Despite the research on opportunities to automatically measure lameness in cattle, lameness detection systems are not widely available commercially and are only used on a few dairy farms. However, farmers need to be aware of the lame cows in their herds in order treat them properly and in a timely fashion. Many papers have focused on the automated measurement of gait or behavioral cow characteristics related to lameness. In order for such automated measurements to be used in a detection system, algorithms to distinguish between non-lame and mildly or severely lame cows need to be developed and validated. Few studies have reached this latter stage of the development process. Also, comparison between the different approaches is impeded by the wide range of practical settings used to measure the gait or behavioral characteristic (e.g., measurements during normal farming routine or during experiments; cows guided or walking at their own speed) and by the different definitions of lame cows. In the majority of the publications, mildly lame cows are included in the non-lame cow group, which limits the possibility of also detecting early lameness cases. In this review, studies that used sensor technology to measure changes in gait or behavior of cows related to lameness are discussed together with practical considerations when conducting lameness research. In addition, other prerequisites for any lameness detection system on farms (e.g., need for early detection, real-time measurements) are discussed.

Due to its detrimental effect on cow welfare, health and production, lameness in dairy cows has received quite a lot of attention in the last few decades-not only in terms of prevention and treatment of lameness but also in terms of detection, as early treatment might decrease the number of severely lame cows in the herds as well as decrease the direct and indirect costs associated with lameness cases. Generally, lame cows are detected by the herdsman, hoof trimmer or veterinarian based on abnormal locomotion, abnormal behavior or the presence of hoof lesions during routine trimming. In the scientific literature, several guidelines are proposed to detect lame cows based on visual interpretation of the locomotion of individual cows (i.e., locomotion scoring systems). Researchers and the industry have focused on automating such observations to support the farmer in finding the lame cows in their herds, but until now, such automated systems have rarely been used in commercial herds. This review starts with the description of normal locomotion of cows in order to define 'abnormal' locomotion caused by lameness. Cow locomotion (gait and posture) and behavioral features that change when a cow becomes lame are described and linked to the existing visual scoring systems. In addition, the lack of information of normal cow gait and a clear description of 'abnormal' gait are discussed. Finally, the different set-ups used during locomotion scoring and their influence on the resulting locomotion scores are evaluated.

Monte Carlo methods commonly used in tissue optics are limited to a layered tissue geometry and thus provide only a very rough approximation for many complex media such as biological structures. To overcome these limitations, a Meshed Monte Carlo method with flexible phase function choice (fpf-MC) has been developed to function in a mesh. This algorithm can model the light propagation in any complexly shaped structure, by attributing optical properties to the different mesh elements. Furthermore, this code allows the use of different discretized phase functions for each tissue type, which can be simulated from the microstructural properties of the tissue, in combination with a tool for simulating the bulk optical properties of polydisperse suspensions. As a result, the scattering properties of tissues can be estimated from information on the microstructural properties of the tissue. This is important for the estimation of the bulk optical properties that can be used for the light propagation model, since many types of tissue have never been characterized in literature. The combination of these contributions, made it possible to use the MMC-fpf for modeling the light porapagation in plant tissue. The developed Meshed Monte Carlo code with flexible phase function choice (MMC-fpf) was successfully validated in simulation through comparison with the Monte Carlo code in Multi-Layered tissues (R2 > 0.9999) and experimentally by comparing the measured and simulated reflectance (RMSE = 0.015%) and transmittance (RMSE = 0.0815%) values for tomato leaves.

