1. AesExpert: Towards Multi-modality Foundation Model for Image Aesthetics Perception

Abstract

The highly abstract nature of image aesthetics perception (IAP) poses significant challenge for current multimodal large language models (MLLMs). The lack of human-annotated multi-modality aesthetic data further exacerbates this dilemma, resulting in MLLMs falling short of aesthetics perception capabilities. To address the above challenge, we first introduce a comprehensively annotated Aesthetic Multi-Modality Instruction Tuning (AesMMIT) dataset, which serves as the footstone for building multi-modality aesthetics foundation models. Specifically, to align MLLMs with human aesthetics perception, we construct a corpus-rich aesthetic critique database with 21,904 diverse-sourced images and 88K human natural language feedbacks, which are collected via progressive questions, ranging from coarse-grained aesthetic grades to fine-grained aesthetic descriptions. To ensure that MLLMs can handle diverse queries, we further prompt GPT to refine the aesthetic critiques and assemble the large-scale aesthetic instruction tuning dataset, i.e. AesMMIT, which consists of 409K multi-typed instructions to activate stronger aesthetic capabilities. Based on the AesMMIT database, we fine-tune the open-sourced general foundation models, achieving multi-modality Aesthetic Expert models, dubbed AesExpert. Extensive experiments demonstrate that the proposed AesExpert models deliver significantly better aesthetic perception performances than the state-of-the-art MLLMs, including the most advanced GPT-4V and Gemini-Pro-Vision. Abstract

Dialog Structure Induction (DSI) is the task of inferring the latent dialog structure (i.e., a set of dialog states and their temporal transitions) of a given goal-oriented dialog. It is a critical component for modern dialog system design and discourse analysis. Existing DSI approaches are often purely data-driven, deploy models that infer latent states without access to domain knowledge, underperform when the training corpus is limited/noisy, or have difficulty when test dialogs exhibit distributional shifts from the training domain. This work explores a neural-symbolic approach as a potential solution to these problems. We introduce Neural Probabilistic Soft Logic Dialogue Structure Induction (NEUPSL DSI), a principled approach that injects symbolic knowledge into the latent space of a generative neural model. We conduct a thorough empirical investigation on the effect of NEUPSL DSI learning on hidden representation quality, few-shot learning, and out-of-domain generalization performance. Over three dialog structure induction datasets and across unsupervised and semi-supervised settings for standard and cross-domain generalization, the injection of symbolic knowledge using NEUPSL DSI provides a consistent boost in performance over the canonical baselines.

Abstract

In this report, we introduce the Gemini 1.5 family of models, representing the next generation of highly compute-efficient multimodal models capable of recalling and reasoning over fine-grained information from millions of tokens of context, including multiple long documents and hours of video and audio. The family includes two new models: (1) an updated Gemini 1.5 Pro, which exceeds the February version on the great majority of capabilities and benchmarks; (2) Gemini 1.5 Flash, a more lightweight variant designed for efficiency with minimal regression in quality. Gemini 1.5 models achieve near-perfect recall on long-context retrieval tasks across modalities, improve the state-of-the-art in long-document QA, long-video QA and long-context ASR, and match or surpass Gemini 1.0 Ultra's state-of-the-art performance across a broad set of benchmarks. Studying the limits of Gemini 1.5's long-context ability, we find continued improvement in next-token prediction and near-perfect retrieval (>99%) up to at least 10M tokens, a generational leap over existing models such as Claude 3.0 (200k) and GPT-4 Turbo (128k). Finally, we highlight real-world use cases, such as Gemini 1.5 collaborating with professionals on completing their tasks achieving 26 to 75% time savings across 10 different job categories, as well as surprising new capabilities of large language models at the frontier; when given a grammar manual for Kalamang, a language with fewer than 200 speakers worldwide, the model learns to translate English to Kalamang at a similar level to a person who learned from the same content.

