The generation of a current by light is a key process in optoelectronic and photovoltaic devices. In band semiconductors, depletion fields associated with interfaces separate long-lived photo-induced carriers. However, in systems with strong electron-electron and electron-phonon correlations it is unclear what physics will dominate the photoresponse. Here, we investigate photocurrent in VO2, an exemplary strongly correlated material known for its dramatic metal-insulator transition at Tc ? 68 °C, which could be useful for optoelectronic detection and switching up to ultraviolet wavelengths. Using scanning photocurrent microscopy on individual suspended VO2 nanobeams we observe a photoresponse peaked at the metal-insulator boundary but extending throughout both insulating and metallic phases. We determine that the response is photothermal, implying efficient carrier relaxation to a local equilibrium in a manner consistent with strong correlations. Temperature-dependent measurements reveal subtle phase changes within the insulating state. We further demonstrate switching of the photocurrent by optical control of the metal-insulator boundary arrangement. Our work shows the value of applying scanning photocurrent microscopy to nanoscale crystals in the investigation of strongly correlated materials, and the results are relevant for designing and controlling optoelectronic devices employing such materials. Abstract: A feasible way to perform evolutionary analyses is to compare characters divergent enough to observe significant differences, but sufficiently similar to exclude saturation of the differences that occurred. Thus, comparisons of invertebrate mitochondrial (mt) genomes at low taxonomic levels can be extremely helpful in investigating patterns of variation and evolutionary dynamics of genomes, as intermediate stages of the process may be identified. Fortunately, in this study, we newly sequenced the mt genome of the eighth member of Asian Crassostrea oysters which can provide necessary intermediate characters for us to believe that the variation of Crassostrea mt genomes is considerably greater than previously acknowledged. Several new features of Asian Crassostrea oyster mitochondrial genomes were revealed, and our results are particularly significant as they 1) suggest a novel model of alloacceptor tRNA gene recruitment, namely “vertical” tRNA gene recruitment, which can be successfully used to explain the origination of the unusually additional trnK and trnQ genes (annotated as trnK 2 and trnQ 2 respectively) in the mt genomes of the five Asian oysters, and we speculate that this recruitment progress may be a common phenomenon in the evolution of the tRNA multigene family; 2) reveal the existence of two additional, lineage-specific, mtDNA-encoded genes that may originate from duplication of nad2 followed by rapid evolutionary change. Each of these two genes encodes a unique amino terminal signal peptide, thus each might possess an unknown function; and 3) identify for the first time the atp8 gene in oysters. The present study thus gives further credence to the comparison of congeneric bivalves as a meaningful strategy to investigate mt genomic evolutionary trends in genome organization, tRNA multigene family, and gene loss and/or duplication that are difficult to undertake at higher taxonomic levels. Abstract: Yb-doped PbClF crystals were obtained by a modified Bridgman method. Broadband emission (FWHM=58nm) with long lifetime (5.33ms) was observed in 2.0% Yb3+-doped PbClF crystals. The Stark splitting of Yb3+ 2F7/2 level was calculated to be 801cm−1 in 0.5% Yb3+-doped PbClF crystals, and the corresponding population of lower laser level is about 2.13%. All the results indicated that Yb-doped PbClF crystals should be a promising material to ultrashort pulses output at 1.0μm. [Copyright &y& Elsevier]

Circadian clocks generate endogenous rhythms in most organisms from cyanobacteria to humans and facilitate entrainment to environmental diurnal cycles, thus conferring a fitness advantage. Both transcriptional and posttranslational mechanisms are prominent in the basic network architecture of circadian systems. Posttranscriptional regulation, including mRNA processing, is emerging as a critical step for clock function. However, little is known about the molecular mechanisms linking RNA metabolism to the circadian clock network. Here, we report that a conserved SNW/Ski-interacting protein (SKIP) domain protein, SKIP, a splicing factor and component of the spliceosome, is involved in posttranscriptional regulation of circadian clock genes in Arabidopsis thaliana. Mutation in SKIP lengthens the circadian period in a temperature-sensitive manner and affects light input and the sensitivity of the clock to light resetting. SKIP physically interacts with the spliceosomal splicing factor Ser/Arg-rich protein45 and associates with the pre-mRNA of clock genes, such as PSEUDORESPONSE REGULATOR7 (PRR7) and PRR9 , and is necessary for the regulation of their alternative splicing and mRNA maturation. Genome-wide investigations reveal that SKIP functions in regulating alternative splicing of many genes, presumably through modulating recognition or cleavage of 5′ and 3′ splice donor and acceptor sites. We recently described the discovery of oxygenated N-alkyl deoxynojirimycin (DNJ) derivative 7(CM-10–18) with antiviral activity against dengue virus (DENV)infection both in vitro and in vivo. This imino sugar was promisingbut had an EC50against DENV in BHK cells of 6.5 μM,which limited its use in in vivo. Compound 7presentedstructural opportunities for activity relationship analysis, whichwe exploited and report here. These structure–activity