This article proposes functional median polish, an extension of univariate median polish, for one-way and two-way functional analysis of variance (ANOVA). The functional median polish estimates the functional grand effect and functional main factor effects based on functional medians in an additive functional ANOVA model assuming no interaction among factors. A functional rank test is used to assess whether the functional main factor effects are significant. The robustness of the functional median polish is demonstrated by comparing its performance with the traditional functional ANOVA fitted by means under different outlier models in simulation studies. The functional median polish is illustrated on various applications in climate science, including one-way and two-way ANOVA when functional data are either curves or images. Specifically, Canadian temperature data, U.S. precipitation observations and outputs of global and regional climate models are considered, which can facilitate the research on the close link between local climate and the occurrence or severity of some diseases and other threats to human health. Trends in monthly heavy precipitation, defined by a return period of one year, are assessed for statistical significance in observations and Global Climate Model (GCM) simulations over the contiguous United States using Monte Carlo non-parametric and parametric bootstrapping techniques. The results from the two Monte Carlo approaches are found to be similar to each other, and also to the traditional non-parametric Kendall's τ test, implying the robustness of the approach. Two different observational data-sets are employed to test for trends in monthly heavy precipitation and are found to exhibit consistent results. Both data-sets demonstrate upward trends, one of which is found to be statistically significant at the 95% confidence level. Upward trends similar to observations are observed in some climate model simulations of the twentieth century, but their statistical significance is marginal. For projections of the twenty-first century, a statistically significant upwards trend is observed in most of the climate models analyzed. The change in the simulated precipitation variance appears to be more important in the twenty-first century projections than changes in the mean precipitation. Stochastic fluctuations of the climate-system are found to be dominate monthly heavy precipitation as some GCM simulations show a downwards trend even in the twenty-first century projections when the greenhouse gas forcings are strong. The Taylor hypothesis (TH) as applied to rainfall is a proposition about the space–time covariance structure of the rainfall field. Specifically, it supposes that if a spatiotemporal precipitation field with a stationary covariance Cov(r, τ) in both space r and time τ moves with a constant velocity v, then the temporal covariance at time lag τ is equal to the spatial covariance at space lag r = vτ that is, Cov(0, τ) = Cov(vτ, 0). Qualitatively this means that the field evolves slowly in time relative to the advective time scale, which is often referred to as the frozen field hypothesis. Of specific interest is whether there is a cutoff or decorrelation time scale for which the TH holds for a given mean flow velocity v. In this study, the validity of the TH is tested for precipitation fields using high-resolution gridded Next Generation Weather Radar (NEXRAD) reflectivity data produced by the WSI Corporation by employing two different statistical approaches. The first method is based on rigorous hypothesis testing, while the second is based on a simple correlation analysis, which neglects possible dependencies between the correlation estimates. Radar reflectivity values are used from the southeastern United States with an approximate horizontal resolution of 4 km × 4 km and a temporal resolution of 15 min. During the 4-day period from 2 to 5 May 2002, substantial precipitation occurs in the region of interest, and the motion of the precipitation systems is approximately uniform. The results of both statistical methods suggest that the TH might hold for the shortest space and time scales resolved by the data (4 km and 15 min) but that it does not hold for longer periods or larger spatial scales. Also, the simple correlation analysis tends to overestimate the statistical significance through failing to account for correlations between the covariance estimates. [ABSTRACT FROM AUTHOR]

Abstract: Research and development into whole organism malaria vaccines is progressing rapidly thanks to the major investments over recent years from several funders, and the commitment and interest of many leading researchers. Progress includes the discovery of potential new candidate vaccines and the start of the first phase 1/2a clinical trial of the radiation attenuated sporozoite approach for Plasmodium falciparum, under US Food and Drug Administration regulatory oversight. A group of leading scientists, clinical trialists and stakeholders, together with representatives of regulatory authorities including some from African countries, met recently to document the issues that will require detailed consideration to assess this promising approach. Questions related to scale-up, quality, purity and consistency of a manufacturing process using mosquitoes to generate a commercial product, and demonstration of the stability of attenuated sporozoites will need further work. Should a high level of efficacy be demonstrated in clinical challenge studies, it will become a priority to agree in which populations and age groups questions about strain-transcendence and duration of efficacy should be answered, and how clinical development can progress with an approach based on cryopreservation in liquid nitrogen.
