Abstract

The size of the human head is highly heritable, but genetic drivers of its variation within the general population remain unmapped. We perform a genome-wide association study on head size (N = 80,890) and identify 67 genetic loci, of which 50 are novel. Neuroimaging studies show that 17 variants affect specific brain areas, but most have widespread effects. Gene set enrichment is observed for various cancers and the p53, Wnt, and ErbB signaling pathways. Genes harboring lead variants are enriched for macrocephaly syndrome genes (37-fold) and high-fidelity cancer genes (9-fold), which is not seen for human height variants. Head size variants are also near genes preferentially expressed in intermediate progenitor cells, neural cells linked to evolutionary brain expansion. Our results indicate that genes regulating early brain and cranial growth incline to neoplasia later in life, irrespective of height. This warrants investigation of clinical implications of the link between head size and cancer.

Abstract

Cortical thickness, surface area and volumes vary with age and cognitive function, and in neurological and psychiatric diseases. Here we report heritability, genetic correlations and genome-wide associations of these cortical measures across the whole cortex, and in 34 anatomically predefined regions. Our discovery sample comprises 22,824 individuals from 20 cohorts within the Cohorts for Heart and Aging Research in Genomic Epidemiology (CHARGE) consortium and the UK Biobank. We identify genetic heterogeneity between cortical measures and brain regions, and 160 genome-wide significant associations pointing to wnt/β-catenin, TGF-β and sonic hedgehog pathways. There is enrichment for genes involved in anthropometric traits, hindbrain development, vascular and neurodegenerative disease and psychiatric conditions. These data are a rich resource for studies of the biological mechanisms behind cortical development and aging.

12. Investigating the impact of microbiome-changing interventions on food decision-making: MIFOOD study protocol.

Abstract

Background: Obesity is a multifactorial disease reaching pandemic proportions with increasing healthcare costs, advocating the development of better prevention and treatment strategies. Previous research indicates that the gut microbiome plays an important role in metabolic, hormonal, and neuronal cross-talk underlying eating behavior. We therefore aim to examine the effects of prebiotic and neurocognitive behavioral interventions on food decision-making and to assay the underlying mechanisms in a Randomized Controlled Trial (RCT). Method: This study uses a parallel arm RCT design with a 26-week intervention period. We plan to enroll 90 participants (male/diverse/female) living with overweight or obesity, defined as either a Waist-to-Hip Ratio (WHR) ≥ 0.9 (male)/0.85 (diverse, female) or a Body Mass Index (BMI) ≥ 25 kg/m2. Key inclusion criteria are 18–60 years of age and exclusion criteria are type 2 diabetes, psychiatric disease, and Magnetic Resonance Imaging (MRI) contraindications. The interventions comprise either a daily supplementary intake of 30 g soluble fiber (inulin), or weekly neurocognitive behavioral group sessions, compared to placebo (equicaloric maltodextrin). At baseline and follow-up, food decision-making is assessed utilizing task-based MRI. Secondary outcome measures include structural MRI, eating habits, lifestyle factors, personality traits, and mood. Further, we obtain fecal and blood samples to investigate gut microbiome composition and related metabolites. Discussion: This study relies on expanding research suggesting that dietary prebiotics could improve gut microbiome composition, leading to beneficial effects on gut-brain signaling and higher-order cognitive functions. In parallel, neurocognitive behavioral interventions have been proposed to improve unhealthy eating habits and metabolic status. However, causal evidence on how these "bottom-up" and "top-down" processes affect food decision-making and neuronal correlates in humans is still scarce. In addition, microbiome, and gut-brain-axis-related mediating mechanisms remain unclear. The present study proposes a comprehensive approach to assess the effects of these gut-brain-related processes influencing food decision-making in overweight and obesity. Abstract

We organized 10Kin1day, a pop-up scientific event with the goal to bring together neuroimaging groups from around the world to jointly analyze 10,000+ existing MRI connectivity datasets during a 3-day workshop. In this report, we describe the motivation and principles of 10Kin1day, together with a public release of 8,000+ MRI connectome maps of the human brain.

