Abstract: In this study, we consider an environment composed of a heterogeneous cluster of multicore-based machines used to analyze satellite data. The workload involves large data sets and is subject to a deadline constraint. Multiple applications, each represented by a directed acyclic graph (DAG), are allocated to a dedicated heterogeneous distributed computing system. Each vertex in the DAG represents a task that needs to be executed and task execution times vary substantially across machines. The goal of this research is to assign the tasks in applications to a heterogeneous multicore-based parallel system in such a way that all applications complete before a common deadline, and their completion times are robust against uncertainties in execution times. We define a measure that quantifies robustness in this environment. We design, compare, and evaluate five static resource allocation heuristics that attempt to maximize robustness. We consider six different scenarios with different ratios of computation versus communication, and loose and tight deadlines. [Copyright &y& Elsevier]

Abstract: Managing seagrass environments and understanding and responding to coastal impacts such as floods or cyclones, requires assessment of seagrass distribution and its biophysical properties in time and space. Comparable assessments of seagrass distribution over time are often lacking as the information is present for separate dates, or created following different mapping approaches, and this makes it difficult to conduct quantitative comparisons. We provide an assessment of available data sets and approaches, and their suitability for monitoring and quantifying change in seagrass percentage cover and extent for a large coastal embayment (Moreton Bay, Australia, 1582 km2). Seagrass percentage cover maps were created for 2011 and 2004 and compared to map and measure the extent of seagrass percentage cover change, and changes in the extent of seagrass environments. Total extent of seagrass was shown to be higher in 2011 compared to 2004. Potential sources of these differences may be: mapping inaccuracy; actual change in extent and cover; and, monthly to seasonal variations in seagrass cover. A qualitative comparison of the 2004 and 2011 maps was performed against maps of seagrass extent maps from 1975, 1986 and 1999, which were created using a range of different methods and data. The output maps show changes in seagrass extent, but a lack of detail arising from variable mapping methods and differing mapping extents prevented a reliable comparison. We conclude that robust mapping of seasonal and inter-annual variation in seagrass percentage cover distribution or extent, as well as impacts of episodic and stochastic disturbance events, requires a thorough understanding of the mapping approaches used so that data sets can be compared. • Coordinated conservation requires clear indicator interrelationships. • We analyse and interrogate global ecosystem indicators for coastal wetlands. • We develop spatially explicit typologies of ecosystem status at the global scale. • Typologies identify diverse conservation and management needs of coastal wetlands. • Simplifying the complex global mosaic of status informs management and conservation. Global-scale conservation initiatives and policy instruments rely on ecosystem indicators to track progress towards targets and objectives. A deeper understanding of indicator interrelationships would benefit these efforts and help characterize ecosystem status. We study interrelationships among 34 indicators for mangroves, saltmarsh, and seagrass ecosystems, and develop data-driven, spatially explicit typologies of coastal wetland status at a global scale. After accounting for environmental covariates and gap-filling missing data, we obtained two levels of clustering at 5 and 18 typologies, providing outputs at different scales for different end users. We generated 2,845 cells (1° (lat) × 1° (long)) globally, of which 29.7% were characterized by high land- and marine-based impacts and a high proportion of threatened species, 13.5% by high climate-based impacts, and 9.6% were refuges with lower impacts, high fish density and a low proportion of threatened species. We identify instances where specific actions could have positive outcomes for coastal wetlands across regions facing similar issues. For example, land- and marine-based threats to coastal wetlands were associated with ecological structure and function indicators, suggesting that reducing these threats may reduce habitat degradation and threats to species persistence. However, several interdimensional relationships might be affected by temporal or spatial mismatches in data. Weak relationships mean that global biodiversity maps that categorize areas by single indicators (such as threats or trends in habitat size) may not be representative of changes in other indicators (e.g., ecosystem function). By simplifying the complex global mosaic of coastal wetland status and identifying regions with similar issues that could benefit from knowledge exchange across national boundaries, we help set the scene for globally and regionally coordinated conservation. Animals can respond to human impacts by favouring different morphological traits or by exploiting different food resources. We quantified the morphology and diet of four functionally different fish species (n = 543 fish) from 13 estuaries with varying degrees of human modification in Queensland, Australia. We found differences in the responses of trophic groups to the environmental conditions of estuaries; principally the extent of seagrass in the estuary, and the amount of