Abstract

This report introduces a new family of multimodal models, Gemini, that exhibit remarkable capabilities across image, audio, video, and text understanding. The Gemini family consists of Ultra, Pro, and Nano sizes, suitable for applications ranging from complex reasoning tasks to on-device memory-constrained use-cases. Evaluation on a broad range of benchmarks shows that our most-capable Gemini Ultra model advances the state of the art in 30 of 32 of these benchmarks - notably being the first model to achieve human-expert performance on the well-studied exam benchmark MMLU, and improving the state of the art in every one of the 20 multimodal benchmarks we examined. We believe that the new capabilities of the Gemini family in cross-modal reasoning and language understanding will enable a wide variety of use cases. We discuss our approach toward post-training and deploying Gemini models responsibly to users through services including Gemini, Gemini Advanced, Google AI Studio, and Cloud Vertex AI.

9. Aperture Diffraction for Compact Snapshot Spectral Imaging

Abstract

We demonstrate a compact, cost-effective snapshot spectral imaging system named Aperture Diffraction Imaging Spectrometer (ADIS), which consists only of an imaging lens with an ultra-thin orthogonal aperture mask and a mosaic filter sensor, requiring no additional physical footprint compared to common RGB cameras. Then we introduce a new optical design that each point in the object space is multiplexed to discrete encoding locations on the mosaic filter sensor by diffraction-based spatial-spectral projection engineering generated from the orthogonal mask. The orthogonal projection is uniformly accepted to obtain a weakly calibration-dependent data form to enhance modulation robustness. Meanwhile, the Cascade Shift-Shuffle Spectral Transformer (CSST) with strong perception of the diffraction degeneration is designed to solve a sparsity-constrained inverse problem, realizing the volume reconstruction from 2D measurements with Large amount of aliasing. Our system is evaluated by elaborating the imaging optical theory and reconstruction algorithm with demonstrating the experimental imaging under a single exposure. Ultimately, we achieve the sub-super-pixel spatial resolution and high spectral resolution imaging. 10. Efficient Rotating Synthetic Aperture Radar Imaging via Robust Sparse Array Synthesis

Abstract

Rotating Synthetic Aperture Radar (ROSAR) can generate a 360$^\circ$ image of its surrounding environment using the collected data from a single moving track. Due to its non-linear track, the Back-Projection Algorithm (BPA) is commonly used to generate SAR images in ROSAR. Despite its superior imaging performance, BPA suffers from high computation complexity, restricting its application in real-time systems. In this paper, we propose an efficient imaging method based on robust sparse array synthesis. It first conducts range-dimension matched filtering, followed by azimuth-dimension matched filtering using a selected sparse aperture and filtering weights. The aperture and weights are computed offline in advance to ensure robustness to array manifold errors induced by the imperfect radar rotation. We introduce robust constraints on the main-lobe and sidelobe levels of filter design. The resultant robust sparse array synthesis problem is a non-convex optimization problem with quadratic constraints. An algorithm based on feasible point pursuit and successive convex approximation is devised to solve the optimization problem. Extensive simulation study and experimental evaluations using a real-world hardware platform demonstrate that the proposed algorithm can achieve image quality comparable to that of BPA, but with a substantial reduction in computational time up to 90%.

11. TaskLAMA: Probing the Complex Task Understanding of Language Models

Abstract

Structured Complex Task Decomposition (SCTD) is the problem of breaking down a complex real-world task (such as planning a wedding) into a directed acyclic graph over individual steps that contribute to achieving the task, with edges specifying temporal dependencies between them. SCTD is an important component of assistive planning tools, and a challenge for commonsense reasoning systems. We probe how accurately SCTD can be done with the knowledge extracted from Large Language Models (LLMs). We introduce a high-quality human-annotated dataset for this problem and novel metrics to fairly assess performance of LLMs against several baselines. Our experiments reveal that LLMs are able to decompose complex tasks into individual steps effectively, with a relative improvement of 15% to 280% over the best baseline. We also propose a number of approaches to further improve their performance, with a relative improvement of 7% to 37% over the base model. However, we find that LLMs still struggle to predict pairwise temporal dependencies, which reveals a gap in their understanding of complex tasks.

