A group of 20 patients with probable Alzheimer disease (AD) and a control group were tested in a verb generation task, in a verb synonym task and several cognitive tests. Three types of verbs and novel verbs were presented in simple sentence frames, in two different conditions. In one condition participants were presented with the verb in the infinitive, providing information about the conjugation of the verb and the most likely type of past participle mapping required. The second was an ambiguous condition in which the suffix of the input verb did not provide any clue to the conjugation. The aim was to investigate if different types of verbs and input mapping affected patients' performance, and to what extent the deficit increased when the illness became more severe. Dependent measures were accuracy rates, rates of different morpho-phonological transformations and error type rates. Patients showed a more marked deficit in verb generation, when input was ambiguous. Correlation between the verb synonym test and accuracy in verb generation indicated that a deficit in lexical-semantic memory was partially responsible for impaired performance. Data suggest that patients maintained information about frequency distribution of different verb types and verb classes in each conjugation, but were impaired in operating complex phonological transformations.

MADS-box genes have been shown to play a role in the formation of fruits, both in Arabidopsis and in tomato. In peach, two C-class MADS-box genes have been isolated. Both of them are expressed during flower and mesocarp development. Here a detailed analysis of a gene that belongs to the PLENA subfamily of MADS-box genes is shown. The expression of this PLENA-like gene (PpPLENA) increases during fruit ripening, and its ectopic expression in tomato plants causes the transformation of sepals into carpel-like structures that become fleshy and ripen like real fruits. Interestingly, the transgenic berries constitutively expressing the PpPLENA gene show an accelerated ripening, as judged by the expression of genes that are important for tomato fruit ripening. It is suggested that PpPLENA might interfere with the endogenous activity of TAGL1, thereby activating the fruit ripening pathway earlier compared with wild-type tomato plants.

Ovules have an important role during the life cycle of the plant, and they provide an excellent model for studying organogenesis in plants. As such, the molecular control of ovule development has been studied for many years. Recent studies in Arabidopsis have revealed important new data concerning ovule primordia formation, ovule identity determination, and patterning. Furthermore, interesting results about ovule development in other species, such as Petunia and rice, have been published recently. In this review, we discuss these recent findings in reference to ovule development in Arabidopsis. We compare available data with those of other species to investigate the evolutionary conservation of the regulatory pathways.

MADS-box transcription factors are key regulators of plant developmental processes. While the function of MIKC (type II) MADS-box genes has been intensively studied, only limited data are available for the other more recently identified classes of MADS-box genes, despite these latter comprising more than 60% of the Arabidopsis MADS-box gene family. Here we describe the function of AGL23, an Arabidopsis type I MADS-box gene belonging to the Malpha subfamily. We show that AGL23 plays an important role during development of the female gametophyte and embryo. The agl23-1 mutant forms a functional megaspore. However, at this stage female gametophyte development is arrested and the megaspore persists during subsequent phases of ovule development. Despite the incomplete penetrance of the female gametophyte defect, plants homozygous for the agl23-1 mutation were never identified. Analysis of developing seeds showed that embryos homozygous for the agl23-1 allele are albino and unable to give rise to viable plants. Electron microscopy analysis revealed that this phenotype is due to the absence of chloroplasts, strongly suggesting that AGL23 is involved in controlling the biogenesis of organelles during embryo development.

Ovules are the organs in which the female gametophyte develops. They develop in Arabidopsis and many other plant species from carpel tissue as new meristematic formations. In ovules we can distinguish three major parts which are the funiculus that attaches the ovule to the placenta, the integuments and the nucellus which contains the female gametophyte. Little is known about the molecular genetic regulatory cues that control the development of these ovule tissues. In the August issue of The Plant Cell, we have shown that there are genetic and molecular interactions between BELL1 and the MADS-box genes AGAMOUS, SEEDSTICK, SHATTERPROOF1 and SHATTERPROOF2 to control integument identity.1 We have shown that BEL1 directly interacts with a MADS-box dimer composed of AG and SEPALLATA3 and we proposed that this interaction is essential to prevent that integuments turn into carpels. Furthermore, we have shown that during ovule development BEL1 is important for the regulation of the stem cell maintenance gene WUSCHEL. In this Addendum we discuss addition literature data that sustain our model for integument development in Arabidopsis.