relationshipstudies led to analogues 2h, 2l, 3j, 3l, 3v, and 4b–4cwith nanomolar antiviral activity (EC50= 0.3–0.5μM) against DENV infection, while maintaining low cytotoxicity(CC50> 500 μM, SI > 1000). Abstract: In the present study, AA6061-T4 T-joints were successfully fabricated by friction stir welding (FSW) in three different combination ways of skins and stringers. Distributions and formation mechanisms of tunnel defects, kissing bond defects, original joint line with severe plastic deformation (OJLwSPD) defects, and zigzag line defects in T-joints were investigated by macro- and micro-observations. Influences of defects distributions and welding parameters on the tensile behaviors of T-joints were examined. Nd-doped PbClF crystals were grown with modified Bridgman method. Broad and flat emission band at 1.0 μm was observed. The FWHM value of 0.0091 wt.% Nd3+-doped PbClF crystal at 1064 nm reached up to 30 nm with the peak emission cross-section of 1.05×10-19 cm2. Second order optical nonlinear processes involve the coherent mixing of two electromagnetic waves to generate a new optical frequency, which plays a central role in a variety of applications, such as ultrafast laser systems, rectifiers, modulators, and optical imaging. However, progress is limited in the mid-infrared (MIR) region due to the lack of suitable nonlinear materials. It is desirable to develop a robust system with a strong, electrically tunable second order optical nonlinearity. Here, we demonstrate theoretically that AB-stacked bilayer graphene (BLG) can exhibit a giant and tunable second order nonlinear susceptibility χ(2) once an in-plane electric field is applied. χ(2) can be electrically tuned from 0 to ∼105 pm/V, 3 orders of magnitude larger than the widely used nonlinear crystal AgGaSe2. We show that the unusually large χ(2) arise from two different quantum enhanced two-photon processes thanks to the unique electronic spectrum of BLG. The tunable electronic bandgap of BLG adds additional tunability on the resonance of χ(2), which corresponds to a tunable wavelength ranging from ∼2.6 to ∼3.1 μm for the up-converted photon. Combined with the high electron mobility and optical transparency of the atomically thin BLG, our scheme suggests a new regime of nonlinear photonics based on BLG. [ABSTRACT FROM AUTHOR]

The combination of its high electron mobility, broadband absorption and ultrafast luminescence make graphene attractive for optoelectronic and photonic applications, including transparent electrodes, mode-locked lasers and high-speed optical modulators. Photo-excited carriers that have not cooled to the temperature of the graphene lattice are known as hot carriers, and may limit device speed and energy efficiency. However, their roles in charge and energy transport are not fully understood. Here, we use time-resolved scanning photocurrent microscopy to demonstrate that hot carriers, rather than phonons, dominate energy transport across a tunable graphene p-n junction excited by ultrafast laser pulses. The photocurrent response time varies from 1.5 ps at room temperature to 4 ps at 20 K, implying a fundamental bandwidth of ?500 GHz (refs 12, 13, 21). Gate-dependent pump-probe measurements demonstrate that both thermoelectric and built-in electric field effects contribute to the photocurrent, with the contribution from each depending on the junction configuration. The photocurrent produced by a single pulsed laser also displays multiple polarity reversals as a function of carrier density, which is a possible signature of impact ionization. Abstract: The Mn-, Cr-doped and Mn, Cr-co-doped MgAl2O4 powders have been synthesized via a gel–solid reaction method. Energy transfer from Mn2+ to Cr3+ has been observed for the first time in the co-doped MgAl2O4 phosphors. When excited with blue light with a wavelength of 450nm at room temperature, both green emission from Mn2+ around 520nm and red emission from Cr3+ around 675and 693nm were generated. Moreover, the color of the emission can be modified by controlling the doping concentrations of Mn2+ and Cr3+. Therefore, MgAl2O4: Mn2+, Cr3+ could be used as a single-phased phosphor for white LED with a blue LED chip. Abstract: Molecular beacon (MB) is especially suited for detection of single nucleotide polymorphism (SNP), and the type of MB immobilized on the surface of microarray in particular, may detect multi-sample and multi-locus. However, the majority of MB needs to be labeled with fluorescence and quenching molecules on the two ends of the probe, and observed the reaction of fluorescence or complicated electrochemical signal produced hybridization of MB and target sequence by complex and expensive instruments. The “molecular beacon” and microarray designed appropriately in our study can produce visible light response signal induced by amplification effect of enzymatic color, and are avoided with the marker of fluorescence and quenching molecules and expensive instruments. The “molecular beacon” without fluorescence and quenching molecules is entitled as “hairpin DNA probe” by us for only the “hairpin” structure of traditional molecular beacon is adopted. The merits of two techniques, molecular beacon and amplification effect of enzymatic color, are successfully combined, and the technique is simple, sensitive and specific, to detect and compare the methylenetetrahydrofolate reductase (MTHFR) Gene C677T mutation of subjects between coronary heart disease (CHD) and control group. Abstract: Friction stir spot welding (FSSW) is a newly-developed solid state joining technology. In this study, two types of FSSW, normal FSSW and walking FSSW, are applied to join the 5052-H112 aluminum alloy sheets with 1mm thickness and