shoreline and catchment urbanisation. Here, seagrass and urbanisation extent correlated with the diet and morphology of zooplanktivores and detritivores; thereby indicating that human modifications may modify these species functional roles. Conversely, environmental variables did not correlate with the diet or morphology of zoobenthivores or piscivores thereby indicating that human modifications may have less an effect on these species functional roles. Our findings demonstrate that anthropogenic impacts to coastal ecosystems might extend from the traditionally measured metrics of abundance and diversity. • Human modifications to ecosystems can change animal diet and morphology. • We quantified diet and morphology of four fish species from 13 Australian estuaries. • Detritivores and zooplanktivores were affected by area of seagrass and urbanisation. • Zoobenthivores and piscivores were not affected by human modifications. • Human impacts to ecosystems extend from traditional metrics of abundance. Human-induced habitat clearing and pollution are leading drivers of biodiversity loss. Ecosystem assessments are required to identify ecosystems at risk of collapse, but they should account for cross-system linkages and dynamics where necessary. This is particularly true for coastal wetlands (e.g. seagrass, mangroves and saltmarsh), which exhibit high ecological connectivity and have individually suffered global declines over the last century. We use the coastal wetlands of Moreton Bay, Queensland, Australia, as a model system to examine how integrating outcomes of multiple, simultaneously conducted, ecosystem assessments can assist in identifying appropriate management and conservation strategies. We simultaneously conducted separate assessments of seagrass, mangrove and saltmarsh ecosystems against the IUCN Red List of Ecosystems criteria. Despite substantial human population growth in the region, seagrass and mangroves were assessed as Least Concern. Mangroves were found to be rapidly encroaching on saltmarsh. This process, together with past clearing, were the major drivers behind saltmarsh being assessed as Endangered. Given the importance of connectivity among these connected ecosystems, collapse in any one ecosystem can have seascape-wide consequences, highlighting the benefit of conducting multi-ecosystem assessments. Consequently, a fully integrated assessment of the coastal wetlands as a single entity would miss key processes, such as mangrove encroachment, potentially underestimating overall risks. Our study highlights the plight of saltmarsh and the value of conducting simultaneous RLE assessments for multiple ecosystems comprising seascapes. The Logan-Albert estuary in southeast Queensland, Australia, has high biodiversity and supports multiple economic and recreational services. Elevated nutrient and sediment loads have been a longstanding management issue for the estuary. We investigated the spatial and seasonal patterns of nutrients and turbidity along the Logan-Albert estuary and assessed the effects of a recently constructed upstream dam. Nutrient concentrations and turbidity levels were analysed using 15 years of monitoring data from 19 water quality sites throughout the estuary. We hypothesised that the construction of Wyaralong Dam would act as a nutrient and sediment sink which may have positive effects on downstream water quality. Long-term trends of water quality constituents were evaluated using a non-parametric seasonal Mann-Kendall test and the effect of upstream impoundment was assessed with a Before-After Control-Impact (BACI) test. Nutrient concentrations and turbidity levels declined significantly with time in the upper Logan estuary and, to a lesser extent, in the lower Albert estuary. The general improvement of water quality in the upper Logan estuary was attributed to construction of the Wyaralong Dam. Significant decreases in concentrations of total phosphorus (TP) and oxidised nitrogen (NO x -N) along the lower Albert were principally attributed to wetter conditions over the 15-year dataset, which diluted point-source loads from a nearby wastewater treatment plant (WWTP). Estuaries are focal points for coastal cities worldwide, their habitats frequently transformed into engineered shorelines abutting waters with elevated nutrients in an urbanised landscape. Here we test for relationships between shoreline armouring and nutrients on the diversity and trophic composition of fish assemblages across 22 estuaries in eastern Australia. Urbanisation was associated with fish diversity and abundance, but there were differences in the effects of shoreline armouring and nutrient level on the trophic composition of fish assemblages. Fish diversity and the abundance of most trophic groups, particularly omnivores, zoobenthivores and detritivores, was greatest in highly urban estuaries. We show that estuarine fish assemblages are associated with urbanisation in more nuanced ways than simple habitat transformation would suggest, but this depends on the broader environmental context. Wetting of leaf litter accumulated in riparian zones during rainfall events provides pulses of dissolved organic matter (DOM) to rivers. Restoring riparian vegetation aims to reduce sediment and nutrient transport into rivers, however DOM from leaf litter can stimulate phytoplankton growth and interfere with water treatment processes. Improved understanding of the loads and chemical composition of DOM leached from leaf litter of different plant species, and how subsequent leaching through soils affects DOM retention or transformation, is needed to predict the outcomes of riparian revegetation. To investigate