During floral induction and flower development plants undergo delicate phase changes which are under tight molecular control. MADS-box transcription factors have been shown to play pivotal roles during these transition phases. SHORT VEGETATIVE PHASE (SVP) and AGAMOUS LIKE 24 (AGL24) are important regulators both during the transition to flowering and during flower development. During vegetative growth they exert opposite roles on floral transition, acting as repressor and promoter of flowering, respectively. Later during flower development they act redundantly as negative regulators of AG expression. In rice, the orthologues of SVP and AGL24 are OsMADS22, OsMADS47, and OsMADS55 and these three genes are involved in the negative regulation of brassinosteroid responses. In order to understand whether these rice genes have maintained the ability to function as regulators of flowering time in Arabidopsis, complementation tests were performed by expressing OsMADS22 and OsMADS47 in the svp and agl24 mutants. The results show that the rice genes are not able to complement the flowering-time phenotype of the Arabidopsis mutants, indicating that they are biologically inactive in Arabidopsis. Nevertheless, they cause floral reversions, which mimic the SVP and AGL24 floral overexpressor phenotypes. Yeast two-hybrid analysis suggests that these floral phenotypes are probably the consequence of protein interactions between OsMADS22 and OsMADS47 and other MADS-box proteins which interfere with the formation of complexes required for normal flower development.

The influence of lexical-semantic impairment and of executive dysfunction on word naming performance was investigated in a group of patients with probable Alzheimer dementia (AD). The patients, who varied in the severity of the illness, were tested in a word naming task where they had to read aloud Italian three-syllable words with a dominant or subordinate stress pattern. These types of words have been shown to interact with frequency in normal adults [J. Exp. Psychol.: Hum. Percept. Perform. 18 (4) (1992) 987], so that the effect of the subordinate stress pattern (slower reading times) is only apparent for low frequency words. The frequency and stress effects on accuracy increased across dementia severity levels. Regression analyses showed that the impairment in reading low frequency words with subordinate stress depended largely on the level of lexical-semantic impairment, measured by a test of semantic memory and comprehension. Implications for the current reading models are discussed.

MADS-box proteins are transcription factors present in different eukaryotic kingdoms. In contrast to plants, for mammalian and yeast MADS-box proteins ternary complex formation with unrelated transcription factors was reported. We show here the first identification of such ternary interaction in plants. A rice seed-specific NF-YB was identified as partner of OsMADS18 by two-hybrid screening. NF-YB contains a histone fold motif, HFM,(1) and is part of the trimeric CCAAT-binding NF-Y complex. OsMADS18, alone or in combination with a natural partner, interacts with OsNF-YB1 through the MADS and I regions. The mouse NF-YB also associates with OsMADS18 in vivo and in vitro as a NF-YB-NF-YC dimer. Other rice MADS-box proteins do not interact in these assays, indicating specificity for the interaction. OsNF-YB1 is capable of heterodimerizing with NF-YC, but not trimerizing with NF-YA, thus precluding CCAAT binding. Mutation of the variant Asp at position 99 of the HFM alpha2-helix into a conserved serine recovers the capacity to interact with NF-YA, but not with DNA. This is the first indication that members of the NF-YB family work through mechanisms independent of the CCAAT box.

In the Italian language there is a higher number of inflectional suffixes in verbs than in nouns, and this might imply that verbs are more likely to undergo a morphological analysis in terms of root and suffix as compared to nouns (Traficante & Burani, unpublished observations). Moreover, verbs tend to be more abstract than nouns, and this aspect might make verb processing more difficult. Finally, the developmental gap in the production of nouns and verbs suggests that age of acquisition might affect noun and verbs differently. Nouns and verbs were presented in a lexical decision and in a naming task. The morphological variable root frequency in addition to word frequency, length and word age of acquisition, and the semantic variables concreteness and context availability (Schwanenflugel, Harnishfeger, & Stowe, 1988) were used as predictors in multiple-regression analyses in which lexical decision and naming latencies were the dependent variables. The results showed that age of acquisition, context availability, and root frequency are all important in predicting both lexical decision and naming latencies for nouns and verbs, but age of acquisition and root frequency are better predictors of the differences in processing Italian nouns and verbs., (Copyright 2001 Elsevier Science (USA).)
