1. Smart Skin Envelope

Abstract

The Smart Skin Envelope research analyses the recent revolution that has taken place in the sector of planning and production of smart skin components, made up of dynamic layers. The aim is to identify the technological, functional, qualitative and performance parameters that guide the decisions of the actors in the innovation process. It explores the factors that drive them to develop solutions and proposals designed to transform the envelope of the building from a static to a dynamic element, featuring interoperable components that can interact with the input from the outdoor and indoor environments, in relation to which the smart skin acts as a system of boundary and delimitation. The proposed research programme explores in particular the sector of Smart Envelopes, setting as its priority objective the identification and definition of the energy performance, both through analysis of the state of the art and through the development of a facade component that is dynamic in terms of the adaptive variability of its performance.

Abstract

Genome editing of human pluripotent stem cells (hPSCs) provides powerful opportunities for in vitro disease modeling, drug discovery, and personalized stem cell-based therapeutics. Currently, only small edits can be engineered with high frequency, while larger modifications suffer from low efficiency and a resultant need for selection markers. Here, we describe marker-free genome editing in hPSCs using Cas9 ribonucleoproteins (RNPs) in combination with AAV6-mediated DNA repair template delivery. We report highly efficient and bi-allelic integration frequencies across multiple loci and hPSC lines, achieving mono-allelic editing frequencies of up to 94% at the HBB locus. Using this method, we show robust bi-allelic correction of homozygous sickle cell mutations in a patient-derived induced PSC (iPSC) line. Thus, this strategy shows significant utility for generating hPSCs with large gene integrations and/or single-nucleotide changes at high frequency and without the need for introducing selection genes, enhancing the applicability of hPSC editing for research and translational uses.

Abstract

The CRISPR-Cas9 system is a powerful tool for genome editing, which allows the precise modification of specific DNA sequences. Many efforts are underway to use the CRISPR-Cas9 system to therapeutically correct human genetic diseases1-6. The most widely used orthologs of Cas9 are derived from Staphylococcus aureus and Streptococcus pyogenes5,7. Given that these two bacterial species infect the human population at high frequencies8,9, we hypothesized that humans may harbor preexisting adaptive immune responses to the Cas9 orthologs derived from these bacterial species, SaCas9 (S. aureus) and SpCas9 (S. pyogenes). By probing human serum for the presence of anti-Cas9 antibodies using an enzyme-linked immunosorbent assay, we detected antibodies against both SaCas9 and SpCas9 in 78% and 58% of donors, respectively. We also found anti-SaCas9 T cells in 78% and anti-SpCas9 T cells in 67% of donors, which demonstrates a high prevalence of antigen-specific T cells against both orthologs. We confirmed that these T cells were Cas9-specific by demonstrating a Cas9-specific cytokine response following isolation, expansion, and antigen restimulation. Together, these data demonstrate that there are preexisting humoral and cell-mediated adaptive immune responses to Cas9 in humans, a finding that should be taken into account as the CRISPR-Cas9 system moves toward clinical trials.

20. Life cycle thinking strategies for constructing nZEB schools

Abstract

By 2050, Italy, too, will have to achieve climate neutrality. Compared to current standards, almost 75% of the existing building stock is energy inefficient, and since 17% consists of school buildings, intervening in this type of building is essential to develop virtuous paths to achieve 2030 decarbonisation targets. Vertical envelopes are among the technological units that contribute the most to achieving better energy-environmental performance of the building, but at the same time, they have a certain weight in terms of carbon and embodied energy; moreover, LCA studies dealing with the vertical building envelope mainly focus on ‘cradle-to-grave’ impacts, neglecting end-of-life scenarios based on the recycling of selective demolition materials. Le chiusure verticali sono tra le unità tecnologiche che contribuiscono maggiormente al raggiungimento di migliori performance energetico-ambientali dell’edificio, ma allo stesso tempo hanno un certo peso in termini di carbonio ed energia incorporata; inoltre gli studi LCA che trattano l’involucro verticale dell’edificio si concentrano principalmente sugli impatti ‘dalla culla alla tomba’, trascurando scenari di fine vita basati sul riciclo dei materiali provenienti da demolizione selettiva. 33. nearly Zero Energy Modules – Low-impact modular housing models for the city of the future

Abstract

This paper presents some of the outcomes of the nZEM (nearly Zero Energy Module) research, aimed at promoting the development and prototyping of modular and flexible building elements in platform frames characterised by the integration of innovative building envelope and system solutions. These modules are designed to address various functional and technical-regulatory needs, including energy savings, energy efficiency, indoor comfort, accessibility, and structural safety while adhering to environmental compatibility. The project, funded by the Tuscany region under the calls for POR CREO 2020-2024, was developed by a consortium comprising research institutions and companies. This paper shows some results of the research activities carried out in the spoke five of the National Biodiversity Future Center aimed at testing the effectiveness of urban regeneration projects, including the creation of Pocket Parks as adaptation strategies to climate change. The methodological approach adopted to validate the design of small scale resilient urban spaces implemented by the Municipality of Florence will be presented starting from the definition of state-of-the-art. Furthermore, it will analyse how the metaplanning model (based on the integration of NbS) and the predictive tools used in the experimentation can be used to develop future regulatory tools for planning and controlling the transformation of the built environment. Abstract

Cultural heritage is recognized as a driver of the sustainable development. The UNESCO’s 1972 World Heritage Convention has addressed key topics that link heritage practices to sustainability objectives. Historic buildings make up a considerable part of EU building stock, and their renovation and safeguarding require retrofit solutions that balance safety, energy efficiency, human comfort, heritage value preservation and environmental sustainability. This article tries to throw light on innovative techniques for heritage restoration based on sustainability, reuse, and recycle principles through non-invasive interventions. The work outlines the progress in the field of advanced materials obtained through the reuse of recycled materials, showing outcomes from MIRACLE research carried out by University of Florence on the development of an innovative fiber-reinforced bio-composite matrix to be applied for energy plus seismic retrofitting of existing masonry buildings built before 1945.

In recent years, the development of digital technologies has opened the field to the possibility of transforming prefabricated building production facilities into highly integrated, controlled, environmentally and economically efficient Industry 4.0 systems. CARES research is part of the ongoing debate, developing and implementing an innovative production model of construction elements made of corrugated cardboard, based on the principle of product-process digitalization through the use of BIM tools, aimed at rationalizing the use of resources and reducing the environmental burden of the construction phase., Le filiere di produzione edile stanno affrontando una profonda trasformazione in chiave ecologica e digitale con l’obiettivo di sostenere la competitività e la resilienza del settore nei confronti delle sfide ambientali e socioeconomiche in atto. Negli ultimi anni lo sviluppo delle tecnologie digitali ha aperto il campo alla possibilità di trasformare gli impianti per la produzione edilizia prefabbricata in sistemi di Industria 4.0 altamente integrati, controllati ed efficienti sotto il profilo economico e ambientale. La ricerca CARES si inserisce nel dibattito in atto sviluppando e implementando un innovativo modello di produzione di elementi costruttivi realizzati in cartone ondulato, basato sul principio della digitalizzazione di prodotto-processo, attraverso l’utilizzo di strumenti BIM, finalizzato a razionalizzare l’uso delle risorse e a ridurre il carico ambientale della fase realizzativa.