The size of colloidal particles in food products has a considerable impact on the product's physicochemical, functional and sensory characteristics. Measurement techniques to monitor the size of suspended particles could, therefore, help to further reduce the variability in production processes and promote the development of new food products with improved properties. Visible and near-infrared (Vis/NIR) spectroscopy is already widely used to measure the composition of agricultural and food products. However, this technology can also be consulted to acquire microstructure-related scattering properties of food products. In this study, the effect of the fat globule size on the Vis/NIR bulk scattering properties of milk was investigated. Variability in fat globule size distribution was created using ultrasonic homogenization of raw milk. Reduction of the fat globule size resulted in a higher wavelength-dependency of both the Vis/NIR bulk scattering coefficient and the scattering anisotropy factor. Moreover, the anisotropy factor and the bulk scattering coefficients for wavelengths above 600 nm were reduced and were dominated by Rayleigh scattering. Additionally, the bulk scattering properties could be well (R(2) ≥ 0.990) estimated from measured particle size distributions by consulting an algorithm based on the Mie solution. The study of animal feeding behaviour is of interest to understand feeding, to investigate the effect of treatments and conditions or to predict illness. This paper reviews the different steps to undertake when studying animal feeding behaviour, with illustrations for group-housed pigs. First, one must be aware of the mechanisms that control feeding and the various influences that can change feeding behaviour. Satiety is shown to largely influence free feeding (ad libitum and without an operant condition) in animals, but 'free' feeding seems a very fragile process, given the many factors that can influence feeding behaviour. Second, a measurement method must be chosen that is compatible with the goal of the research. Several measurement methods exist, which lead to different experimental set-ups and measurement data. Sensors are available for lab conditions, for research on group-housed pigs and also for on-farm use. Most of these methods result in a record of feeding visits. However, these feeding visits are often found to be clustered into meals. Thus, the third step is to choose which unit of feeding behaviour to use for analysis. Depending on the situation, either meals, feeding visits, other raw data, or a combination thereof can be suitable. Meals are more appropriate for analysing short-term feeding behaviour, but this may not be true for disease detection. Further research is therefore needed. To cluster visits into meals, an appropriate analysis method has to be selected. The last part of this paper provides a review and discussion of the existing methods for meal determination. A variety of methods exist, with the most recent methods based on the influence of satiety on feeding. More thorough validation of the recent methods, including validation from a behavioural point of view and uniformity in the applied methods is therefore necessary., (Copyright © 2014 Elsevier Inc. In this study, a flexible tool to simulate the bulk optical properties of polydisperse spherical particles in an absorbing host medium is described. The generalized Mie solution for Maxwell's equations is consulted to simulate the optical properties for a spherical particle in an absorbing host, while polydispersity of the particle systems is supported by discretization of the provided particle size distributions. The number of intervals is optimized automatically in an efficient iterative procedure. The developed tool is validated by simulating the bulk optical properties for two aqueous nanoparticle systems and an oil-in-water emulsion in the visible and near-infrared wavelength range, taking into account the representative particle sizes and refractive indices. The simulated bulk optical properties matched closely (R2 ≥ 0.899) with those obtained by reference measurements.

Partial Least Squares (PLS) regression is one of the most used methods for extracting chemical information from Near Infrared (NIR) spectroscopic measurements. The success of a PLS calibration relies largely on the representativeness of the calibration data set. This is not trivial, because not only the expected variation in the analyte of interest, but also the variation of other contributing factors (interferents) should be included in the calibration data. This also implies that changes in interferent concentrations not covered in the calibration step can deteriorate the prediction ability of the calibration model. Several researchers have suggested that PLS models can be robustified against changes in the interferent structure by incorporating expert knowledge in the preprocessing step with the aim to efficiently filter out the spectral influence of the spectral interferents. However, these methods have not yet been compared against each other. Therefore, in the present study, various preprocessing techniques exploiting expert knowledge were compared on two experimental data sets. In both data sets, the calibration and test set were designed to have a different interferent concentration range. The performance of these techniques was compared to that of preprocessing techniques which do not use any expert knowledge. Using expert knowledge was found to improve the prediction performance for both data sets. For data set-1, the prediction error improved nearly 32% when pure component spectra of the analyte and the interferents were used in the Extended Multiplicative Signal Correction framework. Similarly, for data set-2, nearly 63% improvement in the prediction error was observed when the interferent information was utilized in Spectral Interferent Subtraction preprocessing., (Copyright © 2014 Elsevier B.V. The effect of dependent scattering on the bulk scattering properties of intralipid phantoms in the 600-1850 nm wavelength range has been investigated. A set of 57 liquid optical phantoms, covering a wide range of intralipid concentrations (1-100% v/v), was prepared and the bulk optical properties were accurately determined. The bulk scattering coefficient as a function of the particle density could be well described with Twersky's packing factor (R(2) > 0.990). A general model was elaborated taking into account the wavelength dependency and the effect of the concentration of scattering particles (R(2) = 0.999). Additionally, an empirical approach was followed to characterize the effect of dense packing of scattering particles on the anisotropy factor (R(2) = 0.992) and the reduced scattering coefficient (R(2) = 0.999) of the phantoms. The derived equations can be consulted in future research for the calculation of the bulk scattering properties of intralipid dilutions in the 600-1850 nm range, or for the validation of theories that describe the effects of dependent scattering on the scattering properties of intralipid-like systems.