then the effect of the rotational speed and dwell time on microstructure and mechanical properties is discussed. The lower sheet material underneath the hook didn’t flow into the upper sheet due to the concave surface in the shoulder and groove in the anvil. The hardness profile of the welds exhibited a W-shaped appearance and the minimum hardness was measured in the HAZ. The results of tensile/shear tests and cross-tension tests indicate that the joint strength decreases with increasing rotational speed, while it’s not affected significantly by dwell time. At the rotational speed of 1541rpm, the tensile/shear strength and cross-tension strength reached the maximum of 2847.7N and 902.1N corresponding to the dwell time of 5s and 15s. Two different fracture modes were observed under both tensile/shear and cross-tension loadings: shear fracture and tensile/shear mixed fracture under tensile/shear loadings, and nugget debonding and pull-out under cross-tension loadings. The performance of the welds plays a predominant role in determining the type of fracture modes. In addition, the adoption of walking FSSW brings unremarkable improvements in weld strength. Abstract: In this work, the ion transfer at the dual micro-liquid/liquid interfaces formed at the orifice of a dual micropipette is reported. The goal is to use one channel (“generator”) to support a liquid/liquid interface where ions can be transferred from the inner phase (water) to the outer phase (1,2-dichloroethane, DCE), and another micropipette (“collector”) in vicinity to collect the ions from the outer phase back into the inner phase. Generation/collection voltammetry has been employed and pyridine is chosen as a model compound to study the dependence of collection efficiency upon applied potentials and pH of generator and collector, respectively. The applied potential of the generator has been found no obvious influence on collection efficiency (η). Collection efficiency decreases from maximum to zero as applied potential of collector increases, and the relationship between η and applied potential and pH is discussed. We also demonstrate that the dependence of collection efficiency upon collector pH is rather complicated due to different mechanisms involved at below, near, or above pK a of pyridine. The present study investigated the protective effect of l-citrulline on gastric mucosal injury induced by ischemia-reperfusion (IR) in rats. Under anesthesia, the celiac artery was clamped for 30 min, and then the clamp was removed for 60 min reperfusion. Sixty minutes before ischemia, l-citrulline was administered intragastrically at doses of 300, 600, and 900 mg/kg. After the experiment, the stomachs were removed for biochemical and histological examinations. Pretreatment with l-citrulline (300, 600, and 900 mg/kg) significantly ameliorated the gastric damage caused by IR. Moreover, l-citrulline prevented the production of lipid peroxidation and inhibited the increase of myeloperoxidase activity. The elevation in total nitric oxide synthase (NOS) activity, inducible NOS activity, and inducible NOS protein expression as well as the decrease in constitutive NOS activity and gastric mucus level in the gastric mucosa induced by IR were significantly prevented. However, the protective effect mediated by l-citrulline was significantly antagonized by coadministration of l-nitroarginine methyl ester (10 mg/kg, s.c.). These results suggest that part of the mechanism of gastric protection by l-citrulline might be through inhibiting neutrophil infiltration and preserving gastric mucus synthesis and secretion in rats, functions that are closely related to the maintenance of constitutive NOS activity. Abstract: Cellular α-glucosidases I and II are enzymes that sequentially trim the three terminal glucoses in the N-linked oligosaccharides of viral envelope glycoproteins. This process is essential for the proper folding of viral glycoproteins and subsequent assembly of many enveloped viruses, including dengue virus (DENV). Imino sugars are substrate mimics of α-glucosidases I and II. In this report, we show that two oxygenated alkyl imino sugar derivatives, CM-9-78 and CM-10-18, are potent inhibitors of both α-glucosidases I and II in vitro and in treated animals, and efficiently inhibit DENV infection of cultured human cells. Pharmacokinetic studies reveal that both compounds are well tolerated at doses up to 100mg/kg in rats and have favorable pharmacokinetic properties and bioavailability in mice. Moreover, we showed that oral administration of either CM-9-78 or CM-10-18 reduces the peak viremia of DENV in mice. Interestingly, while treatment of DENV infected mice with ribavirin alone did not reduce the viremia, combination therapy of ribavirin with sub-effective dose of CM-10-18 demonstrated a significantly enhanced antiviral activity, as indicated by a profound reduction of the viremia. Our findings thus suggest that combination therapy of two broad-spectrum antiviral agents may provide a practically useful approach for the treatment of DENV infection. [ABSTRACT FROM AUTHOR]

