Abstract

The extraction method plays a crucial role in obtaining high-quality DNA samples, which is indispensable for various molecular biology techniques and analyses, enabling a deeper comprehension of genetic information and biological processes. The objectives of the study were: a) to optimize the chloroplast DNA extraction protocol by comparing modified CTAB methods and GeneAid for both leaf and wood samples of Shorea leprosula, a major commercial timber species, and b) to identify a suitable cpDNA region that exhibits variability and universality across taxa. Total DNA was analyzed by gel electrophoresis followed by Sanger sequencing to determine the amplification success. The results revealed that trnL intron, trnL-trnF, and trnG yielded readable sequences of the expected length (maximum 586 bp, 480 bp, and 908 bp, respectively), while the rps16 intron failed to assemble a contig. The petL-psbE region provided long readability for reverse sequences (769 bp) but not for the forward sequence (195 bp). Higher successful DNA extraction was achieved from the leaves compared to the woods. The lower sequencing quality may be attributed to suboptimal primer design, the structural features of the regions resulting from extensive repetitive sequences, and the suboptimal condition of the extraction method in eliminating wood chemical compounds. Abstract

Gunung Leuser National Park (Gunung Leuser NP) is a conservation region in Indonesia with a relatively high diversity of flora and fauna. One of the potential species found in Gunung Leuser NP is Keruing. Keruing or Dipterocarpus spp. known as potential of wood and non wood producers with a high economical value and is often a target for exploitation. Habitat destruction and regeneration problems also threatened the species, including associated species with the keruing. The objective of this study were to determine woody species stand structure and regeneration status in Tangkahan forest, Gunung Leuser NP. This research was conducted by purposive sampling method which the plot located based on the presence of keruing. A total of 110 plots were employed for this purpose. Based on the results, the stands in Tangkahan forest formed an inverted J pattern, which is a common structure in natural forests. Individuals with a diameter of 10 cm dominated stands, whereas trees with a diameter lower that 60 cm were the least frequent. The dominance of individual trees in the medium to low diameter class indicates that the forest is still in the early stages of development The vertical structure of the trees at the research location shows that they are mostly from stratum C with 171 individuals, that indicated forest condition is in good condition. The status of woody species regeneration status in the research location is quite variable, with 23 species (34.84 %) in good status, four species (6.06%) in fair status, ten species (15.15%) with poor status, 21 species (31.81 %) in none status, and eight species (12.12%) in new regeneration status. The population structure and regeneration patterns show significant forest degradation due to anthropogenic disturbances. 15. Genetic Structuring of the Wild Aquilaria malaccensisPopulation from Indonesia Inferred by trnL-trnF and trnH-psbA

Abstract

Aquilaria malaccensisLam. is known to produce high-grade agarwood, which is naturally distributed in Indonesia, and their trading has been regulated under the CITES Appendix II. This study aimed to analyze the genetic diversity of wild A. malaccensisfrom Indonesia. Forty-one samples of A. malaccensiswere gathered from various wild populations originating from different islands, including Sumatra mainland, adjacent islands (Bintan and Lingga), West Kalimantan, and Bunguran Island—the northernmost island adjacent to West Kalimantan. Sequencing was performed using trnL-trnF and trnH-psbA. The total sequence (1417 bp) revealed six distinct haplotypes, categorized based on their phytoregion: Sumatra and its adjacent islands Bintan and Lingga, West Kalimantan, and Bunguran Island. Notably, Bunguran displayed the most distinct haplotype, featuring two unique haplotypes and one shared haplotype with the populations of Sumatra and its adjacent islands, Bintan and Lingga. The shared haplotype among Bintan, Lingga, and Bunguran with Sumatra mainland suggests a potential connection to the geographical history of these islands, which were formerly part of Sundaland. The nucleotide diversity (π) was notably low in Bunguran Island (π = 0.00215) and the West Kalimantan population (π = 0.00067), in contrast to Sumatra and its adjacent islands characterized by a nucleotide diversity of 0. Despite this low nucleotide diversity value, the phytoregional populations were genetically differentiated, with the highest differentiation between Bunguran Island and Western Borneo (FST= 0.83686). It is suggested that the individual trees represent each haplotype as genetic sources for ex situ conservation and cultivation.

Abstract

This study aimed to analyze the basic properties (chemical composition and physical and mechanical properties) of belangke bamboo (Gigantochloa pruriens) and its potential as a particleboard reinforcement material, aimed at increasing the mechanical properties of the boards. The chemical composition was determined by Fourier transform near infrared (NIR) analysis and X-ray diffraction (XRD) analysis. The physical and mechanical properties of bamboo were evaluated following the Japanese standard JIS A 5908 (2003) and the ISO 22157:2004 standard, respectively. The results showed that this bamboo had average lignin, holocellulose, and alpha-cellulose content of 29.78%, 65.13%, and 41.48%, respectively, with a degree of crystallinity of 33.54%. The physical properties of bamboo, including specific gravity, inner and outer diameter shrinkage, and linear shrinkage, were 0.59%, 2.18%, 2.26%, and 0.18%, respectively. Meanwhile, bamboo’s mechanical properties, including compressive strength, shear strength, and tensile strength, were 42.19 MPa, 7.63 MPa, and 163.8 MPa, respectively. Markedly, the addition of belangke bamboo strands as a reinforcing material (surface coating) in particleboards significantly improved the mechanical properties of the boards, increasing the modulus of elasticity (MOE) and bending strength (MOR) values of the fabricated composites by 16 and 3 times.

37. Genetic Diversity and Community Structure of Macrozoobenthos from Five Mangrove Forests in North Sumatra and Aceh, Indonesia.

Abstract

Mangrove forests play an important role in macrozoobenthos ecology, assisting them in foraging, sheltering, and reproduction. In order to better understanding the genetic diversity and population genetic structure of macrozoobenthos, the mitochondrial 16S subunit (16S rRNA) gene was used. Present study aimed to determine genetic diversity and community structure of macrozoobenthos in the North Sumatra and Aceh mangrove forests, Indonesia. The reliaprep gDNA tissue miniprep system kit was used to extract 50 samples. To determine the PCR product's molecular weight, UVITEX software was employed. Genetic polymorphism was examined with GenAlex version 6.502, and each community underwent principal coordinates analysis (PCoA). MVSP 3.2.2 software and the unweighted pair group method with arithmetic mean (UPGMA) were used to construct the dendrogram. The 16S rRNA gene revealed a band size in the range of 370–500 bp. The PCoA showed that approximately all individual macrozoobenthos from each community structure were localized in the same quadrant. Genetic variation was varied by 3% between populations and 97.5% within individuals according to the analysis of molecular variance (AMOVA), with the dendrogram classifying the populations into two major clusters. Dendrogram analysis showed low genetic differentiation between macrozoobenthos populations of North Sumatra and the Aceh mangroves, implying the low ability of individuals in a population to adapt. The high polymorphic information content (PIC) value (0.886) reported was consistent with the number of alleles and size of the population's heterozygosity value. Present findings provide important information that will assist in formulation of mangrove conservation and restoration approaches.
