The deuteron is the simplest compound nucleus, composed of one proton and one neutron. Deuteron properties such as the root-mean-square charge radius rd and the polarizability serve as important benchmarks for understanding the nuclear forces and structure. Muonic deuterium μd is the exotic atom formed by a deuteron and a negative muon μ(-). We measured three 2S-2P transitions in μd and obtain r(d) = 2.12562(78) fm, which is 2.7 times more accurate but 7.5σ smaller than the CODATA-2010 value r(d) = 2.1424(21) fm. The μd value is also 3.5σ smaller than the r(d) value from electronic deuterium spectroscopy. The smaller r(d), when combined with the electronic isotope shift, yields a "small" proton radius r(p), similar to the one from muonic hydrogen, amplifying the proton radius puzzle., (Copyright © 2016, American Association for the Advancement of Science.)

This research aims at identifying the main deleterious effects of Cr(VI) on the photosynthetic apparatus and at selecting the most sensitive endpoints related to photosynthesis. To achieve this goal, we used lettuce (Lactuca sativa), a sensible ecotoxicological crop model. Three-week-old plants were exposed to 0, 50, 150 and 200 mg L(-1) of Cr(VI). These concentrations ranged from levels admitted in irrigation waters to values found in several Cr industry effluents and heavily contaminated environments. After 30 days of exposure, plants accumulated Cr preferably in roots and showed nutritional impairment, with decreases of K, Mg, Fe and Zn in both roots and leaves. Cr(VI)-exposed plants showed decreased levels of chlorophyll (Chl) a and anthocyanins, as well as decreased effective quantum yield of photostystem II (ΦPSII) and photochemical Chl fluorescence quenching (qp), but increases in the non-photochemical Chl fluorescence quenching (NPQ) and in the de-epoxidation state (DEP) of the xanthophyll cycle. Net CO2 assimilation rate (P N ) and RuBisCO activity were mostly impaired in the highest Cr(VI) concentration tested. Concerning the final products of photosynthesis, starch content was not affected, while soluble sugar contents increased. These alterations were accompanied by a reduction in protein content and in plant growth. Our results support that endpoints related to the photosynthesis photochemical processes (ΦPSII and the qp) and the content of anthocyanins are sensitive predictors of Cr(VI) toxicity. The advantages of using these parameters as biomarkers for Cr toxicity in plants are discussed. Finally, we report that, despite showing physiological disorders, L. sativa plants survived and accumulated high doses of Cr, and their use in environmental/decontamination studies is open to debate.

Understanding the impact of training sessions on the immune response is crucial for the adequate periodization of training, to prevent both a negative influence on health and a performance impairment of the athlete. This study evaluated acute systemic immune cell changes in response to an actual swimming session, during a 24-h recovery period, controlling for sex, menstrual cycle phases, maturity, and age group. Competitive swimmers (30 females, 15 ± 1.3 years old; and 35 males, 16.5 ± 2.1 years old) performed a high-intensity training session. Blood samples were collected before, immediately after, 2 h after, and 24 h after exercise. Standard procedures for the assessment of leukogram by automated counting (Coulter LH 750, Beckman) and lymphocytes subsets by flow cytometry (FACS Calibur BD, Biosciences) were used. Subjects were grouped according to competitive age groups and pubertal Tanner stages. Menstrual cycle phase was monitored. The training session induced neutrophilia, lymphopenia, and a low eosinophil count, lasting for at least 2 h, independent of sex and maturity. At 24 h postexercise, the acquired immunity of juniors (15-17 years old), expressed by total lymphocytes and total T lymphocytes (CD3(+)), was not fully recovered. This should be accounted for when planning a weekly training program. The observed lymphopenia suggests a lower immune surveillance at the end of the session that may depress the immunity of athletes, highlighting the need for extra care when athletes are exposed to aggressive environmental agents such as swimming pools.

The aim of this study was to compare the redox balance in competitive adult swimmers against recreational practitioners, controlling for Mg intake. Fifteen, competitive swimmers and 16 recreational practitioners, all male and aged 18-25years, were recruited into the study. Oxidative and muscle damage markers, and antioxidant enzymes and non-enzymatic antioxidants were evaluated by photometry (except for thiobarbituric reactive substances (TBARS), which was assessed by fluorimetry). Controlling for the level of exercise, inverse correlations were observed for uric acid and glutathione reductase (GR) or susceptibility of red blood cells to peroxidation (RBCPx); plasma adrenaline oxidase activity (AdOx) and carotenoids; TBARS and GR or Vit E; and direct correlations were observed between AdOx and creatine kinase (CK) or TBARS; CK and superoxide dismutase activity; GR and RBCPx. Controlling for Mg intake in addition to exercise level revealed new inverse correlations: between carotenoids and TBARS or lactate, and new direct correlations between lactate and AdOx or TBARS; cortisol and AdOx, CK, lactate dehydrogenase, or methemoglobin reductase. The associations between uric acid and RBCPx; AdOx and CK or TBARS; and GR and RBCPx lost their significance. All others remained significant. These outcomes suggest that the coordination between antioxidant defences may be partially modulated by Mg, which may be the result of its ability to stabilize cell membranes and oxidation targets, such as adrenaline.