Abstract

The volume of the lateral ventricles (LV) increases with age and their abnormal enlargement is a key feature of several neurological and psychiatric diseases. Although lateral ventricular volume is heritable, a comprehensive investigation of its genetic determinants is lacking. In this meta-analysis of genome-wide association studies of 23,533 healthy middle-aged to elderly individuals from 26 population-based cohorts, we identify 7 genetic loci associated with LV volume. These loci map to chromosomes 3q28, 7p22.3, 10p12.31, 11q23.1, 12q23.3, 16q24.2, and 22q13.1 and implicate pathways related to tau pathology, S1P signaling, and cytoskeleton organization. We also report a significant genetic overlap between the thalamus and LV volumes (ρgenetic = -0.59, p-value = 3.14 × 10-6), suggesting that these brain structures may share a common biology. These genetic associations of LV volume provide insights into brain morphology.

Abstract

Objective: Previous studies have shown that socioeconomically deprived groups exhibit higher lesion load of the white matter (WM) in aging. The aim of this study was to (i) investigate to what extent education and income may contribute to differences in white matter hyperintensities (WMHs) and (ii) identify risk profiles related to a higher prevalence of age-associated WMH. Design and Setting: Population-based adult study of the Leipzig Research Centre for Civilization Diseases (LIFE) in Leipzig, Germany. Participants: Dementia-free sample aged 40–80 years (n = 1,185) derived from the population registry. Measurements: Information was obtained in standardized interviews. WMH (including the derived Fazekas scores) were assessed using automated segmentation of high-resolution T1-weighted anatomical and fluid-attenuated inversion recovery (FLAIR) MRI acquired at 3T. Results: Despite a significant association between income and WMH in univariate analyses, results from adjusted models (age, gender, arterial hypertension, heart disease, and APOE e4 allele) indicated no association between income and WMH. Education was associated with Fazekas scores, but not with WMH and not after Bonferroni correction. Prevalence of some health-related risk factors was significantly higher among low-income/education groups. After combining risk factors in a factor analysis, results from adjusted models indicated significant associations between higher distress and more WMH as well as between obesity and more deep WMH. Conclusions: Previously observed differences in WMH between socioeconomically deprived groups might stem from differences in health-related risk factors. Abstract

The AgeWell.de-MRI study aims to assess the effects of a multicomponent intervention on brain structure and function in older adults at risk for Alzheimer’s disease (AD). The study includes Leipzig-based participants from the AgeWell.de study, a cluster-randomized trial performed in 4 study centers over Germany. The AgeWell.de study investigates the effect of a multicomponent intervention targeting modifiable risk factors for dementia similar to the finnish FINGER study. In the AgeWell.de-MRI study, participants undergo a MRI session before and after the two-year intervention period of the AgeWell.de study. Abstract

This project aims to investigate the neural correlates of long COVID using magnetic resonance imaging (MRI). Anatomical, diffusion-weighted, fluid-attenuated and quantitative susceptibility imaging will be performed. We will test the association of SarsCov2 infection and long COVID symptoms with gray matter volume of olfactory, gustatory and memory-related brain regions as well as white matter microstructure of relevant limbic tracts. Further, we will explore other neuroimaging measures in relation to the disease status as well as cognitive, smell and taste function.