Abstract: Recombinant expression systems differ in the type of glycosylation they impart on expressed antigens such as the human immunodeficiency virus type 1 (HIV-1) envelope glycoproteins, potentially affecting their biological properties. We performed head-to-head antigenic, immunogenic and molecular profiling of two distantly related Env surface (gp120) antigens produced in different systems: (a) mammalian (293 FreeStyle™ cells; 293F) cells in the presence of kifunensine, which impart only high-mannose glycans; (b) insect cells (Spodoptera frugiperda, Sf9), which confer mainly paucimannosidic glycans; (c) Sf9 cells recombinant for mammalian glycosylation enzymes (Sf9 Mimic™), which impart high-mannose, hybrid and complex glycans without sialic acid; and (d) 293F cells, which impart high-mannose, hybrid and complex glycans with sialic acid. Molecular models revealed a significant difference in gp120 glycan coverage between the Sf9-derived and wild-type mammalian-cell-derived material that is predicted to affect ligand binding sites proximal to glycans. Modeling of solvent-exposed surface electrostatic potentials showed that sialic acid imparts a significant negative surface charge that may influence gp120 antigenicity and immunogenicity. Gp120 expressed in systems that do not incorporate sialic acid displayed increased ligand binding to the CD4 binding and CD4-induced sites compared to those expressed in the system that do, and imparted other more subtle differences in antigenicity in a gp120 subtype-specific manner. Non-sialic-acid-containing gp120 was significantly more immunogenic than the sialylated version when administered in two different adjuvants, and induced higher titers of antibodies competing for CD4 binding site ligand–gp120 interaction. These findings suggest that non-sialic-acid-imparting systems yield gp120 immunogens with modified antigenic and immunogenic properties, considerations that should be considered when selecting expression systems for glycosylated antigens to be used for structure–function studies and for vaccine use. Abstract: A laser crystal Er3+:YbVO4 has been grown by the Czochralski method with excellent quality. The rocking curve from the (004) diffraction plane of the as-grown Er3+:YbVO4 crystal was measured and the full-width at half-maximum value was found to be 19.80in. for the (004) face. The effective segregation coefficient of Er3+ was studied by X-ray fluorescence and the crystal structure was determined by means of X-ray diffraction analysis. The polarized absorption spectra and the fluorescence spectra of Er3+:YbVO4 were measured at room temperature. The spectral parameters were calculated based on the Judd–Ofelt theory, and the intensity parameters Ω 2, Ω 4 and Ω 6 are found to be 5.50×10−20, 1.96×10−20 and 2.34×10−20 cm2, respectively. The emission cross-section has been calculated by the Fuechtbauer–Ladenbury method. The spectroscopic parameters of Er3+:YbVO4 are compared with other typical laser hosts. [Copyright &y& Elsevier]

Abstract: The aminoglycoside, geneticin (G418), was recently shown to have antiviral activity against bovine viral diarrhea virus (BVDV). Since BVDV, dengue virus (DENV) and yellow fever virus (YFV) all belong to the Flaviviridae family, it seemed possible that a common step in their life cycle might be affected by this aminoglycoside. Here it is shown that geneticin prevented the cytopathic effect (CPE) resulting from DENV-2 infection of BHK cells, in a dose-dependent manner with an 50% effective concentration (EC50) value of 3±0.4μg/ml. Geneticin had no detectable effect on CPE caused by YFV in BHK cells. Geneticin also inhibited DENV-2 viral yield with an EC50 value of 2±0.1μg/ml and an EC90 value of 20±2μg/ml. With a CC50 value of 165±5μg/ml, the selectivity index of anti-DENV activity of geneticin in BHK cells was established to be 66. Furthermore, 25μg/ml of geneticin nearly completely blocked plaque formation induced by DENV-2, but not YFV. In addition, geneticin, inhibited DENV-2 viral RNA replication and viral translation. Gentamicin, kanamycin, and the guanidinylated geneticin showed no anti-DENV activity. Neomycin and paromomycin demonstrated weak antiviral activity at high concentrations. Finally, aminoglycoside-3′-phosphotransferase activity of neomycin-resistant gene abolished antiviral activity of geneticin. [Copyright &y& Elsevier]

Abstract: Using 10-year lightning localization data observed by the TRMM (Tropical Rainfall Measuring Mission) Lightning Imaging Sensor (LIS), the relationship between lightning activity and a series of convective indices was investigated over nine monsoon-prone areas of China in which high-impact weather (HIW) events are frequently observed. Two methods were used to verify and reconstruct LIS lightning data. First, LIS lightning flash data were verified by both surface thunderstorm reports and ground-based lightning detection data. Seasonal, monthly, and 5-day distributions of LIS observed lightning activity agree well with the surface reports and ground-based lightning observations. Second, due to LIS''s low sampling frequency, a data reconstruction and compensation scheme for LIS lightning observations was designed using both LIS lightning seasonal diurnal cycles and surface thunderstorm reports. After data reconstruction, five lightning products were derived: daily mean and maximum LIS flash rate, daily mean and maximum LIS lightning cell rate, and number of lightning days per five day period. Then, a series of convective indices describing convection conditions were derived from radiosonde data according to atmospheric instability and convective potential analysis. Correlation analysis for each study region was done between 10-year lightning derived products and corresponding convective indices by 5-day periods. The correlation analysis results show that higher lightning flash rate and lightning probability are associated with more unstable air and smaller vertical wind shear in a nearly saturated lower layer in most of the study regions. But the correlation varies from region to region. The best correlation between lightning activity and convective indices was found in eastern and southern China, whereas the correlation is lowest in some inland or basin topography regions in which topographic effects are more significant. Moreover, ambient