this, we simulated rapid leaching of rainfall through the leaf litter of two riparian tree species with and without subsequent leaching through soil, comparing dissolved organic carbon (DOC) and nitrogen (DON) loads, and DOM chemical composition (via spectroscopic and novel NMR-fingerprinting techniques). Plant source affected the load and composition of DOM leaching, with Eucalyptus tereticornis leaching more DOC than Casuarina cunninghamiana. Additionally, E. tereticornis DOM had a higher sugar, myo-inositol, benzoic acid, flavonoid and oxygenated aromatic content. More than 90% of leaf litter DOM was retained in the soil under simulated repeated heavy rainfall. The DOM chemistry of these species determined the total loads and changes in DOM composition leaching through soil. Less E. tereticornis DOM was retained by the soil than C. cunninghamiana DOM, with sugars, myo-inositol and amino acids being poorly retained compared to fatty acids and aromatic compounds. It also appears that DOM from E. tereticornis litter primed the soil, resulting in more DON being leached compared with bare soil. In comparison, C. cunninghamiana litter resulted in greater retention of DON, oxygenated aromatic compounds and the amino acid tryptophan. This study provides new information on how a range of DOM sources and transformations affect the DOM ultimately leached into waterways, key to developing improved models of DOM transformations in catchments. Restoring woody vegetation to riparian zones helps to protect waterways from excessive sediment and nutrient inputs. However, the associated leaf litter can be a major source of dissolved organic matter (DOM) leached into surface waters. DOM can lead to the formation of disinfection by-products (DBPs) during drinking water treatment. This study investigated the DBPs formed during chlorination of DOM leached from leaf litter and assessed the potential toxicity of DBPs generated. We compared the leachate of two native Australian riparian trees, Casuarina cunninghamiana and Eucalyptus tereticornis, and a reservoir water source from a catchment dominated by Eucalyptus species. Leachates were diluted to dissolved organic carbon concentrations equivalent to the reservoir (~9 mg L -1 ). E. tereticornis leachates produced more trihalomethanes (THMs), haloacetic acids (HAAs), and haloketones after chlorination, while C. cunninghamiana produced more chloral hydrate and haloacetonitriles. Leachate from both species produced less THMs and more HAAs per mole of carbon than reservoir water. This may be because reservoir water had more aromatic, humic characteristics while leaf leachates had relatively more protein-like components. Using in vitro bioassays to test the mixture effects of all chemicals, chlorinated E. tereticornis leachate induced oxidative stress in HepG2 liver cells and bacterial toxicity more frequently and at lower concentrations than C. cunninghamiana and reservoir water. Overall, this study has shown that the DOM leached from litter of these species has the potential to generate DBPs and each species has a unique DBP profile with differing bioassay responses. E. tereticornis may pose a relatively greater risk to drinking water than C. cunninghamiana as it showed greater toxicity in bioassays. Human activities in coastal catchments can cause the accumulation of pollutants in seafood. We quantified the concentration of heavy metals, pesticides and PFASs in the flesh of the fisheries species yellowfin bream Acanthopagrus australis (n = 57) and mud crab Scylla serrata (n = 65) from 13 estuaries in southeast Queensland, Australia; a region with a variety of human land uses. Pollutants in yellowfin bream were best explained by the extent of intensive uses in the catchment. Pollutants in mud crabs were best explained by the extent of irrigated agriculture and water bodies. No samples contained detectable levels of pesticides, and only six samples contained low levels of PFASs. Metals were common in fish and crab flesh, but only mercury in yellowfin bream from the Mooloolah River breached Australian food safety standards. Seagrass ecosystems are inherently dynamic, responding to environmental change across a range of scales. Habitat requirements of seagrass are well defined, but less is known about their ability to resist disturbance. Specific means of recovery after loss are particularly difficult to quantify. Here we assess the resistance and recovery capacity of 12 seagrass genera. We document four classic trajectories of degradation and recovery for seagrass ecosystems, illustrated with examples from around the world. Recovery can be rapid once conditions improve, but seagrass absence at landscape scales may persist for many decades, perpetuated by feedbacks and/or lack of seed or plant propagules to initiate recovery. It can be difficult to distinguish between slow recovery, recalcitrant degradation, and the need for a window of opportunity to trigger recovery. The response of seagrass systems to a severe disturbance provides an opportunity to quantify the degree of resilience in different meadows, and subsequently to test whether there is a genetic basis to resilience. We used existing data on levels of long-standing disturbance from poor water quality, and the responses of seagrass ( Zostera muelleri ) after an extreme flood event in Moreton Bay, Queensland, Australia. Sites were grouped into high and low disturbance categories, in which seagrass showed high and low resilience, respectively, as determined by measuring rates of key feedback processes (nutrient removal, suppression of sediment resuspension, and algal grazing), and physiological and morphological