Abstract

Automated reasoning with unstructured natural text is a key requirement for many potential applications of NLP and for developing robust AI systems. Recently, Language Models (LMs) have demonstrated complex reasoning capacities even without any finetuning. However, existing evaluation for automated reasoning assumes access to a consistent and coherent set of information over which models reason. When reasoning in the real-world, the available information is frequently inconsistent or contradictory, and therefore models need to be equipped with a strategy to resolve such conflicts when they arise. One widely-applicable way of resolving conflicts is to impose preferences over information sources (e.g., based on source credibility or information recency) and adopt the source with higher preference. In this paper, we formulate the problem of reasoning with contradictory information guided by preferences over sources as the classical problem of defeasible reasoning, and develop a dataset called BoardgameQA for measuring the reasoning capacity of LMs in this setting. BoardgameQA also incorporates reasoning with implicit background knowledge, to better reflect reasoning problems in downstream applications. We benchmark various LMs on BoardgameQA and the results reveal a significant gap in the reasoning capacity of state-of-the-art LMs on this problem, showing that reasoning with conflicting information does not surface out-of-the-box in LMs. While performance can be improved with finetuning, it nevertheless remains poor.

19. Microstructure and high-temperature wear performance of Zr-Y modified silicide coating deposited on TC4 alloy

Abstract

Zr-Y modified silicide coating was deposited on TC4 alloy by the pack cementation process. The microstructure， friction and wear performance， and wear mechanisms of the coating were investigated. The results show that the Zr-Y modified silicide coating has a multi-layer structure， an outer layer consists of TiSi2 and a small amount of ZrSi2， a thin middle layer is TiSi， and an inner layer is a mixture of Ti5Si4 and Ti5Si3. The micro-hardness of the coating is much higher than that of the TC4 substrate and exhibits an obvious decrease tendency from the surface to the interior. The Zr-Y modified silicide coating possesses good anti-wear performance when wearing against GCr15 and Al2O3 balls at a high-temperature of 600 ℃. When wearing against GCr15， the wear rate of the coating is about 3.59×10-5 mm3/（N·m）， which is about 36.6% of that of the TC4 substrate， and the main wear mechanisms are characterized by the adhesive of GCr15 on the coating surface and oxidation wear. When wearing against the Al2O3 ball， the wear rate of the coating is about 9.75×10-5 mm3/（N·m）， which is about 18.9% of that of the TC4 substrate， and the wear mechanisms are fatigue wear， oxidation wear， and adhesion wear.

Abstract

The sea level in coastal areas of China reached the second highest in 2021, just after that recorded in 2022. External force and dynamic analyses based on tide gauges, satellite observations, reanalysis data and regional numerical outputs were conducted to understand these abnormally high sea levels and determine their possible causes. Results show that the coastal sea level of China had increased at an annual rate of 3.4 ± 0.3 mm during 1980–2021, with an acceleration of 0.06 ± 0.02 mm per year2. The superposition of significant oscillations of quasi-2, 3–7, quasi-9, quasi-11, quasi-19 and 20–30 years contributed to the anomalously high sea levels. The negative-phased El Niño/Southern Oscillation was correlated with the anomalously high sea level and the north‒south anti-phase pattern of the coastal sea level in 2021. Meanwhile, phase lags of 1–4 months occurred with the sea-level response. On a decadal timescale, the Pacific Decadal Oscillation (PDO) was negatively correlated with the anomalous mean sea level (MSL), and the negative-phased PDO contributed to the anomalous sea-level change in 2021. Particularly, the monthly MSL peaked in April and July, and the contribution of wind stress to the anomalously high sea level was 38.5% in the south of the Taiwan Strait in April and 30% along the coast of China in July. These results were consistent with the tide gauge and satellite data. Close agreement was also observed between the coastal sea-level fingerprint and the air and sea surface temperatures.

The relationship between power transmission and reception in AC/DC interconnected power grids is complex， with numerous constraints on grid operation and regulation. Therefore， relying solely on manual expertise to assess the adjustment of power transmission and reception plans presents significant challenges. In response to the precise requirements of market-oriented transactions for adjusting interprovincial power transmission and reception plans， a method for analyzing the feasible domain of interprovincial transmission power is proposed. Firstly， based on the operational characteristics of AC/DC interconnected power grids， the modeling of operational constraints within the feasible scheduling domain is expanded. Subsequently， a model for the feasible domain of interprovincial transmission power is constructed， and solved using the ordered vertex search method. Finally， case studies are conducted based on the actual operational scenarios of the regional power grid in the south， and the effectiveness of the feasible domain for interprovincial transmission power is validated using the computational results of the entire network scheduling and optimization model. The calculation results of the feasible domain provide insight into the limit range of interprovincial transmission power under specific transmission and reception scenarios， thereby providing a reference basis for the verification and adjustment of medium to long-term power transmission and reception plans.

