilized to examine any non-linear correlation., Results: The study comprised 6,742 participants with an average age of 48.30 ± 0.35 years. Among these, 3,236 were males, representing a weighted percentage of 48%. The population's weighted average LE8 score was 68.68 ± 0.37, while the average ePWV was 8.18 ± 0.04. An entirely adjusted model revealed a negative correlation between ePWV and LE8 scores [in the moderate LE8 group, coefficient - 0.17, 95% CI -0.28 to -0.06, p  = 0.004; in the high LE8 group, coefficient - 0.44, 95% CI -0.56 to -0.32, p  < 0.0001]. This negative correlation was consistent with the findings in demographic subgroup analysis, with the effect size being more pronounced among adults under the age of 60, and individuals without hypertension, cardiovascular disease, or diabetes., Conclusion: Our study reveals a negative correlation between LE8 and ePWV in the adult population of the US, suggesting that LE8 could potentially serve as an indicative marker for evaluating the risk of vascular stiffness. This inverse relationship is markedly stronger in adults below 60 years and those without diagnosed vascular diseases. This implies that lifestyle upgrades and risk factor management could be especially advantageous in curbing arterial stiffness within these groups.