moisture plays a much more important role in the convective development of thunderstorms in southern China than other regions. Thunderstorm development mechanism differences among regions were also discussed. Based on the close relationship between lightning activity and convective indices, some regression equations for forecasting 5-day mean or maximum LIS lightning flash rate and lightning area (a thunderstorm cell) rate, and 5-day lightning days for the study regions were developed using convective indices as predictors. The verifications show that the convective index-based lightning forecast methods can provide a reasonable lightning outlook including probability and lightning flash rate forecasts for a 5-day period. A Nd:BGSO crystal with the dimensions of Φ2 × 131 mm3 has been grown successfully by the micro-pulling-down method. The peak absorption cross-section at 808 nm was calculated to 0.98 × 10−20 cm2 with full-width at half-maximum of 17.4 nm. The stimulated emission cross-section was obtained to be 3.15 × 10−20 cm2 at 1063 nm. The fluorescence and radiative lifetimes of 4F 3/2 level were 248 μs and 300 μs respectively. Ho3+-doped GdScO 3 crystal was successfully grown by the edge-defined film-fed growth (EFG) method. The spectroscopic properties of Ho:GdScO 3 crystal were investigated at room temperature. The peak absorption cross-sections were 0.33 × 10−20 cm2 and 1.24 × 10−20 cm2 at 1160 nm and 653 nm with full widths at half maximum of 171.7 nm and 11.5 nm. The peak emission cross-sections of 5I 7 →5I 8 and 5I 6 →5I 7 transitions for the crystal were 0.49 × 10−20 cm2 and 1.24 × 10−20 cm2, respectively. The fluorescence lifetimes of the 5I 6 and 5I 7 multiplets were measured to be 1.12 ms and 6.13 ms, respectively. Tm3+-doped bismuth germanate crystals (Tm:BGO) with various doping concentrations of 0.5, 0.75, 1.0, 1.5, and 2.0 at.% were grown by the micro-pulling-down method (μ-PD). The effect of thulium concentration on the growth, structure, absorption spectra, emission spectra and fluorescence lifetime of 3H 4 and 3F 4 state was studied. Under excitation of 792 nm, Tm:BGO crystals exhibit two fluorescence bands corresponding to the 3H 4 → 3F 4 and the 3F 4 → 3H 6 transitions. High-quality Dy:PbWO 4 crystal has been successfully grown by the Bridgman method. Fluorescence properties of the grown crystal at the yellow-light region were discussed. The absorption cross-section of Dy3+ ions at 454 nm was calculated to be 0.142 × 10−20 cm2, which corresponded to the transition from 6H 15/2 to 4I 15/2 level. Based on Judd-Ofelt theory, the J-O intensity parameters Ω t and the emission cross-section were calculated. Tm3+ doped and Tm3+, Pr3+ co-doped Lu 3 Al 5 O 12 (LuAG) garnet single crystals have been successfully grown by the micro-pulling-down (μ-PD) method. The absorption spectra, fluorescence spectra and lifetimes were investigated at room temperature. In the absorption spectra, the transition centered at 782 nm matches well with the GaAlAs LDs laser pumping. The incorporation of Pr3+ into Tm3+-doped LuAG crystals enhanced the intensity of 1.5 μm emission and decreased the fluorescence lifetime of the lower energy level for 1.5 μm emission from 10.427 ms to 2.603 ms. Abstract: Ytterbium-doped calcium pyroniobate single crystal has been grown for the first time. Spectral properties of Yb: Ca2Nb2O7 were investigated by emission and absorption spectra. Its cooperative luminescence and fluorescence lifetime were also studied. Yb ions in Ca2Nb2O7 showed very broad absorption and emission bandwidth and relatively large absorption and emission cross-sections. Abstract: Nano Y2O3 particles with a spherical shape and narrow size distribution have been prepared by a novel spray combustion method. The experimental procedure is briefly described and the thermodynamical process of the post-heat treatment is investigated in this paper. The precursor fully crystallized when treated at as low as 400 °C. Prepared particles showed spherical shape and well dispersibility under different treating conditions. Abstract: For the first time, a high optical quality Yb3+-doped lutetium pyrosilicate laser crystal Lu2Si2O7 (LPS) was grown by the Czochralski (Cz) method. The segregation coefficient of ytterbium ion in Yb:LPS crystal detected by the inductively coupled plasma atomic emission spectrometer (ICP-AES) method is equal to 0.847. X-ray powder diffraction result confirms the C2/m phase monoclinic space group of the grown crystal and the peaks corresponding to different phases were indexed. The absorption and fluorescence spectra, as well as fluorescence decay lifetime of Yb3+ ion in LPS have been investigated. The absorption and fluorescence cross-sections of the transitions 2F7/2↔2F5/2 of Yb3+ ion in LPS crystal have been determined. The advantages of the Yb:LPS crystal including high crystal quality, quasi-four-level laser operating scheme, high absorption cross-sections (1.33×10−20 cm2) and particularly broad emission bandwidth (∼62nm) indicated that the Yb:LPS crystal seemed to be a promising candidate used as compact, efficient thin chip lasers when LD is pumped at 940 and 980nm due to its low-symmetry monoclinic structure and single crystallographic site. Abstract: 20 at.% Yb:YAG single crystals have been grown by the CZ method and gamma-ray irradiation induced color centers and valence change of Fe3+ and Y b3+ ions in Yb:YAG have been studied. One significant 255 nm absorption band was observed in as-grown crystals and was attributed to Fe3+ ions. Two additional absorption (AA) bands located at 255 nm and 345 nm, respectively, were produced after gamma irradiation. The changes in the AA spectra after gamma irradiation and air annealing are mainly related to the charge exchange of the Fe3+, Fe2+, oxygen vacancies and F-type color centers. Analysis shows that the broad AA band is associated with Fe2+ ions and F-type color centers. The transition Y b3+→Y b2+ takes place as an effect of recharging of one of the Y b3+ ions from a pair in the process of gamma irradiation. [Copyright &y& Elsevier]