Abstract

Obesity and its comorbidities are linked to impaired cognitive performance, accelerated cognitive decline and neurodegenerative pathologies (Dye et al., 2017). Besides frequently reported cortical thinning and alterations in subcortical gray matter volume (Tanaka et al., 2020), obesity seems to have serious repercussions on global white matter coherence and the microstructure of the brain region crucially involved in feeding behavior and metabolism, the hypothalamus (Lampe, et al., 2019; Schwartz et al., 2000; Thomas, et al. 2019). A dominant explanation for obesity-related structural and functional alterations and associated cognitive impairments is that a high-fat fat diet and accumulation of visceral fat set off a metabolic and immunological cascade which may result in systemic chronic low-grade inflammation as well as neuroinflammation attacking the central nervous system (Leigh & Morris, 2020). Bariatric surgery encompasses procedures that surgically modify the stomach and the intestines to induce weight loss by food restriction and nutrients malabsorption. To date, it is the most effective intervention to reliably induce considerable reduction of excess body fat and improve metabolic risk factors. It has been shown to reduce the characteristic low-grade inflammation and, importantly, to exert positive effects on the central nervous system. Widespread effects on brain structure and function are documented by a growing body of literature (Nota et al., 2020). This project aims to determine whether surgical interventions for morbidly obese individuals lead to a recovery in microstructure of major white matter tracts and the hypothalamus, comparing a group of bariatric patients with a waiting-list control group. The overarching goal will be to characterize the mechanisms by which the proposed recovery is driven and the time course in which positive effects unfold.

It is widely established that there is a worldwide increase in the prevalence of obesity. Obesity is problematic as it can cause metabolic disorders such as cardiovascular disease[1]⁠ and type II-diabetes[2]⁠. Often, unhealthy food decisions are one factor leading to obesity. Food decision-making is a very complex topic including not only homeostatic factors but also cognitive processes. One of these processes that plays a role in food decision-making is the food memory. Therefore, it is necessary to get a better understanding of the interplay of food decisions and food memory and the underlying neural systems. We here aim to identify how factors such as food wanting and hunger shape food memory and how the underlying neural anatomy contributes to these memory processes.

Background: Social isolation has been suggested to increase the risk to develop cognitive decline. However, our knowledge on causality and neurobiological underpinnings is still limited. Methods: In this preregistered analysis, we tested the impact of social isolation on central features of brain and cognitive ageing using a longitudinal population-based magnetic resonance imaging (MRI) study. We assayed 1992 cognitively healthy participants (50--82years old, 921women) at baseline and 1409 participants after~6y follow-up. Results: We found baseline social isolation and change in social isolation to be associated with smaller volumes of the hippocampus and clusters of reduced cortical thickness. Furthermore, poorer cognitive functions (memory, processing speed, executive functions) were linked to greater social isolation, too. Conclusions: Combining advanced neuroimaging outcomes with prevalent lifestyle characteristics from a well-characterized population of middle-to older aged adults, we provide evidence that social isolation contributes to human brain atrophy and cognitive decline. Within-subject effects of social isolation were similar to between-subject effects, indicating an opportunity to reduce dementia risk by promoting social networks. Abstract

Over the past few decades, neuroimaging has become a ubiquitous tool in basic research and clinical studies of the human brain. However, no reference standards currently exist to quantify individual differences in neuroimaging metrics over time, in contrast to growth charts for anthropometric traits such as height and weight1. Here we assemble an interactive open resource to benchmark brain morphology derived from any current or future sample of MRI data ( http://www.brainchart.io/ ). With the goal of basing these reference charts on the largest and most inclusive dataset available, acknowledging limitations due to known biases of MRI studies relative to the diversity of the global population, we aggregated 123,984 MRI scans, across more than 100 primary studies, from 101,457 human participants between 115 days post-conception to 100 years of age. MRI metrics were quantified by centile scores, relative to non-linear trajectories2 of brain structural changes, and rates of change, over the lifespan. Brain charts identified previously unreported neurodevelopmental milestones3, showed high stability of individuals across longitudinal assessments, and demonstrated robustness to technical and methodological differences between primary studies. Centile scores showed increased heritability compared with non-centiled MRI phenotypes, and provided a standardized measure of atypical brain structure that revealed patterns of neuroanatomical variation across neurological and psychiatric disorders. In summary, brain charts are an essential step towards robust quantification of individual variation benchmarked to normative trajectories in multiple, commonly used neuroimaging phenotypes.