Green roofs (GRs) are becoming a trend in urban areas, favouring thermal performance of buildings, promoting removal of atmospheric pollutants, and acting as possible water collection spots. Rainwater harvesting systems in buildings can also contribute to the management of stormwater runoff reducing flood peaks. These technologies should be enhanced in Mediterranean countries where water scarcity is increasing and the occurrence of extreme events is becoming very significant, as a result of climate change. An extensive pilot GR with three aromatic plant species, Satureja montana, Thymus caespititius and Thymus pseudolanuginosus, designed to study several parameters affecting rainwater runoff, has been in operation for 12 months. Physico-chemical analyses of roof water runoff (turbidity, pH, conductivity, NH4(+), NO3(-), PO4(3-), chemical oxygen demand) have shown that water was of sufficient quality for non-potable uses in buildings, such as toilet flushing. An innovative approach allowed for the development of an expression to predict a 'monthly runoff coefficient' of the GR system. This parameter is essential when planning and designing GRs combined with rainwater harvesting systems in a Mediterranean climate. This study is a contribution to improving the basis for the design of rainwater harvesting systems in buildings with extensive GRs under a Mediterranean climate.

Adequate magnesium (Mg) levels play a vital role in membrane excitability, cell contractility and metabolism, being a key nutrient for sustaining appropriate muscular contraction and performance levels in athletes. Phase angle (PhA), assessed by bioimpedance analysis (BIA), has been reported to be positively associated with most nutritional markers and is an indicator of membrane integrity and water distribution between intra- and extracellular spaces. The aim of the present study was to verify the association between Mg status and PhA as a predictor of cellular health, in a sample of judo athletes from a period of weight stability to prior to competition. Judo athletes (n = 20) from the national team were evaluated on two occasions: during a period when body weight was stable (M1), and prior to competition (M2). Changes between these occasions were calculated as M2-M1. PhA was obtained by bioelectrical impedance spectroscopy at a frequency of 50 KHz. Mg was measured in serum and red blood cells (RBC) by atomic absorption spectrophotometry, and Mg in the diet was assessed from a 24-h diet record over a seven-day period, after an assessment of body composition. Mean PhA did not differ from M1 to M2. However, individual changes in PhA were positively associated with individual changes in serum (r = 0.62, p = 0.004) and RBC Mg (r = 0.45, p = 0.048). This association was independent of weight changes between assessments, but when adjusted for Mg intake changes, only the association between PhA and serum Mg remained significant. These results highlight that in elite athletes PhA may be an indirect indicator of muscular function.

Avalanche photodiodes are commonly used as detectors for low energy x-rays. In this work, we report on a fitting technique used to account for different detector responses resulting from photoabsorption in the various avalanche photodiode layers. The use of this technique results in an improvement of the energy resolution at 8.2 keV by up to a factor of 2 and corrects the timing information by up to 25 ns to account for space dependent electron drift time. In addition, this waveform analysis is used for particle identification, e.g., to distinguish between x-rays and MeV electrons in our experiment.

Cadmium (Cd) is a widespread heavy metal used in numerous industrial processes. Cd exerts toxicological effects mostly in kidney and liver. Bone is also an important target of Cd, however, the cellular mechanisms of Cd toxicological effects in the bone cells are still poorly understood. Therefore, the present work aimed to investigate the putative cytotoxic and genotoxic effects of Cd to human bone cells. For that, the osteoblast-like MG-63 cells were exposed to 20 and 50μM Cd for 24 and 48h. Results showed a dose-dependent increase in Cd accumulation in cells and a decrease in cell viability, especially after 48h. Cell cycle analysis showed a delay at S phase concomitant with a decrease in cells at G0/G1 phase. After 24h, Cd treatment downregulated the expression of CHEK1, CHEK2 and CDK2 genes and upregulated the expression of CCNE1 gene. After 48h, the expression of ATM and CCNB1 genes were downregulated. Also, a 3.3 fold increase on the expression of gene CCNE1 was detected. Both Cd doses induced DNA fragmentation at 48h, while an increase in micronuclei (MN) and nucleoplasmic bridges (NPBs) together with an increase in the percentage of apoptotic/necrotic cells was detected for both time periods. Overall, our results demonstrate the cytotoxicity and genotoxicity of Cd in human bone cells. Also, the cytokinesis-block micronucleus (CBMN) assay parameters (MN, NPBs and the percentage of cells under apoptosis or necrosis) together with the cell cycle appear as the most sensitive to Cd cyto- and genotoxicity, being early affected even with the lowest Cd dose. Studies examining the immune response to acute intensive swimming have shown increased leukocytosis and lymphocyte populations. However, studies concerning mucosal immunity and sex differences remain controversial. The objective of the study was to examine sex differences on the immune response to maximal incremental swimming exercise in well trained swimmers. Participants (11 females, controlled for menstrual cycle phase effects; 10 males) performed a maximal incremental 7x200 m front crawl set. Fingertip capillary blood samples were obtained after each 200 m swim for lactate assessment. Venous blood and saliva samples were collected before and 5 minutes after the swimming test to determine total numbers of leukocytes, lymphocytes and subpopulations, and serum and salivary immunoglobulin A (IgA) levels. IgA secretion rate was calculated. Menstrual cycle phase did not influence the immune response to exercise. As for sex differences, exercise induced an increase in leukocytes, total lymphocytes, CD3(+), CD4(+), CD8(+), and CD16(+)/56(+) in males. In females, only leukocytosis, of a lower magnitude than was observed in males, occurred. CD19(+) increased and CD4(+)/CD8(+) ratio decreased in both groups following exercise whilst IgA, SIgA concentrations, and srIgA did not change. Both males and females finished the incremental exercise very close to the targeted race velocity, attaining peak blood lactate concentrations of 14.6±2.25 and 10.4±1.99 mmol.L(-1), respectively. The effect of a maximal incremental swimming task on immunity is sex dependent and more noticeable in men. Males, as a consequence of higher levels of immunosurveillance may therefore be at a lower risk of infection than females. Key PointsMaximal exercise induces an immune response.This study investigated the influence of sex over the leukocytes subpopulations and mucosal immune responses to maximal swimming.Male swimmers showed a stronger increase of T helper, T cytotoxic and NK lymphocytes than females, suggesting they may be at a lower risk of infection, due to a higher immunosurveillance.Mucosal immunity remained unchanged in both sexes.

