系统研究了各种因素对复合膜的药物控释、稳定性、药物包埋率等性能的影响.最佳制备条件：pH=5.0、壳聚糖溶液质量分数为0.25%、海藻酸钠溶液质量分数为0.25%、海藻酸钠与壳聚糖的质量比为1：1.当壳聚糖溶液pH = 5.0时，复合膜的药物控释性能最强.通过Ca2+的交联，使替硝唑的包埋率达到96.1%.复合膜的溶胀度研究表明，在pH = 11的介质中具有最大的溶胀度.

喉阻塞中较为常见的病因之一是喉气管瘢痕性狭窄,其造成的呼吸困难、声音嘶哑和吸气性喘鸣严重影响患者生活质量,甚至危及生命。成纤维细胞、肌成纤维细胞、肥大细胞、巨噬细胞等细胞及转化生长因子β、结缔组织生长因子、表皮生长因子受体等细胞因子均参与了喉气管瘢痕性狭窄的纤维化过程中。药物治疗可能成为喉气管瘢痕性狭窄一个重要预防及治疗策略,常见药物有丝裂霉素C、5-氟尿嘧啶/氟羟泼尼松龙、他克莫司和雷帕霉素、甲壳素、壳聚糖及其衍生物等。 [ABSTRACT FROM AUTHOR]

目的　制备关节腔注射用载氯诺昔康(Lnｘｃ)壳聚糖温敏凝胶，观察其理化性质及体外释药性。方法　 以壳聚糖、β-甘油磷酸钠(β-GP)为材料制备载Lnｘｃ温敏凝胶，以胶凝温度、时间、通针性为评价指标，优化处方工 艺；采用椎板法观察凝胶的流变学性质，采用离心试验、温度试验观察凝胶的稳定性，采用动态膜透析法观察载 Lnｘｃ温敏凝胶和Lnｘｃ溶液的体外释药特性。结果　载Lnｘｃ壳聚糖温敏凝胶的最佳处方为Lnｘｃ2mｇ、3％ 壳聚 糖、60％β-GP、ｐH7.2，所制备的温敏凝胶属于非牛顿流体，离心后无分层现象，4℃和25℃条件下呈流动状，37℃ 发生相变形成凝胶。Lnｘｃ溶液8ｈ药物累积释放率86.70％，载Lnｘｃ温敏凝胶72ｈ药物累积释放率86.41％ ，释 药规律符合Hiｇｕｃｈi方程。结论　成功制备载Lnｘｃ壳聚糖关节腔用温敏凝胶，其具有理想的胶凝温度和较好的缓 释效果。 [ABSTRACT FROM AUTHOR]

【目的】探讨含碱性成纤维细胞生长因子（bFGF）的壳聚糖-Ⅰ型胶原复合支架对人牙周膜细胞（HPDLC）生长和增殖的影响。【方法】MTT法观察含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液、不含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液以及正常细胞培养液对HPDLC生长和增殖的影响，利用流式细胞仪（FCM）检测上述3种液体对HPDLC细胞周期的影响，并将HPDLC接种于含bFGF因子壳聚糖-Ⅰ型胶原复合支架和不含bFGF因子壳聚糖-Ⅰ型胶原复合支架内，通过免疫细胞组织化学和激光共聚焦显微镜来检测HPDLC在复合支架内的存活和增殖情况。【结果】含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液对HPDLC生长和增殖的促进作用明显强于不含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液和正常细胞培养液．而后两组间无统计学意义；含bFGF因子壳聚糖-Ⅰ型胶原复合支架浸出液促进HPDLC从G1期进入S期；HPDLC在含bFGF因子壳聚糖-Ⅰ型胶原复合支架内的生长和增殖也明显优于不含bFGF因子壳聚糖-Ⅰ型胶原复合支架。【结论】含bFGF因子壳聚糖-Ⅰ型胶原复合支架可以促进体外培养的HPDLC生长和增殖。

目的探讨低分子壳聚糖(LMCTS)对缺氧复氧大鼠心肌细胞的保护作用,并探讨其作用机制。方法原代培养大鼠心肌细胞,将其分为空白对照组、模型组及低、中、高LMCTS组。空白对照组正常培养,不予任何处理;模型组用真空缺氧处理1 h后复氧24 h;低、中、高LMCTS组分别加入含200、400、600μg/m L LMCTS的培养液,培养24 h后再用真空缺氧处理1 h后复氧24 h。ELISA法测定各组细胞上清液中炎症因子TNF-α、IL-6,Western blot法检测心肌细胞解偶联蛋白-2(UCP-2)表达。结果低、中、高LMCTS组心肌细胞存活率均较模型组升高(P均<0.01)。模型组心肌细胞上清液中TNF-α、IL-6水平高于空白对照组(P均<0.01),低、中、高LMCTS组均低于模型组,以高LMCTS组最低(P均<0.01)。模型组心肌细胞UCP-2表达较空白对照组升高,低、中、高LMCTS组较模型组降低,以高LMCTS组最低(P均<0.01)。结论 LMCTS对缺氧复氧心肌细胞具有保护作用,其机制与抑制炎症反应、降低UCP-2表达有关。 [ABSTRACT FROM AUTHOR]
