Background: We have previously demonstrated that an aldo‐keto reductase (AKR) from Echinochloa colona (EcAKR4‐1) can metabolize glyphosate and confers glyphosate resistance. This study aims to investigate if the EcAKR4‐1 orthologs from Lolium rigidum also play a role in glyphosate resistance in non‐target‐site based, glyphosate‐resistant (R) L. rigidum populations from Western Australia. Results: The full‐length L. rigidum AKR gene (LrAKR4C10) orthologous to EcAKR4‐1, together with a distinct LrAKR1, were cloned from plants of a glyphosate‐susceptible (S) (VLR1) and three glyphosate R L. rigidum populations (WALR50, WALR60 and WALR70). Reverse transcription quantitative polymerase chain reaction (RT‐qPCR) results showed that basal expression levels of the two LrAKR genes did not differ between the R and S populations, but their expression was significantly induced by glyphosate (up to 4.3‐fold) or 2,4‐D treatment (up to 3.4‐fold) in R populations. Escherichia coli cells transformed respectively with LrAKR4C10 and LrAKR1 were more tolerant to glyphosate. Rice (Oryza sativa) seedlings overexpressing each of the two LrAKR gene survived glyphosate rates that were lethal to the green fluorescence protein (GFP) control plants. Structural modeling predicts a similar way of glyphosate binding and detoxification by LrAKR4C10 and EcAKR4‐1, but an alternative way of glyphosate binding by LrAKR1. Relatively lower capacity of the two LrAKRs in conferring glyphosate resistance than the known EcAKR4‐1 was discussed in relation to structural interaction. Conclusion: Glyphosate‐induced higher expression of the two LrAKR genes in L. rigidum populations contributes to a moderate level of glyphosate resistance likely through enhanced glyphosate metabolism. The herbicide 2,4‐D can also induce the LrAKR expression, indicating the potential antagonistic effect of 2,4‐D to glyphosate. © 2022 The Authors. BACKGROUND Trifluralin is widely used in Australia as one of the important pre‐emergence herbicides to control annual ryegrass (Lolium rigidum Gaud.) populations. Trifluralin resistance evolution and mechanisms have been identified in some ryegrass populations. RESULTS: In this study, 21 putative resistant field survey populations from Western Australian were screened with trifluralin, and 90% (19 of 21) contained individuals surviving 480 g ha−1 trifluralin treatment. Twelve populations contained individuals possessing the known α‐tubulin resistance mutations at Val‐202, Thr‐239 and Arg‐243 in TUA4 (alpha‐tubulin 4 n), plus multiple potential resistance mutations in TUA4 pending genetic confirmation. Three populations had only individuals carrying newly identified (but uncharacterized) mutations in TUA3/TUA4. Radioactive work found that six populations evolved metabolic resistance to trifluralin, and at least four of them also possessed the known and/or putative target‐site mutations. CONCLUSION: These results confirm that a high incidence of resistance to the dinitroaniline herbicide (trifluralin) is present, and target‐site tubulin mutations make a major contribution to resistance in these annual ryegrass populations. Glyphosate is the most widely used herbicide in world agriculture and for general vegetation control in a wide range of situations. Global and often intensive glyphosate selection of very large weedy plant populations has resulted in widespread glyphosate resistance evolution in populations of many weed species. Here, working with a glyphosate-resistant (GR) Echinochloa colona population that evolved in a Western Australia agricultural field, we identified an ATP-binding cassette (ABC) transporter (EcABCC8) that is consistently up-regulated in GR plants. When expressed in transgenic rice, this EcABCC8 transporter endowed glyphosate resistance. Equally, rice, maize, and soybean overexpressing the EcABCC8 ortholog genes were made resistant to glyphosate. Conversely, CRISPR/Cas9-mediated knockout of the EcABCC8 ortholog gene OsABCC8 increased rice susceptibility to glyphosate. Subcellular localization analysis and quantification of glyphosate cellular levels in treated ABCC8 transgenic rice plants and isolated leaf protoplasts as well as structural modeling support that EcABCC8 is likely a plasma membrane-localized transporter extruding cytoplasmic glyphosate to the apoplast, lowering the cellular glyphosate level. BACKGROUND The photosystem II (PSII)‐inhibiting herbicides are important for Australian farmers to control Lolium rigidum Gaud. and other weed species in trazine tolerant (TT)‐canola fields. A L. rigidum population (R) collected from a TT‐canola field from Western Australia showed multiple resistance to PSII, acetyl‐coenzyme A carboxylase (ACCase) and acetolactate synthase (ALS) inhibitors. The mechanisms of multiple resistance in this R population were determined. RESULTS: The R population showed a low‐level (about 3.0‐fold) resistance to the PSII‐inhibiting herbicides metribuzin and atrazine. Sequencing of the psbA gene revealed no differences between the R and susceptible (S) sequences. Furthermore, [14C]‐metribuzin experiments found no significant difference in metribuzin foliar uptake and translocation between the R and S plants. However, [14C]‐metribuzin metabolism in R plants was 2.3‐fold greater than in S plants. The cytochrome P450 monooxygenase inhibitor piperonyl butoxide (PBO) enhanced plant mortality response to metribuzin and atrazine in both R and S populations. In addition, multiple resistance to ALS and ACCase inhibitors are due to known resistance mutations in ALS and ACCase genes. CONCLUSION: The results demonstrate that enhanced metribuzin metabolism likely involving cytochrome P450 monooxygenase contributes to metribuzin resistance in Lolium rigidum. BACKGROUND: Relatively new herbicides that target 4‐hydroxyphenylpyruvate dioxygenase (HPPD) are now available for use on the world's great grain crops (rice, wheat, corn and soybean) and for other uses. With widespread and persistent use of HPPD‐inhibiting herbicides, the evolution of HPPD‐inhibiting herbicide resistant weeds is inevitable. Currently, resistance to HPPD‐inhibiting herbicides is known in two weed species, waterhemp and Palmer amaranth. Here, we report a HPPD‐inhibiting herbicide resistant wild radish population from the Western Australia grain belt. This population was not selected with HPPD‐inhibiting herbicides, rather it evolved resistance to earlier used herbicides with different modes of action and exhibits cross‐resistance to HPPD‐inhibiting herbicides. RESULTS: Dose–response experiments showed the resistant (R) population exhibits 4 to 6.5‐fold resistance to the HPPD‐inhibiting herbicides mesotrione, tembotrione and isoxaflutole, compared to the susceptible (S) population. This resistance is not target‐site based as cloning of full coding sequences of the HPPD genes from S and R plants did not reveal resistance‐endowing single nucleotide polymorphisms. The HPPD gene expression levels are similar in S and R plants. In addition, no differences in [14C]‐mesotrione uptake and translocation were observed in the S and R plants. However, the time required for R plants to metabolise 50% [14C]‐mesotrione is 7.7‐fold faster than for the S plants. CONCLUSION: We confirm resistance to HPPD‐inhibiting herbicides exists in a population of the economically damaging global weed wild radish. BACKGROUND: A Lolium rigidum population collected from Western Australia was previously reported as highly resistant to dinitroaniline herbicides mainly due to a Val‐202‐Phe substitution in the target site α‐tubulin protein. To further determine the contribution of the 202 mutation to resistance, two sub‐populations, respectively comprising the 202 mutant and wild‐type (WT) individuals, were isolated from within the same resistant population and subject to dinitroaniline herbicide doses. A rice transgenic study was conducted to demonstrate whether the amino acid substitution at the 202 residue confers resistance. In addition, as indicated in the phenotyping and genotyping study, non‐target enhanced trifluralin metabolism was further examined in the same population. RESULTS: The 202 mutants were more resistant than the wild‐type plants. Rice calli transformed with the L. rigidum mutant α‐tubulin gene (Val‐202‐Phe) were more resistant to dinitroaniline herbicides relative to calli transformed with the wild‐type gene. Also, enhanced trifluralin metabolism was detected in the 202 mutants in comparison to the susceptible seedlings. Avena is a problematic weed of cropping regions of southern Australia and many areas of the world. In 2010, a random survey was conducted across 14 million hectares of the Western Australian grain belt to monitor the change in herbicide resistance levels by comparing resistance frequency results with a survey conducted in 2005. Screening Avena populations with herbicides commonly used to control this weed revealed that 48% of Avena populations displayed resistance to the commonly used acetyl-Co A carboxylase-inhibiting herbicides, which was lower than that found in 2005 (71%). The broad-spectrum herbicides glyphosate and paraquat provided good control of all Avena populations. Resistance to acetolactate synthase-inhibiting herbicides and to flamprop were detected for the first time in Western Australia in this survey. Therefore, a wide range of weed management options that target all phases of the cropping program are needed to sustain these cropping systems in the future. [ABSTRACT FROM AUTHOR]