Background: Aquatic exercise is recommended by the Osteoarthritis Research Society (OARSI), by the American College of Rheumatology (ACR) and by the European League Against Rheumatism (EULAR) as a nonpharmacological method of controlling the knee osteoarthritis (KOA) symptoms. Moreover, given that weight loss results in a reduction of the load that is exerted upon the knee during daily activities, obesity is also considered to be a modifiable risk factor for the development and or exacerbation of KOA. The implementation of an exercise based weight loss program may, however, itself be limited by the symptoms of KOA. The aquatic program against osteoarthritis (termed "PICO" in Portuguese) prioritizes the control of symptoms and the recovery of functionality, with an attendant increase in the patient's physical activity level and, consequently, metabolic rate. Our laboratory is assessing the effectiveness of 3 months of PICO on the symptoms of KOA, on physical function, on quality of life and on gait. In addition, PICO shall examine the effects of said exercise intervention on inflammatory biomarkers, psychological health, life style and body composition., Methods/design: The trial is a prospective, single-blinded, randomized controlled trial, and involves 50 overweight and obese adults (BMI = 28-43.5 kg/m²; age 40-65 yrs) with radiographic KOA. The participants are randomly allocated into either an educational attention (control) group or an aquatic (exercise program) group. This paper describes the experimental protocol that is used in the PICO project., Discussion: The PICO program shall provide insight into the effectiveness of an aquatic exercise program in the control of KOA symptoms and in the improvement of the quality of life. As such, they are likely to prove a useful reference to health professionals who intend to implement any kind of therapeutic intervention based around aquatic exercise., Trial Registration: NCT01832545.

Accurate knowledge of the charge and Zemach radii of the proton is essential, not only for understanding its structure but also as input for tests of bound-state quantum electrodynamics and its predictions for the energy levels of hydrogen. These radii may be extracted from the laser spectroscopy of muonic hydrogen (μp, that is, a proton orbited by a muon). We measured the 2S(1/2)(F=0)-2P(3/2)(F=1) transition frequency in μp to be 54611.16(1.05) gigahertz (numbers in parentheses indicate one standard deviation of uncertainty) and reevaluated the 2S(1/2)(F=1)-2P(3/2)(F=2) transition frequency, yielding 49881.35(65) gigahertz. From the measurements, we determined the Zemach radius, r(Z) = 1.082(37) femtometers, and the magnetic radius, r(M) = 0.87(6) femtometer, of the proton. We also extracted the charge radius, r(E) = 0.84087(39) femtometer, with an order of magnitude more precision than the 2010-CODATA value and at 7σ variance with respect to it, thus reinforcing the proton radius puzzle.

Cadmium is a priority pollutant. Its mechanisms and effects within different plant organs remain unclear. Here, cyto-genotoxicity biomarkers were evaluated in roots and leaves after Cd exposure (0, 1, 10, and 50 μM) of the model crop Lactuca sativa L. (cv. "Reine de Mai"). Overall, superoxide dismutase (SOD) and catalase (CAT) activities were stimulated in leaves, where Cd accumulation was lower in comparison to that in roots. In roots, SOD and peroxidase (POX, APX) activities were stimulated. Moreover, in both organs glutathione reductase (GR) was not affected by Cd. Overall, the H(2)O(2) content increased in both organs, while the total antioxidant capacity decreased in leaves and increased in roots with Cd concentrations. In both organs, lipid and protein oxidation rose with consequent increase of membrane permeability. Simultaneously, the comet assay showed that tail moment, tail length, and % tail DNA were maximum for 1 μM. For 10 μM, shorter tails were found suggesting induced Cd-DNA adducts that lead to DNA-DNA/DNA-protein cross-links, and/or formation of longer DNA fragments, and/or impairment of DNA repair mechanisms, while at 50 μM, nucleoids sensitivity to the technique was evident. This result was consistent with the maximum micronuclei frequency found for the 10 μM Cd dose in roots, suggesting that the surviving cells in this organ had an increase of mitotic catastrophe and that DNA repair systems for blocking cell cycle were dysfunctional. In lower Cd concentrations, root cells might have developed strategies to repair damaged DNA by blocking the cell cycle at specific checkpoints, thus avoiding mitotic catastrophe. Roots at 1 μM showed a cell cycle blockage trend at the G(2) checkpoint, while those at higher concentrations presented S phase delay. We finally discuss a general model of Cd-organ interaction covering these cyto- and genotoxic effects and the potential use of this cultivar in phytoremediation strategies.

Magnesium (Mg) deficiency has been associated with bone disorders. Physical activity is also crucial for bone mineralization. Bone mass loss has been observed to be accelerated in subjects with low Mg intake. We aim to understand if Mg intake mediates the association between bone mineral density (BMD) and lean soft tissue (LST) in elite swimmers. Seventeen elite swimmers (eight males; nine females) were evaluated. Bone mineral content, BMD, LST, and fat mass were assessed using dual energy X-ray absorptiometry. Energy and nutrient intake were assessed during a seven-day period and analyzed with Food Processor SQL. Males presented lower values than the normative data for BMD. Mg, phosphorus (P) and vitamin D intake were significantly lower than the recommended daily allowance. A linear regression model demonstrated a significant association between LST and BMD. When Mg intake was included, we observed that this was a significant, independent predictor of BMD, with a significant increase of 24% in the R(2) of the initial predictive model. When adjusted for energy, vitamin D, calcium, and P intake, Mg remained a significant predictor of BMD. In conclusion, young athletes engaged in low impact sports, should pay special attention to Mg intake, given its potential role in bone mineral mass acquisition during growth.

