Background: In cell-based therapy, the transmission of communicable diseases imposes a substantial threat to recipients. In this study, we investigated whether cell-based screening could detect hepatitis B virus (HBV) in human umbilical cord-derived mesenchymal stem cells (HUMSCs) isolated from HBV-infected donors to understand the susceptibility of HUMSCs to HBV infection. Methods: HBV assay was performed in HUMSCs derived from healthy and HBV-infected donors with enzyme-linked immunosorbent assay (ELISA), fluorescence quantitative PCR (FQ-PCR) assay, and droplet digital PCR (ddPCR) assay. Further, HBV DNA was assayed in HUMSCs derived from healthy donors after incubation with human sera containing a high titer of HBV using FQ-PCR. Results: HBV antigen/antibody and DNA failed to be detected using ELISA, FQ-PCR, and ddPCR. After incubation with HBV infection sera, HBV DNA could be detected, but below the valid titer of the assay kit. The HBV DNA levels in HBV-incubated HUMSCs gradually decreased with medium change every 2 days and then significantly decreased, not even detected after passage. Conclusions: The current cell-based screening methods could not detect HBV in HUMSCs derived from HBV-infected donors, indicating the importance of more stringent donor eligibility to reduce the risk of transmission of communicable diseases in cell-based therapy. To solve the problem of an occult HBV window period in donor eligibility determination, we recommend that the donors undergo another HBV serological test 3 months after the first serological communicable disease screening. Summary The innate RNA sensor RIG-I is critical in the initiation of antiviral type I interferons (IFNs) production upon recognition of “non-self” viral RNAs. Here, we identify a host-derived, IFN-inducible long noncoding RNA, lnc-Lsm3b, that can compete with viral RNAs in the binding of RIG-I monomers and feedback inactivate the RIG-I innate function at late stage of innate response. Mechanistically, binding of lnc-Lsm3b restricts RIG-I protein’s conformational shift and prevents downstream signaling, thereby terminating type I IFNs production. Multivalent structural motifs and long-stem structure are critical features of lnc-Lsm3b for RIG-I binding and inhibition. These data reveal a non-canonical self-recognition mode in the regulation of immune response and demonstrate an important role of an inducible “self” lncRNA acting as a potent molecular decoy actively saturating RIG-I binding sites to restrict the duration of “non-self” RNA-induced innate immune response and maintaining immune homeostasis, with potential utility in inflammatory disease management. Compared with the traditional fluoride fibers and tellurite fibers that can work in the near-infrared region, suspended-core fibers based on chalcogenide glasses have wider transmitting regions and higher nonlinear coefficients, thus the mid-infrared supercontinuum generations can be achieved easily. Rather than adopting the traditional fabrication technique of hole-drilling and air filling, we adopted a totally novel extrusion technique to fabricate As2S3 suspended-core fibers with four holes, and its mid-infrared supercontinuum generation was investigated systematically by integrating theoretical simulation and empirical results. The generalized nonlinear SchrÖdinger equation was used to simulate the supercontinuum generation in the As2S3 suspended-core fibers. The simulated supercontinuum generation in the As2S3 suspended-core fibers with different pump wavelengths (2-5 µm), increasing powers (0.3-4 kW), and various fiber lengths (1-50 cm) was obtained by a simulative software, MATLAB. The experimental results of supercontinuum generation via femtosecond optical parametric amplification (OPA) were recorded by changing fiber lengths (5-25 cm), pump wavelengths (2.9-5 µm), and pump powers (10-200 kW). The simulated consulting spectra are consistent with the experimental results of supercontinuum generation only if the fiber loss is sufficiently low. [ABSTRACT FROM AUTHOR]