Chalcones are naturally occurring aromatic ketones, which consist of an α-, β-unsaturated carbonyl system joining two aryl rings. These compounds are reported to exhibit several pharmacological activities, including antiparasitic, antibacterial, antifungal, anticancer, immunomodulatory, nitric oxide inhibition and anti-inflammatory effects. In the present work, a Quantitative Structure-Activity Relationship (QSAR) study is carried out to classify chalcone derivatives with respect to their antileishmanial activity (active/inactive) on the basis of molecular descriptors. For this purpose, two techniques to select descriptors are employed, the Successive Projections Algorithm (SPA) and the Genetic Algorithm (GA). The selected descriptors are initially employed to build Linear Discriminant Analysis (LDA) models. An additional investigation is then carried out to determine whether the results can be improved by using a non-parametric classification technique (One Nearest Neighbour, 1NN). In a case study involving 100 chalcone derivatives, the 1NN models were found to provide better rates of correct classification than LDA, both in the training and test sets. The best result was achieved by a SPA-1NN model with six molecular descriptors, which provided correct classification rates of 97% and 84% for the training and test sets, respectively., (Copyright © 2013 Elsevier B.V. We review nondestructive techniques for measuring internal and external quality attributes of fruit and vegetables, such as color, size and shape, flavor, texture, and absence of defects. The different techniques are organized according to their physical measurement principle. We first describe each technique and then list some examples. As many of these techniques rely on mathematical models and particular data processing methods, we discuss these where needed. We pay particular attention to techniques that can be implemented online in grading lines.

Near-infrared (NIR) spectra of human blood serum consist of overlapping strong absorption bands of water and serum proteins, which affect the ability of multivariate calibration models to predict glucose. Furthermore, serum proteins such as albumin and globulins undergo a glycation reaction by forming covalent bonds with freely available glucose molecules in the serum. In diabetic individuals with poor glucose control, more and more serum protein molecules react with glucose, resulting in a high glycated protein concentration. The glucose molecules covalently bonded to serum proteins might contribute to the overall glucose signal acquired by NIR spectroscopy. This might affect the prediction ability of multivariate calibration models such as partial least squares regression (PLSR). In this study, we investigated the effect of total protein concentration and the glycated protein concentration in blood serum on the prediction ability of PLSR calibration models. Serum samples were subjected to ultra-filtration, and the PLSR model was built using NIR spectra of filtered serum solutions. Prediction performance was found to improve by 39-42% in absence of serum protein molecules. Various experimental data set designs were generated by carefully varying the glycated serum protein concentration in calibration and test sets of PLSR models. This investigation revealed that the impact of varying glycated protein concentration on the root mean square error of prediction was not drastic. To test the statistical significance of the prediction results, a multiple linear regression model was built. The glycated serum protein concentration was found to be statistically insignificant (p = 0.86) in predicting glucose concentration. Overall, it was concluded that the glycated serum proteins do not affect the glucose prediction accuracy of PLSR models using NIR spectra of human serum.

A metamodeling approach is introduced and applied to efficiently estimate the bulk optical properties of turbid media from spatially resolved spectroscopy (SRS) measurements. The model has been trained on a set of liquid phantoms covering a wide range of optical properties representative for food and agricultural products and was successfully validated in forward and inverse mode on phantoms not used for training the model. With relative prediction errors of 10% for the estimated bulk optical properties the potential of this metamodeling approach for the estimation of the optical properties of turbid media from spatially resolved spectroscopy measurements has been demonstrated.

A supercontinuum laser based double integrating sphere setup in combination with an unscattered transmittance measurement setup was developed and carefully validated for optical characterization of turbid samples in the 500-2250 nm wavelength range. A set of 57 liquid optical phantoms, covering a wide range of absorption and scattering properties, were prepared and measured at two sample thicknesses. The estimated bulk optical properties matched well for both thicknesses, and with theory and literature, without significant crosstalk between absorption and scattering. Equations were derived for the bulk scattering properties μ(s), μ(s)' and g of Intralipid® 20% which can be used to calculate the bulk scattering properties of intralipid-dilutions in the 500-2250 nm range.