Microorganisms isolated from sites contaminated with heavy metals usually possess a higher removal capacity than strains from regular cultures. Heavy metal-containing soil samples from an industrial dumpsite in Northern Portugal were accordingly collected; following enrichment under metal stress, a consortium of wild microalgae was obtained. Their ability to grow in the presence of, and their capacity to recover heavy metals was comprehensively studied; the datasets thus generated were fitted to by a combined model of biomass growth and metal uptake, derived from first principles. After exposure to 15 and 25 mg/L Zn(2+) for 6 days, the microalgal consortium reached similar, or higher cell density than the control; however, under 50 and 65 mg/L Zn(2+), 71% to 84% inhibition was observed. Growth in the presence of Hg(2+) was significantly inhibited, even at a concentration as low as 25 μg/L, and 90% inhibition was observed above 100 μg/L. The maximum amount of Zn(2+) removed was 21.3 mg/L, upon exposure to 25 mg/L for 6 day, whereas the maximum removal of Hg(2+) was 335 μg/L, upon 6 day in the presence of 350 μg/L. The aforementioned mechanistic model was built upon Monod assumptions (including heavy metal inhibition), coupled with Leudeking-Piret relationships between the rates of biomass growth and metal removal. The overall fits were good under all experimental conditions tested, thus conveying a useful tool for rational optimisation of microalga-mediated bioremediation.

Metal contamination of a few aquatic, atmospheric, and soil ecosystems has increased ever since the industrial revolution, owing to discharge of such elements via the effluents of some industrial facilities. Their presence to excessive levels in the environment will eventually lead to serious health problems in higher animals owing to accumulation throughout the food web. Current physicochemical methods available for recovery of metal pollutants (e.g., chemical precipitation, oxidation/reduction, or physical ion exchange) are either expensive or inefficient when they are present at very low concentrations. Consequently, removal of toxic metals by microorganisms has emerged as a potentially more economical alternative. Microalgae (in terms of both living and nonliving biomass) are an example of microorganisms suitable to recover metals and able to attain noteworthy percent removals. Their relatively high metal-binding capacities arise from the intrinsic composition of their cell walls, which contain negatively charged functional groups. Consequently, microalgal cells are particularly efficient in uptake of those contaminants when at low levels. Self-defense mechanisms developed by microalgal cells to survive in metal-containing media and environmental factors that affect their removal (e.g., pH, temperature, and biomass concentration) are reviewed here in a comprehensive way and further discussed in attempts to rationalize this form of remediation vis-a-vis with conventional nonbiological alternatives., (Copyright © 2012 American Institute of Chemical Engineers (AIChE).)

Extended-spectrum β-lactamases (ESBL) are plasmid-mediated enzymes that hydrolyze cephalosporins and monobactams. The lack of a standard method to detect ESBL in Enterobacter spp. has led to underestimating its frequency. The aim of this study was to evaluate ESBL detection in Enterobacter spp. By the double-disk synergy test (DDST) and combined disk test (CDT) assay using cefepime, cefotaxime, and ceftazime as substrates for ESBL, plus AmpC inhibitors in different associations. A total of 83 Enterobacter spp. ESBL and 31 non-ESBL Enterobacter spp. were tested, and a cutoff point ≥3 mm was defined using a receiver operating characteristic (ROC) curve for combined disc methods. All tests showed 100% specificity. The sensitivity was 89.2% for DDST and CDT without AmpC inibitor, 90.4% in the combined disc test in Mueller-Hinton agar containing phenylboronic acid (CDT-PBAA), and 94% in the combined disc test in Mueller-Hinton agar containing cloxacillin (CDT-CLXA). Cefepime was the best substrate, mainly when AmpC inhibitors were not used. However, superior results were achieved when all cephalosporins were evaluated together. In conclusion, to improve ESBL detection in Enterobacter spp., some modifications in phenotypic tests are needed, such as to reduce the distance between the discs to 20 mm in DDST, to use a cutoff point for ≥3 mm on the CDT, and to include a cefepime disk or an inhibitor of AmpC in all tests.

Magnesium plays significant roles in promoting strength. Surveys of athletes reveal that intake of magnesium is often below recommended levels. We aimed to understand the impact of magnesium intake on strength in elite male basketball, handball, and volleyball players. Energy and nutrient intake were assessed from seven-day diet record. Strength tests included maximal isometric trunk flexion, extension, and rotation, handgrip, squat and countermovement Abalakov jump, and maximal isokinetic knee extension and flexion peak torques. Linear regression models were performed with significance at p<0.1. Mean magnesium intake was significantly lower than the recommended daily allowance. Regression analysis indicated that magnesium was directly associated with maximal isometric trunk flexion, rotation, and handgrip, with jumping performance tests, and with all isokinetic strength variables, independent of total energy intake. The observed associations between magnesium intake and muscle strength performance may result from the important role of magnesium in energetic metabolism, transmembrane transport and muscle contraction and relaxation.

Prevalence of H. pylori infection was determined using cultures of gastric biopsy samples of patients attended at the academic hospital of the Federal University of Paraná, Curitiba, Paraná, Brazil. Molecular methods were used to characterize the cagA and vacA genes from bacterial isolates associated with different diseases presented by patients. Out of a total of 81, forty-two gastric biopsy samples tested were positive for H. pylori, with a prevalence of 51.9%. No significant difference was found with regard to the gender (p=0.793) and age (p=0.183) of the patients. Genotype s1m1 vacA gene was found in 67% of the cases of peptic ulcer investigated (p=1.0), despite the limited number of patients with this disease (n=3). A correlation between the presence of less virulent strains (s2m2) and reflux esophagitis was found in the majority of the cases (45%), but without statistical significance. An association between the prevalence of cagA gene, found in 92% of isolates, and peptic ulcer was not observed (p=1.0), suggesting that this gene cannot be considered a specific marker of severity in our environment. The results reinforce the importance of conducting regional studies and the need to characterize H. pylori virulence genes associated with different diseases.