Abstract: For the first time, a quaternary doping system of Er3+, Yb3+, Ce3+, Na+: CaF2 single crystal was demonstrated to have high fluorescence yield in the 1.5μm region. Through a simplified model, the branching ratio of the Er3+ 4I11/2→4I13/2 transition was estimated to be improved more than 40 times by the deactivating effect of Ce3+ in the quaternary-doped CaF2 crystal with dopants of 0.2-at% Er3+ and 2.0-at% Ce3+. Owing to the broad emission bandwidth and large emission cross-section, the quaternary-doped CaF2 system shows the great potential to achieve the high laser performance in the 1.5μm region. Bioluminescence resonance energy transfer (BRET) is a natural biophysical phenomenon that underlies an emerging technique to monitor protein–protein interactions in living cells in real time. Here, we present a series of technical advances to enhance the utility of the BRET assay in plants. A series of recombination cloning vectors was generated to accelerate the expression of proteins tagged with Renilla luciferase or yellow fluorescent protein under transient assay conditions and in stable transgenic plants. Working in stably transformed Arabidopsis or tobacco, we then detected BRET between three pairs of candidate interaction partners: dimerization of the E3 ubiquitin ligase COP1, interaction between COP1 and the B-box protein STH, and interaction between the light regulatory bZip transcription factors HY5 and HYH. A codon-optimized version of the Renilla luciferase gene resulted in improved expression in Arabidopsis. Renilla luciferase was active in a variety of subcellular organelles, including plastids, mitochondria, peroxisomes and Golgi stacks. In a survey of the Arabidopsis light signaling machinery as a model system, we estimated the likelihood that a known protein–protein interaction can be documented using BRET. Abstract: Gd2SiO5 (GSO) single crystal codoped with Yb3+ and Er3+ (Abbr. as Er:Yb:GSO) was successfully grown by the Czochralski (CZ) method for the first time and the spectral characteristics were investigated. The absorption and fluorescence spectra were measured. The emission lifetime of the 4I13/2–Er-level was measured to be 5.84ms and the emission cross-section at 1529nm was calculated to be 1.03×10−20 cm2. Abstract: The effects of Na+ doping level on the thermal conductivities, absorption and emission spectra, and fluorescence lifetimes of Yb3+,Na+:CaF2 crystals were systematically studied. Sites structure, covalent force, and crystal field strength of Yb3+:CaF2 crystals were markedly varied by codoping Na+ as charge compensator. The 2.0at% Yb3+ and 3.0at% Na+-codoped CaF2 crystal was demonstrated to operate in diode-pumped passively mode-locking scheme. Bioluminescence resonance energy transfer (BRET) between Renilla luciferase and yellow fluorescent protein has been adapted to serve as a real-time reporter on protein-protein interactions in live plant cells by using the Arabidopsis Constitutive photomorpho- genesis 1 (COP1) protein as a model system. COP1 is a repressor of light signal transduction that functions as part of a nuclear E3 ubiquitin ligase. COP1 possesses a leucine-rich nuclear-exclusion signal that resides in a domain implicated in COP1 dimerization. BRET was applied in conjunction with site-directed mutagenesis to explore the respective contributions of the nuclear-exclusion and dimerization motifs to the regulation of COP1 activity in vivo. One specific mutant protein, COP1L105, showed increased nuclear accumulation but retained the ability to dimerize, as monitored by BRET, whereas other mutations inhibited both nuclear exclusion and COP1 dimerization. Accurate measurement of forest growing stem volume (GSV) is important for forest resource management and ecosystem dynamics monitoring. Optical remote sensing imagery has great application prospects in forest GSV estimation on regional and global scales as it is easily accessible, has a wide coverage, and mature technology. However, their application is limited by cloud coverage, data stripes, atmospheric effects, and satellite sensor errors. Combining multi-sensor data can reduce such limitations as it increases the data availability, but also causes the multi-dimensional problem that increases the difficulty of feature selection. In this study, GaoFen-2 (GF-2) and Sentinel-2 images were integrated, and feature variables and data scenarios were derived by a proposed adaptive feature variable combination optimization (AFCO) program for estimating the GSV of coniferous plantations. The AFCO algorithm was compared to four traditional feature variable selection methods, namely, random forest (RF), stepwise random forest (SRF), fast iterative feature selection method for k-nearest neighbors (KNN-FIFS), and the feature variable screening and combination optimization procedure based on the distance correlation coefficient and k-nearest neighbors (DC-FSCK). The comparison indicated that the AFCO program not only considered the combination effect of feature variables, but also optimized the selection of the first feature variable, error threshold, and selection of the estimation model. Furthermore, we selected feature variables from three datasets (GF-2, Sentinel-2, and the integrated data) following the AFCO and four other feature selection methods and used the k-nearest