Quantitative structure-activity relationship (QSAR) modeling was performed for imidazo[1,5-a]pyrido[3,2-e]pyrazines, which constitute a class of phosphodiesterase 10A inhibitors. Particle swarm optimization (PSO) and genetic algorithm (GA) were used as feature selection techniques to find the most reliable molecular descriptors from a large pool. Modeling of the relationship between the selected descriptors and the pIC50 activity data was achieved by linear [multiple linear regression (MLR)] and non-linear [locally weighted regression (LWR) based on both Euclidean (E) and Mahalanobis (M) distances] methods. In addition, a stepwise MLR model was built using only a limited number of quantum chemical descriptors, selected because of their correlation with the pIC50 . The model was not found interesting. It was concluded that the LWR model, based on the Euclidean distance, applied on the descriptors selected by PSO has the best prediction ability. However, some other models behaved similarly. The root-mean-squared errors of prediction (RMSEP) for the test sets obtained by PSO/MLR, GA/MLR, PSO/LWRE, PSO/LWRM, GA/LWRE, and GA/LWRM models were 0.333, 0.394, 0.313, 0.333, 0.421, and 0.424, respectively. Partial Least Squares (PLS) is by far the most popular regression method for building multivariate calibration models for spectroscopic data. However, the success of the conventional PLS approach depends on the availability of a 'representative data set' as the model needs to be trained for all expected variation at the prediction stage. When the concentration of the known interferents and their correlation with the analyte of interest change in a fashion which is not covered in the calibration set, the predictive performance of inverse calibration approaches such as conventional PLS can deteriorate. This underscores the need for calibration methods that are capable of building multivariate calibration models which can be robustified against the unexpected variation in the concentrations and the correlations of the known interferents in the test set. Several methods incorporating 'a priori' information such as pure component spectra of the analyte of interest and/or the known interferents have been proposed to build more robust calibration models. In the present study, four such calibration techniques have been benchmarked on two data sets with respect to their predictive ability and robustness: Net Analyte Preprocessing (NAP), Improved Direct Calibration (IDC), Science Based Calibration (SBC) and Augmented Classical Least Squares (ACLS) Calibration. For both data sets, the alternative calibration techniques were found to give good prediction performance even when the interferent structure in the test set was different from the one in the calibration set. The best results were obtained by the ACLS model incorporating both the pure component spectra of the analyte of interest and the interferents, resulting in a reduction of the RMSEP by a factor 3 compared to conventional PLS for the situation when the test set had a different interferent structure than the one in the calibration set., (Copyright © 2013 Elsevier B.V. As a model is only an abstraction of the real system, unmodeled dynamics, parameter variations, and disturbances can result in poor performance of a conventional controller based on this model. In such cases, a conventional controller cannot remain well tuned. This paper presents the control of a spherical rolling robot by using an adaptive neuro-fuzzy controller in combination with a sliding-mode control (SMC)-theory-based learning algorithm. The proposed control structure consists of a neuro-fuzzy network and a conventional controller which is used to guarantee the asymptotic stability of the system in a compact space. The parameter updating rules of the neuro-fuzzy system using SMC theory are derived, and the stability of the learning is proven using a Lyapunov function. The simulation results show that the control scheme with the proposed SMC-theory-based learning algorithm is able to not only eliminate the steady-state error but also improve the transient response performance of the spherical rolling robot without knowing its dynamic equations.

Owing to spectral variations from other sources than the component of interest, large investments in the NIR model development may be required to obtain satisfactory and robust prediction performance. To make the NIR model development for routine active pharmaceutical ingredient (API) prediction in tablets more cost-effective, alternative modelling strategies were proposed. They used a massive amount of prior spectral information on intra- and inter-batch variation and the pure component spectra to define a clutter, i.e., the detrimental spectral information. This was subsequently used for artificial data augmentation and/or orthogonal projections. The model performance improved statistically significantly, with a 34-40% reduction in RMSEP while needing fewer model latent variables, by applying the following procedure before PLS regression: (1) augmentation of the calibration spectra with the spectral shapes from the clutter, and (2) net analyte pre-processing (NAP). The improved prediction performance was not compromised when reducing the variability in the calibration set, making exhaustive calibration unnecessary. Strong water content variations in the tablets caused frequency shifts of the API absorption signals that could not be included in the clutter. Updating the model for this kind of variation demonstrated that the completeness of the clutter is critical for the performance of these models and that the model will only be more robust for spectral variation that is not co-linear with the one from the property of interest., (Copyright © 2012 Elsevier B.V. In this paper a fast, yet accurate method to estimate the spectral and angular distribution of the scattered radiation of a fluorescent material is described. The proposed method is an extension of the adding-doubling algorithm for non-fluorescent samples. The method is validated by comparing the spectral and angular transmittance and reflectance characteristics obtained with the extended algorithm with the results obtained using Monte Carlo simulations. The agreement using both methods is within 2%. However, the adding-doubling method achieves a reduction of the calculation time by a factor of 400. Due to the short calculation time, the extended adding-doubling method is very useful when fluorescent layers have to be optimized in an iterative process.