Introduction: Candida yeasts are commensals; however, if the balance of normal flora is disrupted or the immune defenses are compromised, Candida species can cause disease manifestations. Several attributes contribute to the virulence and pathogenicity of Candida, including the production of extracellular hydrolytic enzymes, particularly phospholipase and proteinase. This study aimed to investigate the in vitro activity of phospholipases and acid proteinases in clinical isolates of Candida spp., Methods: Eighty-two isolates from hospitalized patients collected from various sites of origin were analyzed. Phospholipase production was performed in egg yolk medium and the production of proteinase was verified in a medium containing bovine serum albumin. The study was performed in triplicate., Results: Fifty-six (68.3%) of isolates tested were phospholipase positive and 16 (44.4%) were positive for proteinase activity. C. tropicalis was the species with the highest number of positive isolates for phospholipase (91.7%). Statistically significant differences were observed in relation to production of phospholipases among species(p<0,0001) and among the strains from different sites of origin (p=0.014). Regarding the production of acid protease, the isolates of C. parapsilosis tested presented a larger number of producers (69.2%). Among the species analyzed, the percentage of protease producing isolates did not differ statistically (χ2=1.9 p=0.5901 (χ2=1.9 p=0.5901)., Conclusions: The majority of C. non-albicans and all C. albicans isolates were great producers of hydrolytic enzymes and,consequently, might be able to cause infection under favorable conditions.

On May 2009, both the gas and particulate fractions of smoke from a wildfire in Sever do Vouga, central Portugal, were sampled. Total hydrocarbons and carbon oxides (CO(2) and CO) were measured using automatic analysers with flame ionisation and non-dispersive infrared detectors, respectively. Fine (PM(2.5)) and coarse (PM(2.5-10)) particles from the smoke plume were analysed by a thermal-optical transmission technique to determine the elemental and organic carbon (EC and OC) content. Subsequently, the particle samples were solvent extracted and fractionated by vacuum flash chromatography into different classes of organic compounds. The detailed organic speciation was performed by gas chromatography-mass spectrometry. The CO, CO(2) and total hydrocarbon emission factors (g kg(-1) dry fuel) were 170 ± 83, 1485 ± 147, and 9.8 ± 0.90, respectively. It was observed that the particulate matter and OC emissions are significantly enhanced under smouldering fire conditions. The aerosol emissions were dominated by fine particles whose mass was mainly composed of organic constituents, such as degradation products from biopolymers (e.g. levoglucosan from cellulose, methoxyphenols from lignin). The compound classes also included homologous series (n-alkanes, n-alkenes, n-alkanoic acids and n-alkanols), monosaccharide derivatives from cellulose, steroid and terpenoid biomarkers, and polycyclic aromatic hydrocarbons (PAHs). The most abundant PAH was retene. Even carbon number homologs of monoglycerides were identified for the first time as biomarkers in biomass burning aerosols., (Copyright © 2010 Elsevier B.V. Aim of the Study: Products derived from the palm tree Orbignya phalerata Mart, a species popularly known in Brazil as babassu, are used for the treatment of different diseases such as rheumatism, ulcers and inflammatory processes. The objective of this study was to perform a sociodemographic survey among communities of babassu nut breakers in the region of Médio Mearim, Esperantinópolis, Maranhão, Brazil, and to evaluate the frequency and main forms of therapeutic use of babassu products and subproducts., Materials and Methods: A semiquantitative method was used for the survey, with the application of semistructured interviews consisting of closed and semi-open questions. The data were analyzed using the SPSS 16.0 software for Windows (2007)., Results: The results showed that 82% of female nut breakers had a household income of less than US$ 280.00 and only 2% earned more than US$ 500.00. With respect to race and educational level, 61% of the interviewed women were mulattoes and 33% had a low educational level. The babassu-derived products most frequently used for medicinal purposes were the mesocarp, a residue called "borra", and oil. The mesocarp flour was used by 90% of nut breakers for the treatment of gastritis and inflammation and by 77% for leukorrhea. Babassu residue was mainly used for the treatment of wounds (60%) and the oil was used for woundhealing (16%) and leukorrhea (8%)., Conclusions: In conclusion, the products derived from babassu are used for the treatment of gastritis, leukorrhea and wounds. Magnesium (Mg) deficiency strongly affects muscle performance. In judo, many athletes often undergo impressive weight changes associated with severe dehydration. Common practices used by athletes to achieve a target weight can lead to Mg deficit. This study aimed to understand the impact of Mg changes on strength from periods of weight stability to prior to competition in a sample of elite judo athletes who differentially changed their intracellular water (ICW). The sample consisted of 20 elite male judo athletes. Subjects were divided according to ICW changes: losses below 2% and losses equal to or above 2%. Mg was measured in serum, red blood cells and urine by atomic absorption spectrophotometry. ICW was calculated as the difference between total-body water and extracellular water using dilution techniques. Maximal handgrip strength was evaluated using Jamar Hydraulic Hand Dynamometer. Upper-body power was determined in a bench press. Higher ICW decreases were associated with higher strength reductions, though our results suggest that an increase in red blood cell Mg might attenuate those strength reductions in athletes who decrease the ICW compartment. As Mg losses can be considerable and intake is frequently insufficient, athletes should consider supplementation, especially during periods of weight reduction.