neighbors (KNN) and random forest regression (RFR) to estimate the GSV of coniferous plantations in northern China. The results indicated that the integrated data improved the GSV estimation accuracy of coniferous plantations, with relative root mean square errors (RMSErs) of 15.0% and 19.6%, which were lower than those of GF-2 and Sentinel-2 data, respectively. In particular, the texture feature variables derived from GF-2 red band image have a significant impact on GSV estimation performance of the integrated dataset. For most data scenarios, the AFCO algorithm gained more accurate GSV estimates, as the RMSErs were 30.0%, 23.7%, 17.7%, and 17.5% lower than those of RF, SRF, KNN-FIFS, and DC-FSCK, respectively. The GSV distribution map obtained by the AFCO method and RFR model matched the field observations well. This study provides some insight into the application of optical images, optimization of the feature variable combination, and modeling algorithm selection for estimating the GSV of coniferous plantations. • Diode-pumped CW Pr,Mg:SRA lasers at 643 nm, 724 nm and 622 nm are demonstrated. • The highest output power of the diode-pumped Pr,Mg:SRA laser is 675 mW. • Passively Q-switched Pr,Mg:SRA lasers are also demonstrated, for the first time. • The shortest pulse width is 105.6 ns for the 622 nm laser, while 211.7 ns for the 643 nm laser. We report on compact blue-diode-pumped Pr,Mg:SrAl 12 O 19 (SRA) lasers in continuous-wave and passively Q-switched regimes. For continuous-wave mode, three lasers have been achieved with maximum output powers of 675 mW, 566 mW and 312 mW at wavelengths of 643 nm, 724 nm and 622 nm, respectively. The maximum slope efficiencies with respect to absorbed power for the three lasers are 18.7%, 16.6% and 11.1%. Using a uniaxial Co2+-doped strontium, lanthanum and magnesium aluminate (Co:ASL) crystal as saturable absorber with remarkable absorptions at 643 nm and 622 nm, we have achieved passively Q-switched Pr,Mg:SRA lasers at the two emissions with maximum output powers of 175 mW and 118 mW, respectively, for the first time to our knowledge. The shortest pulse widths for the two lasers are 211.7 ns and 105.6 ns. Correspondingly, we estimated that pulse energies and peak power are 2.13 μJ and 10.1 W for the 643 nm laser as well as 0.98 μJ and 9.27 W for the 622 nm laser. Nd:SrAl 11 O 19 (Nd:SRA) crystal has been grown by the Czochralski method. Polarized absorption spectra, polarized fluorescence spectra and fluorescence lifetime were investigated. For σ-polarization, the peak absorption cross section at 798 nm is 2.08 × 10−20 cm2 with full width at half maximum (FWHM) of 10.2 nm and the peak emission cross section is 3.36 × 10−20 cm2 at 1048.5 nm with FWHM of 4.80 nm. The Judd-Ofelt parameters Ω 2,4,6 were obtained to be 0.51 × 10−20 cm2, 2.08 × 10−20 cm2, and 2.12 × 10−20 cm2, respectively. Continuous-wave laser operation of the a-cut and c-cut Nd:SRA sample has been demonstrated. Yb,Pr:CaF 2 crystals have been grown successfully by the temperature gradient technique(TGT). The absorption spectra, fluorescence spectra, fluorescence decay lifetime of Pr3+ ions in CaF 2 crystal sensitized by different Yb3+ ions concentration have been measured. The effective J-O intensity parameters were fitted to be Ω 2 =0.36 × 10−20cm2, Ω 4 =2.89 × 10−20cm2, Ω 6 =6.69 × 10−20cm2 for 0.3 at.%Pr,1.5 at.%Yb:CaF 2. And the emission cross sections of 3P 1 →3H 5 (535 nm), and 3P 0 →3F 4 (750 nm) are calculated to be 2.02 × 10−20cm2, and 2.50 × 10−20cm2 with the FWHM of 17.2 nm, and 10.5 nm, respectively. The fluorescence lifetime of 3P 0 has achieved great increasing by sensitizing of Yb3+ ion. Not only the up-conversion luminescence can be achieved, but also the broad band emission from 1D 2 energy level can be observed. Long, robust hypocotyls are important for facilitating greenhouse transplant production. The use of far-red light at the end of the day (end-of-day far-red, EOD-FR) is known to prompt hypocotyl elongation, but the mechanism of EOD-FR-mediated hypocotyl elongation in pumpkin remains unclear. Here, we found that hypocotyl length, parenchymal cell size in hypocotyls, and plant IAA levels were significantly greater in pumpkin after EOD-FR treatment. This effect was counteracted by the application of the polar auxin transport inhibitor 1-N-naphthylphthalamic acid. Integrated transcriptomic and metabolomic analysis of pumpkin hypocotyls revealed that the expression of auxin-related genes changed significantly after EOD-FR treatment, and the contents of the auxin biosynthetic precursors tryptophan and indole were also significantly higher. Our results show that auxin plays an essential role in EOD-FR-mediated hypocotyl elongation, shed light on the mechanisms of EOD-FR mediated hypocotyl elongation, and provide a theoretical basis for the use of EOD-FR in facility cultivation. Simple Summary: To reduce feed costs, the exploration of feed resources is currently the main research direction. In the past, fiber feed was generally regarded as anti-nutritional factors. Recently, fiber has received special attention due to its many beneficial effects. Therefore, this study selected five common household and production wastes as fiber sources and added them to the diet of pregnant sows to explore the impact on sows and piglets. The results of this study indicated that adding corn gluten feed (CG) significantly improved the digestibility of sows and body weight (BW) of piglets, which provide evidence and insight for the application of corn gluten feed in gestation sow diets. The purpose of this study was to determine the effect of increasing dietary neutral detergent fiber (NDF) levels on pregnant sows, and to select the