BACKGROUND In Australia, glyphosate has been used routinely to control wild radish ( Raphanus raphanistrum L.) for the past 40 years. This study focuses on two field-evolved glyphosate-resistant populations of wild radish collected from the grainbelt of Western Australia. RESULTS Two wild radish biotypes were confirmed to be glyphosate resistant by comparing R/S of two suspected populations. Based on R/S from dose-response curves, the R1 and R2 populations were 2.3 and 3.2 times more resistant to glyphosate respectively. Dose response on glyphosate-selected progeny (>1080 g ha−1) demonstrated that the glyphosate resistance mechanism was heritable. When compared with the pooled mortality results of three known susceptible populations ( S1, S2 and S3), the R1 and R2 subpopulations were 3.4-fold and 4.5-fold more resistant at the LD50 level respectively. Both populations were found to have multiple resistance to the phytoene desaturase inhibitor; diflufenican, the synthetic auxin; 2,4-D and the ALS inhibitors; chlorsulfuron, sulfometuron-methyl, imazethapyr and metosulam. CONCLUSIONS This is the first report confirming glyphosate resistance evolution in wild radish and serves to re-emphasise the importance of diverse weed control strategies. BACKGROUND: Bromus rigidus is a common weed species that has increased in cropping fields owing to limited control options. During a random field survey in Western Australia, six B. rigidus populations that had survived in-crop weed control programmes were collected. The study aimed to determine the resistance profile of these six populations. RESULTS: Based on dose-response studies, all six B. rigidus populations had a low-level resistance to sulfosulfuron and sulfometuron (both sulfonylurea herbicides) while remaining susceptible to herbicides with other modes of action. ALS in vitro activity assays revealed no differences in enzyme sensitivity between susceptible and resistant populations, while the use of malathion (a cytochrome P450 inhibitor) in combination with sulfosulfuron caused the resistant populations to behave like the susceptible population. CONCLUSION: This study established that these six B. rigidus populations have a low-level resistance to the ALS-inhibiting sulfonylurea herbicides, but are able to be controlled by other herbicide modes of action. The low-level, malathion-reversible resistance, together with a sensitive ALS, strongly suggest that a non-target-site enhanced metabolism is the mechanism of resistance. The increasing number of weedy species resistant to dinitroaniline herbicides warrants studies on the evolutionary factors contributing to resistance evolution, including genetic inheritance of resistance traits. In this study, we investigated the genetic control of trifluralin resistance in a well-characterised Lolium rigidum Gaud. population from Western Australia. This population was purified to contain plants homozygous for the Val-202-Phe α-tubulin mutation, and used as the resistant (R) parents and crossed with susceptible (S) parents to produce eight reciprocal F1 families. Trifluralin dose response curves of the eight F1 families indicate that trifluralin resistance in this population is inherited as an incomplete recessive nuclear trait. The F1 plants were crossed within each families to establish eight pseudo-F2 (ψ -F2) families. Segregation ratio of resistance and susceptibility in ψ -F2 families were determined using the discriminating trifluralin rates of 120 and 480 g a.i. ha−1. At 480 g a.i. ha−1 trifluralin, the segregation ratio in almost all ψ -F2 families (except one) was fit to 1:3 (resistance: susceptibility) one recessive gene control model. However, at 120 g a.i. ha−1 trifluralin, the segregation ratios in half of the families did not fit this model, indicating involvement of one or more genes in resistance at the lower rate. These results showed complexity of genetic inheritance of trifluralin resistance in this L. rigidum population possessing the Val-202-Phe α-tubulin mutation. [ABSTRACT FROM AUTHOR]