Abstract

Abstract Background Human patients often experience an episode of serious seizure activity, such as status epilepticus (SE), prior to the onset of temporal lobe epilepsy (TLE), suggesting that SE can trigger the development of epilepsy. Yet, the underlying mechanisms are not fully understood. The low-density lipoprotein receptor related protein (Lrp4), a receptor for proteoglycan-agrin, has been indicated to modulate seizure susceptibility. However, whether agrin-Lrp4 pathway also plays a role in the development of SE-induced TLE is not clear. Methods Lrp4 f/f mice were crossed with hGFAP-Cre and Nex-Cre mice to generate brain conditional Lrp4 knockout mice (hGFAP-Lrp4 −/− ) and pyramidal neuron specific knockout mice (Nex-Lrp4 −/− ). Lrp4 was specifically knocked down in hippocampal astrocytes by injecting AAV virus carrying hGFAP-Cre into the hippocampus. The effects of agrin-Lrp4 pathway on the development of SE-induced TLE were evaluated on the chronic seizure model generated by injecting kainic acid (KA) into the amygdala. The spontaneous recurrent seizures (SRS) in mice were video monitored. Results We found that Lrp4 deletion from the brain but not from the pyramidal neurons elevated the seizure threshold and reduced SRS numbers, with no change in the stage or duration of SRS. More importantly, knockdown of Lrp4 in the hippocampal astrocytes after SE induction decreased SRS numbers. In accord, direct injection of agrin into the lateral ventricle of control mice but not mice with Lrp4 deletion in hippocampal astrocytes also increased the SRS numbers. These results indicate a promoting effect of agrin-Lrp4 signaling in hippocampal astrocytes on the development of SE-induced TLE. Last, we observed that knockdown of Lrp4 in hippocampal astrocytes increased the extracellular adenosine levels in the hippocampus 2 weeks after SE induction. Blockade of adenosine A1 receptor in the hippocampus by DPCPX after SE induction diminished the effects of Lrp4 on the development of SE-induced TLE. Conclusion These results demonstrate a promoting role of agrin-Lrp4 signaling in hippocampal astrocytes in the development of SE-induced development of epilepsy through elevating adenosine levels. Targeting agrin-Lrp4 signaling may serve as a potential therapeutic intervention strategy to treat TLE.

In order to clarify the controlling effect of different packaging materials on common storage pests, the penetration and survival of 7 kinds of high density polyethylene films with different concentrations of deltamethrin by the larvae of 3 common species of stored grain pests (Tribolium castaneum (Herbst), Tribolium confusum Duval and Lasioderma serricorne (Fabricius)) were investigated after being treated by two penetration methods for different times at 27~29 ℃ and 70%~80% RH. The results showed that all the tested new insect-proof films had certain insect resistance capabilities, which were different based on species of stored grain insects. In addition, the insect resistance capability of the 6 kinds of new films containing chemicals was higher than that of the control films after 9 days of treatment, and the anti-penetration rate of 1% A (HDPE particle containing 5% deltamethrin by mass) film, 3% A film, 1% B (HDPE particle containing 5% deltamethrin and 2% isoethers by mass) film and 3% B film was 100% under two penetration modes. The survival rate of all three species larvae decreased with increasing treatment time. After 9 days of parallel penetration treatment, the survival rate of three species of larvae on 3%B film was significantly lower than that of other films, and the survival rate of L. serricorne larvae reduced to 10% at the lowest. According to the penetration resistance and larvae survival rate of the test insects on the films, 3% B film had the best insect control effectiveness.