traits. Theoretically, meadows with higher genotypic diversity would be expected to have greater resilience. However, because the more resilient meadows occur in areas historically exposed to high disturbance, the alternative is also possible, that selection will have resulted in a narrower, less diverse subset of genotypes than in less disturbed meadows. Levels of genotypic and genetic diversity (allelic richness) based on 11 microsatellite loci, were positively related ( R 2 = 0.58). Genotypic diversity was significantly lower at highly disturbed sites ( R = 0.49) than at less disturbed sites ( R = 0.61). Genotypic diversity also showed a negative trend with two morphological characteristics known to confer resilience on seagrass in Moreton Bay, leaf chlorophyll concentrations and seagrass biomass. Genetic diversity did not differ between disturbed and undisturbed sites. We postulate that the explanation for these results is historical selection for genotypes that confer protection against disturbance, reducing diversity in meadows that contemporarily show greater resilience.

Seagrass meadows are vital ecosystems in coastal zones worldwide, but are also under global threat. One of the major hurdles restricting the success of seagrass conservation and restoration is our limited understanding of ecological feedback mechanisms. In these ecosystems, multiple, self-reinforcing feedbacks can undermine conservation efforts by masking environmental impacts until the decline is precipitous, or alternatively they can inhibit seagrass recovery in spite of restoration efforts. However, no clear framework yet exists for identifying or dealing with feedbacks to improve the management of seagrass ecosystems. Here we review the causes and consequences of multiple feedbacks between seagrass and biotic and/or abiotic processes. We demonstrate how feedbacks have the potential to impose or reinforce regimes of either seagrass dominance or unvegetated substrate, and how the strength and importance of these feedbacks vary across environmental gradients. Although a myriad of feedbacks have now been identified, the co-occurrence and likely interaction among feedbacks has largely been overlooked to date due to difficulties in analysis and detection. Here we take a fundamental step forward by modelling the interactions among two distinct above- and belowground feedbacks to demonstrate that interacting feedbacks are likely to be important for ecosystem resilience. On this basis, we propose a five-step adaptive management plan to address feedback dynamics for effective conservation and restoration strategies. Major flood events can dramatically alter the coastal sediment environment. This study established the current sediment distribution in a large sub-tropical embayment, Moreton Bay, Australia, and examined the effect of three recent floods on modifying this distribution. In 2015, surface sediment samples were collected from 223 sites across the study area and analysed for particle size distribution with the resultant sediment distribution mapped. In addition, sampling of flood waters during two major events in 2011 and 2013 was undertaken and particle size distribution of suspended sediment was determined. Data was compared to the result of an earlier large-scale survey completed in 1970, with three large flood events occurring between the two surveys. The sediment environment has undergone a dramatic change with muddy sediments now covering an estimated area of over 860km 2 , more the double the area found in 1970. The relationship between ecological impact and ecosystem structure is often strongly nonlinear, so that small increases in impact levels can cause a disproportionately large response in ecosystem structure. Nonlinear ecosystem responses can be difficult to predict because locally relevant data sets can be difficult or impossible to obtain. Bayesian networks (BN) are an emerging tool that can help managers to define ecosystem relationships using a range of data types from comprehensive quantitative data sets to expert opinion. We show how a simple BN can reveal nonlinear dynamics in seagrass ecosystems using ecological relationships sourced from the literature. We first developed a conceptual diagram by cataloguing the ecological responses of seagrasses to a range of drivers and impacts. We used the conceptual diagram to develop a BN populated with values sourced from published studies. We then applied the BN to show that the amount of initial seagrass biomass has a mitigating effect on the level of impact a meadow can withstand without loss, and that meadow recovery can often require disproportionately large improvements in impact levels. This mitigating effect resulted in the middle ranges of impact levels having a wide likelihood of seagrass presence, a situation known as bistability. Finally, we applied the model in a case study to identify the risk of loss and the likelihood of recovery for the conservation and management of seagrass meadows in Moreton Bay, Queensland, Australia. We used the model to predict the likelihood of bistability in 23 locations in the Bay. The model predicted bistability in seven locations, most of which have experienced seagrass loss at some stage in the past 25 years providing essential information for potential future restoration efforts. Our results demonstrate the capacity of simple, flexible modeling tools to facilitate collation and synthesis of disparate information. This approach can be adopted in the initial stages of conservation programs as a low-cost and relatively straightforward way to provide preliminary assessments of.nonlinear dynamics in ecosystems.