Over the last decade, the poultry sector has sought to develop ways to monitor chicken embryonic development as to optimize the incubation conditions. One of the parameters of development which may change under different incubation conditions is the angiogenesis in the chorioallantoic membrane (CAM). To be able to quantify these changes in the angiogenesis and detect long-term effects on health, a non-destructive technique is necessary. In this article, the first steps toward such a non-destructive technique are successfully taken. A spatially resolved spectroscopy set-up is built and tested for its potential to measure changes in angiogenesis with incubation time, and differences between a normal and hypercapnic incubation. In this first study, reflectance measurements are performed directly on the CAM as the eggshell considerably complicates the analysis. This issue should be addressed in future research to come to a really non-destructive technique. An experiment was conducted in which one group was incubated under normal conditions, and another under early prenatal hypercapnic conditions (i.e., increased CO(2) concentrations). The angiogenesis in the CAM was measured at embryonic day (ED) 10, 13, and 16. The measurements showed a clear blood spectrum with an increasing amount of blood in time, and significant differences in the reflectance as function of the source-detector distances. Milk production is a dominant factor in the metabolism of dairy cows involving a very intensive interaction with the blood circulation. As a result, the extracted milk contains valuable information on the metabolic status of the cow. On-line measurement of milk components during milking two or more times a day would promote early detection of systemic and local alterations, thus providing a great input for strategic and management decisions. The objective of this study was to investigate the potential of mid-infrared (mid-IR) spectroscopy to measure the milk composition using two different measurement modes: micro attenuated total reflection (μATR) and high throughput transmission (HTT). Partial least squares (PLS) regression was used for prediction of fat, crude protein, lactose and urea after preprocessing IR data and selecting the most informative wavenumber variables. The prediction accuracies were determined separately for raw and homogenized copies of a wide range of milk samples in order to estimate the possibility for on-line analysis of the milk. In case of fat content both measurement modes resulted in an excellent prediction for homogenized samples (R(2)>0.92) but in poor results for raw samples (R(2)<0.70). Homogenization was however not mandatory to achieve good predictions for crude protein and lactose with both μATR and HTT, and urea with μATR spectroscopy. Excellent results were obtained for prediction of crude protein, lactose and urea content (R(2)>0.99, 0.98 and 0.86 respectively) in raw and homogenized milk using μATR IR spectroscopy. These results were significantly better than those obtained by HTT IR spectroscopy. However, the prediction performance of HTT was still good for crude protein and lactose content (R(2)>0.86 and 0.78 respectively) in raw and homogenized samples. However, the detection of urea in milk with HTT spectroscopy was significantly better (R(2)=0.69 versus 0.16) after homogenization of the milk samples. Based on these observations it can be concluded that μATR approach is most suitable for rapid at line or even on-line milk composition measurement, although homogenization is crucial to achieve good prediction of the fat content., (Copyright © 2011 Elsevier B.V. This paper proposes a method based on near-infrared hyperspectral imaging for discriminating between terrestrial and fish species in animal protein by-products used in livestock feed. Four algorithms (Mahalanobis distance, Kennard-Stone, spatial interpolation, and binning) were compared in order to select an appropriate subset of pixels for further partial least squares discriminant analysis (PLS-DA). The method was applied to a set of 50 terrestrial and 40 fish meals analyzed in the 1000-1700 nm range. Models were then tested using an external validation set comprising 45 samples (25 fish and 20 terrestrial). The PLS-DA models obtained using the four subset-selection algorithms yielded a classification accuracy of 99.80%, 99.79%, 99.85%, and 99.61%, respectively. The results represent a first step for the analysis of mixtures of species and suggest that NIR-CI, providing valuable information on the origin of animal components in processed animal proteins, is a promising method that could be used as part of the EU feed control program aimed at eradicating and preventing bovine spongiform encephalopathy (BSE) and related diseases.