Chemotherapeutics can induce immunogenic cell death (ICD) in tumor cells, offering new possibilities for cancer therapy. However, the efficiency of the immune response generated is insufficient due to the inhibitory nature of the tumor microenvironment (TME). Here, we developed a pH/reactive oxygen species (ROS) dual-response system to enhance chemoimmunotherapy for melanoma. The system productively accumulated in tumors by specific binding of phenylboronic acid (PBA) to sialic acids (SA). The nanoparticles (NPs) rapidly swelled and released quercetin (QUE) and doxorubicin (DOX) upon the stimulation of tumor microenvironment (TME). The in vitro and in vivo results consistently demonstrated that the NPs improved anti-tumor efficacy and prolonged survival of mice, significantly enhancing the effects of the combination. Our study revealed DOX was an ICD inducer, stimulating immune responses and promoting maturation of dendritic cells (DCs). Additionally, QUE served as a TME regulator by inhibiting the cyclooxygenase-2 (COX2)-prostaglandin E2 (PGE2) axis, which influenced various immune cells, including increasing cytotoxic T cells (CLTs) infiltration, promoting M1 macrophage polarization, and reducing regulatory T cells (Tregs) infiltration. The combination synergistically facilitated chemoimmunotherapy efficacy by remodeling the immunosuppressive microenvironment. This work presents a promising strategy to increase anti-tumor efficiency of chemotherapeutic agents.

Graph data is used in a wide range of applications, while analyzing graph data without protection is prone to privacy breach risks. To mitigate the privacy risks, we resort to the standard technique of differential privacy to publish a synthetic graph. Abstract

Standard empirical risk minimization (ERM) training can produce deep neural network (DNN) models that are accurate on average but under-perform in under-represented population subgroups, especially when there are imbalanced group distributions in the long-tailed training data. Therefore, approaches that improve the accuracy-group robustness trade-off frontier of a DNN model (i.e. improving worst-group accuracy without sacrificing average accuracy, or vice versa) is of crucial importance. Uncertainty-based active learning (AL) can potentially improve the frontier by preferentially sampling underrepresented subgroups to create a more balanced training dataset. However, the quality of uncertainty estimates from modern DNNs tend to degrade in the presence of spurious correlations and dataset bias, compromising the effectiveness of AL for sampling tail groups. In this work, we propose Introspective Self-play (ISP), a simple approach to improve the uncertainty estimation of a deep neural network under dataset bias, by adding an auxiliary introspection task requiring a model to predict the bias for each data point in addition to the label. We show that ISP provably improves the bias-awareness of the model representation and the resulting uncertainty estimates. Three-phase PWM voltage-source rectifier (VSR) systems have been widely used in various energy conversion systems, where current sensors are the key component for state monitoring and system control. The current sensor faults may bring hidden danger or damage to the whole system; therefore, this paper proposed a random forest (RF) and current fault texture feature-based method for current sensor fault diagnosis in three-phase PWM VSR systems. First, the three-phase alternating currents (ACs) of the three-phase PWM VSR are collected to extract the current fault texture features, and no additional hardware sensors are needed to avoid causing additional unstable factors. Then, the current fault texture features are adopted to train the random forest current sensor fault detection and diagnosis (CSFDD) classifier, which is a data-driven CSFDD classifier. Finally, the effectiveness of the proposed method is verified by simulation experiments. A three-phase pulse-width modulation (PWM) rectifier can usually maintain operation when open-circuit faults occur in insulated-gate bipolar transistors (IGBTs), which will lead the system to be unstable and unsafe. Aiming at this problem, based on random forests with transient synthetic features, a data-driven online fault diagnosis method is proposed to locate the open-circuit faults of IGBTs timely and effectively in this study. Firstly, by analysing the open-circuit fault features of IGBTs in the three-phase PWM rectifier, it is found that the occurrence of the fault features is related to the fault location and time, and the fault features do not always appear immediately with the occurrence of the fault. Secondly, different data-driven fault diagnosis methods are compared and evaluated, the performance of random forests algorithm is better than that of support vector machine or artificial neural networks. Meanwhile, the accuracy of fault diagnosis classifier trained by transient synthetic features is higher than that trained by original features. Also, the random forests fault diagnosis classifier trained by multiplicative features is the best with fault diagnosis accuracy can reach 98.32%. Finally, the online fault diagnosis experiments are carried out and the results demonstrate the effectiveness of the proposed method, which can accurately locate the open-circuit faults in IGBTs while ensuring system safety., Comment: IET Power Electronics

