The bactericidal action of rifampicin was compared with that of chloramphenicol in growing and in sporulating cultures of Bacillus subtilis 168. Chloramphenicol kills cells only very slowly, but exposure to rifampicin kills over 95% of cells in a few minutes, causing gross physical damage, which is visible in both phase-contrast and electron microscopy. This is accompanied by a fall in O(2) consumption and by lysis. Experiments with synchronized cultures showed that susceptibility to the lethal effect of rifampicin is greater when the cells are dividing. The results suggest that the synthesis of some species of RNA other than mRNA may be necessary for the maintenance of cell integrity, although experiments with actinomycin D do not altogether fit this interpretation. However, we conclude that rifampicin is too toxic to use as an antibiotic for assessing the lifetime of mRNA.

1. The synthesis of sulpholactic acid in sporulating cultures of Bacillus subtilis was studied. 2. Sulpholactic acid was first detected about 4h after the initiation of sporulation and 1h before refractility. The rate of synthesis paralleled that of the other events of sporulation examined. 3. Sulpholactic acid accounted for 1.7% of the material of the spore. 4. Because the addition of chloramphenicol in the earlier stages of sporulation inhibited formation of the compound, it is likely that the enzymes concerned are synthesized de novo during sporulation. 5. In asporogenous mutants only those blocked at a late stage and showing partial refractility were able to produce sulpholactic acid. This correlation makes sulpholactic acid a useful marker event in sporulation.

A comparison was made of morphological changes and successive, mainly biochemical, marker events for sporulation in 14 asporogenous mutants. The morphological and biochemical sequences are linked so that arrested development in one is accompanied by corresponding effects in the other. Thus mutants that fail to produce both protease and antibiotic do not progress beyond stage 0, formation of alkaline phosphatase appears to be associated with the transition from stage II to stage III and glucose dehydrogenase with that from stage III to stage IV. Stage II mutants may produce ;pygmy' cells or other bizarre cell-division forms. The biochemical sequence is dependent in the sense that if the occurrence of any one event is blocked that of all the succeeding events is also blocked. This has implications for biochemical models that have been proposed to explain the temporal sequence observed in spore development.

Since canonical correlations (Rc) reflect the relationships between linear combinations of two sets of items, variance shared by both sets may or may not be high. This paper examines a procedure providing needed clarification in the interpretation of Rc. Three canonical solutions disclosed that highly sig- nificant Rcs may actually represent very little overlapping variance. A redundancy index proved to be an invaluable aid for placing the Rc in proper perspective. The efficiency for extracting shared variance was found to decrease steadily with each successive canonical variate.

An alkali-extractable protein fraction comprising 6% of the dry weight of the spore can be removed from spores of Bacillus subtilis 168. Three different extraction procedures each yield at least one similar protein. Extracted protein behaved as a single species on ion-exchange chromatography or gel filtration, but two polypeptides were found on electrophoresis. Comparison of molecular weights on electrophoresis and by sucrosegradient analysis suggests that the protein(s) undergo aggregation into multimers. Extracted spores remain viable, but are altered in density and lysozyme sensitivity and they aggregate together. Electron microscopy of extracted spores shows that loss of material seems to occur from the outer coat layers. Extraction therefore probably removes a specific fraction of the spore-coat protein, but without impairment to the spore protoplast. This protein can first be detected immunologically 4h after the initiation of sporulation, and the synthesis of this protein is sensitive to chloramphenicol, actinomycin D and rifamycin. Labelling experiments also show that the protein begins to be synthesized early in sporulation. Examination of the ability of asporogenous mutants to produce cross-reacting material indicates that some event in stage II of sporulation determines its production.