In this paper we demonstrate how a limited amount of a priori knowledge about spectral variability can be used in extended multiplicative scattering correction (EMSC) to remove disturbing effects such as light scattering variation in visible and near-infrared spectra prior to data modeling. Two different datasets were studied. In the first dataset, pigment concentrations (astaxanthin) were estimated in a model system with different concentrations of the scattering agent intralipid. Different cases were created by including varying levels of intralipid in the calibration set and then applying the models on sample sets with scattering properties both within and outside the calibration range. Including the most accurate estimate of light scattering in the EMSC model gave root mean square errors of prediction (RMSEP) that were similar to a cross-validated global model including all samples, even for extreme extrapolation with regard to scattering properties. Less accurate estimates gave on average RMSEPs half of what could be achieved using EMSC without any a priori knowledge, suggesting that the method also has potential in cases where the accurate light scattering spectrum is difficult to obtain. In the second dataset carbohydrate concentrations (sucrose, fructose, and glucose) were estimated in orange-juice mixtures where unwanted spectral variation was caused by a change in distance between transmittance fiber-optic probes. This caused two different interfering phenomena due to path length variation and saturation in the detection system. The prediction results for a model based on spectra collected at one specific probe distance treated with EMSC with a correction spectrum were comparable to what could be achieved by a global model including spectra collected at three different distances. The corresponding RMSEPs for models using EMSC with no correction term were in the worst cases 4, 22, and 36 times higher for sucrose, fructose, and glucose, respectively.

The analysis of carotenoids is complicated by the tendency of these compounds to react with radical species, leading to oxidative breakdown and isomerization during extraction. Therefore, protocols should be rapid and avoid unnecessary exposure to heat, acids, and so forth. Here, we evaluate the use of visible and near infrared reflectance spectroscopy (Vis/NIRS) to measure carotenoid contents in fruit from 28 Musa (banana and plantain) varieties. Carotenoid contents were first quantified using standardized RP-HPLC protocols, and these results were then used to develop algorithms to predict carotenoid contents from Vis/NIR spectra of the same samples. Cross-validation of the predictive algorithms across a genetically diverse group of varieties demonstrated that correlation coefficients between the HPLC measurements and the Vis/NIRS predictions varied from good for the total carotenoids and beta-carotene fractions (r(2)(cv), 0.84, 0.89) to reasonable for alpha-carotene and cis-carotenes (r(2)(cv), 0.61, 0.66), but there was only a poor correlation (r(2)(cv), 0.30) for the minor lutein component. Nonetheless, since approximately 90% of the Musa carotenoids consist of only alpha- and beta-carotene, results indicate that Vis/NIRS can be used for the high-throughput screening of fruit pulp samples for vitamin A nutritional content on the basis of their total carotenoids content.

A class of multivariate calibration methods called augmented classical least squares (ACLS) has been proposed which combines an explicit linear additive model with the predictive power of inverse models, such as principal component regression (PCR) and partial least squares (PLS). Because of its use of the explicit linear additive model, ACLS provides an interesting framework to incorporate different sources of prior information, such as measured pure component spectra, in the model. In this study, the predictive power of ACLS models incorporating different amounts of prior information has been compared to that of PCR and PLS using two examples, a designed experiment and one with biological samples. In both cases, the ACLS models showed predictive power comparable to PLS under idealized validation conditions. When a different interferent structure was present in the validation samples, the predictive power of the inverse models (PCR and PLS) dramatically decreased, with an increase in root-mean-squared error of prediction by a factor of 3.5 for the first example and a factor of 2 in the second example. The incorporation of prior information in the ACLS framework was found to considerably reduce or even completely remove these dramatic effects, especially when the pure component contributions for the interferents were taken into account.

Optical measurement of fruit quality is challenging due to the presence of a skin around the fruit flesh and the multiple scattering by the structured tissues. To gain insight in the light-tissue interaction, the optical properties of apple skin and flesh tissue are estimated in the 350-2200 nm range for three cultivars. For this purpose, single integrating sphere measurements are combined with inverse adding-doubling. The observed absorption coefficient spectra are dominated by water in the near infrared and by pigments and chlorophyll in the visible region, whose concentrations are much higher in skin tissue. The scattering coefficient spectra show the monotonic decrease with increasing wavelength typical for biological tissues with skin tissue being approximately three times more scattering than flesh tissue. Comparison to the values from time-resolved spectroscopy reported in literature showed comparable profiles for the optical properties, but overestimation of the absorption coefficient values, due to light losses.