Background The number of chronic obstructive pulmonary disease (COPD) patients in China is huge, and respiratory rehabilitation training, as an important part of the management of COPD patients in the stabilization period, can effectively improve their lung function and quality of life, as well as reduce the burden on their families and society. Current data from Europe and the United States have shown that the implementation of respiratory rehabilitation under telemedicine management can improve the lung function and QOL of patients, however, there is a lack of relevant practice in China, especially in the west. Objective To assess the impact of respiratory rehabilitation training via telemedicine management in combination with conventional therapy on improving ventilatory capacity and lung function in elderly patients with moderate-to-severe COPD. Methods This study was a prospective randomized controlled study, enrolling consecutive COPD patients who attended the Fourth People's Hospital of Sichuan Province and five joint community clinics from June 2021 to June 2022. The included patients were randomly divided into the experimental group and control group by simple randomized grouping method using random number table. The control group received traditional long-term regular inhalation bronchodilator and oral medication, and the experimental group was guided by telemedicine on the basis of the treatment plan of the control group. A six-month study was conducted on two groups of patients, lung function, Borg score, 6MWT, and quality of life score (QOL score) were recorded at 1 month before and 1, 3, 6 months after intervention. Results The study subjects were divided into 72 cases in the control group and 73 cases in the experimental group, and there was no significant difference in gender, age and lung function at baseline [the forced expiratory volume in one second/predicted value ratio (FEV1%pred) , and the ratio of the forced expiratory volume in one second to the forced vital capacity (FEV1/FVC) ] between the two groups (P>0.05) . In this study, the open-circuit faults diagnosis and location issue of the neutral-point-clamped (NPC) inverters are analysed. A novel fault diagnosis approach based on knowledge driven and data driven was presented for the open-circuit faults in insulated-gate bipolar transistors (IGBTs) of NPC inverter, and Concordia transform (knowledge driven) and random forests (RFs) technique (data driven) are employed to improve the robustness performance of the fault diagnosis classifier. First, the fault feature data of AC in either normal state or open-circuit faults states of NPC inverter are analysed and extracted. Second, the Concordia transform is used to process the fault samples, and it has been verified that the slopes of current trajectories are not affected by different loads in this study, which can help the proposed method to reduce overdependence on fault data. Moreover, then the transformed fault samples are adopted to train the RFs fault diagnosis classifier, and the fault diagnosis results show that the classification accuracy and robustness performance of the fault diagnosis classifier are improved. Abstract

Car dependence has been threatening transportation sustainability as it contributes to congestion and associated externalities. In response, various transport policies that restrict the use of private vehicle have been implemented. However, empirical evaluations of such policies have been limited. To assess these policies' benefits and costs, it is imperative to accurately evaluate how such policies affect traffic conditions. In this study, we compile a refined spatio-temporal resolution data set of the floating-vehicle-based traffic performance index to examine the effects of a recent nonlocal vehicle driving restriction policy in Shanghai, one of most populous cities in the world. Specifically, we explore whether and how the policy impacted traffic speeds in the short term by employing a quasi-experimental difference-in-differences modeling approach. We find that: (1) In the first month, the policy led to an increase of the network-level traffic speed by 1.47% (0.352 km/h) during evening peak hours (17:00-19:00) but had no significant effects during morning peak hours (7:00-9:00). (2) The policy also helped improve the network-level traffic speed in some unrestricted hours (6:00, 12:00, 14:00, and 20:00) although the impact was marginal. (3) The short-term effects of the policy exhibited heterogeneity across traffic analysis zones. The lower the metro station density, the greater the effects were. We conclude that driving restrictions for non-local vehicles alone may not significantly reduce congestion, and their effects can differ both temporally and spatially. However, they can have potential side effects such as increased purchase and usage of new energy vehicles, owners of which can obtain a local license plate of Shanghai for free.

40. Hepatic artery pseudoaneurysm: three case reports and literature review

Abstract

Laparoscopic surgery is extensively applied in the treatment of hepatobiliary diseases. Hepatic artery pseudoaneurysm (HAP) is a rare complication following hepatic biliary surgery through laparoscopy. The clinical manifestations of HAP are diverse and can be fatal. Given its severity, rapid assessment and management are crucial to ensuring a good prognosis. Here, we report three cases of HAP; two underwent laparoscopic surgery due to cholelithiasis, and another caused by trauma. The first case exhibited a pseudoaneurysm involving the distal portion of the right hepatic artery main trunk. The second patient had a pseudoaneurysm at the bifurcation of the left and right hepatic arteries. The third case involved a patient with a pseudoaneurysm involving a branch of the right hepatic artery. The main clinical manifestations of all three cases were bleeding from the biliary tract (the first two cases showed postoperative bleeding in the T-tube, while the third case exhibited gastrointestinal bleeding). The final diagnosis was obtained through digital subtraction angiography. The three patients underwent successful transcatheter arterial embolization operation and a follow-up revealed they were disease-free and alive. This article aims to highlight a rare complication of laparoscopic hepatobiliary surgery and share our experience in early diagnosis and treatment of HAP.