The proton is the primary building block of the visible Universe, but many of its properties-such as its charge radius and its anomalous magnetic moment-are not well understood. The root-mean-square charge radius, r(p), has been determined with an accuracy of 2 per cent (at best) by electron-proton scattering experiments. The present most accurate value of r(p) (with an uncertainty of 1 per cent) is given by the CODATA compilation of physical constants. This value is based mainly on precision spectroscopy of atomic hydrogen and calculations of bound-state quantum electrodynamics (QED; refs 8, 9). The accuracy of r(p) as deduced from electron-proton scattering limits the testing of bound-state QED in atomic hydrogen as well as the determination of the Rydberg constant (currently the most accurately measured fundamental physical constant). An attractive means to improve the accuracy in the measurement of r(p) is provided by muonic hydrogen (a proton orbited by a negative muon); its much smaller Bohr radius compared to ordinary atomic hydrogen causes enhancement of effects related to the finite size of the proton. In particular, the Lamb shift (the energy difference between the 2S(1/2) and 2P(1/2) states) is affected by as much as 2 per cent. Here we use pulsed laser spectroscopy to measure a muonic Lamb shift of 49,881.88(76) GHz. On the basis of present calculations of fine and hyperfine splittings and QED terms, we find r(p) = 0.84184(67) fm, which differs by 5.0 standard deviations from the CODATA value of 0.8768(69) fm. Our result implies that either the Rydberg constant has to be shifted by -110 kHz/c (4.9 standard deviations), or the calculations of the QED effects in atomic hydrogen or muonic hydrogen atoms are insufficient.

The antibacterial activity of plant extracts obtained from Bixa orellana L., Chamomilla recutita L., Ilex paraguariensis A. St.-Hil., Malva sylvestris L., Plantago major L. and Rheum rhaponticum L. has been evaluated against two reference strains and eleven clinical isolates of Helicobacter pylori. All the plant species chosen are used in popular Brazilian cuisine and folk medicine in the treatment of gastrointestinal disorders. Initial screening was made by the disk diffusion test and then minimum inhibitory concentration was determined by the agar dilution method. The results presented in this work demonstrated that among the plant preparations analyzed, B. orellana L., C. recutita L., I. paraguariensis A. St.-Hil. and M. sylvestris L. were capable of inhibiting the in vitro growth of H. pylori.

Objectives: Erythrocytes may play an important role in regulating blood pressure as storage sites for nitric oxide (NO). The objective of this work was to determine whether factors related to variations in erythrocyte metabolism associated with NO bioavailability, such as the activity of two enzymes--methemoglobin reductase (MHbR) and glutathione reductase (GSHR)--may help explain age-related increased blood pressure., Methods: The sample consisted of 468 individuals of both sexes, 237 hypertensive (HT) and 231 normotensive (NT), aged between 18 and 98 years (48.81 +/- 19.46). The activity of MHbR (micromol.g Hb-1.min-1) and of GSHR (micromol.g Hb-1.min-1) was determined in erythrocytes by spectrophotometry. The statistical methods used were the Mann-Whitney test, Spearman's correlation coefficient and binary logistic regression., Results: In this population, age was a risk factor for hypertension (OR=1.055, 95% CI = 1.045-1.065, p < 0.001). There was a significant difference in erythrocyte activity of these enzymes between normotensive and hypertensive subjects, with lower values in hypertensives: MHbR-NT = 16.97 (3.82-34.63), HT = 16.26 (3.26-37.10), p = 0.012; and GSHR-NT=57.60 (21.59-96.58), HT = 39.26 (23.07-90.27), p < 0.001. Enzyme activity was inversely correlated with age (MHbR: r = -0.193, p < 0.001; GSHR: r = -0.757, p < 0.001). MHbR correlated directly with GSHR only in hypertensive patients (r = 0.343, p = 0.034), which was not observed in normotensives., Conclusions: Age was a risk factor for hypertension. The erythrocyte activity of glutathione and metahemoglobin reductases, essential for redox balance and nitric oxide bioavailability in erythrocytes, may contribute only partially to the increased prevalence of age-related hypertension, and other factors should be taken into consideration, such as nutrition and antihypertensive medication.

Erythrocyte acid phosphatase (ACP locus 1), also known as low-molecular-weight protein tyrosine phosphatase, has previously been associated to glycemia, dyslipidemia, and obesity. In this study, ACP1 genotype and activity were tested in 318 women aged 19 to 83 (mean, 51.74 +/- 13.44) years. ACP1 genotype was found to directly correlate to glutathione reductase activity (P < .001) and levels of low-density lipoprotein cholesterol (P = .038). Glutathione reductase activity was in turn found to correlate to a series of cardiovascular risk factors such as systolic arterial pressure (P < .001), total cholesterol levels (P = .018), and low-density lipoprotein cholesterol levels (P = .039). A possible protective effect of ACP1 genotype AA against these cardiovascular risk factors was observed in this study. Furthermore, this work hypothesizes that nutritional riboflavin uptake becomes more crucial as body mass index increases, to counteract oxidative stress and minimize cardiovascular risk. This might be especially true in ACP1 genotypes AC, BC, and CC, which might possibly show the least endogenous protection against oxidative stress.

Microalgae have been proven efficient biological vectors for heavy metal uptake. In order to further study their biosorption potential, a strain of Desmodesmus pleiomorphus (L) was isolated from a strongly contaminated industrial site in Portugal. Under different initial Zn(2+) concentrations, metal removal by that strain reached a maximum of 360 mg Zn/g biomass after 7 days, at 30 mg Zn/l, after an initial rapid phase of uptake. Comparative studies were carried out using a strain of the same microalgal species that is commercially available (ACOI 561): when exposed to 30 mg Zn/l, it could remove only 81.8 mg Zn/g biomass. Biosorption experiments using inactivated biomass of the isolated strain reached a maximum Zn(2+) uptake of 103.7 mg/g. Metal removal at various initial pH values was studied as well; higher removal was obtained at pH 5.0. The microalga strain L, isolated from the contaminated site, exhibited a much higher removal capacity than the commercial strain, and the living biomass yielded higher levels of metal removal than its inactivated form.

