The title compound (+/-)-5 (R = Pro) (LY141865) has been resolved into a (-) isomer and a (+) isomer as the D- and L-tartrate salts, respectively. Biological studies have shown that dopamine agonist activity is a property of only the (-) isomer. Crystallographic analysis has proven that the absolute configuration of the active (-) isomer is the same as that of the natural ergolines.

Fluoxetine (Li-ly 110140) caused a 63 percent inhibition of [3H]serotonin uptake into platelets obtained from normal volunteers to whom the drug was administered daily for 7 days. This dose had no effect on the usual pressor response produced by injections of norepinephrine or tyramine.

The secretory antibody response to primary and secondary infection with respiratory syncytial virus (RSV) in Igg, IgM and 11S IgA immunoglobulin classes was determined using an indirect immunofluorescence antibody technique. Nasopharyngeal secretions were collected after naturally acquired primary or secondary infection with RSV. Immunoglobulin A responses to RSV were significantly greater in infants older than 6 months of age at the time of infection than in younger infants. Reinfection with RSV resulted in enhanced antibody production in all three immunoglobulin classes, and virus-specific antibody persisted for longer periods after secondary infection. Repeated immunizations with live RSV vaccine may be necessary for the development of immunity to severe illness.

Fluoxetine, a selective inhibitor of serotonin uptake, is clinically useful in treating depression and may be useful for management of a variety of other psychiatric and metabolic derangements. Using X-ray crystallography, we have determined the three-dimensional structure of fluoxetine hydrochloride. A total of 2394 unique reflections were measured, and full-matrix least-squares refinement of all non-hydrogen coordinates and thermal parameters gave a final discrepancy index of 0.074 for 1759 observed reflections. In the solid state, the planes defined by the two aromatic rings are skewed, precluding the possibility of intramolecular ring-ring interactions. The methylene units of the methylpropanamine moiety adopt the anticipated conformational relationships to minimize torsional strain. An exact antiperiplanar relationship exists between N11 and C3; the N11-C1-C2-C3 dihedral angle is -180 degrees. The C1-C2-C3-O4 dihedral angle is 60.6 degrees, indicating that the propanamine side-chain folds toward the phenoxy moiety rather than adopting a fully extended conformation. This folded three-dimensional relationship may be necessary for high-affinity interaction with the serotonin-uptake carrier and confers considerable structural homology between this portion of fluoxetine and the phenylcyclohexylamine substructure of sertraline and EXP-561. However, the nature of substituents on the phenoxy portion of fluoxetine is also critical in determining potency and selectivity in this series of compounds.

In order to determine the natural history of parainfluenza virus infection in early life, we followed prospectively 130 infants and children from birth or a few months of age for evidence of infection with PV. Using rapid diagnostic techniques, standard tissue culture infectivity, and serologic techniques we were able to document primary PV infection in 92% of these infants, and repeated infection with heterotypic or homotypic PV strains in 49% by 30 months of age. Increasing patient age had no significant effect in reducing the incidence of lower respiratory tract illness as a result of PV infection. Infection with one PV serotype provided no protection against LRTI at the time of subsequent infection with a heterotypic PV strain. In contrast, primary PV infection provided a brief period of immunity to LRTI upon homotypic reinfection. Secretory IgA responses to PV were determined by immunofluorescent techniques. Antibody response to PV strains causing primary infection and heterotypic repeated infection were transient and of low magnitude. Homotypic reinfection resulted in significantly enhanced production of secretory antibody to PV. At least in early life, repeated exposures to PV appear to be essential for maintaining immunity to severe forms of illness caused by PV infection.

We recently prepared a series of 3- and 4-(phosphonoalkyl)pyridine- and -piperidine-2-carboxylic acids as antagonists of neurotransmission at N-methyl-D-aspartate (NMDA) preferring receptors. NMDA antagonists may prove to be useful therapeutic agents, for instance, as anticonvulsants, in the treatment of neurodegenerative disorders such as Alzheimer's disease and in the prevention of neuronal damage that occurs during cerebral ischemia. The compounds prepared were evaluated for their ability to displace [3H]CPP binding (an assay shown to be selective for compounds that bind at the NMDA receptor) and for their ability to block NMDA-induced lethality in mice (an assay that is also specific for competitive and noncompetitive NMDA antagonists). Two of the compounds, cis-4-(phosphonomethyl)piperidine-2-carboxylic acid (11a) and cis-4-(3-phosphonoprop-1-yl)piperidine-2-carboxylic acid (11c) proved to be potent NMDA antagonists. 11a and 11c displaced [3H]CPP binding with IC50's of 95 and 120 nM, respectively, and both protected mice from NMDA-induced lethality, with MEDs (minimum effective dose, the dose at which three of the five animals tested survived) of 10 and 40 mg/kg ip, respectively. The rest of the compounds prepared were weakly active or inactive in these assays. The pattern of activity observed for this series parallels that observed for the acyclic series of omega-phosphono-alpha-amino acids, where AP5 and AP7 possessed NMDA antagonist activity while AP6 and AP8 were inactive. Reduction of conformational mobility by incorporation of the piperidine ring led to enhanced potency relative to the acyclic analogues.