41. Evolutionary screening of precision oncology biomarkers and its applications in prognostic model construction

Summary: Biomarker screening is critical for precision oncology. However, one of the main challenges in precision oncology is that the screened biomarkers often fail to achieve the expected clinical effects and are rarely approved by regulatory authorities. Considering the close association between cancer pathogenesis and the evolutionary events of organisms, we first explored the evolutionary feature underlying clinically approved biomarkers, and two evolutionary features of approved biomarkers (Ohnologs and specific evolutionary stages of genes) were identified. Subsequently, we utilized evolutionary features for screening potential prognostic biomarkers in four common cancers: head and neck squamous cell carcinoma, liver hepatocellular carcinoma, lung adenocarcinoma, and lung squamous cell carcinoma. Finally, we constructed an evolution-strengthened prognostic model (ESPM) for cancers. These models can predict cancer patients’ survival time across different cancer cohorts effectively and perform better than conventional models. In summary, our study highlights the application potentials of evolutionary information in precision oncology biomarker screening.

Abstract

Abstract As a new data management paradigm, knowledge graphs can integrate multiple data sources and achieve quick responses, reasoning and better predictions in drug discovery. Characterized by powerful contagion and a high rate of morbidity and mortality, porcine reproductive and respiratory syndrome (PRRS) is a common infectious disease in the global swine industry that causes economically great losses. Traditional Chinese medicine (TCM) has advantages in low adverse effects and a relatively affordable cost of application, and TCM is therefore conceived as a possibility to treat PRRS under the current circumstance that there is a lack of safe and effective approaches. Here, we constructed a knowledge graph containing common biomedical data from humans and Sus Scrofa as well as information from thousands of TCMs. Subsequently, we validated the effectiveness of the Sus Scrofa knowledge graph by the t-SNE algorithm and selected the optimal model (i.e., transR) from six typical models, namely, transE, transR, DistMult, ComplEx, RESCAL and RotatE, according to five indicators, namely, MRR, MR, HITS@1, HITS@3 and HITS@10. Based on embedding vectors trained by the optimal model, anti-PRRSV TCMs were predicted by two paths, namely, VHC-Herb and VHPC-Herb, and potential anti-PRRSV TCMs were identified by retrieving the HERB database according to the pharmacological properties corresponding to symptoms of PRRS. Ultimately, Dan Shen's (Salvia miltiorrhiza Bunge) capacity to resist PRRSV infection was validated by a cell experiment in which the inhibition rate of PRRSV exceeded 90% when the concentrations of Dan Shen extract were 0.004, 0.008, 0.016 and 0.032 mg/mL. In summary, this is the first report on the Sus Scrofa knowledge graph including TCM information, and our study reflects the important application values of deep learning on graphs in the swine industry as well as providing accessible TCM resources for PRRS.

Abstract

Abstract Innate sensors initiate the production of type I interferons (IFN-I) and proinflammatory cytokines to protect host from viral infection. Several innate nuclear sensors that mainly induce IFN-I production have been identified. Whether there exist innate nuclear sensors that mainly induce proinflammatory cytokine production remains to be determined. By functional screening, we identify 40 S ribosomal protein SA (RPSA) as a nuclear protein that recognizes viral nucleic acids and predominantly promotes proinflammatory cytokine gene expression in antiviral innate immunity. Myeloid-specific Rpsa-deficient mice exhibit less innate inflammatory response against infection with Herpes simplex virus-1 (HSV-1) and Influenza A virus (IAV), the viruses replicating in nucleus. Mechanistically, nucleus-localized RPSA is phosphorylated at Tyr204 upon infection, then recruits ISWI complex catalytic subunit SMARCA5 to increase chromatin accessibility of NF-κB to target gene promotors without affecting innate signaling. Our results add mechanistic insights to an intra-nuclear way of initiating proinflammatory cytokine expression in antiviral innate defense.