Introduction: Marked changes occur in the collagen framework of the heart following acute ischemia, which is associated with adverse ventricular remodelling. Plasma markers of collagen turnover are useful in the assessment of remodelling and have predictive value, but their exact temporal dynamics following ischemia are unclear., Objective: To characterize the early temporal dynamics of plasma markers of collagen turnover in a human model of coronary artery occlusion., Methods: Fourteen patients undergoing elective percutaneous coronary intervention (PCI) to a single coronary artery were recruited in addition to a control group of eight patients undergoing elective diagnostic coronary arteriography. Sequential assessment of plasma levels of procollagen type I carboxyterminal propeptide and C-telopeptide for type I collagen (CITP) as markers of synthesis and degradation, respectively, was performed over a 16 h period., Results: The ischemic burden in the PCI group was high, with 13 of the 14 patients demonstrating transient ST segment shift or positive troponin. Mean plasma levels of CITP on admission were 3.1 ng⁄mL and 3.0 ng⁄mL in the PCI and control groups, respectively (P value nonsignificant). There was a sequential increase in plasma CITP following PCI, peaking at 4.7 ng⁄mL at 16 h (P<0.01), with no change in the control group. There were no significant changes in plasma levels of procollagen type I carboxyterminal propeptide in either group., Conclusions: Plasma levels of CITP demonstrated early temporal dynamics of collagen degradation following transient coronary artery occlusion supporting the use of plasma markers of collagen turnover as an early tool in the assessment of the remodelling process following myocardial ischemia.

Stable nitrogen isotopes were used to assess the effects of wastewater treatment plant (WWTP) upgrades on the utilisation of sewage-N by estuarine biota in Moreton Bay, Australia. We measured delta(15)N of filamentous algae, mangrove leaves and shore crabs at the Brisbane and Logan Rivers before and after scheduled WWTP upgrades, and at two reference rivers where WWTPs had been upgraded >4 years previously. The total N discharged into Brisbane River decreased by >80% after the upgrades had occurred, but N loads remained similar at Logan River despite the upgrade. In Brisbane River, delta(15)N values of algae and crabs decreased and were comparable to the reference rivers within 1-2 years but no changes occurred at Logan River. The delta(15)N of mangrove leaves remained elevated in all rivers, indicating that sewage-N remained a major source to mangroves either from residual WWTP discharges or from N accumulated in the sediments over many years.

Introduction: Non-ST elevation acute coronary syndrome (ACS) represents a spectrum of risk, with electrocardiographic (ECG) changes and a positive troponin being associated with higher morbidity and mortality. Ischaemia produces alterations in the collagenous component of the heart, even in the absence of myocyte necrosis. Collagen turnover can be assessed biochemically with C-propeptide for type I collagen (PICP) and C-telopeptide for type I collagen (CITP) being markers of collagen synthesis and degradation respectively. Tissue inhibitor of matrix metalloproteinase-1 (TIMP-1) is a marker of inhibition of degradation., Methods: Fifty-two patients with non-ST elevation acute ACS were recruited and dichotomised into high- and low-risk groups based on ECG and troponin level. Sequential measurements of plasma PICP, CITP and TIMP-1 were performed over a 48 hour period., Results: Twenty were classified as low-risk (negative troponin and normal ECG) and 32 as high-risk. PICP was within the normal range at all time points in both groups. However, admission CITP was higher in the high-risk group (3.7 vs. 2.6 ng/ml, p<0.001) and, unlike the low-risk group, demonstrated a further rise over 48 h. Similarly, mean TIMP-1 displayed a sequential change over time in the high-risk group only, and admission level was higher compared to the low-risk group (302 vs. 221 ng/ml, p<0.01)., Discussion: There is serological evidence of time-dependent altered collagen metabolism in high-risk ACS, which is not present in the low-risk group. This may reflect a degree of remodeling and may aid risk stratification of patients presenting with non-ST elevation ACS.