Background: Wild radish (Raphanus raphanistrum) is a globally important weed of crops. Two atrazine-resistant wild radish populations (R1 and R2), collected from the Western Australia grain belt, were investigated for resistance to photosystem II (PSII) herbicides., Results: Sequencing of the full-length psbA gene revealed the well-known Ser264-Gly substitution in population R1, whereas population R2 displayed a novel Phe274-Val substitution. Herbicide dose-response studies confirmed that the population with the Ser264-Gly mutation exhibited high-level resistance to atrazine, but super-sensitivity to bromoxynil. Plants possessing the novel Phe274-Val mutation exhibited a modest level of resistance to atrazine, metribuzin and diuron, and were bromoxynil susceptible. Structural modelling of the mutant D1 proteins predicts that the Ser264-Gly mutation endows atrazine resistance by abolishing H-bonds, but confers bromoxynil super-sensitivity by enhancing hydrogen bonding. The Phe274-Val substitution provides resistance to atrazine and diuron by indirectly affecting H-bond formation between the Ser264 residue and the herbicides., Conclusion: The results demonstrate that the Phe274-Val mutation is likely responsible for resistance to PSII-inhibiting triazine and urea herbicides. Background: Lolium rigidum (annual ryegrass) is a widespread annual crop weed that has evolved high levels of resistance to selective herbicides. Anecdotal evidence suggests that intensive cropping also leads to higher seed dormancy in L. rigidum. This was quantified by measuring dormancy levels in L. rigidum populations collected from paired sites (one with nil to low cropping intensity, the other intensively cropped) located throughout the Western Australian grain belt., Results: Populations from non-cropped fields or those with low cropping intensity showed higher and faster germination than populations from fields with a medium- or high-intensity cropping regime. Resistance to selective herbicides was also higher in the medium- and high-intensity cropping fields than in the low-intensity cropping fields., Conclusion: High-intensity cropping systems are likely to impose greater selection pressures for seed dormancy and selective herbicide resistance, because late-emerging seedlings avoid preplanting weed control practices (tillage and non-selective herbicide application) but are exposed to selective in-crop herbicides., (© 2014 Society of Chemical Industry.)

Background: Hordeum populations are becoming increasingly difficult to control in cropping fields. Two herbicide-resistant H. leporinum populations were identified during a random crop survey after herbicides were applied. The study aimed to determine the herbicide resistance profile of these H. leporinum biotypes to a range of herbicides used for their control., Results: Based on dose-response studies, one H. leporinum population was very highly resistant to sulfosulfuron and sulfometuron (both sulfonylurea herbicides) and also displayed low-level resistance to imazamox (an imidazolinone herbicide). Reduced sensitivity of the ALS enzyme was identified with in vitro activity assays. Gene sequence analysis revealed a proline-to-threonine substitution at amino acid position 197 of ALS, which is likely to be the molecular basis for resistance in this population. Background and Aims: Karrikinolide (KAR(1)) is a smoke-derived chemical that can trigger seeds to germinate. A potential application for KAR(1) is for synchronizing the germination of weed seeds, thereby enhancing the efficiency of weed control efforts. Yet not all species germinate readily with KAR(1), and it is not known whether seemingly non-responsive species can be induced to respond. Here a major agronomic weed family, the Brassicaceae, is used to test the hypothesis that a stimulatory response to KAR(1) may be present in physiologically dormant seeds but may not be expressed under all circumstances., Methods: Seeds of eight Brassicaceae weed species (Brassica tournefortii, Raphanus raphanistrum, Sisymbrium orientale, S. erysimoides, Rapistrum rugosum, Lepidium africanum, Heliophila pusilla and Carrichtera annua) were tested for their response to 1 µm KAR(1) when freshly collected and following simulated and natural dormancy alleviation, which included wet-dry cycling, dry after-ripening, cold and warm stratification and a 2 year seed burial trial., Key Results: Seven of the eight Brassicaceae species tested were stimulated to germinate with KAR(1) when the seeds were fresh, and the remaining species became responsive to KAR(1) following wet-dry cycling and dry after-ripening. Light influenced the germination response of seeds to KAR(1), with the majority of species germinating better in darkness. Germination with and without KAR(1) fluctuated seasonally throughout the seed burial trial., Conclusions: KAR(1) responses are more complex than simply stating whether a species is responsive or non-responsive; light and temperature conditions, dormancy state and seed lot all influence the sensitivity of seeds to KAR(1), and a response to KAR(1) can be induced. Three response types for generalizing KAR(1) responses are proposed, namely inherent, inducible and undetected. Given that responses to KAR(1) were either inherent or inducible in all 15 seed lots included in this study, the Brassicaceae may be an ideal target for future application of KAR(1) in weed management.