Our aim was to evaluate the pulmonary changes induced by Leptospira interrogans infection in hamsters, and the gene expression of endogenous mediators in lung fragments during 28 days of observation. The animals were euthanized on days 4, 7, 14, 21, and 28 post-inoculation. Histopathologic lung analysis showed hemorrhage, pneumonia, alveolar congestion, and infiltrated cellular areas, with increasing severity until day 21 post-inoculation. Tumor necrosis factor (TNF)-alpha mRNA expression enhanced in first days with peak on day 4 and slightly decreased in the final phase. The interleukin (IL)-10 remained relatively constant throughout the period, with the exceptions of days 4 and 14. The endothelial nitric-oxide synthesis (eNOS) showed an increased expression on day 4, followed by an augment on days 7 and 14, and remaining constant up to day 28 post-infection. Our results demonstrate that inoculation of L. interrogans sorovar Icterohaemorrhagiae induced pulmonary lesions, including pulmonary hemorrhage, supporting that the lung is a target organ.

Physical exercise may deplete magnesium, which together with a marginal dietary magnesium intake may impair energy metabolism, muscle function, oxygen uptake and electrolyte balance. Consequently, the ability to perform physical work may be compromised. Many aspects of immune function can be depressed temporarily by either a single bout of very severe exercise or a longer period of excessive training. Although the disturbance is usually quite transient, it can be sufficient to allow a clinical episode of infection, particularly upper respiratory tract infections. However, regular and moderate exercise has been reported to improve the ability of the immune system to protect the host from infection. Magnesium also has a strong relation with the immune system in both non specific and specific immune responses and magnesium deficit has been shown to be related to impaired cellular and humoral immune function. Magnesium deficiency leads to immunopathological changes that are related to the initiation of a sequential inflammatory response. Although in athletes magnesium deficiency has not been investigated regarding alterations in the immune system, the possibility exists that magnesium deficiency could contribute to the immunological changes observed after strenuous exercise.

Objective: To evaluate the effects of pentoxifylline on hydrochloric acid-induced lung lesions in rats subjected to mechanical ventilation., Methods: Twenty male, adult Wistar-EPM-1 rats were anesthetized and randomly grouped (n=5 animals per group) as follows: control-MV (mechanical ventilation, MV group); bilateral instillation of HCl (HCl group); bilateral instillation of HCl followed by pentoxifylline (50 mg/kg bw) infusion (HCl+PTX group) and pentoxifylline infusion followed by bilateral instillation of HCl (PTX+HCl group). At 20, 30, 90 and 180 min after treatments, the blood partial pressures of CO2 and O2 were measured. The animals were euthanized, and bronchoalveolar lavages were taken to determine the contents of total proteins, corticosterone [corrected] and TNF-alpha. Samples of lung tissue were used for histomorphometric studies and determining the wet-to-dry (W/D) lung weight ratio., Results: In the MV group, rats had alveolar septal congestion, and, in the HCl group, a remarkable recruitment of neutrophils and macrophages into the alveoli was noticed; these events were reduced in the animals with PTX+HCl. The partial pressure of oxygen increased in PTX+HCl animals (121+/-5 mmHg) as compared with the HCl (62+/-6 mmHg) and HCl+PTX (67+/-3 mmHg) groups within 30 minutes. TNF-alpha levels in bronchoalveolar lavage were significantly higher in the HCl group (458+/-50 pg/mL), reduced in the HCl+PTX group (329+/-45 pg/mL) and lowest in the PTX+HCl group (229+/-41 pg/mL). The levels of corticosterone [corrected] in bronchoalveolar lavage were significantly lower in the HCl (8+/-1.3 ng/mL) and HCl+PTX group (16+/-2 ng/mL) and were highest in the PTX+HCl (27+/-1.9 ng/mL)., Conclusion: Pretreatment with PTX improves oxygenation, reduces TNF-alpha concentration and increases the concentration of corticosterone [corrected] in bronchoalveolar lavage upon lung lesion induced by HCl.

Isolation and identification of etiological agents found in body fluids can be of critical importance for the recovery of patients suffering from potentially-severe infections, which are often followed by serious sequels. Eighty-two samples of different body fluids were analyzed using two different methods: (1) the conventional culture method (agar plating) and (2) the enrichment culture technique, using the Bact/Alert blood culture bottle. The number of positive cultures increased on average from 9.7% to 23.1% with the enrichment culture technique. Pseudomonas aeruginosa, Escherichia coli and Staphylococcus aureus were the most frequently isolated bacteria. The enrichment method could provide a more accurate means the identifying etiological agents.

Objective: This study was conducted to determine the association between magnesium (Mg), body composition and insulin resistance in 136 sedentary postmenopausal women, 50 to 77 years of age., Methods: Diabetics, hypertensives and women on hormonal replacement therapy were excluded and the remaining 74 were divided according to BMI> or =25 (obese: OG) and BMI<25 kg/m(2) (non-obese: NOG). Nutritional data disclosed that intakes were high for protein and saturated fat, low for carbohydrates, polyunsaturated fat and Mg and normal for the other nutrients, according to recommended dietary allowances (RDA). Mg values in red blood cells (RBC-Mg) and plasma (P-Mg), were determined, as were fasting glucose, and insulin levels, Homeostasis Model Assessment (HOMA), body mass index (BMI), body fat percent (BF %), abdominal fat (AF) and free fat mass (FFM)., Results: RBC-Mg values were low in both groups when compared with normal values. There were significant differences in body composition parameters, HOMA and insulin levels, with higher basal insulin levels in OG. RBC-Mg was directly correlated with insulin, HOMA and FFM in both groups, according to Pearson correlations. HOMA in OG was also directly correlated with BMI, FFM and AF. In NOG, HOMA was only correlated with FFM. The low RBC-Mg levels observed were probably due to low Mg intake and to deregulation of factors that control Mg homeostasis during menopause., Conclusions: Both Mg deficit and obesity may independently lead to a higher risk for insulin resistance and cardiovascular disease.