Hepatic damage associated with chronic hepatitis B (CHB) relies on measurement of serum transaminases and asssessment of hepatic histology. We determined if serum hepatic function tests, including alpha-glutathione-S-transferase ((GST), were of value in monitoring or predicting the effect of lamivudine therapy for CHB. Thirty-nine patients received orally 100 mg of lamivudine daily for 48 weeks. Blood samples were obtained at baseline and at 24 and 48 weeks. At the end of the treatment period the patients were then divided into four groups according to the pattern of HBs and HBe antigens. At baseline and at 24 weeks ALT, AST, and (GST had lower values in the complete response compared to the complete failure groups. Using ROC analysis, only ALT at 24 weeks (area under the curve = 0.803) had significant diagnostic ability in detecting responders. These results reaffirm the value of measuring serum ALT as an indicator of treatment response and provide information on the potential use of (GST as an additional prognostic biomarker in this patient group.

Background: Infarct expansion characterises early ventricular remodeling following myocardial infarction (AMI) and is a product of the balance between collagen degradation and synthesis. Serological markers of collagen turnover may help in predicting those at risk of remodeling. C-propeptide for type-I collagen (PICP) and C-telopeptide for type-I collagen (CITP) are markers of collagen synthesis and degradation, respectively., Methods: Fifty-one patients with AMI were recruited and dichotomised by echocardiographic wall motion index (WMI). Sequential measurements of plasma PICP and CITP were correlated to this and other echocardiographic variables of remodeling., Results: Twenty-three normal WMI, 28 abnormal WMI. Both groups showed increases in PICP and CITP over time. However, mean admission CITP higher in abnormal WMI group, 4.5 vs. 3.1 ng/ml (p<0.05) as was peak, 6.3 vs. 4.8 ng/ml (p<0.05). Conversely, admission PICP was lower in abnormal WMI group 114 vs. 143 ng/ml (p<0.05). Admission CITP correlated with WMI, r=0.53, p<0.001. CITP>3.2 ng/ml (normal mean+2S.D.) had 74% positive predictive value for abnormal WMI, negative predictive value 65%. Admission CITP negatively correlated with mitral deceleration time (Dt), r=-0.38, p=0.01. CITP>3.2 was associated with lower Dt-183 vs. 221 ms, p<0.05., Conclusion: There is serological evidence of sequential increases in both collagen synthesis and degradation following AMI. However, the balance between these differs in patients who undergo remodeling, manifested by abnormal WMI and reduced Dt, compared to those with no evidence. They have relatively increased degradation and reduced synthesis, favouring net collagen breakdown. These changes occur early with evidence of increased breakdown on admission predicting early remodeling and support the role of serological markers to identify patients at risk of this.

Background/aims: The role of transforming growth factor-beta 1 (TGF-beta 1) in liver fibrosis is in part related to impairment of extracellular matrix breakdown by stimulation of tissue inhibitor of metalloproteinases-1 (TIMP-1) gene. The aim of the study was to evaluate association between TGF-beta 1 and TIMP-1 in relation to liver injury in chronic viral hepatitis B and C., Methodology: Association between plasma TGF-beta 1 and TIMP-1 was evaluated in 28 consecutive patients undergoing liver biopsy for chronic viral hepatitis B and C (CH-B, CH-C) and these tests were correlated with hepatic fibrosis, inflammation and liver function tests. Moreover carboxyterminal cross-linked telopeptide of type 1 procollagen (ICTP) and carboxyterminal propeptide of type 1 collagen (PICP) were also measured for assessment of extracellular matrix breakdown or synthesis, respectively., Results: Chronic viral hepatitis B and C resulted in a significant increase in plasma TIMP-1 levels but not TGF-beta 1. Among biochemical markers of liver injury, significant correlation with TGF-beta 1 and TIMP-1 was demonstrated in respect to aminotransferase activities in both groups. TIMP-1 showed significant correlation with ICTP levels in both CH-B (r = 0.59) and CH-C (r = 0.62), whereas TGF-beta 1 was correlated with ICTP only in CH-C patients (r = 0.75). PICP did not demonstrate any correlation with either TGF-beta 1 or TIMP-1. Hepatic fibrosis, but not inflammation, correlated significantly with TGF-beta 1 (CH-B: r = 0.73; CH-C: r = 0.79) and TIMP-1 (CH-B: r = 0.66; CH-C: r = 0.71) in both groups and there was a significant correlation between TIMP-1 and TGF-beta 1 in the CH-B group (r = 0.83) and CH-C group (r = 79)., Conclusions: These results support the role of TIMP-1 in a TGF-beta 1-dependent mechanism for liver fibrosis and suggest their plasma levels can be used as a possible early non-invasive marker of liver fibrosis useful for chronic hepatitis management.