Human lung cells (ChaGo) derived from a bronchogenic carcinoma produce human chorionic gonadotropin (hCG), predominantly the alpha subunit of the glycoprotein hormone, under culture conditions. Treatment of the cells with the polycyclic aromatic hydrocarbons, benzo(a)pyrene (BaP) or dimethylbenzanthracene, at concentrations which do not affect cell growth or macromolecular synthesis, stimulates the production of hCG in these cells. The levels of alpha hCG-specific mRNA (mRNA alpha hCG) sequences in total poly(A)+ RNA isolated from control and drug-treated ChaGo cells are determined by the dot hybridization technique using 32P-labeled, cloned cDNA alpha hCG probe. A concentration-dependent increase in the levels of mRNA alpha hCG sequences in BaP or dimethylbenzanthracene-treated ChaGo cells has been observed. The increase in the level of mRNA alpha hCG sequences can be detected after treatment of the cells with either of the drugs for 24 h, and this level attains its maximum within 48-72 h following drug treatment. A comparative study of the restriction endonuclease (MspI/HpaII) digestion patterns of the control and BaP-treated cell DNA suggests that the internal "C" residues of the -CCGG- sequences in the alpha hCG gene of untreated cells are highly methylated; whereas the internal C residues of the same MspI/HpaII recognition sequences in the alpha hCG gene are comparatively less methylated in BaP-treated cell DNA.

A series of experiments were undertaken to compare the in vitro behaviour of the medial edge epithelium (MEE) of hamster, in which palatal shelves normally fuse, and chick, in which they do not fuse. Homotypic pairs of hamster and chick embryo palatal processes, single palatal processes, and heterotypic palatal shelves of both animals were grown in vitro. The results indicated that contact between palatal shelves may not be crucial for MEE differentiation in mammals. The ability to acquire pre-fusion characteristics may be present in mammalian palatal tissue from their early development and may be expressed by cessation of DNA synthesis in the MEE, elevation of cAMP, and MEE cell death. Isolated chick palatal shelf cultured under identical conditions did not express these mammalian pre-fusion characteristics. When MEE of hamster and chick palatal shelves were placed in contact with one another, the intervening epithelia underwent cytolysis. This could be due to either the destruction of chick MEE by lysosomal enzymes liberated from adjacent degenerating hamster MEE cells, or by induction of cell death in chick MEE by hamster mesenchyme. Heterotypic palatal tissue combinations also suggest that release of lysosomal enzymes in the hamster MEE, which leads to its dissolution, may be the terminal event in epithelial differentiation prior to the establishment of mesenchymal continuity. It is suggested that an inverse relationship exists between DNA synthesis and cAMP levels during palatogenesis: when palate closes (as in mammals) the MEE is eliminated by increasing cAMP levels, whereas when palate remains open (as in birds) low level of cAMP preserve the integrity of MEE by supporting DNA synthesis.

Pyrrolnitrin has been reported to inhibit Bacillus megaterium primarily by forming complexes with phospholipids and to block electron transfer of Saccharomyces cerevisiae between succinate or reduced nicotinamide adenine dinucleotide (NADH) and coenzyme Q. We found that pyrrolnitrin inhibited respiration of conidia of Microsporum gypseum. In mitochondrial preparations, pyrrolnitrin strongly inhibited respiration and the rotenone-sensitive NADH-cytochrome c reductase. The rotenone-insensitive NADH-cytochrome c reductase, the succinate-cytochrome c reductase, and the reduction of dichlorophenolindophenol by either NADH or succinate were inhibited to a lesser extent. However, the activity of cytochrome oxidase was not affected by pyrrolnitrin. The extent of reduction of flavoproteins by NADH and succinate, measured at 465 - 510 nm, was unaltered; however, the reduction of cytochrome b, measured at 560 - 575 nm, was partially inhibited by pyrrolnitrin. The level of totally reduced cytochrome b was restored with antimycin A. We, therefore, concluded that the primary site of action of this antifungal antibiotic is to block electron transfer between the flavoprotein of the NADH-dehydrogenase and cytochrome b segment of the respiratory chain of M. gypseum.