best feed ingredients based on reproductive performance, plasma biochemistry parameters, colostrum and milk composition, and nutrient digestibility. Seventy-two multiparous sows were randomly allotted to six dietary treatment groups (n = 12). The feeding of chicory meal (CM), wheat bran (WB), corn gluten, and rice bran meal (RBM) increased the average weaning weight of piglets compared with the control (CON) group (p < 0.05). Supplementation with CG diet increased the sow BW, weight gain, and back fat thickness compared with WB and RBM on day 107 of gestation (p < 0.05). Furthermore, Supplementation with CG diet resulted in lower plasma urea nitrogen (PUN) and higher total protein (TP) concentrations in plasma (p < 0.05). Feeding CM diet and soybean curd residue (SCR) diet reduced the total protein and globulin, and supplementation with CM diet significantly increased the PUN (p < 0.05). The apparent total tract digestibility (ATTD) of crude protein (CP), crude fat (EE), calcium (Ca), phosphorus (P), neutral detergent fiber (NDF), and acid detergent fiber (ADF) were decreased following the addition of CM, WB, or SCR to the diets (p < 0.05). GdScO 3 single crystal codoped with Tm3+ and Tb3+ ions was successfully grown by the Edge-defined Film-fed Growth Method (EFG) for the first time. Absorption, fluorescence spectrum and fluorescence lifetimes of the grown crystal at room temperature were measured. The absorption cross-section at 789 nm was calculated to be 8.20 × 10−21 cm2. The emission cross-section was 1.35 × 10−21 cm2 at 1504 nm with the FWHM of 95.3 nm. The fluorescence lifetimes of the 3H 4 state and the 3F 4 state were fitted to be 253 μs and 1460 μs, respectively. Compared with Tm3+ single doped GdScO 3 crystal, the codoping of Tb3+ successfully depopulated the 3F 4 energy level and enhanced the emission at 1.5 μm region. Yb,Ho:Y 2 O 3 ceramics have been fabricated by Spark Plasma Sintering method (SPS). The microstructure and transmittance of the ceramics sintered from 1623 K to 1673 K were analyzed. The highest in-line transmittance of Yb,Ho:Y 2 O 3 ceramic sintered at 1673 K reached almost 83% at 750 nm with the thickness of 1 mm. The absorption spectra, fluorescence spectra and fluorescence decay curve were recorded at room temperature. The Judd-Ofelt parameters Ω 2 , Ω 4 and Ω 6 were calculated to be 8.33 × 10−20 cm2, 1.81 × 10−20 cm2, and 0.82 × 10−20 cm2, respectively. The intense emission cross section of Yb,Ho:Y 2 O 3 ceramic was 3.43 × 10−20 cm2 at 2057 nm with full width at half maximum (FWHM) of 208 nm. In this paper, we have demonstrated a continuous-wave (CW) dual-wavelength Yb:LuYAG laser which works at 1029.41 and 1047.16 nm simultaneously. The laser operation state of dual-wavelength can be adjusted continuously by change the angle of the cavity mirror. And passively Q-switched operation of the crystal Yb:LuYAG was proceeded at the wavelength of 1028.99 nm using the MoS 2 saturable absorber. We report on the comparative growth and spectroscopy of Eu3+-doped Y 3 Al 5 O 12 (YAG) and Y 3 ScAl 4 O 12 crystals grown by the micro-pulling-down (μ-PD) method. The structure of the as-grown crystals was determined by X-ray diffraction (XRD) and Raman spectroscopy. The absorption spectra of the crystals are dominated by a strong line centered at 395 nm, matching well with the emission of commercial ultraviolet laser diodes. The Eu:YAG and Eu:Y 3 ScAl 4 O 12 crystals exhibit strongest red emission centered at 710 nm and the full width at half maximum (FWHM) was calculated to be 2.03 and 3.19 nm, respectively. Tm3+- doped GdScO 3 single crystal was successfully grown by the edge-defined film-fed method (EFG) for the first time. The spectroscopic properties of the crystal were investigated at room temperature. The absorption cross section of Tm:GdScO 3 at 795 nm is 1.58 × 10−20 cm2. By applying the Judd–Ofelt approach, the intensity parameters Ω 2,4,6 , radiative transition rates, branching ratios and radiative lifetime were calculated. Tm:GdScO 3 exhibits strong emission at 1460 nm and 2300 nm with stimulated emission cross section of 1.8 × 10−21 cm2 and 2.7 × 10−21 cm2, respectively. The fluorescence lifetime of the 3H 4 level was measured to be 265 μs. All the results indicated that the Tm:GdScO 3 crystal is a potential laser material. • Tm:GdScO 3 crystal was successfully grown by the edge-defined film-fed method (EFG) for the first time. • The structure parameters of Tm:GdScO 3 crystal were investigated by XRD. • Spectral parameters and Judd-Ofelt parameters of Tm:GdScO 3 crystal were obtained and compared. By first-principles calculations, we investigate the geometric stability, electronic and optical properties of the type-II PN-WSe2 and type-I PAs-WSe2 van der Waals heterostructures(vdWH). They are p-type semiconductors with indirect band gaps of 1.09 eV and 1.08 eV based on PBE functional respectively. By applying the external gate field, the PAs-WSe2 heterostructure would transform to the type-II band alignment from the type-I. With the increasing of magnitude of the electric field, two heterostructures turn into the n-type semiconductors and eventually into metal. Especially, PN/PAs-WSe2 vdWH are both high refractive index materials at low frequencies and show negative refractive index at high frequencies. Because of the steady absorption in ultraviolet region, the PAs-WSe2 heterostructure is a highly sensitive UV detector material with wide spectrum. The type-II PN-WSe2 heterostructure possesses giant and broadband absorption in the near-infrared and visible regions, and its solar power conversion efficiency of 13.8% is higher than the reported GaTe–InSe (9.1%), MoS2/p-Si (5.23%) and organic solar cells (11.7%).