This study was designed to document noninvasively the pathological mechanisms responsible for myocardial fibrosis and to assess the clinical utility of plasma markers of collagen synthesis and degradation as screening tools for the assessment of fibrosis in hypertension. We studied 100 never-treated hypertensive patients and 50 normal subjects. Echocardiographic assessment was made of left ventricular (LV) mass and diastolic filling using measurement of E:A ratio, E wave deceleration time (E dec), and isovolumic relaxation time (IVRT). The presence of diastolic dysfunction was taken as a surrogate marker for the presence of myocardial fibrosis. Plasma carboxy-terminal propeptide of collagen type I (PICP), carboxy-terminal telopeptide of collagen type I (CITP), and tissue inhibitor of matrix metalloproteinases type I (TIMP-1) were measured as markers of collagen synthesis, degradation, and inhibition of degradation, respectively. Plasma TIMP-1 was significantly elevated in the hypertensive cohort (358 ng/mL versus 253 ng/mL, P<0.001) as were CITP (5.2 microg/L versus 2.9 microg/L, P<0.001), and PICP (200 microg/L versus 166 microg/L, P<0.05). TIMP-1 was significantly elevated in patients with diastolic dysfunction (421 ng/mL versus 283 ng/mL P<0.01) and correlated with markers of diastolic filling, namely E:A ratio (r=0.26, P<0.05) and E Dec (r=0.41, P<0.01). A plasma TIMP-1 level of >500 ng/mL had a specificity of 97% and a positive predictive value of 96% in predicting diastolic dysfunction. In patients with untreated hypertension, there is evidence of increased collagen synthesis, degradation, and inhibition of degradation resulting in fibrosis. Our results demonstrate that plasma TIMP-1 correlates with markers of LV diastolic filling, is predictive of LV dysfunction, and is a potential noninvasive marker of fibrosis.

Background: Essential hypertensive patients have an increased heart and arterial collagen concentration. Increased collagen synthesis can be assessed using procollagen III N peptide (PIIINP) and reduced collagen degradation measured using tissue inhibitor of metalloproteinase-1 (TIMP-1)., Methods: Plasma TIMP-1 and PIIINP levels were measured in 31 patients with essential hypertension and in 17 normotensive control subjects. The hypertensive patients were either treatment naive (n = 18) or had been without treatment for 1 month (n = 13). Both groups of patients were screened to exclude other fibrotic diseases., Results: In the hypertensive patients, TIMP-1 levels were significantly (P < .0002) elevated (median 380 ng/ mL, range 160 to 1,560 ng/mL) compared with those of the normotensive control subjects (median 178 ng/mL, range 99 to 330 ng/mL). In hypertensive subjects who had never received antihypertensive therapy there were significant correlations between TIMP-1 and left ventricular posterior wall thickness in diastole (LVPWd) (r = 0.58) (P < .02) and left ventricular mass index (r = 0.58) (P < .02). There was no difference in PIIINP levels (mean +/- 2 SD) between the hypertensive (0.56 U/mL +/- 0.3) and normotensive groups (0.52 U/mL +/- 0.2)., Conclusions: The increased tissue collagen III levels found in the heart and vessels of hypertensive patients is due to a reduction in collagen degradation because of high TIMP-1 levels, rather than an increase in synthesis of collagen type III. The tissue source of this TIMP-1 is unclear.