The aim of this paper is to discuss, on the basis of an extensive literature review, the role of magnesium in health and disease. Magnesium is an essential cation playing a crucial role in many physiological functions. It is critical in energy-requiring metabolic processes, in protein synthesis, membrane integrity, nervous tissue conduction, neuromuscular excitability, muscle contraction, hormone secretion, and in intermediary metabolism. Serum magnesium concentration is maintained within a narrow range by the small intestine and kidney which both increase their fractional magnesium absorption under conditions of magnesium deprivation. If magnesium depletion continues, the bone store helps to maintain serum magnesium concentration by exchanging part of its content with extracellular fluid. The abundance of magnesium within cells is consistent with its relevant role in regulating tissue and cell functions. Recent data suggest that large fluxes of magnesium can cross the cell plasma membrane in either direction following a variety of hormonal and non-hormonal stimuli, resulting in major changes in total and, to a lesser extent, in free magnesium content within tissues. Imbalances of magnesium are common and are associated with a great number of pathological situations responsible for human morbidity and mortality. A large part of the population may have an inadequate magnesium intake, and in particular elderly subjects and athletes may be prone to chronic latent magnesium deficiency. Magnesium deficit is frequently observed in alcoholics and diabetic patients, in whom a combination of factors contributes to its pathogenesis. We will discuss some of the aspects of the involvement of magnesium in the etiology of some pathological situations, such as cardiovascular diseases, diabetes, pre-eclampsia, eclampsia, sickle cell disease and chronic alcoholism.

Loxoscelism or the envenoming by the brown spiders (Loxosceles genus spiders), may produce extensive dermonecrosis and hemorrhage at the bite site and, eventually, systemic reactions that may be lethal. Isolation and identification of many different bacteria, among them Clostridium perfringens, of great medical importance due to its involvement in dermonecrotizing and systemic conditions, was carried out from the venomous apparatus (fangs and venom) of spiders obtained directly from nature, through microbiological cultures in aerobic and anaerobic conditions. Working with Loxosceles intermedia venom (alone) and with the venom conjugated with Clostridium perfringens using rabbits as experimental models for dermonecrosis, allowed for the observation that venom and anaerobic bacteria conjugated resulted in a striking increase of the dermonecrotic picture when compared to venom alone, suggesting a role for Clostridium perfringens in the severe dermonecrotic picture of these patients and opening the possibility for the association of antibiotic therapy in treating loxoscelism.

Objective: To correlate, in a sample of healthy children and adolescents, the activity of the enzyme acid phosphatase (ACP) with its different genetic phenotypes and of these with some cardiovascular risk parameters such as body mass index (BMI), percentage of total fat mass (%TFM), trunk fat (TF), insulin resistance, and the arterial blood pressure (BP)., Design and Methods: The sample was composed of 173 healthy children and adolescents, 96 (55.5%) F and 77 (44.5%) M, with ages between 10 and 16 years (mean: 13.04 +/- 1.68). The ACP activity was determined through a spectrophotometric method and its phenotypes through isoelectric focusing electrophoresis. BMI (Kg/m2) and the BP were obtained by standardized methods. Glycemia determined by the glucose oxidase method and insulinemia by RIA method. Insulin resistance based on the homeostasis model assessment (HOMA) was calculated as: [fasting insulin (microU/ml) x fasting glucose (mmol/l)]: 22.5. The %TFM and TF were determined by dual energy x-ray absorptiometry (DXA). The statistical methods used were ANOVA, the Pearson correlation and the Student's test., Results: The distribution of the phenotypes were the following--absolute versus relative frequencies: BB-74 (48.4%), AB-52 (34%), AA-16 (10.5%), BC-7 (4.6%), AC-3 (2%) and CC-1 (0.7%). ACP activities (mean: 321.04 +/- 84.56) were significantly different among the phenotypes (p < 0.001). The smallest activity was observed in the AA individuals, the highest in CC, followed by BC (247.17 +/- 66.52 and 767.30 and 362.44 +/- 91.56 respectively). Glycemia was higher in the AA individuals (4.61 +/- 0.37) compared to CC + BC (4.40 +/- 0.31) (p = 0.08). A direct correlation was verified between HOMA and BP, both diastolic (p = 0.013, r = 0.250) and systolic (p = 0.015, r = 0.246), as well as of these with BMI (mean: 20.57 +/- 3.24) and insulinemia (p = 0.016, r = 0.215; p = 0.004, r = 0.280 and p = 0.007, r = 0.240; p = 0.008, r = 0.261 respectively for diastolic and systolic BP). There were no significant difference of BMI between sexes, nor of this as well as of % TFM and TF among the different genetic phenotypes of ACP., Conclusions: The smallest enzymatic activity of ACP seems to be associated with the AA individuals, where a trend for higher glycemia was verified. BMI, HOMA and insulinemia, due to their significant direct relationship with diastolic and systolic BP in this sample of children and adolescents may warrant more future attention in the evaluation of cardiovascular risk. There were no significant differences of HOMA, BMI, %TFM, TF nor of BP among the different ACP genetic phenotypes.
